CD46-mediated transduction of a species D adenovirus vaccine improves mucosal vaccine efficacy.

The high levels of preexisting immunity against Adenovirus type 5 (Ad5) have deemed Ad5 unusable for translation as a human vaccine vector. Low seroprevalent alternative viral vectors may be less impacted by preexisting immunity, but they may also have significantly different phenotypes from that of Ad5. In this study we compare species D Ads (26, 28, and 48) to the species C Ad5. In vitro transduction studies show striking differences between the species C and D viruses. Most notably, Ad26 transduced human dendritic cells much more effectively than Ad5. In vivo imaging studies showed striking

Vaccine Efficacy Against Naturally Asymptomatic Infections: A Novel Estimand for Quantifying Vaccine Effects.

The naive approach to estimating the effects of a vaccine on asymptomatic infections, which compares the risk of asymptomatic infection among vaccinated and unvaccinated individuals, can be misleading because it comprises two effects: the vaccine preventing asymptomatic infections and the vaccine converting symptomatic to asymptomatic infections. When the latter effect is strong, vaccines can appear harmful with respect to asymptomatic infections. Using a causal principal stratification framework, we formalize an estimand, vaccine efficacy against naturally asymptomatic infection (VE NAI ), th…

Host Factors Impact Vaccine Efficacy: Implications for Seasonal and Universal Influenza Vaccine Programs.

Influenza is a global public health problem. Current seasonal influenza vaccines have highly variable efficacy, and thus attempts to develop broadly protective universal influenza vaccines with durable protection are under way. While much attention is given to the virus-related factors contributing to inconsistent vaccine responses, host-associated factors are often neglected. Growing evidences suggest that host factors including age, biological sex, pregnancy, and immune history play important roles as modifiers of influenza virus vaccine efficacy. We hypothesize that host genetics, the hormo…

Immunomodulatory vaccine demonstrates therapeutic efficacy across cancer types.

Immunosuppression within the tumor microenvironment (TME) profoundly inhibits anti-tumor immunity, presenting a formidable challenge in cancer therapeutics. Despite this recognized obstacle, multi-targeted immunomodulatory strategies remain elusive. Here we developed a novel mRNA-lipid nanoparticle (LNP) vaccine designed to reprogram key cellular mediators of immune suppression within the TME, including C-C motif chemokine ligand 22 (CCL22), transforming growth factor-β (TGF-β), cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), Galectin-3, programmed cell death ligand 1 (PD-L1), indoleamin

RH5: rationally-designed malaria vaccine antigen improving efficacy.

Plasmodium falciparum reticulocyte-binding protein homolog 5 (RH5) is a unique asexual blood-stage malaria vaccine candidate because of its high conservation and essential biological function of binding to basigin on the erythrocyte surface. Recent studies by Barrett et al., Wang et al., and King et al., have brought RH5-based vaccine development a step forward based on a rational antigen design strategy.

Vaccine efficacy and iron deficiency: an intertwined pair?

Vaccines are the most effective measure to prevent deaths and illness from infectious diseases. Nevertheless, the efficacy of several paediatric vaccines is lower in low-income and middle-income countries (LMICs), where mortality from vaccine-preventable infections remains high. Vaccine efficacy can also be decreased in adults in the context of some common comorbidities. Identifying and correcting the specific causes of impaired vaccine efficacy is of substantial value to global health. Iron deficiency is the most common micronutrient deficiency worldwide, affecting more than 2 billion people,…

Efficacy of intracellular immune checkpoint-silenced DC vaccine.

BACKGROUND: DC-based tumor vaccines have had limited clinical success thus far. SOCS1, a key inhibitor of inflammatory cytokine signaling, is an immune checkpoint regulator that limits DC immunopotency. METHODS: We generated a genetically modified DC (gmDC) vaccine to perform immunotherapy. The adenovirus (Ad-siSSF) delivers two tumor-associated antigens (TAAs), survivin and MUC1; secretory bacterial flagellin for DC maturation; and an RNA interference moiety to suppress SOCS1. A 2-stage phase I trial was performed for patients with relapsed acute leukemia after allogenic hematopoietic stem c

[SAFETY AND EFFICACY OF INFLUENZA VACCINE IN PREGNANCY].

UNLABELLED: Only 22.7% of pregnant women are immunized against the flu virus, by the popular belief that the vaccine can cause the onset of the disease or other complications. OBJECTIVE: To review the scientific evidence regarding the safety and efficacy of influenza vaccines in pregnant women. METHODOLOGY: Literature review of the scientific literature. RESULTS: Pregnant women and the fetus can have complications if they contract the flu. The influenza vaccine in pregnant women is effective in reducing flu boxes during pregnancy, and it is not observed any increase in adverse effects of th

Applying biodegradable particles to enhance cancer vaccine efficacy.

One of the primary goals of our group and our collaborators here at the University of Iowa is to develop therapeutic cancer vaccines using biodegradable and biocompatible polymer-based vectors. A major advantage of using discretely packaged immunogenic cargo over non-encapsulated vaccines is that they promote enhanced cellular immunity, a key requirement in achieving antitumor activity. We discuss the importance of co-encapsulation of tumor antigen and adjuvant, with specific focus on the synthetic oligonucleotide adjuvant, cytosine-phosphate-guanine oligodeoxynucleotides. We also discuss our

Virus-encoded ectopic CD74 enhances poxvirus vaccine efficacy.

Vaccinia virus (VV) has been used globally as a vaccine to eradicate smallpox. Widespread use of this viral vaccine has been tempered in recent years because of its immuno-evasive properties, with restrictions prohibiting VV inoculation of individuals with immune deficiencies or atopic skin diseases. VV infection is known to perturb several pathways for immune recognition including MHC class II (MHCII) and CD1d-restricted antigen presentation. MHCII and CD1d molecules associate with a conserved intracellular chaperone, CD74, also known as invariant chain. Upon VV infection, cellular CD74 level

Rapamycin-induced enhancement of vaccine efficacy in mice.

Th1 immunity protects against tuberculosis infection in mice and humans. The widely used BCG vaccine primes CD4 and CD8 T cells through signaling mechanisms from dendritic cells and macrophages. The latter express MHC-II and MHC-I molecules through which peptides from BCG vaccine are presented to CD4 and CD8 T cells, respectively. Since BCG sequesters within a phagosome that does not fuse with lysosomes, generation of peptides within antigen-presenting cells infected with BCG occurs with reduced efficiency. We demonstrate that activation of DCs containing BCG vaccine with rapamycin leads to an

[Experimental evaluation of the brucellosis therapeutic vaccine efficacy].

Use of antibiotics can't completely solve the problem of brucellosis treatment, especially its chronic forms, because antibacterial preparations do not eliminate main pathogenetic factor of the disease--sensibilization of the macroorganism. It makes actual the question about complex immuno- and antibacterial therapy. Long-term clinical experience proved high effectiveness of a therapeutic brucellosis vaccine. Earlier this preparation was manufactured in Research Institute of Vaccines and Sera in Tbilisi (Georgia). To date new composition of components of the vaccine has been developed, and man

Avidity as a parameter for diphtheria vaccine efficacy.

The avidity of antibodies (Abs) to diphtheria Toxin (DT), toxoids (DTo) and the DT binding site (DTBS) was investigated in sera from guinea … nt (K). The avidity of Abs to either the corresponding DTo or the DT could not be correlated to the vaccine efficacy, whereas the vaccine efficacy could be ranked according to its affinity to the DTBS. This may be explained in terms of the specificity of measurement in each procedure. The thiocyanate procedure measures the average avidity to complex antigens with …
