Transient receptor potential melastatin 8 (TRPM8) is a cold-sensing thermoreceptor cation channel; however, its functional role in endotoxin-induced neuroinflammation remains unclear. In the present study, we investigated chronic sickness responses in TRPM8 knockout (KO) mice during lipopolysaccharide (LPS)-induced sepsis. The intraperitoneal administration of 5 mg/kg LPS generated longer-lasting hypothermia in TRPM8 KO mice than in wild-type (WT) mice. TRPM8 KO mice also exhibited longer-lasting declines in locomotor activity, body weight, and food and water intakes than WT mice upon LPS admi

The thermal sensitivity of different parts of the body was investigated by heating large areas of the body surface while the mean skin temperature calculated from Hardy and DuBois ' formula (1938) was always kept constant. The right arm sweating responses recorded under a local thermal clamp were related to changes in segmental skin temperatures of the different parts of the body. The results show that: 1) the various local peripheral signals are projected into integrating structures in the central nervous system; 2) the thermal sensitivity is greater for the head-and-trunk area in comparison

In cats afferent impulses from splanchnic and vagus nerve preparations were studied. In each splanchnic nerve serving the stomach or the adjacent part of the duodenum, cold-sensitive afferents could be recorded. There were also numerous mechanosensitive fibers originating from stomach, intestine, mesentery and the region of blood vessels. No thermal afferents were found in the vagus nerve. The cardiovascular and respiratory myelinated afferents were insensitive to thermal stimuli.

Strongyloides stercoralis, a skin-penetrating nematode parasite of homeotherms, migrates to warmth. In nematodes, the amphids, anteriorly positioned, paired sensilla, each contain a bundle of sensory neurons. In the amphids of the free-living nematode Caenorhabditis elegans, a pair of neurons, each of which ends in a cluster of microvilli-like projections, are known to be the primary thermoreceptors, and have been named the finger cells (class AFD). A similar neuron pair in the amphids of the parasite Haemonchus contortus is also known to be thermosensory. Strongyloides stercoralis lacks finge…

The capacity of cutaneous, including trigeminal endings, to detect chemicals is known as chemesthesis or cutaneous chemosensation. This sensory function involves the activation of nociceptor and thermoreceptor endings and has a protective or defensive function, as many of these substances are irritants or poisonous. However, humans have also developed a liking for the distinct sharpness or pungency of many foods, beverages, and spices following activation of the same sensory afferents. Our understanding of the cellular and molecular mechanisms of chemosensation in the trigeminal system has exp

In mammals, environmental cold sensing conducted by peripheral cold thermoreceptor neurons mostly depends on TRPM8, an ion channel that has … the plasma membrane. Importantly, basal kinase activity negatively modulates TRPM8 function at cold thermoreceptors from male and female mice, an observation accounted for by mathematical modeling. Overall, our findings suggest that cold temperature detection could be rapidly and reversibly fine-tuned by controlling the TRPM8 basal phosphorylation state, a mechan …

Considering the female breast to be a secondary sexual organ the sensibility of nipple and areola gains essentially in functional significance. This sensory function is impaired as well by reduction mammaplasty. Thus all different operation methods propagated in recent years have to be rated as to functional and esthetic results with regard to resection weight. The optimal operation method presupposes: 1. Satisfactory esthetic result, 2. optional reductional possibility up to subcutaneous mastectomy, 3. minor sensory impairment of nipple and areola.

The novel observation of a palatal retial organ in the bowhead whale (Balaena mysticetus) is reported, with characterization of its form and function. This bulbous ridge of highly vascularized tissue, here designated the corpus cavernosum maxillaris, runs along the center of the hard palate, expanding cranially to form two large lobes that terminate under the tip of the rostral palate, with another enlarged node at the caudal terminus. Gross anatomical and microscopic observation of tissue sections discloses a web-like internal mass with a large blood volume. Histological examination reveals l

Axons are the initial sites of thermal signal detection and transduction, but their localized response to heat remains poorly characterized. Here, we present a photothermal nerve-on-a-chip platform that reconstructs in vitro the key elements of axonal thermosensation. By integrating graphene-based photothermal heaters with microelectrode arrays inside axon-guiding microchannels, the platform enables localized, millisecond-scale thermal stimulation and simultaneous extracellular recording of action potentials. Using this platform, we uncovered rapid and reproducible heat-evoked responses in rat

BACKGROUND: The mechanism of nasal airflow sensation is poorly understood. This study aimed to examine the role of nasal mucosal temperature change in the subjective perception of nasal patency and the methods by which it can be quantified.
METHOD: Medline and PubMed database searches were performed to retrieve literature relevant to the topic.
RESULTS: The primary mechanism producing the sensation of nasal patency is thought to be the activation of transient receptor potential melastatin family member 8 ('TRPM8'), a thermoreceptor that is activated by nasal mucosal cooling. Computational fl

Topical capsaicin is reported to be an effective treatment for idiopathic intractable pruritus ani. While both capsaicin and menthol application produce a transient perianal burning sensation, only capsaicin relieves itching. Classical observations on functional desensitisation of nociceptors by capsaicin may explain the beneficial effects but the recent discovery of a range of receptors which respond to capsaicin, menthol, and temperature, and their expression in subsets of sensory nerve fibres, provides an exciting prospect towards advancing our understanding and treatment of sensory dysfunc

This study was designed to further characterize the properties of previously described laryngeal cold receptors (Respir. Physiol. 59:35, 1985). Single unit action potentials were recorded from the internal branch of the superior laryngeal nerve (SLN) in anesthetized, spontaneously breathing dogs. The nervous conduction of fibers originating from 12 laryngeal cold receptors was blocked at a mean (+/- SE) temperature of 18.8 +/- 0.7 degrees C. Twelve receptors were localized on the edge of the vocal folds in correspondence of the vocal process of the arytenoid cartilage. Topical anesthesia (2% l

Temperature homeostasis is controlled by the vagus nerve. Thermal information is collected by thermoreceptors present in the viscera and driven across sensory neurons of the nodose ganglia (NG), which in turn send it to the hypothalamus. While transient receptor potential channels (TRPs) are traditionally considered for thermal transduction, TREK channels belonging to the two-pore domain K+ channels family are emerging as thermosensors, but their role in the NG remains understudied. Patch-clamp recordings revealed that increasing the temperature to physiological levels causes a hyperpolarizati…

Caenorhabditis elegans navigates thermal gradients by using a behavioral strategy that is regulated by a memory of its cultivation temperature (T(c)). At temperatures above or around the T(c), animals respond to temperature changes by modulating the rate of stochastic reorientation events. The bilateral AFD neurons have been implicated as thermosensory neurons, but additional thermosensory neurons are also predicted to play a role in regulating thermotactic behaviors. Here, we show that the AWC olfactory neurons respond to temperature. Unlike AFD neurons, which respond to thermal stimuli with

The aim of the present study was to describe the types and location of sensory nerve endings in the glans penis of the rat on the basis of light and electron microscopy. Free nerve endings are abundant throughout the glans and are derived from either thinly myelinated A delta or unmyelinated C fibers. The free nerve endings derived from A delta fibers often extend into the epidermis as far as the granular layer and exhibit buttonlike terminal thickenings. The distal urethra contains many free nerve endings from C fibers. Numerous lamellated corpuscles are present. Each corpuscle, innervated by

Fully developed eccrine glands distributed over the entire integument are found only in man. The secretory mechanism is based on the active transport of sodium ions; water diffuses passively following the osmotic gradient. The thermoregulatory center in the hypothalamic region receives impulses from external and internal thermoreceptors and is modified by levels of hormones, by emotions, physical activity and pyrogens. Heat loss is controlled by the blood flow through the skin and by activity of eccrine glands. Thermoregulatory sweating occurs mainly on head and trunk. Emotions trigger eccrine…
