Trypanosomatid detection in Culicoides biting midges (Diptera: Ceratopogonidae) from leishmaniasis-endemic Songkhla Province, Southern Thailand: Microscopy and nanopore metabarcoding reveal parasite diversity.

Leishmaniasis caused by Leishmania (Mundinia) spp. has recently emerged in Southeast Asia, including Thailand. Although molecular studies have detected Leishmania DNA in Culicoides midges, complete parasite development, which proves vector competence, remains undemonstrated in field specimens. We collected 215 parous/gravid (n = 195) and blood-engorged (n = 20) female Culicoides representing at least 10 species from two leishmaniasis foci in Songkhla Province, Southern Thailand. Specimens underwent microscopic dissection, culturing in Schneider's medium, ITS1/SSU rRNA-PCR for Leishmania/trypan

Molecular target analysis of stearoyl-CoA desaturase genes of protozoan parasites.

Protozoan parasites can synthesize polyunsaturated fatty acids. They possess stearoyl-CoA desaturase to convert stearate into oleate and linoleate. Stearoyl-CoA desaturase are the key enzymes required for the synthesis of unsaturated fatty acids. It seems attractive to evaluate the possibility of using unsaturated fatty acid biosynthesis pathways as drug targets. In this study, the authors investigate codon usage bias, base composition variations and protein sequence in ten available complete stearoyl-CoA desaturase gene sequences from Toxoplasma gondii, Neospora caninum etc. The results show

Structural basis of step II spliced leader RNA trans-splicing in trypanosomatid parasites.

Messenger RNA (mRNA) synthesis is fundamental to eukaryotic gene expression. In kinetoplastid parasites, including the human pathogens Trypanosoma and Leishmania, mRNAs are produced through a mechanism called Spliced Leader (SL) RNA trans-splicing. In this process, a short SL exon from a small noncoding RNA is joined to the 5' end of every mRNA, ensuring transcript stability and translation. Despite decades of study, the structural and mechanistic basis of SL trans-splicing remains elusive. Here, we report cryogenic electron microscopy structures of step II SL trans-splicing machineries from L

Trypanosomatid histones: the building blocks of the epigenetic code of highly divergent eukaryotes.

Histones play a fundamental role in eukaryotic organisms not only as scaffolding proteins in DNA packaging but also in regulating gene expression. They constitute the protein reel around which DNA wraps forming nucleosomes. This initial packing gives rise to the chromatin fiber which is next folded into three-dimensional arrangements. Additionally, histones have expanded their functions through the emergence of histone variants which have specialized purposes and can deeply affect chromatin organization and dynamics. Moreover, both canonical histones and histone variants comprise the building

On-site DNA Detection of Trypanosomatid Parasites and Nosema ceranae Through Alkaline Lysis Coupled to RPA/CRISPR/Cas12a System.

Early on-site detection of microscopic pathogens is essential for the effective management and health of honeybee colonies. Currently, the gold standard for molecular detection of bee pathogens is qPCR or RT-qPCR. Here, we present a rapid, sensitive, and cost-effective alternative for field-deployable DNA pathogen detection. This method combines direct, amplification-ready cell lysis of worker bee abdomens with recombinase polymerase amplification (RPA), CRISPR/Cas12a-mediated trans-cleavage of reporter probes, and detection via lateral flow assays. We successfully validated this protocol in h

Differences in binding of lectin-gold conjugates by Trypanosoma cruzi and Blastocrithidia triatomae (Trypanosomatidae) in the intestine of Triatoma infestans (Reduviidae).

Two species of trypanosomatid flagellates, Trypanosoma cruzi and Blastocrithidia triatomae which develop in the intestine of Triatoma infestans, were incubated with lectin-gold conjugates to demonstrate sugar components of the cell surface with the electron microscope. T. cruzi, strain "Chile 5" (zymodeme 1), bound SBA-gold, revealing N-acetyl-galactosamine and/or galactose residues. WGA-BSA-gold, specific for N-acetylglucosamine, did not bind to the surface of this flagellate. On the other hand, N-acetyl-glucosamine bound WGA-BSA-gold on the cell membrane of epimastigotes and cysts of B. tria

Leptomonas seymouri narna-like virus 1 and not leishmaniaviruses detected in kala-azar samples from India.

The great majority of kala-azar/visceral leishmaniasis (VL) cases, which are caused by Leishmania donovani (LD), are reported in Asia. We investigated whether leishmaniaviruses (LRVs) are present in LD isolates. These dsRNA viruses contribute to hyperpathogenicity, as observed in the case of other members of the genus Leishmania. However, LRVs could not be detected in 22 Indian LD isolates tested in the present study, while 70% of these original LD isolates harboured a virus that was not of LD but instead of Leptomonas seymouri (LS) origin. LS is another protozoon that parasitizes the sandfly

Selective elimination of Leptomonas from the in vitro co-culture with Leishmania.

Leishmania and Leptomonas are protozoan parasites of the family Trypanosomatidae. Leishmania donovani causes the fatal visceral leishmaniasis (VL; kala-azar) in mammals and is transmitted by sand fly vector. Certain VL-cured human populations in India and Sudan develop post kala-azar dermal leishmaniasis (PKDL) due to the same parasite. Although Leptomonas is parasitic mainly in insects, several recent reports on the clinical isolates of L. donovani from VL and PKDL patients in India confirm co-infection of Leptomonas seymouri, probably due to immune suppression in those individuals. Detection

Detection of Leptomonas sp. parasites in clinical isolates of Kala-azar patients from India.

We report here nine unusual cases of Kala-azar, of which parasites were isolated and found by 18S rRNA gene sequencing to be most similar to Leptomonas species. One of these isolates was used to inoculate Balb/c mice; organs were collected and directly submitted to a genus-specific rDNA-ITS1 PCR analysis: this revealed the presence of both Leptomonas sp. and Leishmania donovani. Therefore, we conclude that there was a mixed infection of Leptomonas sp. and L. donovani in this isolate. We consider that mixed infection may be present in the patients themselves, Leptomonas persisting in them becau

Evolutionary history of trypanosomes from South American caiman (Caiman yacare) and African crocodiles inferred by phylogenetic analyses using SSU rDNA and gGAPDH genes.

In this study, using a combined data set of SSU rDNA and gGAPDH gene sequences, we provide phylogenetic evidence that supports clustering of crocodilian trypanosomes from the Brazilian Caiman yacare (Alligatoridae) and Trypanosoma grayi, a species that circulates between African crocodiles (Crocodilydae) and tsetse flies. In a survey of trypanosomes in Caiman yacare from the Brazilian Pantanal, the prevalence of trypanosome infection was 35% as determined by microhaematocrit and haemoculture, and 9 cultures were obtained. The morphology of trypomastigotes from caiman blood and tissue imprints

Axenic cultivation of a pathogenic Phytomonas species isolated from tomato fruit, and from its phytophagic insect vector, Phthia picta (Hemiptera: Coreidae).

Axenic cultures of Phytomonas sp. were obtained from naturally infected tomatoes and from Phthia picta, a predator of tomato plants, by using a biphasic medium with Roitman's complex medium overlaying rabbit blood-agar slants. Light and electron microscopy of both isolates showed a similarity of morphological characteristics among the flagellates in fresh material or after cultivation. Other properties, including their agglutinability with the haemolymph of Phthia picta, suggest that these isolates are virtually identical.

The integrated stress response in apicomplexan and trypanosomatid parasites: balancing survival and pathogenesis.

Parasites rely on their hosts for survival and replication and therefore face major challenges as they transition between hosts and encounter hostile microenvironments. Over evolution, these organisms have developed complex mechanisms to rapidly adapt to environmental fluctuations and to exploit host cellular machinery for persistence and dissemination. Among the pathways parasites deploy to cope with stress is the Integrated Stress Response (ISR), a conserved mechanism present in both parasites and their hosts, yet built on partially distinct molecular components. This shared but divergent pa

The Phlebotomus papatasi systemic transcriptional response to trypanosomatid-contaminated blood does not differ from the non-infected blood meal.

BACKGROUND: Leishmaniasis, caused by parasites of the genus Leishmania, is a disease that affects up to 8 million people worldwide. Parasites are transmitted to human and animal hosts through the bite of an infected sand fly. Novel strategies for disease control require a better understanding of the key step for transmission, namely the establishment of infection inside the fly. METHODS: The aim of this work was to identify sand fly systemic transcriptomic signatures associated with Leishmania infection. We used next generation sequencing to describe the transcriptome of whole Phlebotomus pap

Non-Leishmania Parasite in Fatal Visceral Leishmaniasis-Like Disease, Brazil.

Through whole-genome sequencing analysis, we identified non-Leishmania parasites isolated from a man with a fatal visceral leishmaniasis-like illness in Brazil. The parasites infected mice and reproduced the patient's clinical manifestations. Molecular epidemiologic studies are needed to ascertain whether a new infectious disease is emerging that can be confused with leishmaniasis.

If host is refractory, insistent parasite goes berserk: Trypanosomatid Blastocrithidia raabei in the dock bug Coreus marginatus.

Here we characterized the development of the trypanosomatid Blastocrithidia raabei in the dock bug Coreus marginatus using light and electron microscopy. This parasite has been previously reported to occur in the host hemolymph, which is rather typical for dixenous trypanosomatids transmitted to a plant or vertebrate with insect's saliva. In addition, C. marginatus has an unusual organization of the intestine, which makes it refractory to microbial infections: two impassable segments isolate the anterior midgut portion responsible for digestion and absorption from the posterior one containing

Trypanosomatidae (Kinetoplastea: Trypanosomatida) diversity infecting the South American coati Nasua nasua (Carnivora: Procyonidae) in urban forest fragments in the Brazilian Midwest.

INTRODUCTION: South American coatis are reported as key-hosts of trypanosomatid species in Brazilian Pantanal, presenting high parasitemia and infection rates, long-lasting infections, ability to bioaccumulate Trypanosoma evansi, T. rangeli and different genotypes of T. cruzi, and biological features that favor these parasites' transmission. Moreover, this mammal species was also reported parasitized by the monoxenous trypanosomatid Crithidia mellificae and included in the epidemiological scenario of Visceral Leishmaniasis in Campo Grande (CG), Brazilian Midwest. Within this context, the aim o

Infectious diseases caused by trypanosomatids, including African trypanosomiasis (sleeping sickness), Chagas disease, and different forms of leishmaniasis, are Neglected Tropical Diseases affecting millions of people worldwide, mainly in vulnerable territories of tropical and subtropical areas. In general, current treatments against these diseases are old-fashioned, showing adverse effects and loss of efficacy due to misuse or overuse, thus leading to the emergence of resistance. For these reasons, searching for new antitrypanosomatid drugs has become an urgent necessity, and different metabol
