Structure-guided discovery of ubiquitin-specific protease 7 inhibitors through integrative quantitative structure-activity relationship modeling, docking, and molecular dynamics.

Ubiquitin-specific protease 7 (USP7) plays a crucial role in cancer progression by regulating key oncogenic proteins, making it a promising therapeutic target. We developed robust QSAR models using 837 curated USP7 inhibitors, 12 fingerprint descriptors, and a random forest algorithm, achieving high predictive accuracy (R2 = 0.96 ± 0.01, Q2 = 0.92 ± 0.02). Virtual screening identified high-potential compounds from NPASS, TCM, and ZINC databases. Redocking of the co-crystallized ligand (GNE6640, PDB ID: 5UQV) validated the docking protocol (RMSD = 0.330 Å). Top hits NPC472846, TCM11676, TCM3672

USP7, negatively regulated by miR-409-5p, aggravates hypoxia-induced cardiomyocyte injury.

Hypoxia-induced apoptosis is linked to the pathogenesis of myocardial infarction (MI) and heart failure. Ubiquitin-specific peptidase 7 (USP7) is related to catabolic/pro-apoptotic signaling. However, its role in cardiomyocyte injury is unclear. In this study, we aimed to investigate the role and the underlying regulatory mechanism of USP7 in MI. H9c2 cardiomyocytes were cultured in hypoxia to establish an in vitro model of myocardial hypoxic/ischemic injury. Sprague-Dawley (SD) rats were used to establish animal models with MI. Quantitative real-time polymerase chain reaction (qRT-PCR) and We

S100A9 modulates USP7-mediated stabilization of NCOA4 to promote ferroptosis in sepsis-associated acute lung injury.

Sepsis-associated acute lung injury (SALI) is driven by dysregulated macrophage activation; however, mechanisms linking innate immune signaling to cell death remain elusive. Emerging evidence implicates ferroptosis, fueled by NCOA4-mediated ferritinophagy, as a critical executioner of macrophage death. Yet, post-translational mechanisms dictating NCOA4 stability and preventing its premature degradation during sepsis are poorly understood. Specifically, how damage-associated molecular patterns (DAMPs) like S100A9 sustain this pro-ferroptotic flux remains unknown. Here, we identify an S100A9-USP

METTL14 Promotes the Osteogenic Differentiation of Human Bone Marrow Stromal Cells via m6A-Dependent Stabilization of USP7 mRNA.

Osteoporosis (OP) is a common clinical bone disease that can cause a high incidence of non-stress fractures and is one of the main degenerative diseases that endangers the health and life of middle-aged and older women. The mechanism underlying the abnormal differentiation and function of human bone marrow stem cells (hBMSCs) remains to be elucidated. Cell proliferation and differentiation were determined using 3-(4,5)-dimethylthiahiazo (-z-y1)-3,5-di-phenytetrazoliumromide (MTT) assay, alkaline phosphatase (ALP) staining, and Alizarin Red Staining. The interaction between insulin-like growth

MLF2 Negatively Regulates P53 and Promotes Colorectal Carcinogenesis.

Inactivation of the p53 pathway is linked to a variety of human cancers. As a critical component of the p53 pathway, ubiquitin-specific protease 7 (USP7) acts as a deubiquitinase for both p53 and its ubiquitin E3 ligase mouse double minute 2 homolog. Here, myeloid leukemia factor 2 (MLF2) is reported as a new negative regulator of p53. MLF2 interacts with both p53 and USP7. Via these interactions, MLF2 inhibits the binding of USP7 to p53 and antagonizes USP7-mediated deubiquitination of p53, thereby leading to p53 destabilization. Functionally, MLF2 plays an oncogenic role in colorectal cancer

The USP7-STAT3-granzyme-Par-1 axis regulates allergic inflammation by promoting differentiation of IL-5-producing Th2 cells.

Uncontrolled type 2 immunity by type 2 helper T (Th2) cells causes intractable allergic diseases; however, whether the interaction of CD4+ T cells shapes the pathophysiology of allergic diseases remains unclear. We identified a subset of Th2 cells that produced the serine proteases granzyme A and B early in differentiation. Granzymes cleave protease-activated receptor (Par)-1 and induce phosphorylation of p38 mitogen-activated protein kinase (MAPK), resulting in the enhanced production of IL-5 and IL-13 in both mouse and human Th2 cells. Ubiquitin-specific protease 7 (USP7) regulates IL-4-indu

Suppression of USP7 induces BCR-ABL degradation and chronic myelogenous leukemia cell apoptosis.

Chronic myelogenous leukemia (CML) is a clonal malignancy of hematopoietic stem cells featured with the fusion protein kinase BCR-ABL. To elicit the mechanism underlying BCR-ABL stability, we perform a screen against a panel of deubiquitinating enzymes (DUBs) and find that the ubiquitin-specific protease 7 (USP7) drastically stabilizes the BCR-ABL fusion protein. Further studies show that USP7 interacts with BCR-ABL and blocks its polyubiquitination and degradation. Moreover, USP7 knockdown triggers BCR-ABL degradation and suppresses its downstream signaling transduction. In line with this fin

Monocyte USP7-p65 axis mediates immune responses to the immunogenicity of nucleus pulposus.

The nucleus pulposus (NP) of the intervertebral disc is an immune-privileged tissue. During intervertebral disc degeneration (IDD), this immune privilege is compromised, resulting in the exposure of NP components to the peripheral immune system, which in turn activates monocytes and elicits an immune response. In this study, we demonstrate that monocytes respond to NP immunogenicity by activating damage-associated molecular patterns (DAMPs), thereby initiating a sustained NF-κB-mediated inflammatory response in NP tissue and ultimately driving a vicious cycle of inflammation and oxidative stre

USP7 Inhibitors Destabilize EBNA1 and Suppress Epstein-Barr Virus Tumorigenesis.

Epstein-Barr virus (EBV) is a ubiquitous human ɣ-herpesvirus implicated in various malignancies, including Burkitt's lymphoma and gastric carcinomas. In most EBV-associated cancers, the viral genome is maintained as an extrachromosomal episome by the EBV nuclear antigen-1 (EBNA1). EBNA1 is considered to be a highly stable protein that interacts with the ubiquitin-specific protease 7 (USP7). Here, we show that pharmacological inhibitors and small interfering RNA (siRNA) targeting USP7 reduce EBNA1 protein levels in a proteosome-dependent manner. Proteomic analysis revealed that USP7 inhibitor G

TRIP12 promotes HIV-1 replication and latency reactivation by stabilizing Tat via USP7-mediated deubiquitination.

HIV-1 Tat functions as a key transcriptional activator that amplifies viral transcription by recruiting host transcription elongation machinery, thereby forming a self-amplifying positive-feedback loop. Thus, cellular levels and post-translational modifications of Tat directly determine the efficiency of viral replication, as well as the establishment and reactivation of proviral latency. Elucidating the regulatory network that maintains Tat protein levels is therefore important for developing HIV cure strategies. Here, we identify the E3 ubiquitin ligase TRIP12 as a novel positive regulator o

Ubiquitin-specific protease 7 promotes the growth and oncogenic potential of acute myeloid leukemia cells through the deubiquitination and upregulation of LRRK2.

Leucine-rich repeat kinase 2 (LRRK2), a large protein with kinase and GTPase activities, regulates various cellular pathways, including autophagy, endocytosis, and mitochondrial dynamics. LRRK2, extensively studied in the context of Parkinson's disease is functionally impaired in other pathological conditions as well, including inflammatory bowel disease, cancer, and cardiovascular diseases. Despite its critical functions, the mechanisms controlling LRRK2 protein stability are not fully understood. Recent studies suggest that the ubiquitin-proteasome system plays a key role in regulating LRRK2

Cardiac fibroblasts-specific USP7 drives post-infarction cardiac fibrosis by deubiquitinating Krüppel-like factor 7 to promote myofibroblast activation.

Although cardiac fibroblast-to-myofibroblast transition (FMT) can critically exacerbate collagen deposition and adverse remodeling after myocardial infarction (MI), the underlying regulatory mechanisms remains unclear. While ubiquitin-specific protease 7 (USP7), a deubiquitinating enzyme, has been implicated in cardiomyocyte ischemia injury, its role in myofibroblast transition following MI is unknown. Here, we identify cardiac fibroblasts-specific USP7 as a key mediator of FMT and fibrosis. USP7 expression was upregulated in infarcted murine hearts and isolated cardiac fibroblasts, and the up

Identification and optimization of a small molecule inhibitor of the ovarian tumor protease of the Crimean-Congo hemorrhagic fever virus.

Crimean-Congo hemorrhagic fever (CCHF) is a viral tick-borne disease with fatality rates of up to 30 %. Currently, there are no vaccines or specific antivirals available. The genome of the CCHF virus (CCHFV) encodes an ovarian tumor (OTU) protease with a deubiquitinating activity that is responsible for the evasion of the innate immune response. Therefore, the inhibition of the OTU protease could provide a strategy for the treatment of CCHFV infections. In this study, we screened for small-molecule inhibitors of CCHFV OTU using a fluorescent ubiquitin rhodamine 110 assay. We identified and val

Ubiquitin-driven regulation of immune checkpoints in lung cancer: Mechanisms and therapeutic implications (Review).

The ubiquitin-proteasome system is a master regulator of anti-tumor immunity in lung cancer, which primarily functions through controlling the stability of immune checkpoint proteins. The present review offers a synthesis concerning how a dynamic balance between E3 ubiquitin ligases (E3s) and deubiquitinases (DUBs) dictates the fate of key checkpoint proteins, including programmed cell death protein 1/programmed death-ligand 1, lymphocyte-activating gene 3 and B7 homolog 4. Although specific E3s are known to promote checkpoint degradation to enhance T-cell function in certain contexts, and DUB
