Xanthine oxidase (XOD) inhibitors are central to hyperuricemia and gout management, but efficient, reusable enzyme-based screening platforms remain limited. In this study, XOD immobilized Fe₃O₄@SiO₂- nanoparticles (Fe₃O₄@SiO₂-XOD NPs) were developed and optimized as a reusable screening platform and applied to identify bioactive XOD inhibitors from Trichosanthes anguina (Snake Gourd). Fe₃O₄@SiO₂-XOD NPs were synthesized via glutaraldehyde-mediated crosslinking and characterized by FTIR, XRD, and SEM. Optimal immobilization was achieved at 15% glutaraldehyde, pH 7.5, 25 °C, and 1 h incubation.

Trichosanthin (TCS) is a toxic protein isolated from a Chinese herbal medicine, the root tuber of Trichosanthes kirilowii Maximowicz of the Curcurbitaceae family. It is now used in China to terminate early and mid-trimester pregnancies. The ribosome inactivating property is thought to be account for its toxicity; it can inactivate the eukaryotic ribosome through its RNA N-glycosidase activity. The interactions of TCS with biological membrane is thought to be essential for its physiological effect, for it must get across the membrane before it can enter the cytoplasm and exert its RIP function.

Trichosanthes kirilowii-Allium macrostemon (Chinese name Gualou and Xiebai, GLXB), a classical herb pair, has significant clinical efficacy in the treatment of myocardial ischemia (MI).
