Treponema denticola immunoinhibitory protein induces irreversible G1 arrest in activated human lymphocytes.

Oral spirochetes may contribute to the pathogenesis of a number of disorders including periodontal and periradicular diseases; however, the mechanism (s) by which these organisms act to cause disease is unknown. We have previously shown that extracts of the oral spirochete, Treponema denticola, contain an immunosuppressive protein (Sip) which impairs human lymphocyte proliferation. The objective of this study was to determine the mechanism by which Sip alters the proliferative response of lymphocytes. Human T-cells were activated by PHA in the presence or absence of Sip and cell cycle progress

Neutrophil-derived oncostatin m contributes to endothelial cell dysfunction during Treponema denticola interaction.

Periodontitis (PD) is a common chronic inflammatory condition and a risk factor for cardiovascular diseases (CVD), yet underlying linking mechanisms remain unclear. The cytokine Oncostatin M(OSM) is elevated in both PD and CVD and has emerged as a potential mediator linking oral inflammation to vascular dysfunction. Neutrophils represent a prominent source of OSM during PD and OSM production is elevated by the periodontal pathobiont Treponema denticola (Td). Expanded Functional Characterization and Optimization of Protein Expression in Treponema denticola Shuttle Plasmids.

Oral spirochetes are among the small group of keystone pathogens contributing to dysregulation of periodontal tissue homeostasis, leading to breakdown of the tissue and bone supporting the teeth in periodontal disease. Of the more than 60 oral Treponema species and phylotypes, Treponema denticola is one of the few that can be grown in culture and the only one in which genetic manipulation is practicable. T. denticola is thus a model organism for studying spirochete behavior, metabolism, and interactions with other microbes and host tissues that are relevant to oral diseases. We recently demons

Stimulation of Human Periodontal Ligament Fibroblasts Using Purified Dentilisin Extracted from Treponema denticola.

Periodontal disease is a chronic multifactorial disease triggered by a complex of bacterial species. These interact with host tissues to cause the release of a broad array of pro-inflammatory cytokines, chemokines, and tissue remodelers, such as matrix metalloproteinases (MMPs), which lead to the destruction of periodontal tissues. Patients with severe forms of periodontitis are left with a persistent pro-inflammatory transcriptional profile throughout the periodontium, even after clinical intervention, leading to the destruction of teeth-supporting tissues. The oral spirochete, Treponema dent

Treponema denticola biofilm-induced expression of a bacteriophage, toxin-antitoxin systems and transposases.

Treponema denticola is an oral spirochaete that has been strongly associated with chronic periodontitis. The bacterium exists as part of a dense biofilm (subgingival dental plaque) accreted to the tooth. To determine T. denticola gene products important for persistence as a biofilm we developed a continuous-culture biofilm model and conducted a genome-wide transcriptomic analysis of biofilm and planktonic cells. A total of 126 genes were differentially expressed with a fold change of 1.5 or greater. This analysis identified the upregulation of putative prophage genes in the T. denticola 35405

Treponema denticola Induces Alzheimer-Like Tau Hyperphosphorylation by Activating Hippocampal Neuroinflammation in Mice.

Alzheimer's disease (AD) is the most common type of dementia. Tau hyperphosphorylation and amyloid β (Aβ) deposition are the key pathological hallmarks of AD. Recent studies have shown that periodontitis is a significant risk factor for AD. The periodontal pathogen Porphyromonas gingivalis and its virulence factors have been shown to initiate and promote the hallmark pathologies and behavioral symptoms of AD. A possible link between Treponema denticola, another main periodontal pathogen, and AD has been reported. However, the role of T. denticola in AD pathogenesis is still unclear, and whethe

Detection of Treponema denticola in saliva obtained from patients with various periodontal conditions.

The aim of the study was to determine the prevalence of Treponema denticola in saliva of periodontally diseased and healthy patients and its relationship with the periodontal status. A 16S rRNA-based polymerase chain reaction detection method was used to determine the prevalence of T. denticola in whole saliva samples from patients with chronic periodontitis (CP, n = 37), aggressive periodontitis (AgP, n = 24), and healthy subjects (n = 28). The periodontal status of each subject was assessed by criteria based on probing depth, clinical attachment loss, and extent of periodontal breakdown. Ris

Neuroinvasion pathways of Treponema denticola and its role in amyloidogenesis and neuroinflammation: A systematic review.

Alzheimer's disease (AD) is a progressive neurodegenerative condition marked by the accumulation of amyloid-β (Aβ) plaques, tau hyperphosphorylation, and persistent neuroinflammation. Emerging research indicates that Treponema denticola (T. denticola), a prominent periodontal pathogen, plays a role in AD development through various neuroinvasive mechanisms. This systematic review synthesizes current evidence regarding T. denticola ability to penetrate the central nervous system via trigeminal and hematogenous routes, disrupt the blood-brain barrier, and create enduring biofilms within neural t

Fusobacterium nucleatum and Treponema denticola are robust biomarkers for gingivitis and periodontitis in small dogs.

INTRODUCTION: Periodontal disease is one of the most common oral diseases in dogs and humans. It starts with gingivitis, a reversible condition, and progresses to an irreversible condition, periodontitis. Unlike humans, the etiology of periodontal disease in dogs has not been widely studied. Many studies suggest that bacteria strongly implicated in human periodontal disease might also play a role in canine periodontal disease. In contrast to studies examining only the prevalence of bacteria, a recent study analyzed 336 gingival crevicular fluid (GCF) samples in dogs to evaluate the prevalence

Diversity of Treponema denticola and Other Oral Treponeme Lineages in Subjects with Periodontitis and Gingivitis.

More than 75 species/species-level phylotypes belonging to the genus Treponema inhabit the human oral cavity. Treponema denticola is commonly associated with periodontal disease, but the etiological roles and ecological distributions of other oral treponemes remain more obscure. Here, we compared the clinical distributions of phylogroup 1 and 2 oral treponemes in subgingival plaque sampled from Chinese subjects with periodontitis (n = 10) and gingivitis (n = 8) via sequence analysis of the highly conserved pyrH housekeeping gene. Two PCR primer sets that targeted oral phylogroup 1 and 2 trepon

Treponema denticola enolase contributes to the production of antibodies against ENO1 but not to the progression of periodontitis.

Autoantibodies against alpha-enolase (ENO1) are often detected in various infectious and autoimmune diseases. Anti-ENO1 antibody titers were reported to be associated with the severity of periodontitis in patients with rheumatoid arthritis. Because the enolase of the periodontal pathogen Treponema denticola (TdEno) has the highest homology with ENO1 among the enolases of human-associated bacteria, we hypothesized that anti-ENO1 autoantibodies produced during the immune response to TdEno may contribute to the progression of periodontitis and tested it in human and mouse systems. In human subjec

A novel glycan modifies the flagellar filament proteins of the oral bacterium Treponema denticola.

While protein glycosylation has been reported in several spirochetes including the syphilis bacterium Treponema pallidum and Lyme disease pathogen Borrelia burgdorferi, the pertinent glycan structures and their roles remain uncharacterized. Herein, a novel glycan with an unusual chemical composition and structure in the oral spirochete Treponema denticola, a keystone pathogen of periodontitis was reported. The identified glycan of mass 450.2 Da is composed of a monoacetylated nonulosonic acid (Non) with a novel extended N7 acyl modification, a 2-methoxy-4,5,6-trihydroxy-hexanoyl residue in whi

Characterization of Treponema denticola mutants defective in the major antigenic proteins, Msp and TmpC.

Treponema denticola, a gram-negative and anaerobic spirochete, is associated with advancing severity of chronic periodontitis. In this study, we confirmed that two major antigenic proteins were Msp and TmpC, and examined their physiological and pathological roles using gene-deletion mutants. Msp formed a large complex that localized to the outer membrane, while TmpC existed as a monomer and largely localized to the inner membrane. However, TmpC was also detected in the outer membrane fraction, but its cell-surface exposure was not detected. Msp defects increased cell-surface hydrophobicity and

Treponema denticola PrcB is required for expression and activity of the PrcA-PrtP (dentilisin) complex.

The Treponema denticola surface protease complex, consisting of PrtP protease (dentilisin) and two auxiliary polypeptides (PrcA1 and PrcA2), is believed to contribute to periodontal disease by degrading extracellular matrix components and disrupting host intercellular signaling. Previously, we showed that transcription of the protease operon initiates upstream of TDE0760 (herein designated prcB), the open reading frame immediately 5' of prcA-prtP. The prcB gene is conserved in T. denticola strains. PrcB localizes to the detergent phase of Triton X-114 cell surface extracts and migrates as a 22

Detection of Treponema denticola in Chronic Periodontitis by Quantitative Real-Time Polymerase Chain Reaction.

Chronic periodontitis constitutes a significant public health issue, particularly in China. Treponema denticola is one of the bacterial species critically involved in the development of this disease. Therefore, an effort was made in this study to design a technique for isolation of DNA from gingival fluid and detection of T. denticola genes by PCR methodology. For this purpose, samples were collected from 30 patients with severe periodontitis and 20 patients with mild periodontitis. A group of 50 healthy individuals served as a control. Following the isolation of DNA from the gingival fluid by

Treponema denticola as a potential pathogen of pneumonia in individuals with poor oral hygiene: a case report.

Treponema denticola is an oral spirochete closely related to periodontal disease. However, the association between Treponema denticola and pneumonia has seldom been reported. In this report, we present a case wherein pneumonia attributable to Treponema denticola was identified through the application of metagenomic next-generation sequencing analysis. The patient had a history of oral operation and poor oral hygiene, which were likely the risk factors of the infection. The results indicated the association between oral microbes and pneumonia and highlighted the importance of maintaining optima

Dentipain, a Streptococcus pyogenes IdeS protease homolog, is a novel virulence factor of Treponema denticola.

Treponema denticola is a major pathogen of chronic periodontitis. Analysis of the T. denticola genome revealed a gene orthologous with a cysteine protease-encoding gene from Streptococcus pyogenes (IdeS). IdeS interferes with IgG-dependent opsonophagocytosis by specific cleavage of IgG molecules. Analysis of this gene (termed ideT) revealed it to encode a two-domain protein whose N-terminus is composed of tandem immunoglobulin-like domains followed by a C-terminal IdeS-like protease domain. In this study we show that during secretion the IdeT protein is processed into an N-terminal fragment wh

The unique histidine kinase, AtcS, regulates motility and pathogenicity of the periodontal pathobiont, Treponema denticola.

Treponema denticola is an obligate colonizer of the human gingival crevice and, along with other pathobionts, is highly associated with the development of periodontal disease. As periodontal disease develops, significant environmental changes occur in the subgingival crevice and oral microbiome. The ability to sense and respond to changing environmental conditions is essential to the ability of T. denticola to thrive and cause disease. Yet, our understanding of T. denticola sensory transduction and gene regulatory mechanisms is nearly absent. The AtcSR two-component system has been predicted t

Dioxygen and nitric oxide scavenging by Treponema denticola flavodiiron protein: a mechanistic paradigm for catalysis.

Flavodiiron proteins (FDPs) contain a unique active site consisting of a non-heme diiron carboxylate site proximal to a flavin mononucleotide (FMN). FDPs serve as the terminal components for reductive scavenging of dioxygen (to water) or nitric oxide (to nitrous oxide), which combats oxidative or nitrosative stress in many bacteria. Characterizations of FDPs from spirochetes or from any oral microbes have not been previously reported. Here, we report characterization of an FDP from the anaerobic spirochete, Treponema (T.) denticola, which is associated with chronic periodontitis. The isolated
