Evaluation of systemic and mucosal immune responses in Nile tilapia following intraperitoneal and immersion vaccination with inactivated tilapia lake virus (TiLV) vaccine.

Tilapia lake virus (TiLV), an orthomyxovirus, poses a significant threat to global tilapia aquaculture. However, there has been limited success in developing commercial vaccines. The current study assessed the protective efficacy of a cell culture-derived, formalin-inactivated whole virus vaccine for Nile tilapia (Oreochromis niloticus) administered via injection or immersion. Upon viral challenge, the vaccinated fish yielded relative percent survivals of 100% and 83.88% to injection and immersion vaccinations, respectively. Immune responses were assessed through standardized IgM and IgZ indir

Characterization of Staphylococcus aureus isolated from tilapia and utensils used in the commerciali- zation of tilapia in the street markets of a semi-arid Brazilian municipality.

BACKGROUND: The production and commercialization of tilapia (Oreochromis niloticus) is fundamentally important to the semi-arid region of northeastern Brazil. In this region, one of the main forms of commercialization occurs in street markets (fairs). A high incidence of methicillin-resistant Staphylococcus aureus (MRSA) strains was previously detected in different food-related environments in Brazil. However, limited data is available about the presence of MRSA in street markets commercializing tilapias. In this study, we identified Staphylococcus aureus from tilapia and utensils used in the

Molecular characterization of TRADD in GIFT tilapia (Oreochromis niloticus).

Tumor necrosis factor receptor type 1-associated death domain protein (TRADD) is a crucial adaptor in the TNF signaling pathway. The role of fish TRADD remains less understood, compared with that of mammals. In the present study, TRADD of genetically improved farmed tilapia (GIFT, Oreochromis niloticus) was characterized. The GIFT TRADD encoded a putative protein of 291 amino acids, containing an N-terminal domain and C-terminal death domain. qPCR analysis revealed that GIFT TRADD was ubiquitously expressed in all selected tissues and upregulated in spleen following lipopolysaccharide (LPS) or

Gastrointestinal dysbiosis induced by Nocardia sp. infection in tilapia.

Nocardiosis has caused high mortalities among fish cultures; however, the effects of Nocardia infections in the fish gastrointestinal microbiota are unknown. In this research, tilapia was infected with Nocardia sp., to analyze the effect of infection on the gastrointestinal microbiota. Tilapia infected with Nocardia sp. reported a 46 % survival (100 % in non-infected). Moreover, the infection caused severe damage to the stomach microbiota, with a loss of diversity and a significant increase of Proteobacteria (94.8 %), resulting in a negative correlation network between Proteobacteria and other

Localized lymphatic sporotrichosis after fish-induced injury (Tilapia sp.).

Localized lymphatic sporotrichosis generally develops after the fungus Sporothrix schenckii is traumatically introduced into skin or mucosa by contaminated plant material. An 18-year-old male fisherman was injured by spines of the dorsal fin of a fish on the left third finger. The lesion became ulcerated, edematous and suppurative and did not respond to tetracycline and cephalexin. Fifteen days after the accident, a nodular lymphangitic pattern of swelling was observed. Histopathological findings and an intradermal test were suggestive of sporotrichosis and mycological cultures confirmed the d

Transcriptome analysis of tilapia streptococcus agalactiae in response to baicalin.

Streptococcus agalactiae (S. agalactiae) is a highly pathogenic bacterial pathogen in aquatic animals. Our previous study has demonstrated the significant inhibitory effect of baicalin on β-hemolytic/cytolytic activity, which is a key virulence factor of S. agalactiae. In this study, we aimed to elucidate the mechanism underlying baicalin's inhibition of S. agalactiae β-hemolytic/cytolytic activity by transcriptomic analysis. Bacteria were exposed to 39.06 µg/mL baicalin for 6 h, and their β-hemolytic/cytolytic activities were assessed using blood plates. Then, the differentially expressed gen

Involvement of galanin in reproduction of male tilapia, Oreochromis mossambicus.

Galanin(GAL) is recognised for its involvement in various physiological, behavioural, and neuroendocrine functions in vertebrates. Yet, the impact of this peptide on spermatogenesis in fish remains understudied. In this investigation, the administration of 5 µg of GAL receptor agonist (GAL-A) to the tilapia Oreochromis mossambicus did not significantly alter the numbers of different germ cells except for a significant rise in the number of late spermatids compared to the control group. By contrast, treatment with 20 µg of GAL-A caused a significant rise in all germ cell counts and tubule and l

dmrt1 Is Responsible for Androgen-Induced Masculinization in Nile Tilapia.

17α-Methyltestosterone (MT) is a widely used androgen for all-male fish production in aquaculture. However, the molecular mechanism underlying MT-induced masculinization remains unclear. In this study, we aim to identify the key gene responsible for MT-induced masculinization using the Nile tilapia (Oreochromis niloticus) amhy, dmrt1, and gsdf mutants, which exhibit male-to-female sex reversal. Nile tilapia fry from these three mutant lines were treated with 50 μg/g MT from 5 to 30 days after hatching (dah). The results showed that amhy and gsdf mutants, but not dmrt1 mutants, were masculinize

Characterization of the somatostatin system in tilapia: implications for growth and reproduction.

Somatostatin (SST) plays diverse physiological roles in vertebrates, particularly in regulating growth hormone secretion from the pituitary. While the function of SST as a neuromodulator has been studied extensively, its role in fish and mammalian reproduction remains poorly understood. To address this gap, we investigated the involvement of the somatostatin system in the regulation of growth and reproductive hormones in tilapia. RNA sequencing of mature tilapia brain tissue revealed the presence of three SST peptides: SST6, SST3, and low levels of SST1. Four different isoforms of the somatost

Neovaginoplasty with tilapia fish skin: a series of eleven cases.

INTRODUCTION AND HYPOTHESIS: Mayer-Rokitansky-Küster-Hauser syndrome affects about 1 in 5000 live female births and is associated with gonadal dysgenesis and primary amenorrhea. Neovaginoplasty has been established as an appropriate treatment option for patients who have failed or denied dilation therapy. In search of accessible, economical material with low risk of complications, the team proposed the use of Nile tilapia fish skin (NTFS) as an innovative biomaterial in the neovaginoplasty procedure for vaginal agenesis management. NTFS has noninfectious microbiota, morphologic structure compa

Identification and characterization of CD5 in Nile tilapia (Oreochromis niloticus).

CD5 is a type I transmembrane glycoprotein acting as a pleiotropic functional receptor in the mammalian immune response system, mainly presents on the surface of cells associated with the immune system, and is essential for the classification of B cells. In this study, we identify a CD5 homologue in Nile tilapia (Oreochromis niloticus). The open reading frame of OnCD5 is 507 bp, encoding 168 amino acids. The deduced amino acid sequence contains a signal peptide region, a transmembrane region and a conserved portion of the cytoplasmic region. Expression analysis indicates that the OnCD5 exhibit

Prolactin signaling in the highly osmotolerant Mozambique tilapia, Oreochromis mossambicus.

Teleost fishes maintain hydromineral balance through the hormonal regulation of epithelial ion transport. In the euryhaline, and remarkably osmotolerant Mozambique tilapia (Oreochromis mossambicus), two prolactin (Prl) isoforms, Prl188 and Prl177, are released from the pituitary in response to hyposmotic stimulation to promote branchial ion absorption. Prl188 and Prl177 bind two Prl receptors (Prlrs), Prlr1 and Prlr2, which are expressed in the pituitary and key ionoregulatory organs, including the gill. To understand how Prl signaling operates across a range of salinities that reflect their s

Gata2a Mutation Causes Progressive Microphthalmia and Blindness in Nile Tilapia.

The normal development of lens fiber cells plays a critical role in lens morphogenesis and maintaining transparency. Factors involved in the development of lens fiber cells are largely unknown in vertebrates. In this study, we reported that GATA2 is essential for lens morphogenesis in Nile tilapia (Oreochromis niloticus). In this study, Gata2a was detected in the primary and secondary lens fiber cells, with the highest expression in primary fiber cells. gata2a homozygous mutants of tilapia were obtained using CRISPR/Cas9. Different from fetal lethality caused by Gata2/gata2a mutation in mice a

Hypoxia-induced HIF-1α promotes Listeria monocytogenes invasion into tilapia.

Listeria monocytogenes is a zoonotic food-borne bacterial pathogen with a solid pathogenicity for humans. After ingestion of highly contaminated food, L. monocytogenes is able to cross the intestine invading phagocytic and nonphagocytic cells and causes listeriosis. China is the world's largest supplier of tilapia. The contamination rate of L. monocytogenes to tilapia products was as high as 2.81%, causing a severe threat to public health. This study revealed the underlying regulatory mechanisms of HIF-1α in the invasion of L. monocytogenes into tilapia under hypoxic environments. This study w

LECT2 Protects Nile Tilapia (Oreochromis niloticus) Against Streptococcus agalatiae Infection.

Leukocyte cell-derived chemotaxin 2 (LECT2) is a multifunctional cytokine that especially plays an important role in innate immune. However, the roles of LECT2 in the immune response of the economically important fish Nile tilapia (Oreochromis niloticus) against bacterial infection remains unclear. In this study, a lect2 gene from Nile tilapia (On-lect2) was identified, and its roles in the fish's immune response against bacterial infection were determined and characterised. On-lect2 contains an open reading frame of 456 bp that encodes a peptide of 151 amino acids, as well as the conservative
