[Chronic blockade of angiotensin II type 1 receptor cannot completely prevent structural adaptation in vessels of simulated weightless rats].

Our previous studies suggest that the vascular local renin-angiotensin system (L-RAS) plays a pivotal role in the region-specific vascular adaptation due to simulated weightlessness. The present study was designed to determine whether simulated weightlessness still induced adaptive changes in rat vessels when angiotensin II type 1 receptor (AT(1)R) was chronically blocked by the administration of losartan, and whether the expressions of key elements in the L-RAS in the large arteries would change. Tail suspension for 4 weeks was used to simulate the physiological effect of weightlessness. The

Polygonatum sibiricum Polysaccharides Alleviate Simulated Weightlessness-Induced Cognitive Impairment by Gut Microbiota Modulation and Suppression of NLRP3/NF-κB Pathways.

Background/Objectives: Polygonatum sibiricum (PS), possessing both medicinal and edible dual functions, boasts a long history of application in Chinese traditional practices. As a component of its effectiveness, Polygonatum sibiricum polysaccharides (PSPs) have been reported to exert neuroprotective effects. However, the protective effects of PS on the cognitive deficits induced by simulated weightlessness remain unclear. This study evaluated the therapeutic potential of PSPs to counteract the cognitive deficits induced by simulated weightlessness using the Hindlimb Unloading (HU) method. Meth

Multiple organ injury in septic rats under weightlessness.

INTRODUCTION: As human space exploration advances, the risk of infection faced by astronauts has also become apparent. However, the impact of sepsis on the body under weightlessness remains unclear. METHODS: A rat model of sepsis under simulated weightlessness was preliminarily established by combining hindlimb unloading with cecal ligation and puncture. The effects of simulated weightlessness on the early progression of sepsis in rats were systematically evaluated through assessments of mortality, hematological parameters, inflammatory markers, coagulation function, organ function, and histo

Physiological effects induced by antiorthostatic hypokinesia.

The main purpose of the present investigation was to reproduce some physiological reactions in the sensory system which are similar to those observed during the first day of the adaptation to weightlessness. This was achieved by a 5-day bed rest experiment during which the test subjects were kept in the antiorthostatic position at angles of 0 degrees, -4 degrees, -8 degrees, -12 degrees. Our investigations allowed simulation of the acute stage of adaptation to weightlessness and assessment of gravity-induced blood redistribution in the development of the above physiological reactions.

[Genesis of vestibulo-autonomic disorders in space flight].

Some considerations on the mechanisms of vestibulo-vegetative disturbance in space flight are given on the basis of clinical observations, experience of otoneurological clinic, simulation experiments and examinations of cosmonauts. The starting point in these disturbances was a significant fact of blood redistribution in weightlessness and subsequent changes in hemodynamics, metabolism, etc. The body reactions to weightlessness seem to prepare pronounced vestibulo-vegetative reflexes to threshold and subthreshold vestibular stimuli induced by head and body movements in flight. Some aspects of

An effect of weightlessness following exposure to vibration.

Vibration of germinating wheat seedlings at the levels experienced during the launch of the NASA Biosatellite II increases the frequency of developmental arrest in seedling organs. Severe vibrations lasted approximately 30 sec in two stages. Power spectral density was greatest at frequencies around 15-16 and 19-22 Hz on the entire vehicle. Vibration forces reaching the affected parts of individual seedlings could not be measured. One or more seedling organs may be expected to be absent in 11% of selected Earth-grown wheat plants. If subjected to simulated launch vibration between 12 and 27 hr

[Mechanism of weightlessness osteoporosis and preventive and therapeutic effect of traditional Chinese medicine].

Weightlessness environment can lead to the muscle atrophy and body fluid distribution upward, which can cause the bone calcium metabolism disorder and always accompanied by the loss of bone microstructure and increased rate of bone fracture. Under microgravity, the astronauts are much easier to decrease the Ca2+ ion in bone, which can cause serious osteoporosis. However the bone lost is not equilibrium, it is especially serious in the mechanism loading bone and the recovery process is more difficult. These are very different from the osteoporosis in older people and postmenopausal osteoporosis.

Metabolic Profiles of Encapsulated Chondrocytes Exposed to Short-Term Simulated Microgravity.

The mechanism by which chondrocytes respond to reduced mechanical loading environments and the subsequent risk of developing osteoarthritis remains unclear. This is of particular concern for astronauts. In space the reduced joint loading forces during prolonged microgravity (10-6 g) exposure could lead to osteoarthritis (OA), compromising quality of life post-spaceflight. In this study, we encapsulated human chondrocytes in an agarose gel of similar stiffness to the pericellular matrix to mimic the cartilage microenvironment. We then exposed agarose-chondrocyte constructs to simulated microgra

Ultrasound-based Assessment of Jugular Vein Alterations in Response to Simulated Microgravity and Countermeasures: A Systematic Review.

During spaceflight, assessment of jugular vein (JV) alterations is critical as it reflects systemic responses to fluid redistribution and carries a risk of thrombosis, a potentially fatal condition if undetected. Imaging options during spaceflight are restricted, making ultrasound the only feasible modality. However, available data on JV alterations are from small astronaut cohorts (typically n <12), limiting statistical power and robust validation. As a result, ground-based microgravity simulation studies are required to characterise microgravity-induced physiological changes, including JV al

Pharmacology in space: pharmacotherapy.

This chapter summarized the information available on the pharmacological kits onboard spacecraft and on the use of drugs in space, while the next chapter is dedicated to the impacts of weightlessness on drug pharmacokinetics. The need of a selected group of drugs for the use of astronauts during short-term and long-term spaceflights has been discussed. Recommendations are made for a Space Pharmacopoeia as well as for the areas of research needed to adapt medication to the weightlessness of the space environment. Although the usefulness of these drugs has been clearly demonstrated, their use al

Remains Containment Considerations for Death in Low-Earth Orbit.

BACKGROUND: Maintenance and disposition of decedent remains during spaceflight require the isolation of biohazardous products of decomposition in microgravity and in the absence of refrigeration. Containment and isolation options would preferably offer sufficient time to enable crew and ground support teams to determine appropriate disposition of remains and even potentially return remains to the Earth. The pilot study described herein undertook an effort to develop a postmortem containment unit for the isolation and maintenance of decedent remains in a microgravity environment.METHODS: Commer

Formaldehyde initiates memory and motor impairments under weightlessness condition.

During space flight, prolonged weightlessness stress exerts a range of detrimental impacts on the physiology and psychology of astronauts. These manifestations encompass depressive symptoms, anxiety, and impairments in both short-term memory and motor functions, albeit the precise underlying mechanisms remain elusive. Recent studies have revealed that hindlimb unloading (HU) animal models, which simulate space weightlessness, exhibited a disorder in memory and motor function associated with endogenous formaldehyde (FA) accumulation in the hippocampus and cerebellum, disruption of brain extrace

Significance of vestibular organs in problems of weightlessness.

Two important questions concerning the organs of equilibrium are posed by orbital and space flights. The first is whether exposure to weightlessness may evoke symptoms of vestibular origin, and the second is how to prevent symptoms of vestibular origin should it be decided to abolish weightlessness by causing the spacecraft to spin. It is important to determine whether vestibular symptoms are to be ascribed to the otolith apparatus or to the semicircular canals, or to both. During exposure to weightlessness the usual gravitational stimulus to the otolith apparatus is lost, and this reduction i

Establishment and Validation of a Novel Microgravity Simulation Platform for Ground-Based Animal Experiments.

Ground-based analog models, such as the hindlimb unloading (HU) technique, have been used for several decades in simulating microgravity effects on musculoskeletal studies. However, this model has seen limited technological advancement in several aspects. Here, the design and validation of the Microgravity Simulated Platform (MSP), a refined evolution of the classic HU model, is presented. The multipurpose engineered MSP apparatus is designed to overcome key limitations of existing models by enabling simultaneous study of a larger number of mice in compact space, with fewer lesions, less infla
