Action of diethylcarbamazine in vitro on infective larvae of Wuchereria bancrofti.

Infective larvae of Wuchereia bancrofti were obtained from laboratory infected Culex fatigans and maintained in serum containing diethylcarbamazine. There was no difference in the survival time of these larvae and larvae maintained in control serum without diethylcarbamazine. In one of the three experiments reported the survival time of the larvae was much longer than in the other two experiments. It is suggested that this may have been due to the presence of filarial antibodies which killed the larvae in two of the sera used. In the serum which did not contain antibodies, the larvae survived

Ultrastructural analysis of Wuchereria bancrofti (Nematoda: Filarioidea) body wall.

Bancroftian filariasis constitutes the principal mosquito-borne nematode infection of humans and the surface of adult of Wuchereria bancrofti seems to be especially important in the intricate interplay between host and parasite. The study of the parasite's surface structure might help to understand the localization and function of various organelles. W. bancrofti adult worms were recovered from untreated patients during hydrocele repair surgery and studied by transmission electron microscopy. The body wall of adult parasite is composed of cuticle, hypodermis and muscular layer. Cuticle is the

[Epidemiological study on lymphatic filariasis (Wuchereria bancrofti) in Sénégal].

The purpose of this descriptive study conducted in an area endemic for lymphatic filariasis was to update information on the prevalence, clinical aspects, and awareness of this parasitic disease. All consenting inhabitants over 10 years old in three selected sectors were included. An entomological investigation was carried out to estimate the rate of vector infection. Out of a total of 3.359 subjects examined, 57.3% reported familiarity with the disease and provided an accurate description. The prevalence of clinical manifestations, i.e., mainly adenopathy, attributable to the disease was 14.1

Red blood cell antioxidant levels in Wuchereria bancrofti infection.

The elimination of microfilariae of Wuchereria bancrofti is probably mediated by free radicals. Red cell catalase (C), glutathione peroxidase (GPX), and superoxide dismutase (SOD) activity levels were measured as an indirect method of assessing blood oxidant status in 29 asymptomatic microfilaraemics, 29 "endemic normals", and 29 controls living in a non-endemic area. Changes in the activity of these enzymes were also compared over a one month period in 22 asymptomatic microfilaraemics randomised to receive either single dose or 14 day treatment with diethyl carbamazine citrate (DEC). Red cell

Effectiveness of annual ivermectin treatment for Wuchereria bancrofti infection.

Using estimates for the anthelmintic efficacy of a single dose of ivermectin in the treatment of lymphatic filariasis patients, Anton Plaisier, Wilma Stolk, Gerrit van Oortmarssen and Dik Habbema here present and discuss model predictions of the impact of a five-year programme of annual community treatment on the intensity of infection. They show that the effectiveness of such programmes in terms of reductions in the microfilarial density depends critically on the treatment coverage and the pattern of attendance at repeated mass administrations. Improving these factors will possibly be more im

Periodicity of Wuchereria bancrofti microfilariae in south western Ethiopia.

As part of a clinical and epidemiological study of lymphatic filariasis in southwestern Ethiopia the microfilarial density in the blood of two male and two female volunteers from the village of Ketch (near the town of Gambella) was determined every four hours for a 24-hour period using the counting chamber technique. In the blood of the volunteers the majority of the microfilariae appeared between 20 and 04 hours with peak at 24 hours (range at peak time + 3060-3560 mf/ml blood) depicting a nocturnal periodicity of circulating Wuchereria bancrofti microfilariae. This has important implications

Scanning electron microscopy of adult Wuchereria bancrofti (Nematoda: Filarioidea).

Despite the great importance of Wuchereria bancrofti in causing human lymphatic filariasis, only conventional morphological studies have been completed with adult forms of this filaria. No ultrastructural studies have been carried out, mainly due to the difficulty in obtaining viable parasites from human tissues and the lack of a suitable experimental model or in vitro cultivation. With the recent success in using ultrasound to localize adult worms in living tissues and their surgical recovery from human lymphatic vessels, we show in the present paper the first ultrastructural observations of

[Wuchereria bancrofti and asthmatic pulmonary signs in Upper Egypt].

A biological and clinical survey was conducted in 25 patients with bancroftian filariasis, consulting the El Nikhela Health Centre, Assiout Governorate, Upper Egypt. Eleven patients showed asthmatic bronchitis. According to parasitological and serological data (specific antibodies level, total IgE), those clinical manifestations cannot be considered as a part of the "eosinophilic lung" syndrome. In this area, where Wuchereria bancrofti has been recently introduced and where there is an increasing degree of infection, bancroftian filariasis seems to predispose to allergic manifestations.

Human Migration and the Spread of the Nematode Parasite Wuchereria bancrofti.

The human disease lymphatic filariasis causes the debilitating effects of elephantiasis and hydrocele. Lymphatic filariasis currently affects the lives of 90 million people in 52 countries. There are three nematodes that cause lymphatic filariasis, Brugia malayi, Brugia timori, and Wuchereria bancrofti, but 90% of all cases of lymphatic filariasis are caused solely by W. bancrofti (Wb). Here we use population genomics to reconstruct the probable route and timing of migration of Wb strains that currently infect Africa, Haiti, and Papua New Guinea (PNG). We used selective whole genome amplificat

Plasmodium knowlesi and Wuchereria bancrofti: Their Vectors and Challenges for the Future.

Malaria and filariasis still continue to pose public health problems in developing countries of the tropics. Although plans are in progress for the elimination of both these parasitic vector borne diseases, we are now faced with a daunting challenge as we have a fifth species, Plasmodium knowlesi a simian malaria parasite affecting humans. Similarly in peninsular Malaysia, filariasis was mainly due to Brugia malayi. However, we now see cases of Wuchereria bancrofti in immigrant workers coming into the country. In order to successfully eliminate both these diseases we need to know the vectors i

Assessment of the efficacy of diethylcarbamazine on adult Wuchereria bancrofti in vivo.

To assess directly the effect of various doses of diethylcarbamazine (DEC) on adult Wuchereria bancrofti, 31 infected men were randomly assigned to receive an initial single DEC dose of 1 mg/kg (n = 7), 6 mg/kg (n = 10), or 12 mg/kg (n = 14). Beginning 7 d later, the dosage of DEC and duration of treatment were progressively increased for 7-10 weeks. Physical examinations were performed to detect scrotal nodules and the scrotal area was examined by ultrasound (7.5 MHz transducer) to monitor the 'filaria dance sign' (FDS), the characteristic pattern of adult worm movement. Of 53 adult worm 'nes

A case of probable transplacental transmission of Wuchereria bancrofti microfilariae.

A case of transplacental transmission of Wuchereria bancrofti microfilariae is reported in endemic are of Bauchi State, Nigeria. The diagnosis was based on the history of illness, clinical symptoms, age of the child and presence of sheathed microfilariae of nocturnal periodic form of W. bancrofti in the peripheral blood films. Also the night blood sample collected from the child's mother revealed mf of W. bancrofti in addition to the unilateral elephantiasis of the hindlimb. The implication of this observation is stressed.

Circulating Cell-Free Nucleic Acid Detection for Wuchereria bancrofti Infections.

Human lymphatic filariasis (LF) is primarily caused by the helminth parasites Wuchereria bancrofti (Wb) and Brugia malayi (Bm). Detecting parasitic circulating cell-free nucleic acids (ccfDNA/ccfRNA) in plasma is a promising approach for detection of active infections. Plasma RNA sequencing analyses from individuals with active Wb infection and uninfected controls identified 6 RNA targets that were specific to Wb and/or Bm. Assays developed to detect either ccfDNA and ccfRNA were successfully used in Wb-infected individuals, but DNA-based assays targeting WbTR1 were found to be the most sensit

Structural, molecular, functional and immunological characterization of Wuchereria bancrofti-galectin.

Galectins regulate growth and differentiation of immune cells and inflammation through their carbohydrate-binding function in humans, while also play a role in parasite survival. This study focused on the galectin of lymphatic filarial parasite Wuchereria bancrofti (Wb-Gal). The multiple sequence alignment with other galectins showed that the Wb-Gal belonged to galactoside binding lectin family, particularly tandem repeat type galectin-9. A homology model of Wb-Gal was developed in the I-TASser server using high similarity 3D structures with a quality score of 89.5. Molecular docking and dynam

Periodicity of Wuchereria bancrofti var. pacifica filariasis in French Polynesia.

In 1992, a study on microfilaremia periodicity was carried out on 12 Wuchereria bancrofti carriers in the Marquesas islands. Blood samples were collected simultaneously every 4 hours during a 48 hour period by finger-prick and venipuncture for determination of microfilaremia by both blood film and membrane filtration technique methods, and for determination of antigenemia. The membrane filtration results showed no significant nycthemeral variations between the microfilaria densities at hours 16:00, 20:00, 24:00, 04:00, 08:00 and 12:00.
