Estrogen-like endocrine disrupting chemicals affecting puberty in humans--a review.

Estrogen-like endocrine disrupting chemicals (EEDC) are exogenous, man-made chemicals that alter the functions of the endocrine system and cause various health defects by interfering with the synthesis, metabolism, binding or cellular responses of natural estrogens. EEDCs have been found in various plastic products, flame retardants, pesticides and many other products that are needed for daily use. Some of the greatest effects of EEDCs are on puberty, a period of rapid physiological changes like growth spurt, maturation of the gonads and the brain. Estrogen, one of the key hormones required in

Harnessing the value of reproductive hormones in cattle production with considerations to animal welfare and human health.

The human population is ever increasing while the quality and quantity of natural resources used for livestock production decline. This calls for improved product efficiency and the development of improved and sustainable cattle production methods to produce higher quality products to satisfy the demands of both the modern and transient world. The goal of this review was to summarize the interactions, challenges, and opportunities in cattle production relating to their endocrine system, and how reproductive hormones and others impact economically important traits, animal welfare, and human hea

Metabolic fate of anabolic agents in treated animals and residue levels in their meat.

Anabolic agents applied in animal production can be classified as sex hormones, as far as one aspect of their biological activity is concerned. In respect to their chemical nature the agents can be divided into three sub-groups: (a) steroids natural to the body (endogenous steroids); (b) steroids foreign to the body; (c) other compounds foreign to the body. After exogenous application (oral or parenteral) metabolism of compounds in group (a), e.g. 17beta-estradiol or testosterone, follows the pathways for the identical endogenous hormones; i.e. enzymatic transformation of the biologically acti

Study of Competitive Displacement of Curcumin on α-zearalenol Binding to Human Serum Albumin Complex Using Fluorescence Spectroscopy.

α-zearalenol (α-ZOL) is a mycotoxin with a strong estrogen effect that affects the synthesis and secretion of sex hormones and is transported to target organs through human serum albumin (HSA). Additionally, it has been reported that curcumin can also bind to HSA with high affinity at the same binding site as α-ZOL. Additionally, several studies reported that reducing the bound fraction of α-ZOL contributes to speeding up the elimination rate of α-ZOL to reduce its hazard to organs. Therefore, to explore the influence of a nutrition intervention with curcumin on α-ZOL effects, the competitive

Maternal mycoestrogen exposure in relation to timing of birth and placental corticotropin releasing hormone concentrations in a U.S. pregnancy cohort.

Zearalenone (ZEN) is a mycotoxin that widely contaminates human food supplies. In animal models, exposure to ZEN has extensive impacts on reproduction and pregnancy including contributing to early parturition. Here, we translate that research to humans by examining gestational exposure to ZEN in relation to the timing of birth and placental corticotropin releasing hormone (pCRH), a hormone that regulates the timing of parturition. Utilizing data from a pregnancy cohort (UPSIDE; Rochester, NY; n = 299), we investigated exposure to ZEN and its metabolites in relation to: 1) gestational age at bi

Implant sequence effects in intact male Holstein veal calves: live and slaughter traits.

Seven sequences of growth promotant implants were used in special-fed intact male Holstein veal calves (n = 443). Calves received implants 4 d after arrival at the veal barn, 42, and 84. The following implants were used: placebo (0), Z (36 mg zeranol), ET (20 mg estradiol, 200 mg testosterone), EP/2 (10 mg estradiol, 100 mg progesterone), EP (20 mg estradiol, 200 mg progesterone), and EBA (24 mg estradiol, 120 mg trenbolone acetate). The following sequences were compared: 0-0-0 (negative control), 0-ET-ET, Z-ET-ET, 0-EP-EP, Z-EP-EP, 0-EP/2-EBA, and Z-0-EBA. From 0 to 42 d, Z implants increased

Effects of trenbolone acetate on carcass characteristics and serum testosterone and cortisol concentrations in bulls and steers on different management and implant schemes.

The objective of this study was to determine the effects of different implanting schemes on serum testosterone and cortisol concentrations, and carcass traits of bulls and steers implanted with trenbolone acetate (TBA) and zeranol (Z). Twenty Polled Hereford bulls were randomly assigned to one of three treatments after birth. Five calves served as nonimplanted control bulls (NIB). Nine bulls were implanted (IB) with 140 mg of TBA and 36 mg of Z at about 1 mo of age and reimplanted with both compounds 10 wk later. When IB calves were about 21 wk of age, the TBA implant was removed and calves we

Mycoestrogen Exposure during Pregnancy: Impact of the ABCG2 Q141K Variant on Birth and Placental Outcomes.

BACKGROUND: Zearalenone (ZEN) is an estrogenic mycotoxin ("mycoestrogen") that contaminates global grain crops leading to detectable concentrations of ZEN and its metabolites, including the synthetic version α-zearalanol (also called zeranol; ZER), in human populations. Despite in vitro and in vivo animal evidence of endocrine disruption by ZEN, there has been limited investigation in humans. OBJECTIVES: To examine markers of fetal growth following prenatal exposure to ZEN and evaluate the role of the placental efflux transporter BCRP/ABCG2 in protecting against ZEN's potential fetoplacental

The mycotoxin zearalenone and its metabolite α-zearalenol impair anticancer activity of doxorubicin in estrogen receptor-positive breast cancer cells.

Our environment, especially our diet, exposes us to numerous bioactive compounds that have the potential to interact with anticancer therapies. Although understanding these exposome-drug interactions is crucial for optimizing anticancer therapy, limited knowledge exists on the interference of contaminants, especially mycotoxins, with anticancer drugs. One of the most prevalent mycotoxins in food and feed is zearalenone (ZEN), which demonstrated estrogenic activity that is even stronger in its metabolite α-zearalenol (α-ZEL). Consequently, in this study, we investigated whether these mycoestrog

Phytoestrogen α-zearalanol improves vascular function in ovariectomized hyperhomocysteinemic rats.

Previous studies have showed that phytoestrogen α-zearalanol (α-ZAL) could antagonize homocysteine (Hcy) induced endothelin-1 (ET-1) expression, oxidative stress and apoptosis in human umbilical vein endothelial cells in vitro, however, its effect on vascular function in vivo remains to be determined. This study was designed to investigate the effects of α-ZAL on vascular function in ovariectomized (OVX) hyperhomocysteinemia (HHcy) rats and explore the mechanisms involved primarily. HHcy rat model was induced by diets containing 2.5% methionine (Met) for 12 weeks. Forty adult female Wistar rat

General aspects of the effectiveness of anabolic agents in increasing protein production in farm animals, in particular in bull calves.

The role of farm animals as converters of feed protein into protein edible for humans has often been criticized for the lack of efficiency of this process. In a series of 192 N-balance studies with veal calves it was found, however, that these animals convert at an early age even more than 70 percent of the feed protein into body protein. The deterioration of the N-conversion, so undesirable from a point of view of food production, is found when the animals grow older. With increasing age the conversion ratio went down to approximately 40-50 percent in our studies. This could only be explained

Estrogen-induced hepatic toxicity and hepatic cancer: differences between two closely related hamster species.

AIMS/BACKGROUND: Estrogen is known to affect hepatobiliary function; however, it is unusual for high serum levels of estrogen to actually result in clinically detectable hyperbilirubinemia. Women affected by cholestatic jaundice during pregnancy share this genetic susceptibility with two Cricetulus hamsters, the Armenian hamster (Cricetulus migratorius) and the Chinese hamster (Cricetulus griseus). Nevertheless, the pathophysiologic process responsible for this estrogen induced icterus may be different in women and hamsters. The present study compares various facets of estrogen-induced icterus

Potential health risks of foodborne performance-enhancing drugs in competitive sports.

Athletes need to consume a significant amount of energy during prolonged training and in high-intensity competition. It is necessary for them to take nutritional foods that recharge their bodies. However, in sporting events of recent years, both domestic and international, many positive drug tests are found to be caused by the ingestion of foods that contain performance-enhancing drugs (PEDs). As a result, the prevention and control of PEDs in food supply have drawn increasing attention. For better prevention and control, the first step is to understand the food contaminants -- PEDs. Phytoestrogen α-Zearalanol attenuates homocysteine-induced apoptosis in human umbilical vein endothelial cells.

Hyperhomocysteinemia is an independent risk factor for cardiovascular diseases. The enhanced nitrative stress plays an important role in homocysteine-induced endothelial dysfunction. Previous studies have showed that phytoestrogen α -zearalanol alleviated endothelial injury in ovariectomized hyperhomocysteinemic rats; however, the underlying mechanism remains to be clarified. This study was to investigate the effects of α -zearalanol on homocysteine-induced endothelial apoptosis in vitro and explore the possible role of nitrative stress in these effects. Results showed that homocysteine (500

Physiological data including evaluation of immuno-response in relation to anabolic effects on veal calves.

In a series of experiments with a total of 1480 veal calves, different aspects of treating calves with anabolic steroids were examined. The anabolics used were 17beta-estradiol (E), trenbolone acetate (T), progesterone (P), testosterone (Te), C+T, E+P, E+Te and zeranole (Z). The N-retention was estimated by examining the urea: creatinine ratio in single urine specimens during the course of two feeding trials. Increased gain due to the treatment with E (20 mg implanted/calf) + P (200 mg) and Te (200 mg), respectively, E + T (140 mg) or Z (36 mg) was during the whole experimental period. The ext

Alleviation of plasma homocysteine level by phytoestrogen α-zearalanol might be related to the reduction of cystathionine β-synthase nitration.

Hyperhomocysteinemia is strongly associated with cardiovascular diseases. Previous studies have shown that phytoestrogen α-zearalanol can protect cardiovascular system from hyperhomocysteinemia and ameliorate the level of plasma total homocysteine; however, the underlying mechanisms remain to be clarified. The aim of this research is to investigate the possible molecular mechanisms involved in ameliorating the level of plasma homocysteine by α-zearalanol. By the successfully established diet-induced hyperhomocysteinemia rat models, we found that, after α-zearalanol treatment, the activity of c

Hormones in international meat production: biological, sociological and consumer issues.

Beef and its products are an important source of nutrition in many human societies. Methods of production vary and include the use of hormonal compounds ('hormones') to increase growth and lean tissue with reduced fat deposition in cattle. The hormonal compounds are naturally occurring in animals or are synthetically produced xenobiotics and have oestrogenic (oestradiol-17beta and its esters; zeranol), androgenic (testosterone and esters; trenbolone acetate) or progestogenic (progesterone; melengestrol acetate) activity. Zearalenone and alpha-zearalenol inhibit the synthesis and secretion of pig follicle stimulating hormone via the non-classical estrogen membrane receptor GPR30.

Zearalenone (ZEA) is one of the most popular endocrine-disrupting chemicals and is mainly produced by fungi of the genus Fusarium. The excessive intake of ZEA severely disrupts human and animal fertility by affecting the reproductive axis. However, most studies on the effects of ZEA and its metabolite α-zearalenol (α-ZOL) on reproductive systems have focused on gonads. Few studies have investigated the endocrine-disrupting effects of ZEA and α-ZOL on pituitary gonadotropins, including follicle-stimulating hormone (FSH) and luteinizing hormone (LH). The present study was designed to investigate

Toxicological effects of fumonisin B1 alone and in combination with other fusariotoxins on bovine granulosa cells.

There is now overwhelming evidence of global contamination of commodities with Fusarium mycotoxins. Fumonisin B1 (FB1) is a Fusarium mycotoxin frequently occurring in corn in combination with deoxynivalenol (DON), α-zearalenol (α-ZEA) and β-zearalenol (β-ZEA). The aim of this study was to determine if FB1, alone and combined with DON or α-ZEA or β-ZEA, can affect cell proliferation and steroid production of bovine granulosa cells (GC). A species-specific model with bovine granulosa cells (GC) was used to study the potential endocrine disruptor effects of FB1 alone and in co-exposure. In the pr
