Dosimetric calculations for uranium miners for epidemiological studies.

Epidemiological studies on uranium miners are being carried out to quantify the risk of cancer based on organ dose calculations. Mathematical models have been applied to calculate the annual absorbed doses to regions of the lung, red bone marrow, liver, kidney and stomach for each individual miner arising from exposure to radon gas, radon progeny and long-lived radionuclides (LLR) present in the uranium ore dust and to external gamma radiation. The methodology and dosimetric models used to calculate these organ doses are described and the resulting doses for unit exposure to each source (radon

Nephrotoxicity of uranium: pathophysiological, diagnostic and therapeutic perspectives.

As in the case of other heavy metals, a considerable body of evidence suggests that overexposure to uranium may cause pathological alterations to the kidneys in both humans and animals. In the present work, our aim was to analyze the available data from a critical perspective that should provide a view of the real danger of the nephrotoxicity of this metal for human beings. A further aim was to elaborate a comparative compilation of the renal pathophysiological data obtained in humans and experimental animals with a view to gaining more insight into our knowledge of the mechanisms of action an

Characteristics of the German uranium miners cohort study.

To evaluate the risk of cancer associated with low and high levels of radon exposure one of the largest single cohort studies on uranium miners is being conducted in Germany including 58,721 men who were employed for at least 6 mo between 1946 and 1989 at the former Wismut uranium company in Eastern Germany. Information on job history, smoking, dust, and arsenic was collected from the original payrolls and the medical records. Exposure to radon and its progeny was estimated by using a detailed job-exposure matrix. The first mortality follow-up determining the vital status as of 31 December 199

On depleted uranium: gulf war and Balkan syndrome.

The complex clinical symptomatology of chronic illnesses, commonly described as Gulf War Syndrome, remains a poorly understood disease entity with diversified theories of its etiology and pathogenesis. Several causative factors have been postulated, with a particular emphasis on low level chemical warfare agents, oil fires, multiple vaccines, desert sand (Al-Eskan disease), botulism, Aspergillus flavus, Mycoplasma, aflatoxins, and others, contributing to the broad scope of clinical manifestations. Among several hundred thousand veterans deployed in the Operation Desert Storm, 15-20% have repor

Uranium-series dating and the origin of modern man.

Uranium-series dating is based on measurement of the radioactivity of short-lived daughter isotopes of uranium formed in samples which initially contained only the parent uranium. Materials suitable for U-series dating are found in many prehistoric archaeological sites, and include stalagmitic layers (flowstones), and spring-deposited travertines. Some marls and calcretes are also datable using isochron methods, whereas dates on molluscan shells, bones and teeth are less reliable. Ages obtained using alpha counting to determine isotope ratios have errors greater than 5%, and can range from 1 t

Uranium in drinking water: a public health threat.

Uranium (U) has no known essential biological functions. Furthermore, it is well known for its toxicity, radioactivity, and carcinogenic potency. Impacts on human health due to U exposure have been studied extensively by many researchers. Chronic exposure to low-level U isotopes (radionuclides) may be interlinked with cancer etiology and at high exposure levels, also kidney disease. Other important issues covered U and fertilizers, and also U in soils or human tissues as an easily measurable indicator element in a pathophysiological examination. Furthermore, phosphate fertilization is known as

Uranium toxicity to aquatic invertebrates: A laboratory assay.

Uranium mining is an environmental concern because of runoff and the potential for toxic effects on the biota. To investigate uranium toxicity to freshwater invertebrates, we conducted a 96-h acute toxicity test to determine lethal concentrations (testing concentrations up to 262 mg L-1) for three stream invertebrates: a shredder caddisfly, Schizopelex festiva Rambur (Trichoptera, Sericostomatidae); a detritivorous isopod, Proasellus sp. (Isopoda, Asellidae); and a scraper gastropod, Theodoxus fluviatilis (Gastropoda, Neritidae). Next, we ran a chronic-toxicity test with the most tolerant spec

Effect of DTPA on the nephrotoxicity induced by uranium in the rat.

The only treatment proposed after human contamination with MOX (mixed oxide of uranium and plutonium) is diethylenetriaminepentaacetic acid (DTPA), because plutonium is considered to be the major risk. However, both DTPA and uranium are nephrotoxic at high dosages and DTPA has been shown to increase in vitro the cytotoxicity induced by uranium on cultured epithelial tubular cells. This work aimed to test this effect in vivo. Rats were injected with subtoxic (57 microg kg(-1)) to toxic (639 microg kg(-1)) amounts of uranium as nitrate at 0 h, they received two DTPA injections (30 micromol kg(-1

A cytogenetic study of men occupationally exposed to uranium.

Blood lymphocyte cultures from two groups of workers occupationally exposed to uranium were examined for asymmetrical chromosome aberrations and sister chromatid exchanges (SCEs). Significant increases in both cytogenetic endpoints were seen. For dicentrics this appeared to be particularly associated with exposure to soluble uranium. The external radiation exposure experienced by these men was insufficient to explain the increase in dicentrics, and irradiation of lymphocytes by internally deposited uranium would have been minimal. As the SCEs were also raised, the genotoxic effect is likely to

Angle-resolved photoemission spectroscopy of uranium-based systems.

Uranium-based materials are used primarily in nuclear technologies and are also of fundamental interest due to the dual itinerant-localized nature of their 5f electrons, which gives rise to correlated electronic states such as heavy-fermion behavior, unconventional superconductivity, and band-inversion-driven electronic states. Angle-resolved photoemission spectroscopy (ARPES) provides a direct, momentum-resolved probe of the electronic structure, which governs the electronic, thermal, and optical properties of these systems. Despite the relatively small number of available studies, there is g

Hedgehog pathway negatively regulated depleted uranium-induced nephrotoxicity.

Depleted uranium (DU) retains the radiological toxicities, which accumulates preferentially in the kidneys. Hedgehog (Hh) pathway plays a critical role in tissue injury. However, the role of Hh in DU-induced nephrotoxicity was still unclear. This study was carried out to investigate the effect of Gli2, which was an important transcription effector of Hh signaling, on DU induced nephrotoxicity. To clarify it, CK19 positive tubular epithelial cells specific Gli2 conditional knockout (KO) mice model was exposed to DU, and then histopathological damage and Hh signaling pathway activation was analy

Uranium transport in acidic brines under reducing conditions.

The behavior of uranium in environments, ranging from those of natural systems responsible for the formation of uranium deposits to those of nuclear reactors providing 11% of the world's electricity, is governed by processes involving high-temperature aqueous solutions. It has been well documented that uranium is mobile in aqueous solutions in its oxidized, U6+ state, whereas in its reduced, U4+ state, uranium has been assumed to be immobile. Here, we present experimental evidence from high temperature (>100 °C) acidic brines that invalidates this assumption. Our experiments have identified a

Natural Uranium Tissue Content of Three Caucasian Males.

Uranium content and concentrations were measured in the tissues of three Caucasian male whole body donors to the U.S. Transuranium and Uranium Registries with no known intake other than from natural environmental sources. Average total body uranium content in the three cases was 81.3 ± 22.3 μg, of which 37.2 ± 2.1 μg (46%) was in the skeleton. The skeleton had a mean concentration of 3.79 ± 0.45 μg U kg(-1) wet weight and 11.72 ± 1.49 μg U kg(-1) ash. Distribution was in bone volume and not predominately on bone surfaces. Soft tissue concentrations ranged over about an order of magnitude, aver

Screen-printed electrografted electrode for trace uranium analysis.

This paper reports the interest of the novel 4-carboxyphenyl-grafted screen-printed electrodes (4-CP-SPEs) for sub-nanomolar analysis of uranium in water samples. Electrodes were easily prepared via electrochemically reduction of the corresponding diazonium salt. The stability of the grafted layer has been clearly demonstrated. Uranium detection was then achieved by immersing the grafted electrode into the sample solution, followed by the electrochemical measurement of adsorbed U(VI) by square wave voltammetry. Adsorption time was investigated so as to find the best compromise between analysis

Kidney toxicity of ingested uranium from drinking water.

BACKGROUND: In experimental settings, uranium is toxic to kidneys, but effects on humans are unclear. Ingestion of water from drilled wells is a source of high uranium exposure in some populations. METHODS: Uranium exposure was measured in 95 men and 98 women aged 18 to 81 years who had used drinking water from drilled wells for an average of 16 years. Urinary N-acetyl-gamma-d-glucosaminidase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyltransferase, and glutathione-S-transferase; serum cystatin C; and urinary and serum calcium, phosphate, glucose, and creatinine were measured t

Land application of mine water causes minimal uranium loss offsite in the wet-dry tropics: Ranger Uranium Mine, Northern Territory, Australia.

Ranger Uranium Mine (RUM) is situated in the wet-dry tropics of Northern Australia. Land application (irrigation) of stockpile (ore and waste) runoff water to natural woodland on the mine lease is a key part of water management at the mine. Consequently, the soil in these Land Application Areas (LAAs) presents a range of uranium (U) and other metals concentrations. Knowledge of seasonal and temporal changes in soil U and physicochemical parameters at RUM LAAs is important to develop suitable management and rehabilitation strategies. Therefore, soil samples were collected from low, medium, high

Association of systemic lupus erythematosus with uranium exposure in a community living near a uranium-processing plant: a nested case-control study.

OBJECTIVE: To explore the hypothesis that cases of systemic lupus erythematosus (SLE) would be found more frequently in community members with high prior uranium exposure in the Fernald Community Cohort (FCC). METHODS: A nested case-control study was performed using data from the FCC, a volunteer population of individuals who had resided near a uranium ore-processing plant in Fernald, Ohio during the years of plant operation; uranium plant workers were excluded. Members of the FCC were monitored for 18 years. SLE cases were identified using the American College of Rheumatology 1997 revised cl

Efficacy of a novel chelator BPCBG for removing uranium and protecting against uranium-induced renal cell damage in rats and HK-2 cells.

Chelation therapy is a known effective method to increase the excretion of U(VI) from the body. Until now, no any uranium chelator has been approved for emergency medical use worldwide. The present study aimed to evaluate the efficacy of new ligand BPCBG containing two catechol groups and two aminocarboxylic acid groups in decorporation of U(VI) and protection against acute U(VI) nephrotoxicity in rats, and further explored the detoxification mechanism of BPCBG for U(VI)-induced nephrotoxicity in HK-2 cells with comparison to DTPA-CaNa₃. Chelating agents were administered at various times befo

Insights into uranium enrichment of indigenous electroactive Shewanella putrefaciens.

Bioremediation of uranium-contaminated environments using native bacteria shows great promise. While Shewanella putrefaciens (S. putrefaciens) is a known uranium reducing bacterium, the mechanisms and adaptability of indigenous strains from uranium mine tailings remain unexplored. This study isolated a dominant indigenous strain of S. putrefaciens and employed a combined genomic and spectroscopic approach to elucidate its unique uranium fixation mechanism. Microbial diversity analysis confirmed the dominance of Shewanella in the oligotrophic and radioactive tailings. Whole-genome sequencing re

Polyphenolic polymers for uranium decorporation with radiation protection effect.

The effective decorporation and elimination of excess reactive oxygen species (ROS) are critical for the treatment of uranium internal contamination. Herein, we developed a natural polyphenol-based self-assembled polymer (EGCG-Arg) by the Mannich reaction, which exhibits effective uranium decorporation and ROS scavenging capability. In vivo studies revealed that EGCG-Arg effectively chelate and remove uranium from the kidneys and femurs by efficiencies of 55.7% and 43.5%, and its uranium removal rate in kidneys was approximately 3.5-fold higher than that of CaNa3-DTPA. Simultaneously, EGCG-Arg
