Uptake of verteporfin by articular tissues following systemic and intra-articular administration.

Photodynamic therapy (PDT) using the photosensitizer BPD-Verteporfin (liposomal benzoporphyrin derivative-monoacid ring A) has been shown in previous studies to be effective in the amelioration of inflammatory arthritis in both the MRL-lpr mouse and the New Zealand White (NZW) rabbit models, and could potentially offer alleviation of certain inflammation-related symptoms of rheumatoid arthritis. Time and dose dependency of BPD-MA tissue uptake was carried out in the inflamed synovium and other articular and peri-articular tissues following intravenous and intra-articular administration in the

Verteporfin-mediated on/off photoswitching functions synergistically to treat choroidal vascular diseases.

Choroidal vascular diseases, such as age-related macular degeneration, are the leading cause of vision impairment and are characterized by pathological angiogenesis. Verteporfin-mediated photodynamic therapy is a current strategy that selectively occludes choroidal neovasculature. However, the clinically used large-dose systemic administration increases the risk of systemic adverse events, such as phototoxicity to superficial tissues. In this study, we developed an in situ verteporfin delivery system with a photoswitching synergistic function that disassembles in response to intraocular inflam

Tumor-selective proteotoxicity of verteporfin inhibits colon cancer progression independently of YAP1.

Yes-associated protein 1 (YAP1) is a transcriptional coactivator in the Hippo signaling pathway. Increased YAP1 activity promotes the growth of tumors, including that of colorectal cancer (CRC). Verteporfin, a drug that enhances phototherapy to treat neovascular macular degeneration, is an inhibitor of YAP1. We found that verteporfin inhibited tumor growth independently of its effects on YAP1 or the related protein TAZ in genetically or chemically induced mouse models of CRC, in patient-derived xenografts, and in enteroid models of CRC. Instead, verteporfin exhibited in vivo selectivity for ki

Evaluation of Verteporfin as a Photosensitizer With Anticancer Activity Via System Biology Tools.

Introduction: Verteporfin as a photosensitizer in photodynamic therapy (PDT) is used to inhibit deviant choroidal vascularization. This agent also has an anticancer property. The effective treatment of various tumors such as melanoma by using verteporfin is confirmed. In the present study, the ability of verteporfin in growth inhibition of MKN45 is investigated via protein-protein interaction (PPI) analysis to explore its beneficial role in PDT. Methods: The gene expression profiles of the treated MKN45 cell with verteporfin are mined from the Gene Expression Omnibus (GEO) database and evaluat

Copper-verteporfin coordination nanoparticles to reverse ferroptosis resistance in pancreatic cancer therapy.

Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has attracted attention for cancer treatment. However, ferroptosis resistance in pancreatic cancer, mediated by the hyperactivated system Xc-/SLC7A11/GSH/GPX4 axis and YAP-driven SLC7A11 overexpression, remains a major therapeutic challenge. To overcome this, we screened various metalloporphyrins and identified Cu-based verteporfin as an effective YAP inhibitor, so that a tumor-targeted nanosystem termed CuVP-F127-IKE-Mem is developed, which integrates the YAP inhibitor copper-verteporfin metalloporphyrin

Monocyte-Mimetic Nanoparticle Delivery of Verteporfin Promotes Plaque-Stabilizing Remodeling in Atherosclerosis.

BACKGROUND: Despite advances in lipid-lowering and anti-inflammatory therapies, pharmacological strategies that act directly within established atherosclerotic lesions to suppress plaque progression remain limited. We recently developed a monocyte-mimetic nanoparticle (MoNP) platform that selectively targets inflamed endothelium and delivers Verteporfin (VP), termed MoNP-VP, to suppress YAP/TAZ-associated endothelial activation and plaque development. Here, we extend this biomimetic nanotherapy by defining the pharmacological effects of MoNP-VP within atherosclerotic vessels and evaluating its

Ambient light drives verteporfin-induced protein cross-linking during standard laboratory sample processing.

Verteporfin (VP) is widely used as a light-independent pharmacologic inhibitor of the YAP-TEAD transcriptional complex, despite its potent photosensitizing properties. Here we show that VP-associated high-molecular-weight complexes and apparent depletion of key proteins on immunoblots can be substantially driven by ambient-light exposure during post-lysis handling, more than by light during cellular treatment. In this study, the hypothesis is tested independently using MEL270, HEK293, and MCF-7 cells treated with therapeutic and supra-therapeutic concentrations of VP under four systematically

Mechanisms of action of photodynamic therapy with verteporfin for the treatment of age-related macular degeneration.

Age-related macular degeneration, especially the neovascular form of the disease, is the leading cause of blindness in elderly people in developed countries. Thermal photocoagulation is still the preferred treatment for choroidal neovascularization that does not involve the fovea, but it is suitable for only a small number of patients and it can lead to immediate loss of visual acuity. Photodynamic therapy with use of photochemical light activation of verteporfin as a photosensitizer (verteporfin therapy) has been shown to be effective in treating vascularized tumors, and its potential to trea

High-Performance Photodynamic Therapy of Tongue Squamous Cell Carcinoma with Multifunctional Nano-Verteporfin.

BACKGROUND: The photodynamic therapy (PDT) showed promising potential in treating tongue squamous cell carcinoma (TSCC). The Food and Drug Administration approved Verteporfin (Ver) is a powerful alternative in this field for its penetrating power and high production of reactive oxygen species (ROS). However, its applications in the treatment of TSCC are still rare. METHODS: Ver was loaded onto Poly (lactic-co-glycolic acid) (PLGA) nanoparticles, followed by the modification with RGD peptide as the ligand. The nanostructured was named as RPV.
