Characterizing the role of Tupaia DNA damage inducible transcript 3 (DDIT3) gene in viral infections.

DNA damage inducible transcript 3 (DDIT3, also known as CHOP) belongs to the CCAAT/enhancer-binding protein (C/EBP) family and plays an essential role in endoplasmic reticulum stress. Here, we characterized the potential role of the Chinese tree shrew (Tupaia belangeri chinensis) DDIT3 (tDDIT3) in viral infections. The tDDIT3 protein is highly conserved and has a species-specific insertion of the SQSS repeat upstream of the C-terminal basic-leucine zipper (bZIP) domain. Phylogenetic analysis of DDIT3 protein sequences of tree shrew and related mammals indicated a closer genetic affinity betwee

Zika Virus Infection in Tupaia belangeri Causes Dermatological Manifestations and Confers Protection against Secondary Infection.

Animal models of Zika virus (ZIKV) infection have recently been established in mice, guinea pigs, and nonhuman primates. Tree shrews (Tupaia belangeri) are an emerging experimental animal in biomedical applications, but their susceptibility to ZIKV infection has not been explored. In the present study, we show that subcutaneous inoculation of ZIKV led to rapid viremia and viral secretion in saliva, as well as to typical dermatological manifestations characterized by massive diffuse skin rash on the trunk. Global transcriptomic sequencing of peripheral blood mononuclear cells isolated from ZIKV

Cerebral microvascular architecture in the common tree shrew (Tupaia glis) revealed by plastic corrosion casts.

The vascularization of the cerebrum (cerebral cortex and basal ganglia) in the common tree shrew (Tupaia glis) has been studied in detail using vinyl injection and vascular corrosion cast/SEM techniques. It is found that the arterial supply of the cerebral cortex are from cortical branches of the middle cerebral artery (MCA) and of the anterior cerebral artery (ACA). These arteries are in turn branches of the internal carotid artery (ICA). In addition, the cerebral cortex receives the blood from the cortical branches of the posterior cerebral artery (PCA) that originates from the basilar arter

A study of Epstein-Barr virus infection in the Chinese tree shrew(Tupaia belangeri chinensis).

BACKGROUND: Epstein-Barr virus (EBV) is closely associated with many human diseases, including a variety of deadly human malignant tumours. However, due to the lack of ideal animal models, the biological characteristics of EBV, particularly its function in tumourigenesis, have not been determined. Chinese tree shrews (Tupaia belangeri chinensis), which are similar to primates, have been used to establish a variety of animal models and have recently received much attention. Here, we established tree shrews as a model for EBV infection by intravenous injection. METHODS: Ten tree shrews were inoc

Role of thermal physiology and bioenergetics on adaptation in tree shrew (Tupaia belangeri): the experiment test.

Ambient conditions, as temperature and photoperiod, play a key role in animals' physiology and behaviors. To test the hypothesis that the maximum thermal physiological and bioenergetics tolerances are induced by extreme environments in Tupaia belangeri. We integrated the acclimatized and acclimated data in several physiological, hormonal, and biochemical markers of thermogenic capacity and bioenergetics in T. belangeri. Results showed that T. belangeri increased body mass, thermogenesis capacity, protein contents and cytochrome c oxidase (COX) activity of liver and brown adipose tissue in wint

Lectin binding sites in the vomeronasal organ and the olfactory epithelium of the tree shrew tupaia belangeri.

Lectin binding histochemistry was performed on the peripheral parts of the vomeronasal and olfactory system of Tupaia belangeri to investigate the distribution and density of defined carbohydrate terminals on the cell surface glycoproteins of the specific receptors, nerves and associated glands. Dolichus biflorus lectin staining was identifiable in all cells of the sensory epithelia of both systems, with the exception of the olfactory basal cells. The vomeronasal nerve expressed alpha-N-acetylgalactosamine, the olfactory nerve did not. Differences in the Dolichus biflorus lectin staining patte

Tupaias (tree shrews)--a new animal model for gallstone research; first observations of gallstones.

In tupaias, rat-sized mammals with phylogenetic affinities to insectivores and primates, gallstones can be induced by diet. Twenty per cent butter, 20% sucrose, and 1% cholesterol, added to the standard diet, caused massive stone formation in 11 of 16 male tupaias. In the other five animals the bile contained cholesterol crystals or few stones, liquid crystals, and fat droplets. The stones were composed almost totally of cholesterol and closely resembled those described in squirrel monkeys, which had been fed a similar diet. The diet caused neither a change in body weight nor serious diarrhea

Avian H5N1 influenza virus infection causes severe pneumonia in the Northern tree shrew (Tupaia belangeri).

Avian-origin influenza viruses like H5N1 and H7N9 often cause severe symptoms with high mortality in humans. Animal models are useful for clarification of the mechanisms of pathogenicity of these infections. In this study, to expand the potential utility of the Northern tree shrew (Tupaia belangeri) for influenza virus infection, we assessed the pathogenicity of H5N1 and H7N9 avian influenza viruses in tupaia. Infectious virus was detected continuously from nasal, oral, tracheal, and conjunctival swab samples in the animals infected with these viruses. H5N1 influenza virus infection of tupaia

Pattern of calretinin immunoreactivity in the main olfactory system and the vomeronasal system of the tree shrew, Tupaia belangeri.

The distribution of the calcium-binding protein calretinin was studied in peripheral and central parts of the main olfactory system (MOS) and the vomeronasal system (VNS) of adult tree shrew Tupaia belangeri. The calretinin immunoreaction was carried out with a peroxidase-coupled polyclonal antibody. In the VNS, complete labeling of all receptor cells and vomeronasal nerve fibers was observed, whereas only a subset of the somata and dendrites of receptor cells and of the olfactory nerve fibers of the MOS was immunoreactive. From the immunoreactive dendritic clubs of vomeronasal receptor cells,

Tupaia belangeri better reflects the typical symptoms of Chikungunya virus infection than mice: A comparative study.

Chikungunya virus (CHIKV) infection in humans is typically characterized by acute febrile illness, rash, and polyarthralgia, and often progresses to chronic arthralgia. However, existing small-animal models fail to capture both the acute and chronic phases of the disease. In this study, we compared Chinese tree shrews (Tupaia belangeri) and C57BL/6 mice in terms of infection dynamics, viral characteristics, histopathological changes, and immune responses. Following dual-site inoculation (subcutaneous injection into the abdomen and footpad), Tupaia belangeri exhibited typical symptoms that more

Cerebellar cortical efferents of the posterior lobe vermis in a prosimian primate (Galago) and the tree shrew (Tupaia).

The topographical organization of cerebellar cortical efferents of the posterior lobe vermis was studied in a prosimian primate (Galago senegalensis) and the tree shrew (Tupaia glis). Two patterns emerge; one which shows longitudinal zones of the entire vermis and a second which shows that induvidual lobules within the overall longitudinal pattern terminate in specific areas of the ipsilateral medial cerebellar nucleus (NM) and vestibular complix. The posterior lobe vermis consists of a narrow midline portion which projects bilaterally into the NM and a paramidline zone which projects only int

Characterization of prostaglandin E2 binding to uterine membranes from baboon, rabbit and tree shrew (Tupaia belangeri).

Membrane preparations (100,000 g pellet) of rabbit, baboon and tree shrew (Tupaia belangeri) uteri were studied for binding of [3H]prostaglandin E2 ([3H]PGE2). Unbound [3H]PGE2 was separated by filtration through Whatman GF/F filters. Non-specific binding was determined by the amount of radioactivity associated with the filters in the presence of a 100-fold excess of radioinert PGE2. PGE2 bound to membranes could be displaced by some other prostaglandin (PG) molecules: PGE1, 16,16-dimethyl-PGE2, PGA1 and PGF2 alpha, but not by 6-keto-PGF1 alpha, PGD2 or arachidonic acid. No PGE2 binding was de

Testicular microvascularization in the common tree shrew (Tupaia glis) as revealed by vascular corrosion cast/SEM and by TEM.

Testicular angioarchitecture in lower primates has not been established and the route of androgens from Leydig cells entering the systemic circulation is still a matter of controversy. In the present study, the common tree shrew (Tupaia glis) was used as the model for vascular corrosion cast/SEM and conventional TEM studies. With vascular corrosion cast/SEM, it was revealed that while coursing in the spermatic cord, the testicular artery convoluted and gave off branches to supply the epididymis, the coverings of the spermatic cord and the pampiniform plexus. Upon approaching the testis, it enc

Mapping of the hepatitis B virus attachment site by use of infection-inhibiting preS1 lipopeptides and tupaia hepatocytes.

BACKGROUND & AIMS: Studies on the early steps in the life cycle of hepatitis B virus have been hampered by the lack of readily available target cells. In this study, we mapped a defined virus attachment site to primary hepatocytes that is essential for infection. METHODS: We used purified virus particles from human carrier plasma as an inoculum and primary cultures of tupaia hepatocytes as susceptible target cells and studied the inhibitory effect of amino-terminally acylated preS1-derived lipopeptides on infection interference. RESULTS: Infectivity of virus could be blocked efficiently in t

Endocrine correlates of reproductive development in the male tree-shrew (Tupaia belangeri) and the effects of infantile exposure to exogenous androgens.

The developmental life-history of tree-shrews conforms with the general primate pattern. Consequently, elucidation of the tree-shrew's neuroendocrine reproductive axis could shed light on the mechanisms that underlie human pubertal development. In the present study, we examined plasma gonadotropin concentrations in male tree-shrews from birth to sexual maturity, and related them to changes in the androgenic and gametogenic status of the testis. A hypogonadotropic infantile phase, during which a stable population of primordial cells is established, extended from birth to approximately Day 30. F

Intracortical Microstimulation Maps of Motor, Somatosensory, and Posterior Parietal Cortex in Tree Shrews (Tupaia belangeri) Reveal Complex Movement Representations.

Long-train intracortical microstimulation (LT-ICMS) is a popular method for studying the organization of motor and posterior parietal cortex (PPC) in mammals. In primates, LT-ICMS evokes both multijoint and multiple-body-part movements in primary motor, premotor, and PPC. In rodents, LT-ICMS evokes complex movements of a single limb in motor cortex. Unfortunately, very little is known about motor/PPC organization in other mammals. Tree shrews are closely related to both primates and rodents and could provide insights into the evolution of complex movement domains in primates. The present study
