Latest Advances in Obesity Treatment by Regulating Adipocyte Thermogenesis.

Obesity is driven by chronic energy imbalance and is associated with multiple metabolic diseases. As adipocyte thermogenesis contributes to energy expenditure, it is being actively investigated as a possible target for obesity treatment. This review summarizes recent progress in the molecular regulation of adipocyte thermogenesis, focusing mainly on preclinical mechanistic evidence from cellular and animal studies, while discussing clinical and human data only where available. We review emerging regulators within the cAMP pathway, the PRDM16-PPARγ axis, and mitochondrial quality control and me

Mitochondria-Targeted Liposomes Boost Thermogenesis for Adipose Tissue Regulation.

Mitochondria act as central organelles regulating energy metabolism and lipogenesis in adipocytes. Mitochondrial dysfunction and oxidative stress are closely implicated in the pathogenesis of obesity. Therefore, targeting adipose mitochondrial function to reduce mitochondrial reactive oxygen (mROS) levels can enhance mitochondrial activity and promote thermogenesis, presenting a promising therapeutic strategy for obesity. In this study, we developed cationic liposomes (AE@PEP-Lip) specifically designed for adipose mitochondrial targeting. These liposomes can effectively accumulate in white adi

Adipose tissue-gut microbiome crosstalk in inflammation and thermogenesis.

Previously characterized as inert fat depots, adipocytes are now recognized as dynamic mediators of inflammatory tone, metabolic health, and nutrient homeostasis. As endocrine organs, specialized depots of adipose tissue engage in crosstalk between the gut, liver, pancreas, and brain to coordinate appetite, thermogenesis, and ultimately body weight. These functions are tightly linked to the inflammatory status of adipose tissue, which is in turn influenced by the health of the gut microbiome. Here, we review recent findings linking specific gut microbes and their secreted factors, including re

CLCF1 inhibits energy expenditure via suppressing brown fat thermogenesis.

Brown adipose tissue (BAT) is the main site of nonshivering thermogenesis which plays an important role in thermogenesis and energy metabolism. However, the regulatory factors that inhibit BAT activity remain largely unknown. Here, cardiotrophin-like cytokine factor 1 (CLCF1) is identified as a negative regulator of thermogenesis in BAT. Adenovirus-mediated overexpression of CLCF1 in BAT greatly impairs the thermogenic capacity of BAT and reduces the metabolic rate. Consistently, BAT-specific ablation of CLCF1 enhances the BAT function and energy expenditure under both thermoneutral and cold c

Nutritionally-induced alterations in thyroid hormone metabolism and thermogenesis.

Serum 3,5,3'-triiodothyronine (T3) is increased by overnutrition and decreased by starvation. The production rate of T3 was increased by overfeeding (82%), but T4 production did not change. Feeding a weight maintenance diet with high CHO mimiced serum T3 and rT3 changes of overfeeding, substitution of CHO by FAT those of starvation. VO2 increased during overfeeding and in T4 equilibrated volunteers supplemented with 40 microgram T3 daily for 2 weeks. No weight change occurred in the latter subjects. Yet no decrease in VO2 was found when serum T3 was reduced by the administration of iopanoic ac

Thermogenesis induced by intravenous infusion of hypertonic solutions in the rat.

1. Intravenous administration of 20-60 % glucose, 3.2-9.7 % NaCl or 20 % mannitol solutions (1.66 ml kg(-1)) for 5 min increased oxygen consumption in urethane-anaesthetized rats, whereas administration of physiological saline had no effect. Administration of 7.7-18.3 % urea slightly increased the oxygen consumption, but the increase was significantly smaller than that measured after the administration of other hypertonic solutions. The magnitude of the thermogenic effect correlated with the osmolality of the applied solutions. These results suggest that the thermogenesis was caused mainly by

Thyroid hormones and thermogenesis: the metabolic cost of food and exercise.

To mimic plasma T3 levels observed in a previous overfeeding study, six lean healthy men received replacement amounts of L-thyroxine (200 micrograms/d) to block endogenous thyroid hormone production while consuming their habitual diet. After 4 weeks equilibration on T4, L-triiodothyronine (T3) was given (45 micrograms/d) in addition to T4, to produce mild T3-thyrotoxicosis, for another 2 weeks. At the end of this period T3 was discontinued but the subjects continued to receive T4 for another 2 weeks. Resting metabolic rate, exercise efficiency, and the thermic effect of food were measured usin

Gut bacterial regulation of primate adaptive thermogenesis at high altitude.

BACKGROUND: Adaptive thermogenesis is a fundamental physiological process by which mammals maintain their body temperature through shivering and non-shivering thermogenesis to adapt to environmental changes. The activation of brown adipose tissue (BAT) is the core of non-shivering thermogenesis. However, the specific mechanisms by which gut-derived bacteria trigger BAT to activate adaptive thermogenesis remain poorly understood. Environmental variations across different altitudes create unique temperature gradients, providing an ideal condition for identifying bacteria associated with cold ada

Apolipoprotein A4 Elevates Sympathetic Activity and Thermogenesis in Male Mice.

Long-chain fatty acids induce apolipoprotein A4 (APOA4) production in the small intestine and activate brown adipose tissue (BAT) thermogenesis. The increase in BAT thermogenesis enhances triglyceride clearance and insulin sensitivity. Acute administration of recombinant APOA4 protein elevates BAT thermogenesis in chow-fed mice. However, the physiological role of continuous infusion of recombinant APOA4 protein in regulating sympathetic activity, thermogenesis, and lipid and glucose metabolism in low-fat-diet (LFD)-fed mice remained elusive. The hypothesis of this study was that continuous inf

Central Apolipoprotein A-IV Stimulates Thermogenesis in Brown Adipose Tissue.

Stimulation of thermogenesis in brown adipose tissue (BAT) could have far-reaching health benefits in combatting obesity and obesity-related complications. Apolipoprotein A-IV (ApoA-IV), produced by the gut and the brain in the presence of dietary lipids, is a well-known short-term satiating protein. While our previous studies have demonstrated reduced diet-induced thermogenesis in ApoA-IV-deficient mice, it is unclear whether this reduction is due to a loss of peripheral or central effects of ApoA-IV. We hypothesized that central administration of ApoA-IV stimulates BAT thermogenesis and that

Mitochondrial pyruvate carrier is required for optimal brown fat thermogenesis.

Brown adipose tissue (BAT) is composed of thermogenic cells that convert chemical energy into heat to maintain a constant body temperature and counteract metabolic disease. The metabolic adaptations required for thermogenesis are not fully understood. Here, we explore how steady state levels of metabolic intermediates are altered in brown adipose tissue in response to cold exposure. Transcriptome and metabolome analysis revealed changes in pathways involved in amino acid, glucose, and TCA cycle metabolism. Using isotopic labeling experiments, we found that activated brown adipocytes increased

Rat behavioral thermoregulation integrates with nonshivering thermogenesis during postnatal development.

Rat pups are capable of behavioral thermoregulation, both in the nest and on a thermocline, as early as the 1st week of postnatal life, and these pups can also produce heat metabolically without shivering. The rat pup's primary source of nonshivering thermogenesis is the sympathetically mediated metabolism of brown adipose tissue (BAT). BAT is well formed in newborns and functions shortly after birth. While infant behavioral thermoregulation and BAT thermogenesis have been extensively studied, little is known about the extent to which thermoregulatory behavior can be influenced by BAT thermoge

DUSP4 activates adipocyte thermogenesis via CRTC3 dephosphorylation-dependent UCP1 expression.

Obesity arises when a chronic energy surplus overwhelms the storage capacity of adipocytes and subsequently causes systemic metabolic dysfunction. Adaptive thermogenesis, primarily mediated by uncoupling protein 1 (Ucp1) in adipocytes, serves to counteract obesity by dissipating excess energy as heat. Despite extensive studies on thermogenic regulation, the phosphatases that regulate this protective metabolic pathway remain poorly understood. Here we identify dual-specificity phosphatase 4 (Dusp4) as a critical modulator of adipocyte thermogenesis via direct modulation of CREB-regulated transc

STAU1 fine-tunes adipose thermogenesis by regulating Ucp1 mRNA stability.

STAU1, an RNA-binding protein central to the STAU1-mediated mRNA decay (SMD) pathway, promotes adipogenesis. This study reveals that STAU1 protein levels positively correlate with obesity severity. Adipose specific STAU1 deficient mice on a high-fat diet showed reduced weight, enhanced thermogenesis, and improved glucose tolerance. Deleting Stau1 gene in brown adipose tissue upregulated UCP1 protein. We further identified that STAU1 binds to the 3' UTR of Ucp1 mRNA. The β3 adrenergic receptor pathway enhances SMD activity, while inhibiting the cAMP-PKA pathway downregulates STAU1. Our findings

ASB3 limits adipocyte thermogenesis and energy expenditure through p62 ubiquitination.

BACKGROUND: Adipose thermogenesis increases energy expenditure and protects against obesity. However, the endogenous mechanisms that restrain this process are not fully understood. METHODS: Adipose tissue- and adipocyte-specific Asb3-deficient mice were exposed to cold or treated with the β3-adrenergic receptor agonist CL-316,243. Primary thermogenic adipocytes were used to assess adipogenic differentiation and β3-adrenergic/cAMP-PKA responses. Proteomic/phosphoproteomic profiling, co-immunoprecipitation, ubiquitination assays and domain mapping were performed to define the mechanism. Adipose

Nicotinamide enhances lipid deposition and thermogenesis in goat brown adipocytes.

Brown adipose tissue (BAT) serves as a crucial thermogenic tissue in newborn goats, playing an important role in maintaining their core body temperature through adaptive thermogenesis. Initially, we found that nicotinamide (NAM) content was significantly higher in BAT (0.047 μg/g, n = 6 biological replicates) than that in white adipose tissue (WAT, 0.021 μg/g, n = 6 biological replicates). However, the effect of NAM on goat brown adipocytes remains unknown. The experimental results revealed that NAM enhanced lipid deposition, mitochondrial DNA content and the expression of uncoupling protein 1

Β 2 AR Agonists Sustain Thermogenesis and Leanness via Sympathofacilitation.

UNLABELLED: Human thermogenesis depends on β2-adrenoceptors (β2AR) expressed in thermogenic adipocytes, which are activated by norepinephrine released from sympathetic neurons. Whether β2AR also modulates thermogenesis via direct presynaptic action within sympathetic neurons has remained unclear. Here, we identify Adrb2 expression in human and rodent cervical sympathetic neurons. β2AR agonism exerts neurotrophic effects and facilitates cholinergic responsiveness in mouse sympathetic neurons, indicating a sympathofacilitatory role. Selective deletion of β2AR in sympathetic neurons leads to impa

From Muscle-Based Nonshivering Thermogenesis to Malignant Hyperthermia in Mammals.

For physiological processes in the vital organs of eutherian mammals to function, it is important to maintain constant core body temperature at ∼37°C. Mammals generate heat internally by thermogenesis. The focus of this review is on heat generated in resting skeletal muscles, using the same cellular components that muscles use to regulate cytoplasmic calcium concentrations [Ca2+] and contraction. Key to this process, known as muscle-based nonshivering thermogenesis (MB-NST), are tiny Ca2+ movements and associated ATP turnover coordinated by the plasma membrane, sarcoplasmic reticulum (SR), and
