Ciliary vasoconstriction after topical adrenergic drugs.

Rabbits underwent the single-dose or long-term therapeutic administration of the adrenergic drugs phenylephrine hydrochloride, timolol maleate, and betaxolol hydrochloride. After a single dose, all three drugs caused substantial, localized constriction in the arterioles that supply the ciliary processes but did not affect the downstream bore of the same vessels. After seven weeks of a daily dose, tolerance reduced the response to betaxolol to insignificant levels and that to phenylephrine substantially, whereas timolol maleate continued to produce identical levels of vasoconstriction to those

Enhanced hypoxic pulmonary vasoconstriction in hypertension.

In this study, we tested the hypothesis that hypoxic pulmonary vasoconstriction may be enhanced in systemic hypertension. The hypothesis took origin from the following two considerations: alveolar hypoxia constricts the pulmonary vessels by enhancing the Ca2+ penetration across sarcolemma of the smooth muscle cells and systemic high blood pressure is associated with an elevation of tone and reactivity of the lung vessels, which seems to depend on an excessive cytosol free Ca2+ concentration due to alterations in sodium handling and in the Na+-Ca2+ exchange system. These considerations suggest

Hypothesis: vasoconstriction contributes to amaurosis fugax.

Platelets play a critical role in the pathophysiology of amaurosis fugax. Emboli to retinal vessels apparently produce amaurosis but, in addition, we propose that augmented vasoconstrictor responses and vasospasm may contribute to amaurosis. In this study we tested the hypothesis that constrictor responses of retinal vessels to serotonin, which is released when platelets aggregate, are potentiated in experimental atherosclerosis. Blood flow to the retina was measured in normal and atherosclerotic cynomolgus monkeys. In normal monkeys, infusion of serotonin did not alter flow to the retina. In

Naloxone: effects on hypoxic pulmonary vasoconstriction.

To determine the effect of naloxone on the hypoxic pulmonary vasoconstrictive response, six mongrel dogs were rendered hypoxic with 10% oxygen and were given either saline or naloxone. Following hypoxia all dogs had significant increases in mean pulmonary artery pressure (PAP) and pulmonary arterial resistance index (PARI) without changes in cardiac output or systemic blood pressure. Beta endorphins did not change at any time following hypoxia. Dogs receiving naloxone had significant lowering of PAP and PARI without changes in plasma beta endorphin levels. We conclude that naloxone attenuates

Niclosamide potentiates TMEM16A and induces vasoconstriction.

The TMEM16A calcium-activated chloride channel is a promising therapeutic target for various diseases. Niclosamide, an anthelmintic medication, has been considered as a TMEM16A inhibitor for treating asthma and chronic obstructive pulmonary disease, but was recently found to possess broad-spectrum off-target effects. Here we show that, under physiological conditions, niclosamide acutely potentiates TMEM16A without having any inhibitory effect. Our computational and functional characterizations pinpoint a putative niclosamide binding site on the extracellular side of TMEM16A. Mutations in this

Marijuana-related Reversible Cerebral Vasoconstriction Syndrome.

The legalization of recreational marijuana in some countries has been accompanied by an increased number of case reports of serious cardiovascular and cerebrovascular complications. However, there have been few studies describing the detailed clinical course of reversible cerebral vasospasm syndrome (RCVS) associated with marijuana use. We herein report a unique case of recurrent bi-fronto-parietal subcortical (watershed) infarction in the setting of chronic daily marijuana use for several years, with evidence of bilateral anterior cerebral artery vasoconstriction. The quick resolution of symp

[Skin vasoconstriction effect of contact antiphlogistics].

Five contact antiphlogistics were evaluated and compared to hydrocortisone by the human skin vasoconstriction assay in 20 volunteers. The test concentrations ranged from 10(-2) to 10(-6) w/w; the duration of occlusive application was 16 h. The substances could be ranked as follows in ascending order of activity (index values in relation to hydrocortisone).

Vanadate-induced oliguria and vasoconstriction in the cat.

The effect of sodium orthovanadate (vanadate) on kidney function and hemodynamics was tested in chloralose anesthetized, fasting cats. Vanadate given i.v. caused a reversible, marked decrease in glomerular filtration rate and urine production without significantly affecting the fractional water excretion rate. In the recovery period, after administration of vanadate, urine flow and the urinary excretion rate of electrolytes were only slightly different from control values. This action of vanadate appears to be due mainly to constriction of the renal blood vessels leading to a fall in glomerula

Pattern A personality and noise-induced vasoconstriction.

A perplexing question about noise-induced hearing loss is why some persons seem to be more affected by high intensity noise than others. Hawkins (1971) has shown noise-induced vasoconstriction to be implicated in noise-induced hearing loss in animals as evidenced by vascular changes within the inner ear and he asks the question: Are these changes caused by the noise itself or mediated by the autonomic nervous system (ANS)? Our research employed a plethysmograph to measure ANS mediated vasoconstriction during noise exposure. The subjects were stress prone males and females (Pattern A) whose beh

Effect of vasoconstriction by dexmedetomidine in the mandible of rat.

BACKGROUND: In recent years, dexmedetomidine (DEX) has been proposed as a useful vasoconstrictor for local anesthesia because it is less effective in circulation than clonidine of antihypertensive drugs. In addition, DEX is expected to act as a vasoconstrictor during local anesthesia. However, histomorphometric studies demonstrating that DEX exerts vasoconstrictive effects are lacking. This study aimed to clarify whether DEX exerts a histomorphologically vasoconstrictive effect on blood vessels in the mandible of rats. METHODS: A total of 12 male Wistar rats were used. General anesthesia was

Ropivacaine in peribulbar anesthesia - vasoconstrictive properties.

BACKGROUND AND OBJECTIVES: Peribulbar anesthesia can reduce ocular blood flow (OBF) by increasing intraocular pressure (IOP) or due to the action of drugs. Ropivacaine has low toxicity and intrinsic vasoconstrictive properties, yet to be proven on the ocular vasculature. Measurements of ocular pulse amplitude (OPA) allow the indirect evaluation of the OBF. The objective of the present study was to evaluate through the OBF the vasoconstrictive properties of ropivacaine in peribulbar anesthesia. METHODS: Forty eyes undergoing peribulbar anesthesia with 7 mL of anesthetic solution without vasoco

Vasoconstriction as the etiology of hypercalcemia-induced seizures.

PURPOSE: Reversible cerebral vasoconstriction has been hypothesized to be the etiology of seizures due to hypercalcemia, but angiographic studies documenting vasoconstriction have not previously been available. METHODS: We present a 43-year-old woman who had frequent seizures that later evolved to status epilepticus with marked hypercalcemia at the time of the seizures. RESULTS: Magnetic resonance imaging (MRI) of the patient's brain revealed high signal changes in T(2)-weighted imaging, fluorescence-attenuated inversion recovery (FLAIR), and diffusion-weighted imaging (DWI) over the bilater

Two forms of vasoconstriction in systemic hypertension.

Clinical, pharmacologic and biochemical evidence characterizes essential hypertension as a heterogeneous spectrum of pathophysiologic substances rather than the single entity it has long been presumed to be. Although the causes of essential hypertension remain obscure, 2 different mechanisms for long-term vasoconstriction that sustain diastolic hypertension in the experimental and clinical forms of primary aldosteronism and renovascular hypertension can also be identified and quantified among patients with essential hypertension. The first mechanism is renin independent, requires antecedent so

Volume and vasoconstriction in experimental renovascular hypertension.

An analysis is presented of volume and vasoconstrictor factors in experimental renovascular hypertension. Volume expansion and increased cardiac output produced by renal retention of salt and water are not essential for chronic renovascular hypertension to develop. When volume expansion and increased cardiac output do occur, however, it appears that the increased cardiac output contributes directly to the hypertensive process without triggering myogenic alterations in peripheral resistance predicted by the whole-body autoregulation theory of hypertension. Activation of the renin-angiotensin va

Reversible cerebral vasoconstriction syndrome: A narrative review.

OBJECTIVES/BACKGROUND: This review summarizes current insights into Reversible cerebral vasoconstriction syndrome (RCVS) diagnosis, management, and outcomes. RCVS is a cerebrovascular disorder characterized by recurrent thunderclap headaches and transient segmental vasoconstriction of cerebral arteries, typically resolving within 3 months. METHODS: A comprehensive database search was performed across MEDLINE (via PubMed), Embase, Scopus, and the Cochrane Library. RESULTS: Although often self-limiting, RCVS may cause complications such as subarachnoid hemorrhage, ischemic stroke, and cerebral

Perfusion dynamics in reversible cerebral vasoconstriction syndrome.

BACKGROUND: Elucidating the spatiotemporal dynamics of cerebral blood flow (CBF) may resolve the enigmatic pathophysiology of reversible cerebral vasoconstriction syndrome (RCVS), a condition with variable clinical and vascular manifestations, where the current understanding of perfusion is limited to inconsistent case report findings. We aimed to determine the spatiotemporal perfusion dynamics and their clinical and vascular correlations in RCVS. METHODS: We prospectively recruited RCVS patients and age- and sex-matched healthy controls (HCs) over a 5-year period (June 2018 to April 2023) in

[Reversible cerebral vasoconstriction syndrome and ischemic stroke].

The article describes a 45-year-old female patient with recurrent transient ischemic attacks and ischemic stroke due to spontaneous spasm of the middle cerebral artery, the anterior cerebral artery and distal part of the internal cerebral artery on the left, verified by MR angiography and CT angiography. It is assumed that the spasm caused damage to the vascular wall, an increase in its permeability, the development of edema, inflammation and subsequent fibrosis, therefore a complete restoration of the arterial lumen did not occur.
