Honey bee (Apis mellifera L.) breeders seek to enhance disease resistance by selecting for behavioral resistance traits such as hygienic behavior and Varroa-sensitive hygiene. However, traditional phenotypic assays for these traits are labor-intensive, limiting scalability. Antennal protein biomarkers correlated with these behaviors offer a promising tool for marker-assisted selection (MAS), potentially increasing phenotyping efficiency and accelerating genetic gains. This study evaluated the performance of honey bee colonies from 4 stocks: those selected using field-based phenotypic assays (F

Parasitic mites Varroa destructor and Tropilaelaps mercedesae represent major threats to global honey bee (Apis mellifera) health and productivity, yet comparative molecular insights into host responses remain limited. To address this, we systematically compiled published studies (2015-2025) reporting genes associated with honey bee interactions with V. destructor (11 studies, 87 genes), T. mercedesae (4 studies, 35 genes), and hygienic behavior (6 studies, 44 genes). Gene identifiers were harmonized to the Amel_HAv3.1 genome assembly, yielding three non-redundant sets: 64 Varroa-associated, 3

The parasitic mite Varroa destructor is the primary factor contributing to global honeybee (Apis mellifera) colony losses, posing a sustainable challenge to apiculture and pollination services. Its intricate life cycle adaptive reproductive strategies, and advanced sensory mechanisms have facilitated its emergence as the most destructive honeybee parasite. V. destructor uses highly specialized feeding strategies that extract essential nutrients from their hosts and introduce various pathogens, causing honeybee health problems. The mite functions as a viral vector, particularly in the case of t

The Varroa destructor represents a significant threat to honey bees, leading to substantial yield losses and declines in colony health. Defense behaviors, including grooming (auto and allogrooming), serve as crucial mechanisms against Varroa infestations, yet the genetic basis of these behaviors remains elusive. This study examines the expression levels of hymenoptaecin (hym), neurexin-1 (AmNrx1), and CYP9Q3, potentially associated with defense behavior, in colonies of the Muğla honey bee ecotype (Apis mellifera anatoliaca) subjected to a Varroa selection program. Worker bees from 23 control g

A 2-year field experiment was performed to test lithium chloride, LiCl, application in a normal beekeeping management system. The effect of LiCl on bee larval mortality, beehive weight (honey production) and Varroa mite mortality were tested. Spectrometric quantification of Li on honey and the larval body were made to test the effectiveness of the presence of LiCl. Li was detected in bee larval bodies and in honey over 2 years, from 2018 to 2019. According to the results, no effect of LiCl on mite mortality or bee larval mortality was detected in the first year of application. By assessing the

Understanding the diversity and consequences of viruses present in honey bees is critical for maintaining pollinator health and managing the spread of disease. The viral landscape of honey bees (Apis mellifera) has changed dramatically since the emergence of the parasitic mite Varroa destructor, which increased the spread of virulent variants of viruses such as deformed wing virus. Previous genomic studies have focused on colonies suffering from infections by Varroa and virulent viruses, which could mask other viral species present in honey bees, resulting in a distorted view of viral diversit

Varroa destructor mites (Acari: Varroidae) are harmful ectoparasites of Apis mellifera honey bees. Female foundresses of wax-capped pupal host cells and their daughters feed on host fluids from open wounds on the host's integument. Details of V. destructor mite nutrition are forthcoming, and little is known about the potential physical effects on hosts from mite feeding. Chemical analysis of waste excretions can infer details of animals' nutrition. Here, chemical analysis by high-performance liquid chromatography/mass spectrometry (HPLC-MS/MS) of mite excretions showed that the purine content

Varroa destructor mites are a leading cause of honey bee (Apis mellifera) colony losses worldwide. The acaracide amitraz has been among the most preferred Varroa treatments for more than a decade because of its high effectiveness and convenience of application. As a result, over reliance on amitraz has led to Varroa resistance to amitraz. Here, we evaluated U.S. registered Varroa treatments with differing active ingredients on Varroa infestation rates and their impact on amitraz resistant Varroa to identify alternative treatment options for commercial beekeepers just prior to winter, a critica

Honey bee (Apis mellifera) colonies are essential for agricultural production and ecosystem services but are increasingly threatened by multiple stressors, particularly the parasitic mite Varroa destructor, one of the major causes of global colony losses. Early and reliable detection of infestations is therefore critical for effective colony management. In this study, we investigated continuous hive acoustics as a non-invasive approach for the early detection of Varroa infestations. Approximately 960,000 timestamped acoustic recordings were collected from 53 colonies, of which only recordings

USDA-ARS Bee Research Laboratory received symptomatic honey bee (Apis mellifera L.) samples across the United States for disease diagnosis. Here, we present a retrospective study and cartography of ectoparasite Varroa destructor and intracellular microsporidia parasite Nosema spp. These two major parasites were identified in the diseased honey bee samples between 2015 and 2022. Varroa infestation level (VIL) was examined by a wash technique (Mites/100 bees) and calculated as a percentage, while Nosema infection was quantified by microscopical spore count (Million Spores/Bee). Data were analyze

Humanitarian demining is a worldwide effort and the range of climates and environments prevent any one detection method being suitable for all sites, so more tools are required for safe and efficient explosives sensing. Landmines emit a chemical flux over time, and honeybees can collect the trace residues of explosives (as particles or as vapour) on their body hairs. This capability was exploited using a passive method allowing the honeybees to freely forage in a mined area, where trace explosives present in the environment stuck to the honeybee body, which were subsequently transferred onto a

We developed a rapid method for extraction of DNA from honey bees, Apis mellifera, and from the parasitic bee mite, Varroa destructor. The advantages include fast processing and low toxicity of the substances that are utilized. We used lysis buffer with nonionic detergents to lyse cell walls and proteinase K to digest proteins. We tested whole thorax, thoracic muscle mass, legs, and antennae from individual bees; the mites were processed whole (1 mite/sample). Each thorax was incubated whole, without cutting, because exocuticle color pigment darkened the extraction solution, interfering with P

The frequency of damaged Varroa destructor Anderson and Trueman (Mesostigmata: Varroidae) found on the bottom board of hives of the honey bee, Apis mellifera L. (Hymenoptera: Apidae) has been used as an indicator of the degree of tolerance or resistance of honey bee colonies against mites. However, it is not clear that this measure is adequate. These injuries should be separated from regular dorsal dimples that have a developmental origin. To investigate damage to Varroa mites and regular dorsal dimples, 32 honey bee (A. mellifera) colonies were selected from four Iranian provinces: Isfahan, M

Tropilaelaps mercedesae is a major ectoparasitic mite threatening the western honey bee (Apis mellifera L.), yet the lack of standardized in vitro rearing methods limits mechanistic studies of its biology and control. Here, we extended a gelatin capsule-based rearing method for Varroa destructor to establish an verage, each reproducingin vitro platform for T. mercedesae. Reproductively primed females were co-cultured with freshly capped worker bee larvae (≤ 24 h post-capping) in gelatin capsules until honey bee emergence. To evaluate the efficacy of this method, the fitness of the original fou

Given recent pathogen-mediated pollinator declines, there is concern regarding virus spillover from managed honeybees to wild pollinators. While the parasitic mite Varroa destructor specializes on honeybees, the viruses it transmits are widespread throughout pollinator communities. Here, we investigated whether beekeeper control of varroa mites influences the magnitude of deformed wing virus (DWV) spillover into wild pollinators. We sampled 27 apiaries along a gradient of mite infestation, screening nearly 1000 pollinators and 350 flowers for DWV via quantitative PCR. We found that controlling

Varroa destructor is one of the main causes of colony losses of the western honey bee (Apis mellifera). Many efforts exist to breed honey bees resistant to V. destructor. Varroa sensitive hygiene (VSH) is a commonly selected behavioural trait; VSH workers remove the pupae of mite infested brood cells with high efficiency, interrupting the reproduction of the mite. The cues and triggers for this behaviour are not yet fully understood. To determine what elicits this removal behaviour, we examined preselected VSH workers´ responses to four different groups of objects inserted into freshly capped

BACKGROUND: Over the last two decades, honey bees (Apis mellifera) have suffered high rates of colony losses that have been attributed to a variety of factors, chief among which are viral pathogens, such as deformed wing virus (DWV), whose virulence has increased because of vector-based transmission by the invasive, ectoparasitic varroa mite (Varroa destructor). A shift in the experimental mode of transmission of the black queen cell virus (BQCV) and sacbrood virus (SBV) from fecal/food-oral (direct horizontal) to vector-mediated (indirect horizontal) transmission also results in high virulenc

Managed honey bee (Apis mellifera) populations play a crucial role in supporting pollination of food crops but are facing unsustainable colony losses, largely due to rampant disease spread within agricultural environments. While mounting evidence suggests that select lactobacilli strains (some being natural symbionts of honey bees) can protect against multiple infections, there has been limited validation at the field-level and few methods exist for applying viable microorganisms to the hive. Here, we compare how two different delivery systems-standard pollen patty infusion and a novel spray-b

BACKGROUND: Varroa destructor mites, and the numerous viruses they vector to their honey bee hosts, are among the most serious threats to honey bee populations, causing mortality and morbidity to both the individual honey bee and colony, the negative effects of which convey to the pollination services provided by honey bees worldwide. Here we use a combination of targeted assays and deep RNA sequencing to determine host and microbial changes in resistant and susceptible honey bee lineages. We focus on three study sets. The first involves field sampling of sympatric western bees, some derived f

The ectoparasitic mite Varroa destructor is responsible for the death of millions of honey bee (Apis mellifera) colonies worldwide. Testing potential miticide compounds with different delivery methods that effectively control V. destructor and have low toxicity for honey bees is crucial to manage this parasite in hives. We determined the varroacide efficacy of three natural compounds delivered to hives with three application methods over a 4-week period. Oxalic acid in a sucrose solution was applied impregnated in cardboard (T1). A mixture of oregano and clove oils in an ethanol-gelatin soluti
