Fluorometric quantification of alkaloids in the homeopathic mother tinctures of Vinca minor L. and Fumaria officinalis L.

To assess the toxic potential of the alkaloids, a quantification method is necessary. An ion pair extraction method was used for quantitative fluorometric determination of vincamine, protopine and all contained alkaloids in the mother tinctures of Vinca minor and Fumaria officinalis. The non-fluorescent alkaloids were transformed into an ion pair with sodium-9,10-dimethoxy-anthracene-sulfonate and then fluorometrically determined and quantified in this study. The applicable ion pair was extracted in a suitable organic solvent, where dichloromethane has proven to be beneficial. Conditions for t

Effect of various classes of drugs on complete ischemia induced by decapitation and cyanide intoxication in mice.

Effects of various classes of drugs on complete ischemia (gasping) induced by decapitation and cyanide intoxication in mice were investigated. The average gasping duration in complete ischemia and survival time in potassium cyanide injection of control mice were 21.0 +/- 0.1 and 26.1 +/- 0.4 sec (mean +/- SEM), respectively. Some antidepressants, neuroleptics and tranquilizers significantly and dose-dependently prolonged the duration of gasping and survival time, and decreased locomotor activity. Vincamine and its analogues are markedly effective for treating cyanide intoxication. It is concei

Assessment of pharmacological effects on cerebral blood flow.

The effect of various drugs on hemispheric and regional cerebral blood flow (CBF) was investigated in a total of 410 patients. While a few drugs (midodrin, proxazole, vincamine, hexobendine, extract of ginkgo biloba, dextran and ouabain) were able to improve hemispheric CBF, only ephedrine combined with xanthines decreased CBF. For vincamine the dependency of the effect on certain plasma levels was established. Only ouabain of the tested cardiac glycosides effected CBF; their similar hemodynamic actions suggest here an influence of ouabain on cerebral vessels. For the evaluation of drug effect

Eburnamine derivatives and the brain.

The Apocynaceae plant family contains a great number of so called eburnamine-vincamine alkaloids. Quite a few of these alkaloids exert varied pharmacological activities on the cell multiplication, cardiovascular system, and brain functions. Many derivatives were also synthesized to find pharmacologically active compounds better characterized and safer to be administered than the natural plant alkaloids themselves. We concentrate on the eburnamine structures with cerebral activities in this review. Vincamine, vinburnine, vindeburnol, apovincaminate, and vinpocetine (cis-ethyl-apovincaminate) al

In vivo antispasmodic activity of Sulcotidil.

Suloctidil was tested in vivo for its antispasmodic activity on peripheral and cerebral circulations. In the perfused dog hind limb preperation, suloctidil was found to inhibit after 2 and 5 min, the vasospasm induced by norepinephrine or angiotensin; at equal dose, and at the game time intervals, it was more potent than cinnarizine and papaverine. BaCl(2) induced spasms of the pial arterial of the rabbit were also rapidly alleviated by i.a. suloctidil (3.5 mug/kg or more), in conditions where papaverine and vincamine were inactive.

APOE ε4 allele drives female-specific Alzheimer's disease progression via vascular dysfunction and tau spreading.

BACKGROUND: Apolipoprotein E (APOE) epsilon4 (ε4) is a major genetic risk factor for late-onset Alzheimer's disease (AD), with women exhibiting heightened vulnerability to APOE ε4-associated cognitive impairment. Despite recognition of this sexual dimorphism, the underlying biological mechanisms remain incompletely understood. METHODS: We performed weighted gene co-expression network analysis (WGCNA) on RNA-seq data from the Mayo Clinic cohort (n = 277 temporal cortex samples) to identify sex- and APOE ε4-linked co-expression modules. 315 hub genes were identified within the most relevant gen

THE ACTIVITY OF ANTIOXIDANT ENZYMES AND THE EXPRESSION OF THEIR GENES IN THE RAT BRAIN AFTER INJECTING OF VINDEBURNOL IN THE MULTIPLE SCLEROSIS MODEL

The article presents the results of a study on the effectiveness of vindeburnol, a structural analogue of the cerebral vasodilator vincamine, in the model of multiple sclerosis. The drug was injected subcutaneously in rats after modeling experimental allergic encephalomyelitis at a dose of 20 μg/kg, from the first manifestation of a neurological deficit over the next 10 days. In the somatosensory cortex of animals, the activity of superoxide dismutase, glutathione peroxidase and glutathione reductase, as well as the expression level of the SOD1, GPX4, GPX6 and GSR genes using real-time polymer

Protective Potential of Vinpocetine on Acrylamide Induced Liver Toxicity in Male Albino Rats.

Acrylamide is a highly reactive carbonyl compound extensively utilized in industrial applications. It shows a significant association with oxidative stress, neurotoxic, and genotoxic effects. This study sought to assess the protective effects of vinpocetine, a synthetic analog of the natural alkaloid vincamine, which exhibits strong antioxidant and anti-inflammatory properties, against ACR-induced hepatic injury. Experimental animals were categorized into four groups and orally treated as follows: control group, Vinpo. group (5 mg/kg bw), Acrylamide group (38.27 mg/kg bw), and Vinpo. plus acry

Vinpocetine inhibits Streptococcus pneumoniae-induced upregulation of mucin MUC5AC expression via induction of MKP-1 phosphatase in the pathogenesis of otitis media.

Mucin overproduction is a hallmark of otitis media (OM). Streptococcus pneumoniae is one of the most common bacterial pathogens causing OM. Mucin MUC5AC plays an important role in mucociliary clearance of bacterial pathogens. However, if uncontrolled, excessive mucus contributes significantly to conductive hearing loss. Currently, there is a lack of effective therapeutic agents that suppress mucus overproduction. In this study, we show that a currently existing antistroke drug, vinpocetine, a derivative of the alkaloid vincamine, inhibited S. pneumoniae-induced mucin MUC5AC upregulation in cul

Antidepressant-like Effect of the Eburnamine-Type Molecule Vindeburnol in Rat and Mouse Models of Ultrasound-Induced Depression.

Vindeburnol (VIND, RU24722, BC19), a synthetic molecule derived from the eburnamine-vincamine alkaloid group, has many neuropsychopharmacological effects, but its antidepressant-like effects are poorly understood and have only been described in a few patents. To reliably estimate this effect, vindeburnol was studied in a model of long-term variable-frequency ultrasound (US) exposure at 20-45 kHz in male Wistar rats and BALB/c mice. Vindeburnol was administered chronically for 21 days against a background of simultaneous ultrasound exposure at a dose of 20 mg/kg intraperitoneally (IP). Using fo

Effects of vinburnine on experimental models of learning and memory impairments.

Retrograde amnesia can be induced experimentally in mice by injecting them with scopolamine (3 mg/kg, IP) or by inducing seizures with pentylenetetrazol (50 mg/kg, IP), and in rats by subjecting them to hypobaric hypoxia (at a barometric pressure of 300 mmHg for 3 min). We have studied the effects of vinburnine (VNB) in these amnesic states compared to vincamine (VNC) and nicergoline (NCG), in order to assess its activity on drug-induced learning and memory impairments. Vinburnine reduced the disrupting effect of both scopolamine and pentylenetetrazol-induced seizures on the retention of a ste

Flunarizine. A reappraisal of its pharmacological properties and therapeutic use in neurological disorders.

Flunarizine is a class IV calcium antagonist with a pharmacological profile which suggests its therapeutic potential in a number of neurological and cerebrovascular disorders. It is an effective prophylactic treatment for common or classic migraine in children and adults, and it appears at least as effective as a number of other agents which act by different pharmacological mechanisms, including pizotifen (pizotyline), cinnarizine, methysergide, nimodipine, metoprolol, propranolol, aspirin and cyclandelate. Flunarizine is also effective in reducing the frequency of seizures, when used as an 'a

Protective action of vinpocetine against experimentally induced gastric damage in rats.

The efficacy of vinpocetine (CAS 42971-09-5) to prevent gastric mucosal damage induced by several noxious agents and its antisecretory effect were studied in rats. Vinpocetine administered orally or intraperitoneally inhibited the development of gastric lesions induced by 96% ethanol in a dose-dependent way. The highest protective activity was observed when vinpocetine was given intraperitoneally 30 min before ethanol, and its effect was still significant when administered 120 min before ethanol exposure. Oral administration of vincamine also displayed gastroprotective action in this model. Pr

[Pharmacologic modification of the circulation in the brain].

Hemispheric and regional cerebral blood flow were measured in 448 patients before and after application of various drugs. Of the tested substances, only a few affected hCBF: ephedrine-xanthines decreased hCBF, and midodrine, proxazole, vincamine, hexobendine, extract of ginkgo biloba, dextran, dextran-sorbitol, carboanhydrase inhibitor, and ouabain incresaed hCBF. In the rCBF maps different reaction patterns of regional flow were observed which were statistically tested by regression analysis. Homogeneous responses were seen as diffuse increases or decreases of rCBF. Heterogeneous responses oc

Vinpocetine attenuates thioacetamide-induced liver fibrosis in rats.

Liver fibrosis is associated with increased mortality and morbidity. However, there is not effective treatment so far. Vinpocetine (Vinpo) is a synthetic derivative of vinca alkaloid vincamine. Limited previous reports have shown some beneficial effects of Vinpo in different organ fibrosis, but the ability of Vinpo to inhibit liver fibrosis induced by thioacetamide (TAA) has not been reported, that is why we investigate the potential ability of this vinca alkaloid derivative to attenuate liver fibrosis. Hepatic fibrosis was induced in male Sprague Dawley rats by TAA (200 mg/kg; ip; 3 times/wee
