Evidence of intermolecular associations of β-glucan and high-molar mass xylan in a hot water extract of raw oat groat.

High molar mass (HM) β-glucan present in oat products can reduce the risk of diet-related diseases, mainly owing to significantly increased digesta viscosity in human small intestine. To verify a research hypothesis that arabinoxylan (AX) present in oat water extract is associated with HM-β-glucan and thus may influence its functionality, multi-detection HPSEC coupled with enzymatic hydrolysis, sugar and 1H NMR analyses were performed. Isolated cell wall polysaccharide fraction comprised branched AX (arabinose-to-xylose ratio, Ara/Xyl ~ 0.8) and arabinogalactan-protein (AG-P). Nevertheless, it

Comparison of synthetic and post-synthetic molecular genetic methods of reducing xylan glucuronidation in hybrid aspen for biorefinery applications.

BACKGROUND: The pattern of xylan backbone substitution by 4-O-methylglucuronic acid (mGlcA) and O-acetyl groups generated during biosynthesis in the Golgi apparatus plays a key role in molecular interactions between secondary cell wall polymers in hardwoods, thus regulating biorefinery properties of woody biomass. This study aims at comparing the effects of two different approaches to reduce xylan glucuronidation, targeting either xylan biosynthesis by RNAi suppression of GLUCURONIC SUBSTITUTION OF XYLAN1 genes (denoted Gux1RNAi) or by xylan post-synthetic modification in the secondary cell wa

Acrylic acid-based hydrogel with multi-properties endowed by synergistic multifunction of glucurono-dialdehyde xylan and silver nanoparticles.

Conductive hydrogels frequently struggle to balance high conductivity, robust mechanical strength, excellent UV resistance, and antibacterial properties, a limitation that restricts their applications in intelligent wearable sensors. In this work, a comprehensive strategy was developed to fabricate a semi-interpenetrating polymer network (semi-IPN) composite hydrogel. This hydrogel consists of a polyacrylic acid (PAA) network interpenetrated by linear glucurono-dialdehyde xylan (GlcU-DAX) via hydrogen-bonding, reinforced by the synergistic multifunctional effects of GlcU-DAX and silver nanopar

Loss of UBIQUITIN-PROTEIN LIGASE 3 (UPL3) impairs cellulose-xylan structural organization in Arabidopsis seed mucilage.

Arabidopsis thaliana seeds release large capsules of mucilaginous polysaccharides, whose adherence depends not only cellulose and hemicellulose biosynthesis but also their interactions. However, the key proteins regulating the connection within this polysaccharide network in the seed coat mucilage remain unclear and require further investigation. Through reverse genetics, we demonstrate that UBIQUITIN-PROTEIN LIGASE 3 (UPL3), a nucleus-localized, HECT E3 ligase protein, is required for the proper organization of cellulosic rays in seed coat mucilage. Disruption of UPL3 results in an enlarged s

Anti-solvent precipitation of saturated steam autohydrolyzed wheat straw xylan - Regression model-based intensification for biorefineries.

The precipitation of water-soluble carbohydrates using organic solvents offers considerable potential, as separation and purification can be achieved in a single step. Therefore, ethanol precipitation of potential high-value xylan - the main component of wheat straw hemicellulose - is investigated and optimized. Pelletized wheat straw, prepared without chemical or biotechnological auxiliaries, is autohydrolyzed using saturated steam. The pretreatment is designed to effectively solubilize hemicellulose, resulting in a hydrolysate low in impurities that is concentrated and precipitated without f

Engineering a Carbonyl Reductase for High-Efficiency Synthesis of Optically Pure (S)-Pro-Xylane: An Alternative Synthetic Route.

Pro-Xylane is a biologically active C-glycoside compound in cosmetic products. The conventional method involves converting d-xylose from beech trees to 1-C-(β-d-xylopyranosyl)-acetone and reducing it with sodium borohydride. This method produced borate salts as byproducts, requiring a tedious removal process. In addition, (S)-Pro-Xylane shows superior biological activity compared to (R)-Pro-Xylane. Hence, with focus on the second step of the reduction reaction, the carbonyl reductase (CR) from Candida orthopsilosis Co 90-125 (CoCR13) was engineered in this study. Variant R129E/D210F showed a 2

Effects of Resistant Starch Infusion, Solely and Mixed with Xylan or Cellulose, on Gut Microbiota Composition in Ileum-Cannulated Pigs.

Fermentation of dietary fiber (DF) is beneficial for gut health, but its prebiotic effects are often impeded in the distal large intestine because of the fast degradation of fermentable substrates. One way to enhance the prebiotic effect of DF is to deliver fibers to the lower parts of the gut, which can be achieved by mixing different kinds of fiber. Therefore, in the present study, an ileum-cannulated pig model was employed to investigate the fermentation influence in the large intestine by infusing resistant starch solely (RS, fast fermentable fiber) and mixing with other fibers (xylan or c

Engineering a semi-interpenetrating constructed xylan-based hydrogel with superior compressive strength, resilience, and creep recovery abilities.

Recent advances in the area of hydrogel synthesis have been directed to enhance the mechanical properties and biocompatibility, which are critical in their use as functional biomaterials. In this work, a green and facile method is introduced to produce a hydrogel based on xylan, a plant-based heteropolysaccharide, that is shown to successfully form hydrogen-bonded, semi-interpenetrating polymer networks with polyvinyl alcohol. Upon crosslinking with sodium trimetaphosphate, the obtained hydrogels achieved an exceptional compressive strength (up to 84.2 MPa at a fracture strain of 90 %), which

Exploring the Reaction Mechanisms of Fast Pyrolysis of Xylan Model Compounds via Tandem Mass Spectrometry and Quantum Chemical Calculations.

A commercial fast pyrolysis probe coupled with a high-resolution tandem mass spectrometer was employed to identify the initial reactions and products of fast pyrolysis of xylobiose and xylotriose, model compounds of xylans. Fragmentation of the reducing end by loss of an ethenediol molecule via ring-opening and retro-aldol condensation was found to be the dominant pyrolysis pathway for xylobiose, and the structure of the product-β-d-xylopyranosylglyceraldehyde-was identified by comparing collision-activated dissociation of the ionized product and an ionized authentic compound. Xylan acetate ester ameliorates ulcerative colitis through intestinal barrier repair and inflammation inhibition via regulation of macrophage M1 polarization.

Ulcerative colitis (UC) is a chronic inflammatory disease resulting from abnormal immune response to gut microflora translocating through damaged intestinal barrier. Xylan acetate ester (XylA) can increase colon short-chain fatty acids (SCFAs) levels and alleviate kidney disease by inhibiting inflammation through the G protein-coupled receptor pathway. Here, we synthesized and purified XylA, and then the effects and mechanisms of XylA on dextran sodium sulfate-induced UC in mice were investigated. The results showed that in mice, similar to the positive drug 5-aminosalicylic acid, oral adminis

Efficient conversion of xylan to l-arabinose by multi-enzymatic cascade reaction including d-xylulose 4-epimerase as a new stereoselectivity-exchange enzyme.

l-Arabinose has been produced by hydrolyzing arabinan, a component of hemicellulose. However, l-arabinose has limitations in industrial applications owing to its relatively high cost. Here, d-xylulose 4-epimerase as a new-type enzyme was developed from d-tagaturonate 3-epimerase from Thermotoga petrophila using structure-guided enzyme engineering. d-Xylulose 4-epimerase, which epimerized d-xylulose to l-ribulose, d-xylulokinase and sugar phosphatase, which overcame the equilibrium of d-xylose isomerase, were included to establish a new efficient conversion pathway from d-xylose to l-arabinose.

Multi-copy expression of a protease-resistant xylanase with high xylan degradation ability and its application in broilers fed wheat-based diets.

The acidic thermostable xylanase (AT-xynA) has great potential in the feed industry, but its low activity is not conductive to large-scale production, and its application in poultry diets still needs to be further evaluated. In Experiment1, AT-xynA activity increased 3.10 times by constructing multi-copy strains, and the highest activity reached 10,018.29 ± 91.18 U/mL. AT-xynA showed protease resistance, high specificity for xylan substrates, xylobiose and xylotriose were the main hydrolysates. In Experiment2, 192 broilers were assigned into 3 treatments including a wheat-based diet, and the d

Ferrihydrite Addition Activated Geobacteraceae, the Most Abundant Iron-reducing Diazotrophs, and Suppressed Methanogenesis by Heterogeneous Methanogens in Xylan-amended Paddy Soil Microcosms.

Paddy fields are a major emission source of the greenhouse gas methane. In the present study, the addition of ferrihydrite to xylan-amended paddy soil microcosms suppressed methane emissions. PCR-based and metatranscriptomic ana-lyses revealed that the addition of ferrihydrite suppressed methanogenesis by heterogeneous methanogens and simultaneously activated Geobacteraceae, the most abundant iron-reducing diazotrophs. Geobacteraceae may preferentially metabolize xylan and/or xylan-derived carbon compounds that are utilized by methanogens. Geomonas terrae R111 utilized xylan as a growth substr

A family IIb xylan-binding domain has a similar secondary structure to a homologous family IIa cellulose-binding domain but different ligand specificity.

BACKGROUND: Many enzymes that digest polysaccharides contain separate polysaccharide-binding domains. Structures have been previously determined for a number of cellulose-binding domains (CBDs) from cellulases. RESULTS: The family IIb xylan-binding domain 1 (XBD1) from Cellulomonas fimi xylanase D is shown to bind xylan but not cellulose. Its structure is similar to that of the homologous family IIa CBD from C. fimi Cex, consisting of two four-stranded beta sheets that form a twisted 'beta sandwich'. The xylan-binding site is a groove made from two tryptophan residues that stack against the f
