Wolffian duct derivative anomalies: technical considerations when encountered during robot-assisted radical prostatectomy.

BACKGROUND: Although malformations of the genitourinary tract are typically identified during childhood, they can remain silent until incidental detection in evaluation and treatment of other pathologies during adulthood. The advent of the minimally invasive era in urologic surgery has given rise to unique challenges in the surgical management of anomalies of the genitourinary tract. OBJECTIVE: This article reviews the embryology of anomalies of Wolffian duct (WD) derivatives with specific attention to the seminal vesicles, vas deferens, ureter, and kidneys. This is followed by a discussion o

Altered gene expression during rat Wolffian duct development in response to in utero exposure to the antiandrogen linuron.

Linuron is an herbicide with weak androgen receptor (AR) antagonist activity. Exposure to linuron from gestation days (GD) 12 to 21 perturbs androgen-dependent male reproductive development. In utero exposure to 50-mg/kg/day linuron induces malformations of the epididymis and the vas deferens. The objective of this study was to identify alterations in gene expression within the testis and epididymis associated with abnormal Wolffian duct development and to correlate changes in gene expression with the gross morphology of the affected epididymides. Pregnant Sprague-Dawley rats were administered

Right ureterovesical junction cyst associated with ipsilateral renal agenesis: a case report of a possible Wolffian duct maldevelopment.

INTRODUCTION: Right ureterovesical junction cyst associated with ipsilateral renal agenesis is an extremely rare congenital anomaly of the male genitourinary tract, likely resulting from abnormal development of the Wolffian duct and ureteric bud. We report the case of a 35-year-old man with a one-year history of progressive dysuria. Contrast-enhanced computed tomography (CT) revealed right renal agenesis and cystic dilatation of the intramural segment of the right ureter protruding into the bladder cavity, with the proximal ureter showing tortuosity and atresia. The contralateral kidney and ur

Complete androgen insensitivity without Wolffian duct development: the AR-A form of the androgen receptor is not sufficient for male genital development.

BACKGROUND: The androgen receptor (AR) is essential for the differentiation of male external and internal genitalia. It is normally present in two forms, a full-length form B and an N-terminal truncated form A with still unknown function. Mutations in the AR gene cause androgen insensitivity syndrome (AIS), which is divided into subgroups according to the degree of undermasculinization. Patients with completely female external genitalia are classified as complete AIS (CAIS). However, a recent study has shown that some CAIS patients have signs of internal male genital differentiation due to mis

Loss of peri-Wolffian duct stromal Frs2α expression in mice leads to abnormal ureteric bud induction and vesicoureteral reflux.

UNLABELLED: BackgroundFibroblast growth factor receptor 2 (Fgfr2) deletion from murine peri-Wolffian duct stroma (ST) results in aberrant ureteric bud induction, abnormal ureteral insertion into the bladder, and high rates of vesicoureteral reflux (VUR). It is unclear which receptor docking protein(s) is/are responsible for Fgfr2 actions in these tissues. We investigated whether the docking protein, fibroblast receptor substrate 2α (Frs2α), had a role in peri-Wolffian duct ST similar to Fgfr2.MethodsWe conditionally deleted Frs2α in peri-Wolffian duct ST with a Tbx18cre mouse line (Frs2αST-/-)

Deletion of fibroblast growth factor receptor 2 from the peri-wolffian duct stroma leads to ureteric induction abnormalities and vesicoureteral reflux.

PURPOSE: Pax3cre-mediated deletion of fibroblast growth factor receptor 2 (Fgfr2) broadly in renal and urinary tract mesenchyme led to ureteric bud (UB) induction defects and vesicoureteral reflux (VUR), although the mechanisms were unclear. Here, we investigated whether Fgfr2 acts specifically in peri-Wolffian duct stroma (ST) to regulate UB induction and development of VUR and the mechanisms of Fgfr2 activity. METHODS: We conditionally deleted Fgfr2 in ST (Fgfr2(ST-/-)) using Tbx18cre mice. To look for ureteric bud induction defects in young embryos, we assessed length and apoptosis of comm

Analysis of the prostatic central zone in patients with unilateral absence of wolffian duct structures: further evidence of the mesodermal origin of the prostatic central zone.

PURPOSE: We evaluate the status of the prostatic central zone in patients with unilateral absence of the seminal vesicles and vas deferens. MATERIALS AND METHODS: The entire bases of the prostate specimens from 3 patients with unilateral absence of the seminal vesicles and vas deferens who underwent radical prostatectomy for prostatic adenocarcinoma were evaluated on hemotoxylin and eosin stained sections for the distribution of the central zone. Immunohistochemical stains for pepsinogen II were performed on areas thought to be central zone and on corresponding contralateral sections. RESULT

Genital ducts and external genitalia: development and anomalies.

Human wolffian ducts are considered to be of ectodermal origin. Developing müllerian ducts in the area of the mesonephric ducts grow in contact with wolffian ducts. Vaginal differentiation occurs only if müllerian ducts contact the urogenital sinus, inducing proliferation of the entodermal epithelium and if the caudal ends of the müllerian ducts do not regress. Development of the external genitalia and the role of fetal androgens are discussed. Malformations of the external genitalia are classified pathogenetically as steroid-independent and steroid-dependent. Definitions for steroid-dependent…

Cell death serves as a single etiological cause of a wide spectrum of congenital urinary tract defects.

PURPOSE: We genetically disrupted the wolffian duct in mice to study the affected organogenesis processes and to test the hypothesis that cell loss can be the developmental basis for a wide spectrum of congenital anomalies in the kidney and urinary tract. MATERIALS AND METHODS: We used Hoxb7-Cre transgenic lines (HC1 and HC2) to induce diphtheria toxin production from a ROSA(DTA) allele, disrupting the wolffian duct and derivatives. RESULTS: The first set of mutants (HC1;ROSA(DTA/+)) exhibited agenesis of the kidney, ureter and reproductive tracts. The second set of mutants (HC2;ROSA(DTA/+))

Essential roles of inhibin beta A in mouse epididymal coiling.

Testis-derived testosterone has been recognized as the key factor for morphogenesis of the Wolffian duct, the precursor of several male reproductive tract structures. Evidence supports that testosterone is required for the maintenance of the Wolffian duct via its action on the mesenchyme. However, it remains uncertain how testosterone alone is able to facilitate formation of regionally specific structures such as the epididymis, vas deferens, and seminal vesicle from a straight Wolffian duct. In this study, we identified inhibin beta A (or Inhba) as a regional paracrine factor in mouse mesonep

Sexually dimorphic development of the mammalian reproductive tract requires Wnt-7a.

An important feature of mammalian development is the generation of sexually dimorphic reproductive tracts from the Müllerian and Wolffian ducts. In females, Müllerian ducts develop into the oviduct, uterus, cervix and upper vagina, whereas Wolffian ducts regress. In males, testosterone promotes differentiation of Wolffian ducts into the epididymis, vas deferens and seminal vesicle. The Sertoli cells of the testes produce Müllerian-inhibiting substance, which stimulates Müllerian duct regression in males. The receptor for Müllerian-inhibiting substance is expressed by mesenchymal cells underlyi…

Bilateral isolated Epididymal Agenesis in a 32 year old man.

Epididymal agenesis is defined as the absence of the epididymis totally or segmentally, unilateral or bilateral, which is secondary to the Wolffian duct malformation (1). Rete testis, epididymis, vas deferens and seminal vesicle are believed to develop from Wolffian ducts.

Using mouse models to understand normal and abnormal urogenital tract development.

Removal of toxic substances from the blood depends on patent connections between the kidneys, ureters and bladder that are established when the ureter is transposed from its original insertion site in the Wolffian duct, to the bladder, its final insertion site. The Ureteral Bud Theory of Mackie and Stephens suggests that repositioning of the ureter orifice occurs as the trigone forms from the common nephric duct (CND), the caudal-most Wolffian duct segment. According to this model, insertion of the CND into the bladder and its expansion into the trigone both repositions the ureter in the bladd

Post-testicular change in the reptile sperm surface with particular reference to the snake, Natrix fasciata.

Sperm surface changes occurring in the reptile Wolffian duct have been explored with particular references to the snake, Natrix fasciata. In the snake Wolffian duct there are several proteins not present in serum, the pattern of which changes in concert with the seasonal testicular cycle. Whereas testicular spermatozoa did not bind antibody to duct secretions, all Wolffian duct spermatozoa did so over both head and tail, according to immunofluorescence patterns. Thus, on entering the Wolffian duct, the entire surface of N. fasciata spermatozoa acquires one of more of the duct's secretory compo

Ultrastructural study of a female adnexal tumor of probable wolffian origin.

A broad-ligament tumor, of probable wolffian origin, is presented. The ultrastructural features of this histologically benign tumor have not been previously described. The fine structure of this tumor is compared with normal structures and tumors derived from wolffian and müllerian anlage. The findings are consistent with derivation from wolffian duct.

Wolffian Tumor of the Fallopian Tube Mimicking Malignant Adnexal Tumor: A Case Report and Literature Review.

Wolffian tumors are rare neoplasms that arise from remnants of the Wolffian duct, also known as female adnexal tumors of probable Wolffian origin. Owing to their rarity, they are frequently misdiagnosed as malignant adnexal tumors. We present the case of a 61-year-old woman with a Wolffian tumor initially misdiagnosed as malignant tubal cancer based on atypical imaging findings. Diagnostic evaluations included ultrasonography, CT, and MRI.

[The role of MRI in congenital cystic lesions in the pelvis: a case of uterus didelphys with double vagina, hematocolpos, and ipsilateral renal agenesis.].

We report a case of uterus didelphys with blind hemivagina and ipsilateral renal agenesis. This is an uncommon anomaly of the development of the Mullerian ducts in which a defect in one of the Wolffian ducts leads to failed induction in kidney formation and in the fusion of the Mullerian ducts. MRI plays a decisive role in the diagnosis and characterization of the malformation.
