Fine structure of the parathyroid and ultimobranchial glands of the snake, Elaphe quadrivirgata.

The ultrastructure of the parathyroid gland and ultimobranchial gland of the snake, Elaphe quadrivirgata, was studied. The parenchyma of the parathyroid gland was consisted of chief cells arranged in cords. Oxyphil cells and water-clear cells were not recognized. The chief cells contained irregular shaped nucleus, moderately-developed mitochondria, granular endoplasmic reticulum and Golgi complexes. A few secretory granules of 100-300 nm in diameter distributed in the cytoplasm. Small aggregations of glycogen particles and lipid droplets were dispersed widely in the cytoplasm. The ultimobranch

The topographical anatomy and blood supply of the carotid body region of the domestic fowl.

The topographical anatomy of the carotid body region was investigated on both sides in 30 adult domestic fowls (Gallus gallus domesticus). On each side of the body the two parathyroid glands always adhered to each other, the caudal gland being the smaller. The right cranial parathyroid usually touched the thyroid gland, but on the left side the two glands were nearly always separate. The ultimobranchial gland was usually attached to the parathyroid glands on the left side, but was always well separated on the right side. The carotid body, the identity of which was checked by light and electron

Influence of calcitonin administration on ultimobranchial and parathyroid glands of pigeon, Columba livia.

Pigeons were divided into two numerically equal groups (A and B) containing 30 specimens each. Birds from group A were intraperitoneally injected daily with 0.1 mL/100 g body wt of vehicle (0.1 M acetate buffer, pH 4.3 containing 0.1% gelatin). Specimens from group B were intraperitoneally injected daily with 1 microg/100 g body wt of salmon calcitonin. Six birds were sacrificed from each group 2 h after the last injection on 1st, 3rd, 5th, 10th, and 15th day of the experiment. After collection of blood samples, ultimobranchial and parathyroid glands were fixed for histological studies. In cal

The crab-eating frog Rana cancrivora: serum mineral concentrations and histology of the ultimobranchial gland and the parathyroid gland.

In the crab-eating frog Rana cancrivora, taken from a full-strength seawater pond, the in situ levels of serum Na, Cl, K, Ca, Mg, Pi, urea, and osmolarity were examined. The levels were higher than those usually reported for freshwater anurans. However, values for the monovalent salts, urea, and osmolarity were lower than those reported by Gordon et al. (1961, J. Exp. Biol. 38, 659-678) following immersion of this species in 80% seawater for 7 days. The histological features of the ultimobranchial gland and parathyroid gland were coincident with those known in usual freshwater frogs, in spite

Cilia-forming potentials of endocrine organs originating from the foregut.

Cilia were found in intracellular localizations or on the surface of certain cells of frog thymus and chicken parathyroid gland and ultimobranchial body. The experimental observations suggest that ciliogenesis is probably a general property of branchiogenic epithelium, which either persists, or can be activated by certain influences after the functional differentiation of endocrine organs arising from the foregut.

Differentiation of human pluripotent stem cells toward pharyngeal endoderm derivatives: Current status and potential.

The pharyngeal apparatus, a transient embryological structure, includes diverse cells from all three germ layers that ultimately contribute to a variety of adult tissues. In particular, pharyngeal endoderm produces cells of the inner ear, palatine tonsils, the thymus, parathyroid and thyroid glands, and ultimobranchial bodies. Each of these structures and organs contribute to vital human physiological processes, including central immune tolerance (thymus) and metabolic homeostasis (parathyroid and thyroid glands, and ultimobranchial bodies). Thus, improper development or damage to pharyngeal e

Uptake of radioiodine in follicles of dog C-cell complexes studied by autoradiograph and immunoperoxidase staining.

C-cell complexes are special cell groups consisting of a mass of C-cells associated with other epithelial elements and cysts. They are remnants of ultimobranchial bodies retaining fetal characteristics. In the C-cell complexes there are follicular cells in various stages of differentiation, i.e., the cell clusters not yet organized into follicles, primordial follicles with small lumens and comparatively enlarged follicles storing plentiful amounts of colloid. They have a morphology similar to follicular cells of fetal thyroid glands and react to antiserum to 19S thyroglobulin. In order to dete

Morphological and functional aspects of reptilian ultimobranchial gland.

The intention of this review is to compare studies on the morphology and histology (light and electron microscopic) of ultimobranchial glands of various groups of reptiles. Moreover, experiments (including our investigations) on suppression or stimulation of the ultimobranchial gland are included. Adult reptiles possess one (on the left side) or two ultimobranchial glands (UBG). The UBG lie just anterior to the heart. Light as well as electron microscopically, the gland has been shown to contain follicles and cell cords (cell aggregates). The follicular epithelium is lined by simple cuboidal o

Ultimobranchial gland of the domestic fowl. Two types of secretory cells involved in calcitonin metabolism.

The ultimobranchial gland (UBG) of birds is particularly rich in calcitonin, the hypocalcaemic hypophosphataemic hormone, that is secreted by the C-cells of the mammalian thyroid. The principal cells of the UBG have a striking resemblance with the mammalian C-cells, i.e., they possess small intracytoplasmic dense-core secretory granules, 150-300 nm in diameter. The gland also contains a second, morphologically distinct, endocrine cell type with larger granules, 500-800 nm in diameter. A sensitive immunocytochemical reaction was developed with the use of antibodies against salmon calcitonin. By

Growth of the parathyroid glands in omeprazole-treated chickens.

BACKGROUND: Omeprazole, a long-acting inhibitor of gastric acid secretion, is able to increase the circulating concentrations of gastrin. Daily treatment with high doses of omeprazole cause sustained hypergastrinemia. Long-standing hypergastrinemia can be expected to exert numerous effects in the body. For instance, gastrin has been proposed to promote growth in the digestive tract and pancreas. The present study is concerned with the effect of omeprazole on parathyroid glands in the chicken. METHODS: Chickens were treated with omeprazole (400 mumol/kg/day) in methylcellulose (2.5 ml/kg) for

Morphological and molecular evolution of the ultimobranchial gland of nonmammalian vertebrates, with special reference to the chicken C cells.

This review summarizes the current understanding of the nonmammalian ultimobranchial gland from morphological and molecular perspectives. Ultimobranchial anlage of all animal species develops from the last pharyngeal pouch. The genes involved in the development of pharyngeal pouches are well conserved across vertebrates. The ultimobranchial anlage of nonmammalian vertebrates and monotremes does not merge with the thyroid, remaining as an independent organ throughout adulthood. Although C cells of all animal species secrete calcitonin, the shape, cellular components and location of the ultimobr

The role of Hoxa-3 in mouse thymus and thyroid development.

Targeted disruption of Hoxa-3 results in a number of regionally restricted defects in tissues and structures derived from or patterned by mesenchymal neural crest. However, analysis of mutant embryos with injections of a carbocyanine dye or with molecular markers that label these cells indicates that neither the amount nor the migration patterns of this neural crest population are grossly affected. Therefore, it appears that the loss of Hoxa-3 affects the intrinsic capacity of this neural crest cell population to differentiate and/or to induce proper differentiation of the surrounding pharynge

Role of EphA4 receptor signaling in thyroid development: regulation of folliculogenesis and propagation of the C-cell lineage.

Transcriptome analysis revealed that the tyrosine kinase receptor EphA4 is enriched in the thyroid bud in mouse embryos. We used heterozygous EphA4-EGFP knock-in mice in which enhanced green fluorescent protein (EGFP) replaced the intracellular receptor domain (EphA4(+/EGFP)) to localize EphA4 protein in thyroid primordial tissues. This showed that thyroid progenitors originating in the pharyngeal floor express EphA4 at all embryonic stages and when follicles are formed in late development. Also, the ultimobranchial bodies developed from the pharyngeal pouch endoderm express EphA4, but the ult

Co-expression of vimentin and 19S-thyroglobulin in follicular cells located in the C-cell complex of dog thyroid gland.

The localization of vimentin, an intermediate filament protein, in the dog thyroid cells was examined by immunohistochemistry with a monoclonal antibody against vimentin and polyclonal anti-19S-thyroglobulin and anti-calcitonin antibodies. In the dog thyroid gland, C-cells are usually distributed as large cell groups in an interfollicular location. In addition, C-cell complexes that are special C-cell clusters associated with other epithelial elements and cysts are frequently located around the parathyroid gland IV. The C-cell complexes are remnants of embryonic ultimobranchial bodies and reta

Revising the embryonic origin of thyroid C cells in mice and humans.

Current understanding infers a neural crest origin of thyroid C cells, the major source of calcitonin in mammals and ancestors to neuroendocrine thyroid tumors. The concept is primarily based on investigations in quail-chick chimeras involving fate mapping of neural crest cells to the ultimobranchial glands that regulate Ca(2+) homeostasis in birds, reptiles, amphibians and fishes, but whether mammalian C cell development involves a homologous ontogenetic trajectory has not been experimentally verified. With lineage tracing, we now provide direct evidence that Sox17+ anterior endoderm is the o

Medullary carcinoma of the thyroid.

Medullary carcinoma is an unusual malignant neoplasm. The cell of origin is considered to be the C-cell which is derived from the ultimobranchial body and neural crest. The tumor is solid and has characteristic amyloid in the stroma. The lesion usually presents as a neck mass, and multicentricity is not unusual. In some patients diarrhea may be an important presenting symptom. Serum calcitonin is typically elevated and is important in the diagnostic confirmation. About 10 percent of the cases are part of a familial endocrine syndrome which includes pheochromocytomas, mucosal neuromas, and othe

Eya1 is required for the morphogenesis of mammalian thymus, parathyroid and thyroid.

Eyes absent (Eya) genes regulate organogenesis in both vertebrates and invertebrates. Mutations in human EYA1 cause congenital Branchio-Oto-Renal (BOR) syndrome, while targeted inactivation of murine Eya1 impairs early developmental processes in multiple organs, including ear, kidney and skeletal system. We have now examined the role of Eya1 during the morphogenesis of organs derived from the pharyngeal region, including thymus, parathyroid and thyroid. The thymus and parathyroid are derived from 3rd pharyngeal pouches and their development is initiated via inductive interactions between neura

Immunoreactive calcitonin in the central nervous system of the pigeon.

To determine whether calcitonin (CT) is present in avian central nervous system (CNS), as it has been recently found in the CNS of other species, we studied extracts of pigeon (Columbia livia) brain by immunochemical methods. A sensitive and specific radioimmunoassay for salmon CT was used together with gel filtration and high performance liquid chromatography (HPLC). Extracts of hypothalamus contained 31.5 +/- 9.65 ng/g wet weight as salmon CT-like immunoreactivity while extracts of midbrain and brain stem contained 17.0 +/- 5.00 and 2.85 +/- 2.1 ng/g wet weight respectively. [Calcitonin and stanniocalcin. Particular aspects of the endocrine regulation of phospho-calcium metabolism in mammals and fish].

The hypocalcemic and hypophosphatemic peptide calcitonin (CT) is secreted by mammalain thyroid parafollicular cells and fish ultimobranchial body. Over a dozen species of CTs have been cloned and/or sequenced. They can be separated into three classes based on structural and biological similarities: teleost/avian, artiodactyl, and human/rat. In mammals, CT exerts its anti-hypercalcemic and hypophosphatemic effects by inhibiting osteoclastic bone resorption and renal tubular phosphate reabsorption, respectively. CT receptors (CTRs) are members of a subfamily of seven-transmembrane domain, G prot
