Isolation and characterization of Shouchella clausii MJ1G0B, a novel halotolerant bacterium from Madura salt ponds: a putative Valenciaxanthin-like xanthophyll with antioxidant and antibacterial activities.

Shouchella clausii MJ1G0B (GenBank: MW308498.1), a halotolerant bacterium isolated from Madura salt ponds, East Java, Indonesia, was characterized through integrated phenotypic, molecular, pigment, spectroscopic, and bioactivity analyses. The Gram-positive isolate grew at 0-15% NaCl (optimum 0% NaCl), with distinct temporal windows for carotenoid accumulation (84 h) and antibacterial secondary metabolite production (120 h). 16S rRNA sequencing (1,418 bp) confirmed 99.42% identity with S. clausii and 99.27% with S. rhizosphaerae. The dominant yellow-orange xanthophyll (Spot 3; Rf 0.98), purifie

Aflatoxin B1-induced toxicity, oxido-inflammatory damage, and apoptosis in male rat reproductive circuitry were abrogated by co-treating with the xanthophyll-lutein and zeaxanthin.

Reproductive dysfunction is a recognised adverse effect of exposure to aflatoxin B1 (AFB1) in humans and animals. Despite the widely acknowledged health risks, exposure to AFB1 remains unavoidable. Conversely, lutein (LUT) and zeaxanthin (ZEA) are plants' potent antioxidants and anti-inflammatory agents, exhibiting promising potential for modulating inflammatory and apoptotic signalling pathways. This study aimed to investigate the effects of co-treatment with LUT/ZEA on reproductive function in rats intoxicated with AFB1. The study utilised male Wistar rats (n = 20 and n = 4 per cohort). A mutation in the promoter of the chicken β,β-carotene 15,15'-monooxygenase 1 gene alters xanthophyll metabolism through a selective effect on its mRNA abundance in the breast muscle.

A polymorphism in the promoter of the β,β-carotene 15,15'-monooxygenase 1 (BCMO1) gene recently was identified in an experimental cross between 2 chicken lines divergently selected on growth rate and found to be associated with variations in the yellow color of the breast meat. In this study, the effects of the polymorphism on several aspects of carotenoid metabolism were evaluated in chickens sharing the same genetic background except for their genotype at the BCMO1 locus. We confirmed that BCMO1 mRNA abundance varied (P < 0.001) between the 2 homozygous genotypes (GG << AA) and in the pector

More Than Pigments: The Potential of Astaxanthin and Bacterioruberin-Based Nanomedicines.

Carotenoids are natural products regulated by the food sector, currently used as feed dyes and as antioxidants in dietary supplements and composing functional foods for human consumption. Of the nearly one thousand carotenoids described to date, only retinoids, derived from beta carotene, have the status of a drug and are regulated by the pharmaceutical sector. In this review, we address a novel field: the transformation of xanthophylls, particularly the highly marketed astaxanthin and the practically unknown bacterioruberin, in therapeutic agents by altering their pharmacokinetics, biodistrib

Carotenoid transformations underlying the blue absorbance change in flashed leaves during the induction of oxygen evolution.

The blue absorbance change occurring in flashed bean (Phaseolus vulgaris L.) leaves when exposed to continuous light (first observed by Strasser; Strasser, R.J. (1973) Arch. Int. Physiol. Biochem. 81, 935--955) is caused by the conversion of the following xanthophylls: violaxanthine leads to antheraxanthine leads to zeaxanthine. This conclusion is derived from the simultaneous occurrence of both reactions: (a) In flashed leaves, blue absorbance change and xanthophyll conversion take place under strong (2 mW . cm-2) but not under weak (0.02 mW . cm-2) white light. (b) In chloroplasts isolated f

Lutein, Zeaxanthin, and meso-Zeaxanthin in the Clinical Management of Eye Disease.

Lutein, zeaxanthin, and meso-zeaxanthin are xanthophyll carotenoids found within the retina and throughout the visual system. The retina is one of the most metabolically active tissues in the body. The highest concentration of xanthophylls is found within the retina, and this selective presence has generated many theories regarding their role in supporting retinal function. Subsequently, the effect of xanthophylls in the prevention and treatment of various eye diseases has been examined through epidemiological studies, animal studies, and clinical trials. This paper attempts to review the epid

The role of silver ions in the regulation of the senescence process in Triticum aestivum.

The control of senescence has economic importance due to its effects on parameters such as herbal product quality and shelf life. This study is on the control of induced senescence in Triticum aestivum L. 'Gün-91' plants with silver nitrate (AgNO3) treatments. It was observed that some changes that occurred with dark and indole-1-acetic acid (IAA) treatments could be reduced with AgNO3 treatments. After dark-induced senescence, it was observed in plants that seedling length, relative water content (RWC), chlorophyll, β-carotene, xanthophylls, total antioxidant capacity, soluble phenol, total s

Carotenoid, tocopherol, and retinol concentrations in elderly human brain.

BACKGROUND: Antioxidants, such as tocopherols and carotenoids, have been implicated in the prevention of degenerative diseases. Although correlations have been made between diseases and tissue levels of antioxidants, to date there are no reports of individual carotenoid concentrations in human brain. OBJECTIVE: To measure the major carotenoids, tocopherols, and retinol in frontal and occipital regions of human brain. DESIGN: Ten samples of brain tissue from frontal lobe cortex and occipital cortex of five cadavers were examined. Sections were dissected into gray and white matter, extracted w

Impact of antioxidants fortified eggs consumption on eye health in individuals with mild to moderate untreated eczema: a randomized controlled trial.

PURPOSE: Eczema is a systemic condition associated with an increased risk of eye diseases. Xanthophylls and other antioxidants are eye-beneficial, and eggs are an excellent source of xanthophylls, often fortified to enhance their benefits. However, the effects of antioxidant-rich egg consumption on eye health in individuals with eczema have not been clinically validated. This study examined the effects of consuming regular eggs and antioxidant-fortified eggs on eye health in individuals with mild to moderate eczema via a 12 weeks, double-blind, randomized controlled trial. METHODS: Forty adult

Expressing CaCCS encoding capsanthin/capsorubin synthase in tomato increases fruit provitamin A content and yield.

We developed a tomato (Solanum lycopersicum) biofortification strategy by engineering the simultaneous accumulation of β-carotene (provitamin A) and the pepper (Capsicum annuum)-specific ketocarotenoids capsanthin and capsorubin. Capsanthin/capsorubin synthase (CaCCS), an enzyme related to lycopene β-cyclase (LCYB) known for β-carotene biosynthesis, was investigated for its in vivo role in ripening pepper fruit (C. annuum). Silencing CaCCS by virus-induced gene silencing reduced flux from lycopene to β-carotene (the β-branch), altering carotenoid profiles and expression of carotenogenic genes.

Mutant combinations of lycopene ɛ-cyclase and β-carotene hydroxylase 2 homoeologs increased β-carotene accumulation in endosperm of tetraploid wheat (Triticum turgidum L.) grains.

Grains of tetraploid wheat (Triticum turgidum L.) mainly accumulate the non-provitamin A carotenoid lutein-with low natural variation in provitamin A β-carotene in wheat accessions necessitating alternative strategies for provitamin A biofortification. Lycopene ɛ-cyclase (LCYe) and β-carotene hydroxylase (HYD) function in diverting carbons from β-carotene to lutein biosynthesis and catalyzing the turnover of β-carotene to xanthophylls, respectively. However, the contribution of LCYe and HYD gene homoeologs to carotenoid metabolism and how they can be manipulated to increase β-carotene in tetra

Photoinhibition and Photoprotective Responses of a Brown Marine Macroalga Acclimated to Different Light and Nutrient Regimes.

Plants and brown algae avoid photoinhibition (decline in photosystem II efficiency, Fv/Fm) caused by excess light energy and oxidative stress through several photoprotective mechanisms, such as antioxidant xanthophyll production and heat dissipation. The heat dissipation can be measured as non-photochemical quenching (NPQ) and is strongly driven by de-epoxidation of xanthophyll cycle pigments (XCP). Although NPQ is known to increase under high light acclimation and nutrient-deficient conditions, a few studies have investigated the combined effects of the conditions on both NPQ and associated x

Quick conversions and de novo synthesis within the entire α- and β-carotenoid branches during non-steady-state light transients.

Understanding the rapid adjustments of plants to high-light exposure remains challenging, as multiple excitation and de-excitation pathways are simultaneously activated. In this study, we examined carotenoid pigment conversions at the second-scale in three tree species in parallel with high temporal resolution (<1 s) in vivo fluorescence and absorption spectroscopy. Our results reveal that both β-branch (violaxanthin, antheraxanthin, zeaxanthin) and α-branch (lutein, lutein epoxide) xanthophylls exhibit remarkably fast and oscillating pool dynamics within the first 20 seconds of illumination,

Spectrometric investigations in ocular hypertension and early stages of primary open angle glaucoma and of low tension glaucoma--multisubstance analysis.

The approximation of logarithmic difference spectra between the reflectance of the normal fundus and the fundus reflectance in different stages of glaucoma is demonstrated by a model. The influences of fundus pigments like oxihemoglobin, melanin, xanthophyll and rhodopsin as well as the intensity and the exponent of the scattered light are optimized. Glaucomatous alterations in the extinction of these pigments and of the scattering parameters are different in the macula, in the papillo-macular bundle and in the parapapillary region temporal to the optic disc. Carotenoid Extraction from Plant Tissues.

Carotenoids are the natural pigments available in nature and exhibit different colors such as yellow, red, and orange. These are a class of phytonutrients that have anti-cancer, anti-inflammatory, anti-oxidant, immune-modulatory, and anti-aging properties. These were used in food, pharmaceutical, nutraceutical, and cosmetic industries. They are divided into two classes: carotenes and xanthophylls. The carotenes are non-oxygenated derivatives and xanthophylls are oxygenated derivatives. The major source of carotenoids are vegetables, fruits, and tissues. Carotenoids also perform the roles of ph

A Mechanistic Review of β-Carotene, Lutein, and Zeaxanthin in Eye Health and Disease.

Carotenoids are natural lipid-soluble antioxidants abundantly found as colorful pigments in fruits and vegetables. At least 600 carotenoids occur naturally, although about 20 of them, including β-carotene, α-carotene, lycopene, lutein, zeaxanthin, meso-zeaxanthin, and cryptoxanthin, are detectable in the human blood. They have distinct physiological and pathophysiological functions ranging from fetal development to adult homeostasis. β-carotene is a precursor of vitamin A that essentially functions in many biological processes including vision. The human macula lutea and eye lens are rich in l

Effects of the Macular Carotenoid Lutein in Human Retinal Pigment Epithelial Cells.

Retinal pigment epithelial (RPE) cells are central to retinal health and homoeostasis. Oxidative stress-induced damage to the RPE occurs as part of the pathogenesis of age-related macular degeneration and neovascular retinopathies (e.g., retinopathy of prematurity, diabetic retinopathy). The xanthophyll carotenoids, lutein and zeaxanthin, are selectively taken up by the RPE, preferentially accumulated in the human macula, and transferred to photoreceptors. These macular xanthophylls protect the macula (and the broader retina) via their antioxidant and photo-protective activities. This study wa

Photoprotection mechanism in the 'Fuji' apple peel at different levels of photooxidative sunburn.

The xanthophyll cycle, flavonoid metabolism, the antioxidant system and the production of active oxygen species were analyzed in the peel of 'Fuji' apples re-exposed to sunlight after extended periods of fruit bagging treatment, resulting in different levels of photooxidative sunburn. After re-exposing bagged fruits to sunlight, the production of active oxygen species and the photoprotective capacity in apple peels were both significantly enhanced. As sunburn severity increased, the concentration of hydrogen peroxide increased, while xanthophyll cycle pool size decreased. For the key genes inv
