A Human Glucoregulatory Model for the Rational Design and Evaluation of Diabetes Therapeutics.

Diabetes mellitus remains a leading cause of morbidity worldwide, motivating the development of novel diabetes therapeutics. Computational glucoregulatory models that incorporate biochemical signaling and mechanisms have shown utility for preclinical evaluation and even the rational design of such interventions, potentially reducing the need for animal experimentation. In this work, we expand upon our IM3PACT 2.0 computational framework by applying it to human clinical data and demonstrate its utility for translational prediction of glucose-responsive glucagon (GRG) therapeutic performance. Evaluation of Ethyl Cellulose-Ethanol Ablation in a Human-Sized 3D Construct of High-Grade Cervical Dysplasia.

Current preclinical models for cervical dysplasia rely on animal systems that poorly mimic human anatomy. To address this and align with emerging initiatives to reduce animal experimentation, we developed a human-sized, 3D in vitro model of high-grade cervical dysplasia. The construct integrates normal human fibroblasts, keratinocytes, and SiHa cancer cells within a gelatin methacrylate (GelMA) hydrogel, engineered with a cervical "os" and asymmetric lesion to replicate native human cervical architecture. We utilized this platform to evaluate the efficacy and safety of ethyl cellulose-ethanol

Inherited metabolic disease.

Our understanding of the pathophysiology and of new treatments for inherited metabolic diseases that affect the liver continues to grow through the study of gene mutations and their functional effect on the proteins they encode. For genetic hemochromatosis and Wilson's disease, studies focused on the function of their respective gene products provide new insights into metal metabolism. For Crigler-Najjar syndrome, an inherited disorder that results in failure of proper bilirubin glucuronidation, the once futuristic idea of treatment by transplantation of donor hepatocytes has now proven succes

Pharmacology of thrombolytic drugs.

Streptokinase and urokinase have proved to be useful in a limited number of clinical conditions. Mainly because of the risk and unpredictability of bleeding with this first generation of thrombolytic agents, thrombolysis has not been ingrained in medical practice. In the interim, more fibrin-specific thrombolytic agents have been developed such as acylated streptokinase-human plasminogen complex, tissue-type plasminogen activator (t-PA) and single chain urokinase-type plasminogen activator (scu-PA or pro-urokinase). Only the latter two drugs do not induce major systemic fibrinogenolysis at thr

Toward fully exploiting the therapeutic potential of marketed pharmaceuticals: the use of octreotide and chloroquine in oncology.

Pleiotropy in biological systems and their targeting allows many pharmaceuticals to be used for multiple therapeutic purposes. Fully exploiting the therapeutic properties of drugs that are already marketed would be highly advantageous. This is especially the case in the field of oncology, where the ineffectiveness of typical anticancer agents is a common issue, while the development of novel anticancer agents is a costly and particularly time-consuming process. Octreotide and chloroquine are two pharmaceuticals that exhibit profound antitumorigenic activities. However, the current therapeutic

Prion-like properties of the mutant huntingtin protein in living organisms: the evidence and the relevance.

If theories postulating that pathological proteins associated with neurodegenerative disorders behave similarly to prions were initially viewed with reluctance, it is now well-accepted that this occurs in several disease contexts. Notably, it has been reported that protein misfolding and subsequent prion-like properties can actively participate in neurodegenerative disorders. While this has been demonstrated in multiple cellular and animal model systems related to Alzheimer's and Parkinson's diseases, the prion-like properties of the mutant huntingtin protein (mHTT), associated with Huntington

From rodents to chips: preclinical models of inflammatory bowel disease with emphasis on host-microbiome interactions.

Inflammatory bowel disease (IBD), comprising Crohn's disease and ulcerative colitis, is a chronic relapsing inflammatory disorder with steadily increasing global prevalence, significantly impairing patient quality of life. Preclinical research in IBD has traditionally relied on animal models to investigate disease etiology, immunopathogenesis, and therapeutic responses. A wide range of experimental models primarily rodents, along with limited use of larger animals such as pigs and nonhuman primates, have been developed to reproduce key clinical, histological, and immunological features of huma

Neurotoxin-based models of Parkinson's disease.

Animal experimentation in the Parkinson's disease (PD) field is a classic example of how the use of animal models to study diseases can have a significant impact on human health. Among the different neurotoxin-based animal models of PD that are presently available, the 6-hydroxydopamine (6-OHDA) and the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) models have been established and validated as useful models for the development of therapeutic strategies aimed to treat motor symptoms and to study alterations of the basal ganglia that occur in this disease. The 6-OHDA rat model and the MPTP

Stem cell models of Angelman syndrome.

Angelman syndrome (AS) is an imprinted neurodevelopmental disorder that lacks a cure, characterized by developmental delay, intellectual impairment, seizures, ataxia, and paroxysmal laughter. The condition arises due to the loss of the maternally inherited copy of the UBE3A gene in neurons. The paternally inherited UBE3A allele is unable to compensate because it is silenced by the expression of an antisense transcript (UBE3A-ATS) on the paternal chromosome. UBE3A, encoding enigmatic E3 ubiquitin ligase variants, regulates target proteins by either modifying their properties/functions or leadin

AI-integrated human-relevant alternatives to animal experimentation in drug development: Toward a predictive translational ecosystem, from in vitro and in silico models to clinical and real-world evidence.

Animal experimentation has long been associated with biomedical research and drug development. However, ethical concerns, limited translatability to humans and high clinical attrition rates (> 90%) necessitate a transition toward human-relevant approaches. Although diverse in vitro, in silico and artificial intelligence (AI)-enabled methodologies have emerged, their integration into a unified, regulatory-acceptable framework remains limited. This review proposes an AI-centred, multi-tier decision-support framework, to enable a phased shift toward animal-reduced and, ultimately, animal-free dru

Combining machine learning and iterative experiments to keep pace with emerging viral variants of concern.

Modeling and predicting viral mutations before they emerge plays a crucial role in pandemic preparedness, enabling the early identification of emerging variants of concern (VOCs) and guiding timely updates to vaccines, diagnostic tests, and therapeutic strategies. However, existing machine learning models and large-scale experiments lose their predictive power as viral variants evolve further from the original strains in sequence space. Here, we present a scalable framework that integrates random forest and neural network machine learning models with targeted high-throughput experimentation to

Prevention and repair of circulatory shock and cerebral ischemia/injury by various agents in experimental heatstroke.

The current report summarized animal models of heatstroke experimentation that advance our current knowledge of therapeutic effects on cerebrovascular dysfunction, hypercoagulable state and/or systemic inflammation with various agents in the setting of heatstroke. This was a narrative review of selected published primary basic literature from MEDLINE for 1973-2006. It was found that rodents shared with humans almost the same heatstroke reactions such as hyperpyrexia, hypotension, hyperventilation, pulmonary edema, hepatic and renal failure, hypercoagulable state, metabolic acidosis, systemic i

Medical treatment of patients with hypertrophic cardiomyopathy: An overview of current and emerging therapy.

Several treatments have demonstrated safety and effectiveness in the treatment of patients with hypertrophic cardiomyopathy; however, no drug has been shown to modify the natural history of the disease or to decrease maximal wall thickness. Improvement in our knowledge of the physiopathology of the disease has permitted the development of new therapeutical approaches, including sarcomere modulators and gene therapy. A sarcomere modulator - mavacamten - has been shown to improve exercise capacity, left ventricular outflow tract obstruction, New York Heart Association functional class and health

Mesenchymal stem cells in osteobiology and applied bone regeneration.

Bone marrow contains a population of rare progenitor cells capable of differentiating into bone, cartilage, muscle, tendon, and other connective tissues. These cells, referred to as MSCs, can be purified and culture expanded from animals and humans. This review summarizes recent experimentation focused on characterizing the cellular aspects of osteogenic differentiation, and exploration of the potential for using autologous stem cell therapy to augment bone repair and regeneration. The authors have completed an array of preclinical studies showing the feasibility and efficacy of MSC based impl

[A new microfilaricide in the therapeutic arsenal of the ophthalmologist].

With no doubt the Ophthalmologist in Tropical African country is the practician most needed for ocular disorders which are precursors of blindness by onchocerciasis or loasis. So, he has to be well-informed concerning each new product able to bring a better efficacy in the treatment of his patients suffering from one or two of these endemic filariasis. He has to know as well how to use it perfectly to experiment it if needed. Ivermectine, MECTIZAN patented, is nowadays the microfilariacide most tolerated in human medicine and is very easy to use in mass therapeutic campaigns as it is easy to a

Ethical, anatomical and physiological issues in developing vestibular implants for human use.

Effort towards the development of a vestibular implant for human use are being made. This paper will summarize the first important steps conducted in Geneva towards this ambitious goal. Basically, we have faced three major issues. First, an ethical issue. While it was clear that such development would require the collaboration of human volunteers, it was also clear that stimulation of the vestibular system may produce periods of significant incomfort. We know today how to minimize (and potentially eliminate) this type of incomfort. The second issue was anatomical. The anatomical topology of th

Emerging Mitochondrial Therapeutic Targets in Optic Neuropathies.

Optic neuropathies are an important cause of blindness worldwide. The study of the most common inherited mitochondrial optic neuropathies, Leber hereditary optic neuropathy (LHON) and autosomal dominant optic atrophy (ADOA) has highlighted a fundamental role for mitochondrial function in the survival of the affected neuron-the retinal ganglion cell. A picture is now emerging that links mitochondrial dysfunction to optic nerve disease and other neurodegenerative processes. Insights gained from the peculiar susceptibility of retinal ganglion cells to mitochondrial dysfunction are likely to infor

This current review summarized animal models of heatstroke experimentation that promote our current knowledge of therapeutic effects on cerebrovascular dysfunction, coagulopathy, and/or systemic inflammation with human umbilical cord blood cells (HUCBCs) or estrogen in the setting of heatstroke. Accumulating evidences have demonstrated that HUCBCs provide a promising new therapeutic method against neurodegenerative diseases, such as stroke, traumatic brain injury, and spinal cord injury as well as blood disease. More recently, we have also demonstrated that post- or pretreatment by HUCBCs may
