Purification and characterization of a novel recombinant highly enantioselective short-chain NAD(H)-dependent alcohol dehydrogenase from Thermus thermophilus.

The gene encoding a novel alcohol dehydrogenase (ADH) that belongs to the short-chain dehydrogenase/reductase (SDR) superfamily was identified in the extremely thermophilic, halotolerant gram-negative eubacterium Thermus thermophilus HB27. The T. thermophilus ADH gene (adh(Tt)) was heterologously overexpressed in Escherichia coli, and the protein (ADH(Tt)) was purified to homogeneity and characterized. ADH(Tt) is a tetrameric enzyme consisting of identical 26,961-Da subunits composed of 256 amino acids. The enzyme has remarkable thermophilicity and thermal stability, displaying activity at tem

Searching for Frataxin Function: Exploring the Analogy with Nqo15, the Frataxin-like Protein of Respiratory Complex I from Thermus thermophilus.

Nqo15 is a subunit of respiratory complex I of the bacterium Thermus thermophilus, with strong structural similarity to human frataxin (FXN), a protein involved in the mitochondrial disease Friedreich's ataxia (FRDA). Recently, we showed that the expression of recombinant Nqo15 can ameliorate the respiratory phenotype of FRDA patients' cells, and this prompted us to further characterize both the Nqo15 solution's behavior and its potential functional overlap with FXN, using a combination of in silico and in vitro techniques. We studied the analogy of Nqo15 and FXN by performing extensive databa

Studying The Role of Aprotic and Protic Solvents on Extremophilic Laccase From Thermus thermophilus for Solvent-Mediated One-Pot Biocatalysis.

In present times, globally, solvent-mediated biocatalysis is gaining a pivotal role in sustainable biotechnological solutions for organic synthesis. The versatile green catalyst, laccase, a multicopper oxidoreductase, has gained significant importance in aromatic bioconversion in solvent systems. Laccases portray varied behavioral patterns and functionality in solvents. With this interest, the influence of aprotic polar organic solvents and a benchmark out-of-the-ordinary protic solvent, an ionic liquid, i.e., 1-Ethyl-3-methylimidazolium acetate (EmimOAc), on extremophilic laccase from Thermus

UV-induced crosslinks in the 16S rRNAs of Escherichia coli, Bacillus subtilis and Thermus aquaticus and their implications for ribosome structure and photochemistry.

Sixteen long-range crosslinks are induced in Escherichia coli 16S rRNA by far-UV irradiation. Crosslinking patterns in two other organisms, Bacillus subtilis and Thermus aquaticus, were investigated to determine if the number and location of crosslinks in E.coli occur because of unusually photoreactive nucleotides at particular locations in the rRNA sequence. Thirteen long-range crosslinks in B.subtilis and 15 long-range crosslinks in T.aquaticus were detected by gel electrophoresis and 10 crosslinks in each organism were identified completely by reverse transcription analysis. A novel type III pullulan hydrolase from the bacterium Thermus scotoductus: Biochemical characterization and functional analysis of its carbohydrate-binding domains.

A novel type III pullulan hydrolase (PulTS) was identified from the thermophilic bacterium Thermus scotoductus, representing the first bacterial member of this enzyme class. The recombinant PulTS exhibited optimal activity at pH 6.5 and 80 °C, with a specific activity of 46.3 ± 2.4 U/mg toward pullulan. The enzyme hydrolyzed pullulan into maltotriose, panose, maltose, and glucose, and also degraded soluble starch, amylose, amylopectin, dextran, glycogen, and γ-cyclodextrin. PulTS contains sequentially arranged carbohydrate-binding module family 48 (CBM48), CBM34, and glycoside hydrolase family

Immobilized Wild-Type and Mutant L‑Alanine Dehydrogenase from Thermus thermophilus Coupled with Formate Dehydrogenase for Asymmetric Synthesis of Noncanonical Amino Acids.

The asymmetric synthesis of noncanonical amino acids (ncAAs) is of increasing interest due to their importance as precursors for pharmaceuticals, enzyme inhibitors, and functional peptide building blocks. In this study, wild-type and Tyr92Ser mutant l-alanine dehydrogenases from Thermus thermophilus (TtAlaDH) (WT and Mut, respectively) were immobilized using three different strategies: covalent attachment on Eupergit C 250 L (EC250L), cross-linked enzyme aggregates (CLEAs), and entrapment in poly-(vinyl alcohol) (PVA) gel. Following a comparison of immobilization yield and recovered activity,

Observation of ligand transfer in ba3 oxidase from Thermus thermophilus: simultaneous FTIR detection of photolabile heme a3(2+)-CN and transient Cu(B)(2+)-CN complexes.

FTIR and light-minus-dark FTIR spectroscopy have been employed to investigate the reaction of oxidized and fully reduced ba(3) oxidase with cyanide. The characterization of the structures of the bound CN(-) in the binuclear heme Fe-Cu(B) center is essential, given that a central issue in the function of ba(3) oxidase is the extent to which the partially reduced substrates interact with the two metals. In the reaction of oxidized ba(3) oxidase with cyanide the initially formed heme a(3)(3+)-C≡N-Cu(B)(2+) species with ν(CN) frequency at 2152 cm(-1) was replaced by a photolabile complex with a fr

Elucidation of D-alanine biosynthetic pathways in the hyperthermophiles: Characterization of alanine racemase from Thermus thermophilus and the structure and dual physiological functions of lysine racemase from Thermotoga maritima.

The hyperthermophilic bacterium Thermotoga maritima contains a peptidoglycan comprising D-alanine, D-glutamate, and D-lysine. We have previously identified the biosynthetic pathways of D-lysine and D-glutamate; however, the biosynthetic pathway of D-alanine is unclear in T. maritima. The present study elucidated the D-alanine biosynthetic pathway in T. maritima, following the characterization of the TTHA0062 activity, constructed a TTHA0062 (encoding alanine racemase)-deficient Thermus thermophilus (ΔTTHA0062) strain. TTHA0062 exhibited the racemase activity toward 10 amino acids including Ala

The mixed valence state of the oxidase binuclear centre: how Thermus thermophilus cytochrome ba3 differs from classical aa3 in the aerobic steady state and when inhibited by cyanide.

In the aerobic steady state of the classical eukaryotic cytochrome c oxidase, three aa(3) redox metal centres (cytochrome a, CuA and CuB) are partially reduced while the fourth, cytochrome a(3), remains almost fully oxidized. Turnover depends primarily upon the rate of cytochrome a(3) reduction. When prokaryotic cytochrome c-552 oxidase (ba(3)) of Thermus thermophilus turns over, three different metal centres (cytochromes b, a(3) and CuA) share the steady state electrons; it is the fourth, CuB, that apparently remains almost fully oxidized until anaerobiosis. Cytochrome a(3) stays partially re

Reaction of cyanide with cytochrome ba3 from Thermus thermophilus: spectroscopic characterization of the Fe(II)a3-CN.Cu(II)B-CN complex suggests four 14N atoms are coordinated to CuB.

Cytochrome ba3 from Thermus thermophilus reacts slowly with excess HCN at pH 7.4 to create a form of the enzyme in which CuA, cytochrome b, and CuB remain oxidized, while cytochrome a3 is reduced by one electron, presumably with the formation of cyanogen. We have examined this form of the enzyme by UV-visible, resonance Raman, EPR, and electron nuclear double resonance spectroscopies in conjunction with permutations of 13C- and 15N-labeled cyanide. The results support a model in which one CN- binds through the carbon atom to ferrous a3, supporting a low-spin (S = 0) configuration on the Fe; br

Extremely thermophilic gram-negative bacteria from hot tap water.

Two strains of heterotrophic, non-motile, Gram-negative extreme thermophiles have been isolated from hot tap water. These strains (NH and DI) have been characterized and compared with strains of genus Thermus. Few of the single organic compounds tested supported growth in the presence of ammonium salts, and, like Thermus strains, growth on undefined media was restricted to dilute tryptone-yeast extract-mineral salt solutions. Nutrients agar and similar common laboratory media did not support growth. The growth rate was similar to that of Thermus strains, as was the unusual pattern of antibioti

Polymerase resistance to polymerase chain reaction inhibitors in bone*.

Amplification of DNA from aged or degraded skeletal remains can be a challenging task, in part due to naturally occurring inhibitors of the polymerase chain reaction. PCR inhibitors may act by inactivating a polymerase itself, or compete with or bind other reaction components, although various polymerases may be differentially susceptible to such insult. In this study, ten thermostable polymerases from six bacterial species were examined for their ability to amplify DNA in the presence of bone-derived or individual PCR inhibitors. Two polymerases, one from Thermus aquaticus and one from Thermu

Inoculation effect of thermophilic microorganisms on protease production through solid-state fermentation under non-sterile conditions at lab and bench scale (SSF).

The production of protease enzyme was evaluated through the solid state fermentation (SSF) of soy fibre, a waste product that acted as a sole substrate for the fermentation, at a laboratory and bench scale using a 500-mL (batch size 115 g) and 10-L (batch size 2300 g) bioreactors. The objective was to assess the effect of the inoculation of the thermophilic bacteria Thermus sp. on the production of the enzyme when working at laboratory and bench scale under non-sterile conditions, since scaling-up and the need of sterilization are the main challenges of SSF, preventing its industrial developme

Structural Hairpin Anchoring-Mediated TtAgo Activity Regulation for Programmable Biosensing.

Thermus thermophilus Argonaute (TtAgo) has shown exceptional promise in biosensing and molecular diagnostics owing to its programmable and DNA-guided specific cleavage activity. However, the potential of regulating TtAgo activity through precise structural modification of guide DNA (gDNA) remains to be fully utilized, thereby limiting the flexible expansion of TtAgo-based biosensing strategies. In this work, TtAgo achieves optimal cleavage activity strictly with a specific linear single-stranded gDNA (linear gDNA) length range (about 15-21 nt), whereas an excessively long linear gDNA fails to

Genomic adaptation of prokaryotic organisms at high temperature.

One of the central issues of evolutionary genomics is to find out the adaptive strategies of microorganisms to stabilize nucleic acid molecules under high temperature. Thermal adaptation hypothesis gives a link between G+C content and growth temperature if there is a considerable variation of guanine and cytosine content between species. However, there has been a long-standing debate regarding the correlations between genomic GC content and optimal growth temperature (Topt). We urged that adaptation to growth at high temperature requires a coordinated set of evolutionary changes affecting: (i)
