Yohimbine treatment of sexual side effects induced by serotonin reuptake blockers.

BACKGROUND: Preclinical and clinical studies suggest that yohimbine facilitates sexual behavior and may be helpful in the treatment of male impotence. A single case report suggests that yohimbine may be used to treat the sexual side effects of clomipramine. This study evaluated yohimbine as a treatment for the sexual side effects caused by serotonin reuptake blockers. METHOD: Six patients with either obsessive compulsive disorder, trichotillomania, anxiety, or affective disorders who suffered sexual side effects after treatment with serotonin reuptake blockers were given yohimbine on a p.r.n.

Effect of diazepam and yohimbine on neuronal activity in sham and hemiparkinsonian rats.

The prefrontal cortex and the amygdala are critical for the emotional guidance of behavior and are believed to be a site of action for many anxiolytics and anxiogenics. Despite extensive studies examining how these drugs affect behavior, there is little information regarding their effects on neuronal activity. Additionally, with recent recognition of anxiety as a non-motor symptom of Parkinson's disease, it is unknown if activity in the cortex and the amygdala is altered. Previously, we reported that hemiparkinsonian rats had higher baseline anxiety-like behavior and diminished responsiveness

Effects of tryptophan depletion on responses to yohimbine in healthy human subjects.

There is considerable evidence that both the norepinephrine (NE) and serotonin (5-HT) systems are involved in the regulation of human anxiety and fear responses. To assess the modulating effects of central 5-HT levels on NE function, 11 healthy human subjects were studied with placebo-controlled challenge tests involving tryptophan depletion followed by administration of the alpha-2-adrenergic antagonist yohimbine 0.4 mg/kg IV. Five of the 11 subjects reported a marked increase in feelings of nervousness (> or = 25 mm on a 100 mm analog scale) following the combination test, while 1/11 had thi

Antagonism of medetomidine-induced ataxia in rats by yohimbine, aminophylline and caffeine.

The actions of the alpha 2-antagonist yohimbine and methylxanthines aminophylline and caffeine were evaluated in reversing ataxia, increase in landing foot splay (LFS), produced by the alpha 2-agonist medetomidine in male rats. Medetomidine at 0.1 and 0.15 mg/kg, i.p. increased LFS by 42.9 and 69.6%, respectively. The peripherally acting alpha 2-agonist ST91 (0.125 to 0.5 mg/kg, i.p.) did not significantly affect the LFS. Intraperitoneal injection of yohimbine at 0.5 and 1 mg/kg, aminophylline at 25, 50 and 100 mg/kg, and caffeine at 25 and 50 mg/kg significantly antagonized medetomidine (0.15

Interaction of yohimbine with batrachotoxinin binding to mouse brain sodium channels.

To study the local anesthetic properties of yohimbine in more detail, its effect was examined in vitro on the scorpion toxin-enhanced specific binding of [3H]batrachotoxinin A 20-alpha-benzoate [( 3H]BTX-B) to the gating complex in sodium channel preparations from mouse brain cortex. Both equilibrium and kinetic experiments were carried out. Yohimbine inhibited the specific binding of [3H]BTX-B in the vesicular preparation with an IC50 value of 2.2 X 10(-5) M. This is about one order of magnitude higher than the concentration required for antagonism via the alpha 2-adrenoceptors; however, yohi

Xylazine-ketamine-induced anesthesia in rats and its antagonism by yohimbine.

A combination of xylazine and ketamine was used to anesthetize 60 male rats, and then yohimbine was given to evaluate its reversing effect on xylazine-ketamine-induced anesthesia. In experiment A, xylazine (21 mg/kg of body weight) and ketamine (45 mg/kg) were admixed and administered IM to 12 Sprague-Dawley rats. Anesthesia lasted approximately 70 minutes. The xylazine-ketamine combination also induced polyuria, bradycardia, and bradypnea. When yohimbine (2.1 mg/kg) was given intraperitoneally 20 minutes after the xylazine-ketamine injection, the rats regained consciousness and righting refle

Baclofen blocks yohimbine-induced increases in ethanol-reinforced responding in rats.

Chronic or repeated stress increases alcohol consumption. The GABA-B agonist baclofen decreases alcohol consumption and may be most effective for individuals with comorbid anxiety/stress disorders. The present study sought to determine if baclofen blocks stress-induced increases in ethanol self-administration as modeled by repeated yohimbine injections in rats. Rats were trained to respond for 15% w/v ethanol in operant chambers using a method that applies neither water deprivation nor saccharin/sucrose fading. Following training, the rats received 6 injections of 1.25mg/kg yohimbine were give

Yohimbine increases opioid-seeking behavior in heroin-dependent, buprenorphine-maintained individuals.

RATIONALE: In laboratory animals, the biological stressor yohimbine (α(2)-noradrenergic autoreceptor antagonist) promotes drug seeking. Human laboratory studies have demonstrated that psychological stressors can increase drug craving but not that stressors alter drug seeking. OBJECTIVES: This clinical study tested whether yohimbine increases opioid-seeking behavior. METHODS: Ten heroin-dependent, buprenorphine-stabilized (8 mg/day) volunteers sampled two doses of hydromorphone [12 and 24 mg IM in counterbalanced order, labeled drug A (session 1) and drug B (session 2)]. During each of six la

Effects of yohimbine on cerebral blood flow, symptoms, and physiological functions in humans.

OBJECTIVE: Increases in adrenergic activity are associated with stress, anxiety, and other psychiatric, neurological, and medical disorders. To improve understanding of normal CNS adrenergic function, CBF responses to adrenergic stimulation were determined. METHODS: Using PET, the CBF changes after intravenous yohimbine, an alpha2-adrenoreceptor antagonist that produces adrenergic activation, were compared with placebo in nine healthy humans. Heart rate, blood pressure, Paco2, plasma catecholamines, and symptom responses were also determined. RESULTS: Among nonscan variables, yohimbine produ

The anxiogenic agents, yohimbine and FG 7142, disrupt the noradrenergic response to novelty.

Whether or not abnormal noradrenergic transmission can be a causal factor in anxiety is controversial. The present experiments examined this question by comparing the effects of two anxiogenic agents on noradrenaline efflux in the frontal cortex of freely moving rats. A single anxiogenic dose of either yohimbine (2.5 or 5 mg/kg) or FG 7142 (10 or 20 mg/kg) was administered to rats by i.p. injection. Yohimbine increased spontaneous efflux of noradrenaline, but FG 7142 had no effect. However, subsequent exposure of rats to a novel environment increased noradrenaline efflux in vehicle-, but not d

Effects of phentolamine or yohimbine on naloxone's actions during endotoxin shock in rats.

Endogenous opioids are known to mediate some of the cardiovascular sequelae of sepsis. Inhibition of adrenergic action has been implicated as a physiological path by which endogenous opioids cause deleterious changes in cardiovascular function during endotoxin shock, but where and to what extent this accounts for changes in regional vascular resistance remains unclear. In this study, we addressed this question by examining the role of alpha-adrenergic actions in cardiovascular performance and the regional perfusion changes caused by naloxone during endotoxin shock. Rats had catheters inserted

Effects of clonidine and yohimbine on parasympathetic reflex salivation and vasodilatation in cat SMG.

The present experiments were designed to examine the site of action of the alpha 2-adrenoceptor agonist, clonidine, and the alpha 2-antagonist, yohimbine, on the salivary secretion response to parasympathetic and sympathetic stimulation in the cat submandibular gland (SMG). The parasympathetic reflex salivary secretion was significantly affected by both clonidine and yohimbine (clonidine reduced and yohimbine increased the amount of saliva secreted) while the reflex vasodilator response was not affected by either agent. No degree of methacholine-induced salivation was affected by prior treatme

Antagonism of cocaine-induced hepatotoxicity by the alpha adrenergic antagonists phentolamine and yohimbine.

The ability of the alpha adrenoreceptor antagonists phentolamine and yohimbine to antagonize cocaine-induced hepatotoxicity was determined in phenobarbital-induced B6C3/F1 mice. Hepatotoxicity was assessed by the histologic extent of necrosis, incidence of latent lethality and increases in serum alanine aminotransferase activity. The depression of hepatic glutathione levels also were measured. The administration of a single 5-mg/kg dose of phentolamine antagonized the decrease in glutathione levels and the elevation of aminotransferase activity caused by a 60-mg/kg dose of cocaine. Similar res

[Treatment of orthostatic hypotension induced by tricyclic antidepressants with yohimbine (author's transl)].

Following the observation that two patients with depression and essential orthostatic hypotension recovered from the latter affection after yohimbine administration, an open study was conducted to assess the activity of yohimbine in correcting orthostatic hypotension induced by tricyclic antidepressants. Results were conclusive in 10 of the 11 patients treated, and were confirmed in a further 12 cases during a double-blind study. Yohimbine appears to be the most effective treatment for orthostatic hypotension, either essential or induced by thymoanaleptic or neuroleptic therapy.

Yohimbine derivatives containing carboxylic acid and ester bioisosteres as potent ADRA2A antagonists.

The α2A-adrenergic receptor (ADRA2A) has emerged as a promising target for precision medicine in metabolic and vascular disorders. However, the clinical translation of the prototypical antagonist, yohimbine, is hindered by poor subtype selectivity, leading to ADRA1A-mediated cardiovascular adverse events, and rapid, highly variable CYP2D6-mediated hepatic clearance. To overcome these liabilities, we designed and synthesized a novel series of yohimbine analogues by replacing the metabolically labile methyl carboxylate with potentially stable isosteric five-membered heterocycles or a nitrile gro

Respiratory depressant interactions of fentanyl and xylazine and their reversal by naloxone and yohimbine in mice.

BACKGROUND: Opioid misuse remains a critical public health crisis in the U.S., causing over 82,000 opioid-related overdose deaths in 2023. Fentanyl, a potent synthetic opioid acting via μ-opioid receptors (MOR), poses unique challenges for overdose reversal due to its high potency and rapid onset. Increasingly, illicit fentanyl is adulterated with xylazine, an alpha-2 adrenergic receptor agonist (α2AR) used in veterinary medicine. This combination, known as "tranq dope," is associated with polydrug dependence and enhanced respiratory depression that may be more resilient to naloxone reversal.
