Comparative persistence of opportunistic Vibrio alginolyticus and major foodborne Vibrio species (Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus) in ready-to-eat seafood under different storage conditions.

Vibrio alginolyticus is an opportunistic marine bacterium capable of infecting both humans and aquatic animals, in addition to major foodborne pathogenic Vibrio species (Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus). Although these species are increasingly detected in seafood, differences in their persistence under conditions simulating real food systems remain poorly understood. This study investigated the growth, survival, and recovery of V. alginolyticus and compared its behavior with those of the three pathogenic species in ready-to-eat seafood (flatfish, smoked salmon,

The role of rcpA gene in regulating biofilm formation and virulence in Vibrio parahaemolyticus.

Vibrio parahaemolyticus (V. parahaemolyticus) is a common seafood-borne pathogen that can colonize the intestine of host and cause gastroenteritis. Biofilm formation by V. parahaemolyticus enhances its persistence in various environments, which poses a series of threats to food safety. This work aims to investigate the function of rcpA gene in biofilm formation and virulence of V. parahaemolyticus. Deletion of rcpA significantly reduced motility, biofilm biomass, and extracellular polymeric substances, and inhibited biofilm formation on a variety of food and food contact surfaces. In mice infe

Genomic Characterization of Ciprofloxacin Resistance in Laboratory-Derived Mutants of Vibrio parahaemolyticus.

The quinolone ciprofloxacin is a broad-spectrum bactericidal antibiotic used for human medicine as well as the aquaculture industry. The emergence of ciprofloxacin-resistant Vibrio parahaemolyticus strains is currently a global public health concern. However, the mechanism of ciprofloxacin resistance in V. parahaemolyticus is not yet fully clarified. We generated mutants with decreased ciprofloxacin susceptibility using in vitro selection and investigated genes associated with ciprofloxacin resistance on a genetic level. Our selection process yielded mutants that possessed altered minimal inhi

Recombinant expression and immunogenicity evaluation of a TonB-dependent receptor of Vibrio parahaemolyticus.

This study constructed the recombinant plasmid of a TonB-dependent receptor from V. parahaemolyticus and evaluated the immunogenicity of the recombinant protein in mice. The TonB-dependent receptor gene (GI: 28901321) was obtained by PCR amplification and cloned into plasmid pET-32a (+). The recombinant plasmids were transformed into Escherichia coli BL21, and the protein expression was induced by isopropyl-β-d-thiogalactopyranoside (IPTG). The 6 × His-tagged TonB-dependent receptor inclusion bodies were purified by Ni-NTA Agarose column and renatured by gradient urea dialysis. The soluble and

Tetrathionate sensing enables Vibrio parahaemolyticus to repress quorum sensing for intestinal colonization.

Quorum sensing (QS) enables bacterial coordination of group behaviors, yet how enteric pathogens modulate QS during intestinal colonization remains poorly understood. Here, we demonstrated that Vibrio parahaemolyticus represses QS to promote gut colonization through sensing of self‑generated tetrathionate derived from host thiosulfate. We found that the cecal extracts from healthy mouse contained thiosulfate, which could be converted to tetrathionate by V. parahaemolyticus via the thiosulfate dehydrogenase TsdBA. The two‑component system TtrRS then sensed this self‑produced tetrathionate and,

Vibrio parahaemolyticus Induced Necrotizing Fasciitis: An Atypical Organism Causing an Unusual Presentation.

Background necrotizing fasciitis (NF) represents a life-threatening bacterial infection characterized by a rapid necrosis of deep subcutaneous tissue and facia underlying the skin. Despite its lethal nature, NF occurs infrequently, leaving many physicians unfamiliar with the disease process, common pathogens, and treatment strategies. Here we present a case of NF caused by an unlikely organism, Vibrio parahaemolyticus. We highlight the innocuous nature of initial presentation and the potentially devastating sequela.

Isolation and characterization of a novel Schitoviridae phage VipHU7 that infects Vibrio parahaemolyticus.

In recent years, various bacteriophages that infect Vibrio spp. have been isolated and characterized. However, many characteristics concerning their infection mechanisms remain unknown. Here, we isolated and characterized a novel phage, VipHU7, that infects Vibrio parahaemolyticus. The morphology of VipHU7 was examined using transmission electron microscopy, which demonstrated that it has a short, noncontractile tail characteristic of podoviruses. VipHU7 formed clear plaques with halo zones on a bacterial lawn of V. parahaemolyticus MFS 1101, and host range analysis revealed that it had a limi

Serlogical, virulence, and molecular characterization of clinical and seafood Vibrio parahaemolyticus isolates.

In this study, 42 clinical and 114 seafood isolates of V. parahaemolyticus from Shanghai, China, were investigated. Serotypes, Kanagawa phenomenon (KP), potential virulence factors, and phylogenetic relationship (gyrB gene) of these isolates were analyzed. In 156 isolates, a total of 28 serotypes were identified and only 3 predominant serotypes (O1:KUT, O3:K6, and O4:K2) were in common between clinical and seafood isolates. Of these, O1:KUT and O3:K6 were pandemic-associated. Results showed that 95.2% (40/42) clinical and 1.8% (2/114) seafood isolates were tdh+and thus KP. All clinical and sea

Phage Φ170 biocontrol of multidrug-resistant Vibrio parahaemolyticus in a zebrafish model.

Vibrio parahaemolyticus is an important zoonotic pathogen that poses a persistent threat to aquaculture systems and food safety. The rapid emergence of multidrug resistance, driven by increasing antibiotic selection pressure, highlights the urgent need for alternative antimicrobial strategies, among which phage therapy has emerged as a promising candidate. Here, we reported the isolation of a bacteriophage, designated Φ170, targeting multidrug-resistant Vibrio parahaemolyticus from sewage collected at a shrimp farm. Φ170 exhibited a latent period of 10 min, an eclipse period of approximately 6

Regulation of Virulence Factors Expression During the Intestinal Colonization of Vibrio parahaemolyticus.

Colonization and adhesion are the key steps for Vibrio parahaemolyticus to infect human body and cause seafood poisoning. However, at present, there is a lack of systematic review on the regulation of virulence factors expression during the intestinal colonization of V. parahaemolyticus. This review aims to describe the virulence factors associated with the colonization and adhesion of V. parahaemolyticus (multivalent adhesion molecule 7, enolase secretion, use of flagella, biofilm formation, and the action of secretion systems) and focuses on the aspects that affect these processes in V. para

A systematic review of post-harvest interventions for Vibrio parahaemolyticus in raw oysters.

BACKGROUND: Non-cholera Vibrio bacteria are a major cause of foodborne illness in the United States. Raw oysters are commonly implicated in gastroenteritis caused by pathogenic Vibrio parahaemolyticus. In response to outbreaks in 1997-1998, the US Food and Drug Administration developed a nation-wide quantitative microbial risk assessment (QMRA) of V. parahaemolyticus in raw oysters in 2005. The QMRA identified information gaps that new research may address. Incidence of sporadic V. parahaemolyticus illness has recently increased and, as oyster consumption increases and sea temperatures rise, V
