Decoding the thermocyclic solar-driven fermentation: Multi-omics insights into microbial and metabolic dynamics of traditional Chishui river basin soy sauce.

Traditional Chishui River Basin soy sauce is produced through prolonged solar-cycle fermentation under diurnal temperature fluctuations and moisture absorption. Here, we employed integrated metagenomic and metabolomic analyses to investigate microbial and metabolic dynamics throughout fermentation. Results revealed a three-phase microbial succession: initial fungal-hydrolytic phase dominated by Aspergillus oryzae (82.28%), marked by proteolysis and amino acid accumulation; transitional phase enriched with Weissella (23.45%) and Zygosaccharomyces rouxii (5.85%), producing organic acids, esters,

Synergistic improvement of flavor development in fermented chili paste through microbial co-fermentation and enzymatic hydrolysis.

This study evaluated eight fermentation strategies to improve flavor development in fermented chili paste. LAB-yeast co-fermentation with Lactiplantibacillus plantarum LP06 and Zygosaccharomyces rouxii ZR26 increased total acidity and amino acid nitrogen, reduced residual sugar and capsaicinoids, enhanced sourness, and decreased bitterness. Cellulase and flavourzyme supplementation promoted substrate conversion and modified taste responses. Untargeted metabolomics showed that co-fermentation without enzyme (LZ) was mainly associated with amino acid- and sulfur-related metabolites, whereas enzy

Selection of Non-Saccharomyces Yeasts from Extreme Oenological Environments for Potential Use in Winemaking.

This study evaluated the oenological potential of two non-Saccharomyces yeast strains, Hanseniaspora opuntiae TR-5 and Zygosaccharomyces bailii L-25, isolated from extreme winemaking environments in southern Spain. Out of 156 yeast isolates screened from high-sugar musts and flor yeast biofilms, strains were selected based on their β-glucosidase activity, killer phenotype, and ethanol production, traits associated with aroma release and microbial competition. Fermentation trials on sugar-rich synthetic medium showed that both H. opuntiae and Z. bailii achieved ethanol yields of 10% v/v and res

Resistance of Yeast Species to Benzoic and Sorbic Acids and to Sulfur Dioxide.

Effects of sorbic and benzoic acids and SO2 on the growth and survival of 12 strains of yeasts, differing widely in their preservative resistance, were studied. Exponential phase cultures not adapted to preservative were tested under anaerobic conditions at pH 3.5. In general, species tolerant of one preservative were also tolerant of the others, but significant differences in the relative effectiveness of the preservatives were found in some species. Maximum tolerated levels of benzoic acid ranged from 0.5 mM for Hansenula anomala to 4 mM for Zygosaccharomyces bailii . The range in tolerance

Molecular interpretation of flavor improvement for Pixian Douban fermented with Bacillus bioaugmentation in the tank based on whole-genome and metatranscriptomics.

Considering the flavor disadvantages of the strong acidity and whole pungency in Pixian Douban (PXDB) of the tank fermentation, an improvement was obtained by the bioaugmentation of B. licheniformis JY and B. atrophaeus XY, and the underlying mechanism was explored by whole-genome and metatranscriptomics. The key volatiles contents especially for phenethyl alcohol, 1-nonanal, and 4-ethylphenol were increased by Bacillus bioaugmentation while the acid content was reduced. The metatranscriptome analysis indicated that Zygosaccharomyces and Bacillus were the dominant strains in the group of Bacil

Whole-genome analysis guided molecular mechanism of cyanogenic glucoside degradation by yeast isolated from Prunus mume fruit syrup.

Consumption of fermented Prunus mume fruit (maesil) sugar syrup raise a health concern due to the presence of the cyanogenic glucoside amygdalin. The goal of the present study was to investigate the amygdalin degradation potential and genome profile of the native microbes found in maesil syrup. The microbial profile analysis revealed that yeast is the predominant microorganism native to maesil syrup and that the isolated yeast cells showed a remarkable potential for amygdalin reduction (99.7%). Moreover, the reduction in amygdalin was inversely proportional to the growth of the isolated yeast.

Hydrophobic diversification is the key to simultaneously increased antifungal activity and decreased cytotoxicity of two ab initio designed peptides.

The fact that some antimicrobial peptides have been utilized clinically and as food preservatives stimulated the efforts in search of new candidates. In our previous studies, we succeeded in designing potent peptides against methicillin-resistant Staphylococcus aureus (MRSA), severe acute respiratory syndrome coronavirus 2 (SARS-Cov-2), and Ebola viruses based on the database filtering technology. The designed peptides were proved highly potent. However, this ab initio method has not been utilized to design antifungal peptides. This study report two novel antifungal peptides with 21 and 15 ami

Use of the Glycolipopeptid Biosurfactant Produced by Lactiplantibacillus plantarum Tw226 to Formulate Functional Cinnamon Bark Essential Oil Emulsions.

The stabilization of essential oils in emulsions using surfactants of natural origin is of significant interest, and the use of biosurfactants produced by lactic acid bacteria could be an alternative. In this study, the total and partial substitution of Tween 80 in cinnamon bark essential oil emulsions was proposed using a glycolipopeptide biosurfactant produced by Lactiplantibacillus plantarum Tw226. The oil-in-water emulsions formulated contained cinnamon bark oil at a concentration of 5 g/L, with Tween 80, the biosurfactant, or a mixture of both as the surfactant agent, reaching a final con

Evidence for viable but nonculturable yeasts in botrytis-affected wine.

AIMS: In Botrytis-affected wine, high concentrations of SO2 are added to stop the alcoholic fermentation and to stabilize the wine. During maturing in barrels or bottle-ageing, an unwanted refermentation can sometimes occur. However, results of the usual plate count of wine samples at the beginning of maturing suggest wine microbial stability. The aim of this study was to investigate the mode of yeasts survival after the addition of SO2 and to identify surviving yeasts. METHODS AND RESULTS: Using direct epifluorescence technique, we observed the behaviour of cells after SO2 addition and compa
