A Ve homologous gene from Gossypium barbadense, Gbvdr3, enhances the defense response against Verticillium dahliae.

The tomato Ve1 gene and several Ve1 homologues are involved in the resistance to Verticillium dahliae. Here, we report on another Ve homologous gene, Gbvdr3, from a Verticillium wilt-resistant cotton cultivar, Gossypium barbadense Hai7124, which has a 3207-bp region that encodes a predicted receptor-like protein. Transient expression analyses indicated that Gbvdr3 is localized in the plasma membrane, and virus-induced gene silencing of Gbvdr3 compromised the resistance of Hai7124 cotton to a defoliating strain of V. dahliae, V991, but not to a non-defoliating strain, BP2. This resistance patte

Functional characterization of cotton genes responsive to Verticillium dahliae through bioinformatics and reverse genetics strategies.

Verticillium wilt causes dramatic cotton yield loss in China. Although some genes or biological processes involved in the interaction between cotton and Verticillium dahliae have been identified, the molecular mechanism of cotton resistance to this disease is still poorly understood. The basic innate immune response for defence is somewhat conserved among plant species to defend themselves in complex environments, which makes it possible to characterize genes involved in cotton immunity based on information from model plants. With the availability of Arabidopsis databases, a data-mining strate

From isolation to application: Characterization of Talaromyces purpureogenus TS as a biocontrol agent against Verticillium wilt.

Verticillium wilt, caused by Verticillium dahliae Kleb., is a destructive vascular disease that is difficult to control using chemical fungicides and remains a major challenge in plant disease management. Microbial biocontrol represents a sustainable alternative, and the identification of highly antagonistic strains against V. dahliae is of considerable importance. In this study, a strain of Talaromyces purpureogenus TS, which demonstrated strong antagonistic and mycoparasitic activity against V. dahliae, was isolated from soil. Its cell-free supernatant inhibited the growth of V. dahliae by d

Drought-mitigation of Verticillium longisporum colonization in Brassica napus is associated with transcriptome and DNA methylation reprogramming.

Climate is becoming more variable, and the frequency and magnitude of drought and pathogen epidemics are changing. This creates a need to understand how limited water availability influences plant responses to biotic challenges. In Brassica napus L., drought decreases the disease progression and fungal colonization of the vascular pathogen Verticillium longisporum, yet the underlying regulatory mechanisms remain unclear. Here, we used an integrative approach combining transcriptome profiling, co-expression network analysis, and DNA methylation analysis to investigate host responses in hypocoty

Use of natural-based commercial products as an alternative for providing bioprotection against verticillium wilt of olive.

BACKGROUND: As a result of the ineffectiveness of existing control methods against Verticillium dahliae, the causal agent of verticillium wilt of olive (Olea europaea; VWO), it is necessary to search for sustainable and environmentally friendly alternatives, such as bioprotection by products based on plant extracts and other naturally synthesized compounds. Therefore, present study aimed to evaluate the effects of seven natural-based commercial products on the inhibition of mycelial growth, the germination of V. dahliae conidia and microsclerotia, and disease progression in olive plants (cv. Salicylic acid and salicylic acid glucoside in xylem sap of Brassica napus infected with Verticillium longisporum.

Salicylic acid (SA) and its glucoside (SAG) were detected in xylem sap of Brassica napus by HPLC-MS. Concentrations of SA and SAG in xylem sap from the root and hypocotyl of the plant, and in extracts of shoots above the hypocotyl, increased after infection with the vascular pathogen Verticillium longisporum. Both concentrations were correlated with disease severity assessed as the reduction in shoot length. Furthermore, SAG levels in shoot extracts were correlated with the amount of V. longisporum DNA in the hypocotyls. Although the concentration of SAG (but not SA) in xylem sap of infected p

Identification of GASA Protein Family Expression Levels in Cotton (Gossypium hirsutum L.) Affected by Verticillium wilt.

Verticillium wilt, caused by Verticillium dahliae, is a major constraint on cotton production. The GASA (Gibberellic Acid-Stimulated in Arabidopsis) proteins are cysteine-rich peptides induced by gibberellin signaling; however, their systemic role in upland cotton (Gossypium hirsutum) resistance to Verticillium wilt remains elusive. Here, we identified 40 GhGASA members from the G. hirsutum genome, which were clustered into three subfamilies. All members possessed a typical gibberellin-regulated domain, and segmental duplication was the primary driver of family expansion. Transcriptomic analys

Cotton RLP6 Interacts With NDR1/HIN6 to Enhance Verticillium Wilt Resistance via Altering ROS and SA.

Cotton Verticillium wilt (VW) is often a destructive disease that results in significant fibre yield and quality losses in Gossypium hirsutum. Transferring the resistance trait of Gossypium barbadense to G. hirsutum is optional but challenging in traditional breeding due to limited molecular dissections of resistance genes. Here, we discovered a species-diversified structural variation (SV) in the promoter of receptor-like protein 6 (RLP6) that caused distinctly higher expression level of RLP6 in G. barbadense with the SV than G. hirsutum without the SV. Functional experiments showed that RLP6

Antifungal Activity of Chaetoviridin A from Chaetomium globosum CEF-082 Metabolites Against Verticillium dahliae in Cotton.

Cotton Verticillium wilt (CVW) is a severe soilborne disease caused by the pathogen Verticillium dahliae, and it has a great impact on cotton production. Previous studies found that the biocontrol agent Chaetomium globosum CEF-082 and its metabolic filtrate could reduce the incidence of CVW; however, the underlying mechanism remains unclear. The metabolic crude extract of CEF-082 increased the sensitivity of V. dahliae to stress, degraded the cell wall of V. dahliae, and increased the emergence and plant height of cotton. Through separation and purification of the metabolic crude extract of CE

Detection of Multiple Verticillium Species in Soil Using Density Flotation and Real-Time Polymerase Chain Reaction.

Wet sieving of soil samples, followed by plating on semi-selective medium and microscopic analysis, is the most commonly used technique to quantify microsclerotia-forming Verticillium species in soil. However, the method is restricted to small samples, does not allow easy differentiation between species, and takes several weeks to complete. This study describes an alternative method to test 100-g soil samples for three Verticillium species (V. tricorpus, V. dahliae, and V. longisporum) using density flotation-based extraction of microsclerotia followed by new real-time polymerase chain reactio

Exploring the biochemical and biological functions of copper radical oxidases in the vascular wilt phytopathogen Verticillium dahliae.

Plant pathogenic fungi pose a significant threat to global food security. Hence, discovering targeted (bio)chemical methods to combat plant diseases is a critical research avenue. Recent work has implicated copper radical oxidases (CROs) from Auxiliary Activity Family 5 (AA5) in fungal morphogenesis and pathogenesis, including in appressorium penetration by foliar phytopathogens in the genera Colletotrichum and Magnaporthe. We identified orthologous AA5-encoding genes in Verticillium species; however, it was not clear that the functions of the corresponding CROs would be conserved in these vas

Nitrogen dioxide treatment reduces the viability of seedborne Verticillium dahliae and the transmission of Peronospora effusa in spinach.

The global spinach (Spinacia oleracea) seed trade has contributed to the spread of plant pathogens such as Verticillium dahliae and Peronospora effusa. Verticillium dahliae causes Verticillium wilt in a broad range of crops, including lettuce (Lactuca sativa), strawberry (Fragaria x ananassa), and others. Verticillium wilt occurs only in the spinach seed production fields and is not observed in the spinach production fields as the crop is harvested well before symptom manifestation. In contrast, P. effusa exclusively infects spinach in production fields causing destructive downy mildew. Managi

Components of TOR and MAP kinase signaling control chemotropism and pathogenicity in the fungal pathogen Verticillium dahliae.

Filamentous fungi can sense useful resources and hazards in their environment and direct growth of their hyphae accordingly. Chemotropism ensures access to nutrients, contact with other individuals (e.g., for mating), and interaction with hosts in the case of pathogens. Previous studies have revealed a complex chemotropic sensing landscape during host-pathogen interactions, but the underlying molecular machinery remains poorly characterized. Here we studied mechanisms controlling directed hyphal growth of the important plant-pathogenic fungus Verticillium dahliae towards different chemoattract

Molecular evidence for the involvement of cotton GhGLP2, in enhanced resistance to Verticillium and Fusarium Wilts and oxidative stress.

Germin-like proteins (GLPs) are a diverse and ubiquitous family of plant glycoproteins belonging to the cupin super family; they play considerable roles in plant responses against various abiotic and biotic stresses. Here, we provide evidence that GLP2 protein from cotton (Gossypium hirsutum) functions in plant defense responses against Verticillium dahliae, Fusarium oxysporum and oxidative stress. Purified recombinant GhGLP2 exhibits superoxide dismutase (SOD) activity and inhibits spore germination of pathogens. Virus-induced silencing of GhGLP2 in cotton results in increased susceptibility

Synergistic Interactions between Plant-parasitic Nematodes (Meloidogyne hapla and Paratylenchus projectus) and Verticillium dahliae on Peppermint.

Synergistic interactions between soil-borne fungi and plant-parasitic nematodes can cause significantly more damage to a crop than either pathogen alone. For peppermint, synergistic interactions between Verticillium dahliae and plant-parasitic nematodes have only been described for the migratory endoparasite Pratylenchus, whereas there is no information for such interactions for ectoparasitic or sedentary endoparasitic nematodes. To determine whether species of these nematode groups co-occur with V. dahliae on peppermint, and to evaluate whether their interactions with V. dahliae on peppermint

Cotton GhMKK5-GhMPK13 cascade regulates resistance to verticillium wilt by phosphorylating GhBES1-GhPEL75-mediated molecular module.

Verticillium wilt, caused by Verticillium dahliae, is a soil-borne vascular disease that harms many plants, including cotton. The mitogen-activated protein kinase cascade is a highly conserved signal transduction pathway that transmits various extracellular stimuli to downstream targets during plant defense responses. Although there have been many studies on the key role of the mitogen-activated protein kinase cascade in plant disease resistance, mitogen-activated protein kinase signal transduction in cotton remains to be elucidated. Here, we found that GhMKK5 interacted with GhMPK13 and direc

The small GTP-binding protein GhARAC3 is involved in cotton resistance to Verticillium dahliae by regulating lignin metabolism.

Plant Rho/ROP GTPases act as essential molecular switches in disease resistance. However, how their protein stability is regulated and integrated with lignin metabolism remains poorly understood. Here, we demonstrate that the cotton small GTPase GhARAC3 serves as a positive regulator of resistance against the vascular pathogen Verticillium dahliae (V. dahliae). Genetic analyses reveal that overexpression of GhARAC3 enhances defense responses in cotton and Arabidopsis, whereas its silencing significantly compromises plant immunity. Under normal physiological conditions, the E3 ubiquitin ligase
