Application of a monoclonal antibody-based antigen detection enzyme-linked immunosorbent assay (antigen ELISA) for field diagnosis of bovine trypanosomiasis at Nguruman, Kenya.

A monoclonal antibody-based, enzyme immunoassay (antigen ELISA) for the detection of species-specific invariant antigens of Trypanosoma congolense, T. vivax or T. brucei in the serum of infected animals was evaluated as a means of diagnosis using bovine field sera from a trypanosomiasis endemic area, Nguruman, Kenya. Circulating trypanosome antigens were detected in 126 (96.2%) of 131 serum samples from animals with parasitologically confirmed diagnosis: 74.8% were positive for antigens of two or three trypanosome species, while 21.4% tested positive for one trypanosome species. When 70 sera f

Immune depression in bovine trypanosomiasis: effects of acute and chronic Trypanosoma congolense and chronic Trypanosoma vivax infections on antibody response to Brucella abortus vaccine.

Cattle were vaccinated with Brucella abortus (S19) vaccine during acute (25 days) and chronic (25 weeks) Trypanosoma congolense and chronic Trypanosoma vivax (25 weeks) infections in order to determine the effect of such infections on the antibody response to the vaccine. It was found that the specific antibody responses of IgG1 and IgG2 sub-classes were profoundly depressed (80%) in both the acute and chronic infections with T. congolense. Whereas IgM antibody response was also profoundly depressed (90%) in cattle with the acute infection, it was only 50% depressed in those with chronic infec

Epidemiology of bovine trypanosomiasis in the Ghibe valley, southwest Ethiopia. 2. Factors associated with variations in trypanosome prevalence, incidence of new infections and prevalence of recurrent infections.

An average of 840 East African Zebu cattle from nine herds in the Ghibe valley, southwest Ethiopia were monitored from January 1986 to April 1990. Each month blood samples were collected for analysis of packed red cell volume (PCV) and detection of trypanosomes. Animals found to be parasitaemic and with a PCV less than 26% were treated with diminazene aceturate at a dose of 3.5 mg/kg body weight. The majority of infections were associated with Trypanosoma congolense (84% of infections in adult cattle and 71% in cattle less than 24 months of age), and the mean percentage of adult animals detect

The seasonality as a relevant aspect to be considered for differential diagnosis of Trypanosoma vivax infection and co-infections in female cattle.

Bovine Trypanosomiasis and other infectious diseases cause relevant loss for the livestock industry impacting productive/reproductive indices. This study intended to better understand the frequency, seasonality, and profile of infections associated with Bovine Trypanosomiasis. A total of 1443 serum samples were screened for T. vivax infection and other infectious diseases: Neosporosis, Leptospirosis, Bovine Leukosis Virus infection/(BLV), Infectious Bovine Rhinotracheitis/(IBR) or Bovine Viral Diarrhea/(BVD). Distinct methods were used for screening and diagnosis: immunofluorescence assay (Try

Methods Applied to the Diagnosis of Cattle Trypanosoma vivax Infection: An Overview of the Current State of the Art.

Bovine trypanosomiasis caused by Trypanosoma vivax is a relevant disease in domestic ungulates in Latin America, causing different types of livestock losses, particularly in African and South American countries, leading to loss of millions of dollars/year related to dairy and meat production. In addition, T. vivax trypanosomiasis requires intensive veterinary care. While vector control is a feasible measure to manage disease spreading, the search for accurate diagnostic tools still represents a gap in routine veterinary practices and a challenge for the scientific community. The parasite is me

A longitudinal survey of African animal trypanosomiasis in domestic cattle on the Jos Plateau, Nigeria: prevalence, distribution and risk factors.

BACKGROUND: Trypanosomiasis is a widespread disease of livestock in Nigeria and a major constraint to the rural economy. The Jos Plateau, Nigeria was free from tsetse flies and the trypanosomes they transmit due to its high altitude and the absence of animal trypanosomiasis attracted large numbers of cattle-keeping pastoralists to inhabit the plateau. The Jos Plateau now plays a significant role in the national cattle industry, accommodating approximately 7% of the national herd and supporting 300,000 pastoralists and over one million cattle. However, during the past two decades tsetse flies h

Nitric oxide synthesis is depressed in Bos indicus cattle infected with Trypanosoma congolense and Trypanosoma vivax and does not mediate T-cell suppression.

Infection with African trypanosomes causes the diseases sleeping sickness in humans and nagana in cattle in sub-Saharan Africa. Suppression of cellular immune responses is a feature of trypanosomiasis in bovine, human, and murine hosts. Some aspects of immunosuppression in the murine model are mediated by nitric oxide (NO) produced by gamma interferon (IFN-gamma)-activated macrophages. We have investigated whether a similar mechanism is responsible for T-cell unresponsiveness in bovine trypanosomiasis. Bovine monocytes and macrophages from uninfected cattle and activated in vitro with IFN-gamm

Mechanisms of the anemia in trypanosomiasis: studies on the role of the hemolytic fatty acids derived from Trypanosoma congolense.

Trypanosoma congolense releases a number of hemolytic free fatty acids on autolysis of which the most potent is linoleic acid. These fatty acids can lyse washed rat and bovine erythrocytes in vitro. Autolysed T. congoleuse will cause increased fragility of erythrocytes in whole rat blood but not in whole bovine blood. The lack of effect of these fatty acids on bovine erythrocytes tends to be confirmed by the failure of this material to influence the survival of these cells in vivo. Calculations based on the amount of free fatty acids required to cause hemolysis in vivo also confirm that this m

Tenth international meeting on Trypanosoma evansi: report of the working group. Paris, 24 May 1989.

Some epidemiological surveys have provided information on the incidence of Trypanosoma evansi infection among dromedaries in Mali, and among buffaloes in Java and Indonesia. The disease among camels has been reported again from Kazakhstan in the USSR, with the coexistence of T. evansi and Cephalopina titillator in animals which developed acute infection. The disease has been studied among horses in Venezuela and among buffaloes in Vietnam and Indonesia, and suspected among horses in Brazil. Diagnostic kits for rapid and reliable detection of T. evansi are being made available free of charge, u

Infectivity to cattle of metacyclic forms of Trypanosoma (Nannomonas) congolense propagated in vitro. I. Development of localized skin reactions following intradermal inoculation.

Skin reactions similar to those induced by tsetse infected with Trypanosoma congolense were elicited in cattle at sites of intradermal inoculation of in vitro propagated parasites which morphologically resembled metacyclic trypanosomes. The time to detection of the reaction, the time to maximal size and the maximal size attained were dependent on the number of parasites inoculated, although it was possible to induce a skin reaction with as few as 20 trypanosomes. All cattle became infected with the initial detection of the skin reaction preceding parasitaemia by 3 to 7 days.

The impact of nagana.

The disease in cattle, called nagana in Zululand, was linked with trypanosomal parasitaemia and tsetse flies. Nagana occurs in livestock throughout the tsetse belts of Africa. Wild animals are tolerant of trypanosomal infections. Nagana affects individual animals, herds and socio-economic development. In susceptible animals nagana may be acute, but chronic infections are more common. The host-parasite interaction produces extensive pathology and severe anaemia. Clinically affected animals lose condition and become weak and unproductive. Nagana is often fatal and, at herd level, its impact is w

[Experimental infection of the Guyanese zebu with Trypanosoma vivax].

The Guyanese strain of Trypanosoma vivax is pathogenic for the local Brahman zebu; the experimental infection of 19 one-year old cattle was followed by a moderate and transitory fever, a drop in packed cell volume (PCV) and a quick and marked weight loss of 10 to 17 kg one month after as compared to the 16 non-infected animals. Other symptoms were associated with the parasitaemia: diarrhoea, swollen haematic glands on the neck and the flank, lachrymation, weakness. Despite a trypanocidal treatment injected one month post-infection, one animal died and the weight losses of others were not compe
