Discovery of a novel orthototivirus-like virus in patients with vulvovaginal candidiasis.

INTRODUCTION: Vulvovaginal candidiasis (VVC) is a common fungal infection affecting women worldwide. Although the vaginal microbiome has been extensively studied, the diversity of viruses present in the vaginal microenvironment remains poorly characterized. METHODS: Vaginal swab samples from patients diagnosed with VVC were subjected to viral metagenomic sequencing using an Illumina NovaSeq platform. Viral contigs were assembled, annotated, and screened against public databases. Genome organization, pairwise sequence identity, and phylogenetic relationships were analyzed to determine the evol

Investigating links between Trichomonas vaginalis, T. vaginalis virus, Mycoplasma hominis, and metronidazole resistance.

INTRODUCTION: Trichomonas vaginalis (TV) is the etiological agent of the common non-viral sexually transmitted infection (STI), trichomoniasis. TV can inherently harbour Mycoplasma hominis and Trichomonas vaginalis virus (TVV) species. Endosymbiosis of TV with M. hominis and TVV may contribute to metronidazole resistance in this pathogen. This study determined the prevalence of TVVs across clinical isolates of TV, as well as the symbiosis between TV, TVV, and M. hominis in relation to metronidazole resistance. METHODOLOGY: Twenty-one clinical isolates of TV were analysed in this study. The is

Transcriptome profiling of immune responses to cardiomyopathy syndrome (CMS) in Atlantic salmon.

BACKGROUND: Cardiomyopathy syndrome (CMS) is a disease associated with severe myocarditis primarily in adult farmed Atlantic salmon (Salmo salar L.), caused by a double-stranded RNA virus named piscine myocarditis virus (PMCV) with structural similarities to the Totiviridae family. Here we present the first characterisation of host immune responses to CMS assessed by microarray transcriptome profiling. RESULTS: Unvaccinated farmed Atlantic salmon post-smolts were infected by intraperitoneal injection of PMCV and developed cardiac pathology consistent with CMS. From analysis of heart samples a

Genomic Characterization of a Novel Victorivirus Detected in a Sample Derived From a Human-Biting Tick in Japan.

Surveillance of tick-associated viruses may contribute to our understanding of viral diversity and evolution. Here, we identified a novel victorivirus in a sample derived from a human-biting tick, Amblyomma testudinarium, in Japan. The viral sequence was identified by metatranscriptomic sequencing of total RNA extracted from Vero cells treated with tick homogenate. The viral sequence was 4621 bp in length and contained two major open reading frames predicted to encode a putative coat protein (CP) and a putative RNA-dependent RNA polymerase (RdRp), respectively. The two open reading frames over

Complete genome sequence of a novel isolate of Victorivirus nijyusani infecting Botrytis cinerea strain G20-5 associated with strawberry gray mold.

In the study, a novel isolate of Victorivirus nijyusani was isolated from Botrytis cinerea strain G20-5, a causal agent of strawberry gray mold, and tentatively named Botrytis cinerea victorivirus 4 (BcVV4). BcVV4 has a double-stranded RNA (dsRNA) of 5135 bp long and contains two open reading frames (ORFs) 1 and 2, which overlap in the AUGA sequence. They are predicted to encode a coat protein (CP) of 756-amino acid (aa) with a molecular weight of 80.2 kDa, and an RNA-dependent RNA polymerase (RdRp) of 850-aa with a molecular weight of 93.3 kDa, respectively. The aa sequence of CP encoded by O

Complete genome sequence of a novel victorivirus from Colletotrichum gloeosporioides.

A novel virus belonging to the genus Victorivirus was identified in the plant-pathogenic fungus Colletotrichum gloeosporioides. Its genome is a double-stranded RNA molecule of 5217 bp containing two open reading frames (ORFs). The 5'-proximal ORF (ORF1) encodes a coat protein (CP) consisting of 734 amino acids (aa), whereas the 3'-proximal ORF (ORF2) encodes an RNA-dependent RNA polymerase (RdRp) of 875 aa. The two ORFs overlap at the tetranucleotide sequence AUGA, facilitating a coupled termination-initiation mechanism, which is characteristic of members of the genus Victorivirus. The amino a

Profiling of RNA Viruses in Biting Midges (Ceratopogonidae) and Related Diptera from Kenya Using Metagenomics and Metabarcoding Analysis.

Vector-borne diseases (VBDs) cause enormous health burden worldwide, as they account for more than 17% of all infectious diseases and over 700,000 deaths each year. A significant number of these VBDs are caused by RNA virus pathogens. Here, we used metagenomics and metabarcoding analysis to characterize RNA viruses and their insect hosts among biting midges from Kenya. We identified a total of 15 phylogenetically distinct insect-specific viruses. These viruses fall into six families, with one virus falling in the recently proposed negevirus taxon. The six virus families include Partitiviridae,

Metagenomic sequencing reveals viral diversity of mosquitoes from Egypt: co-circulation of multiple insect-specific viruses.

UNLABELLED: Mosquito-borne virus surveillance is pivotal for investigating mosquito viromes, facilitating understanding of viral evolutionary histories and genetic diversity. Natural viral communities in mosquitoes include not only insect-specific viruses (ISVs) but also viruses infecting symbiotic microorganisms. In this study, a total of 654 mosquito samples-encompassing species from the Aedes and Culex genera-were collected from Egypt and subjected to metagenomic sequencing analysis. Over 130 virus species were identified, grouped into 35 families or equivalent taxonomic ranks. Detected ISV

Molecular typing of Trichomonas vaginalis isolates by actin gene sequence analysis and carriage of T. vaginalis viruses.

BACKGROUND: The protozoan parasite Trichomonas vaginalis is the most common non-viral, sexually transmitted pathogen. Although T. vaginalis is highly prevalent among women in Kenya, there is lack of data regarding genetic diversity of isolates currently in circulation in Kenya. METHODS: Typing was performed on 22 clinical isolates of T. vaginalis collected from women attending the antenatal care clinic at Kilifi County Hospital, Kenya, in 2015. Genotyping followed a previously proposed restriction fragment length polymorphism (RFLP) scheme, which involved in silico cleavage of the amplified a

Endobiont viruses sensed by the human host - beyond conventional antiparasitic therapy.

Wide-spread protozoan parasites carry endosymbiotic dsRNA viruses with uncharted implications to the human host. Among them, Trichomonas vaginalis, a parasite adapted to the human genitourinary tract, infects globally ∼250 million each year rendering them more susceptible to devastating pregnancy complications (especially preterm birth), HIV infection and HPV-related cancer. While first-line antibiotic treatment (metronidazole) commonly kills the protozoan pathogen, it fails to improve reproductive outcome. We show that endosymbiotic Trichomonasvirus, highly prevalent in T. vaginalis clinical
