Activation of TLR4 induces severe acute pancreatitis-associated spleen injury via ROS-disrupted mitophagy pathway.

Severe acute pancreatitis (SAP) is complicated by systemic inflammatory response syndrome and multiple organ dysfunction, the disease will eventually result in death in almost half of the case. The spleen, as the largest immune organ adjacent to the pancreas, is prone to damage in SAP, thereby aggravating the damage of other organs and increasing mortality. However, to date, the research on the mechanism and treatment of spleen injury caused by SAP is still in its infancy. Herein, we investigated the mechanism of spleen injury, and explored the application potential of tuftsin for relieving sp

Phagocytosis and microbicidal capacity of human monocytes in the course of HIV infection.

Phagocytosis and bactericidal activity of human monocytes were studied in asymptomatic HIV carriers and in patients with clinically apparent HIV infection, diagnosed as persistent generalised lymphadenopathy (PGL) or AIDS-related complex (ARC). Monocytes of asymptomatic HIV carriers manifested no significant changes while a decreased phagocytosis was shown by monocytes in the majority of patients with PGL or ARC. The latter patients also exhibited a decreased bactericidal activity of the cells. Tuftsin and even serum from healthy donors were found to normalise the disturbed monocyte function,

Splenic autotransplantation after abdominal trauma in childhood. Clinical and experimental data.

Splenectomy is known to increase the risk of overwhelming bacterial infection. There is a decrease in immunoglobulin IgM and T-lymphocytes, primary antibody response to antigen challenge is impaired, altered opsonic function an Tuftsin deficiency are noted. Splenic autotransplantation has been suggested as a method of preserving function and this concept is supported by experiments in animals. Prior to operation on humans the technique was thoroughly elaborated and practised in animal experiments (dogs). After splenectomy, 6-8 thin segments (Furka's "spleen chip") are placed in between the pla

The LRRK2 P1446L mutation triggers dopaminergic neurodegeneration via DAPK1-mediated microglial neuroinflammation and neuronal apoptosis.

Parkinson's disease (PD) involves dopaminergic neuron loss and neuroinflammation, with leucine-rich repeat kinase 2 (LRRK2) mutations identified as major genetic risk factors. However, the pathogenic mechanism of the novel LRRK2-P1446L mutation remains unknown. Here, we designed LRRK2-P1446L mutant mice and demonstrated that the novel LRRK2-P1446L mutation drives neurodegeneration through death-associated protein kinase 1 (DAPK1) dysregulation. This mutation downregulates LRRK2 while upregulating DAPK1, which concurrently triggers microglial PI3K/Akt-dependent NF-κB activation (inducing IL-1β/

In vitro stimulation of macrophages by quadrol [N,N,N',N'-tetrakis(2-hydroxypropyl)ethylenediamine].

Mouse peritoneal macrophages, when exposed to Quadrol [N,N,N',N'-tetrakis(2-hydroxypropyl)ethylenediamine] in vitro, show a dose dependent enhanced spreading over a four-hour period. In vitro Quadrol induced phagocytosis of polystyrene beads was found to be time and concentration dependent. The rate and extent of the enhancement of phagocytosis was comparable to that observed for lipopolysaccharide and tuftsin.

Tetrahedral DNA Nanostructure-Based Biomimetic Nanovesicles Attenuate Sepsis-Associated ARDS by Suppressing Glycolysis via the BMAL1/PFKFB3 Axis.

Sepsis-associated acute respiratory distress syndrome (SA-ARDS) is a life-threatening complication characterized by excessive pulmonary inflammation and pulmonary edema, lacking effective treatments. This study identifies the transcription factor BMAL1 in alveolar macrophages (AMs) as a key therapeutic target. Mechanistically, BMAL1 represses the expression of the glycolytic enzyme PFKFB3 by binding to the Pfkfb3 promoter, thereby inhibiting glycolysis, M1 polarization of AMs, and the generation of pro-inflammatory cytokines and reactive oxygen species (ROS). Based on this regulatory mechanism

Colonic surgery. The splenic connection.

Colonic resections, particularly those that require mobilization of the splenic flexure, occasionally will lead to injury of the spleen. Under these circumstances, the abdominal surgeon has traditionally considered incidental splenectomy to be the only safe alternative. Currently, a better understanding of splenic physiology and its role in sepsis prevention has reversed this trend. These efforts to preserve splenic function have resulted in various options available to the surgeon, herein reviewed. The results obtained in 36 general surgical patients with splenic injuries suggest that the sal

A highly luminescent ZnS/CdSe/ZnS nanocrystals-tetrapeptide biolabeling agent.

Stable colloidal ZnS/CdSe/ZnS nanocrystals capped with hexadecylamine were transferred into water by encapsulation in a protective shell of an amphiphilic polymer. The properties of the products have been investigated by dynamic lighting scattering, absorption, and photoluminescence spectroscopy. These nanocrystals, when conjugated with tuftsin in aqueous suspension, formed an effective labeling reagent for macrophages or lymphocytes. The labeling of cells was demonstrated by confocal microscopy. Hybrid bio-inorganic nanoconjugates are potentially powerful fluorescent tracking tools in biologi

Studies of the influence of different cross-linking reagents on the immune response against a B-epitope.

We have previously shown that the carrier polytuftsin obtained by polycondensation of tuftsin, a naturally occurring macrophage activator, increases significantly the antibody response against a linked B-epitope. In the present work, we have studied the influence of different cross-linking reagents on the quality of the conjugation and on the immune response, at both B-cell and T-cell levels. We observed that the cross-linking method used for coupling the B-epitope to the carrier influences the immune response. A hypothesis is put forward to explain the differences observed.

Neuropilins in Multiple Sclerosis: Dual Roles of NRP-1 in Neuroinflammation and Neuroprotection.

Neuropilins (NRPs), particularly NRP-1, are multifunctional co-receptors involved in neuroinflammatory and neuroprotective processes. Altered NRP expression has been observed in multiple sclerosis (MS) lesions and peripheral circulation, suggesting early involvement in disease progression. This review addresses the dual role of NRPs in MS and experimental autoimmune encephalomyelitis (EAE), emphasizing expression patterns, signaling pathways, and therapeutic interventions. NRP-1 is expressed by endothelial cells, microglia, and macrophages, while Sema3A, a key ligand, is produced by reactive a

An Engineered Multifunctional Fusion Protein Targeting Aβ Oligomers, Microglia and Autophagy Ameliorates Cognitive Deficits and Amyloid Pathology in Alzheimer's Disease Mice.

In Alzheimer's disease (AD), Amyloid-β (Aβ) oligomers function as key neurotoxic agents that underpin the disease's progression. A diverse array of therapeutic entities, including peptides, single-chain variable fragments (scFvs), and small molecules, have demonstrated the ability to interact with Aβ oligomers, thereby suppressing their aggregation and associated neurotoxicity. Despite these advances, such agents frequently struggle to promote the phagocytosis and subsequent breakdown of aggregated Aβ by microglia. Moreover, the dense accumulation of Aβ oligomers may resist enzymatic hydrolysi

Effects of composite yeast culture on the expression of immune-related genes in the splenic tissue of weaned lambs.

The objective of this study was to evaluate the effects of dietary supplementation with a composite yeast culture (CYC) on the immune status and splenic gene expression in weaned lambs. Eighteen healthy, male weaned lambs of similar age and body weight were randomly assigned to three groups, including the control group (Con, basal TMR diet), the CYC1 group (TMR supplemented with 40 g/day/lamb of CYC), and CYC2 group (TMR supplemented with 50 g/day/lamb of CYC). The pre-feeding period lasted for 7 days, followed by a 40-day formal experimental period. Upon completion of the experiment, spleens

Peptide-targeted diagnostics and radiotherapeutics.

Radiotherapeutic drugs and medical imaging agents, although used for different purposes, both benefit from precise targeting. When systemically administered, either would be most useful if designed to find and bind only a tumor, single type of cell, or unique molecular assembly thereon. In this Account, we examine the use of small peptides, natural and synthetic, to create biochemically specific molecular imaging agents and radiotherapeutic pharmaceuticals, discussing three distinct examples. In one project, a small natural peptide known to target members of the bombesin family of receptors wa

Prophylactic role of immunomodulators in treatment of systemic candidiasis in leukopenic mice.

In the present study, we have evaluated prophylactic role of various immunomodulators viz. lipopolysachharide, protein A and tuftsin to impart protection against experimental candidiasis in leukopenic mice. Both free as well as liposomised form of nystatin was not effective enough in offering complete cure against less susceptible isolate of Candida albicans (JNMCR) infection in immunodebilitant mice. Interestingly, the pretreatment of leukopenic mice with immunomodulators before challenging them with C. albicans increased therapeutic efficacy of the nystatin against systemic candidiasis. Effi

Humoral factors in host defense against microbial invaders.

Normal host defense against microbial invaders depends upon an intact humoral and cellular system of inflammation and specific immunity. The major weapons for killing microbes that invade tissue are phagocytic cells, however, humoral factors enable these phagocytic cells to recognize, migrate toward, efficiently ingest, and kill the invaders. Humoral factors essential for host defense include specific antimicrobial immunoglobulin antibodies and multiple proteins which comprise the complement system. The complement system is the oldest phylogenetically and the most important protein in this sys

Effect of leukocyte hydrolases on bacteria. XV. Inhibition by antibiotics, metabolic inhibitors, and ultraviolet irradiation of the release by leukocyte extracts, trypsin, and lysozyme of lipopolysaccharide from gram-negative bacteria.

Leukocyte extracts, trypsin, and lysozyme are all capable of releasing the bulk of the LPS from S. typhi, S. typhimurium, and E. coli. Bacteria which have been killed by heat, ultraviolet irradiation, or by a variety of metabolic inhibitors and antibiotics which affect protein, DNA, RNA, and cell wall synthesis no longer yield soluble LPS following treatment with the releasing agents. On the other hand, bacteria which are resistant to certain of the antibiotics yield nearly the full amount of soluble LPS following treatment, suggesting that certain heatlabile endogenous metabolic pathways coll

Therapeutic effects of a synthetic peptide of C-reactive protein in pre-clinical tumor models.

Previous studies have shown that multilamellar vesicles (MLV) or other carriers containing purified human C-reactive protein (CRP) have therapeutic activity in preclinical tumor models. Here we evaluated the therapeutic effects of MLV containing novel synthetic peptides, derived from the structure of CRP, on the extent of (a) established lung metastases of fibrosarcoma T241 in C57Bl/6 mice, (b) survival of C57Bl/6 mice bearing established liver metastases of colon carcinoma MCA-38, and (c) primary tumor growth of Renca renal carcinoma in Balb/c mice. [Should splenic tissue be re-implanted after splenectomy due to birth trauma?].

Splenectomy results in loss of about 1/4 of the reticulo-endothelial system and 1/3 of the lymphatic tissue. Reduced phagocytosis and "clearing" capacity are reflected in the appearance of Howell Jolly bodies, thrombocytosis and decreased circulating immune-complexes. Reduction of IgM and compensatory increase of IgG and IgA levels further indicate immunological impairment. Transitory reduction of complement activity and the number of T-lymphocytes in the first weeks post-splenectomy constitute a significant limitation of immunological function and are accompanied by low serum tuftsin levels.
