We report a multiplex family with extended multisystem neurological phenotype associated with a CRYAB variant. Two affected siblings were evaluated with whole exome sequencing, muscle biopsy, laser microdissection, and mass spectrometry-based proteomic analysis. Both patients and their mother manifested a combination of early-onset cataracts, cardiomyopathy, cerebellar ataxia, optic atrophy, cognitive impairment, and myopathy. Whole exome sequencing identified a heterozygous c.458C>T variant mapped to the C-terminal extension domain of the Alpha-crystallin B chain, disrupting its function as a

BACKGROUND/AIMS: We searched for factors implicated in early hepatic stellate cell (HSC) activation in diseased liver, by means of suppression subtractive hybridization (SSH).
METHODS: SSH was performed between messenger RNA (mRNA) from normal rat HSC and mRNA from HSC, isolated from rats with acute D-galactosamine (Gal)-induced hepatitis.
RESULTS: One of the potentially upregulated factors which we found was alpha B-crystallin (ABCRYS), a small heat-shock protein and a chaperone known to protect the cell against protein degradation in conditions of cellular stress and known to associate wit

The clarity and refractivity of the eye lens are mediated by the highly soluble crystallin proteins. Post-translational modifications impact solubility and stability of the structural and refractive βγ-crystallins, eventually leading to cataract. Such damaged proteins are kept in solution by the holdase chaperone α-crystallins, maintaining lens transparency over decades despite the absence of protein turnover. It was previously found that a short peptide from human αA-crystallin (mini-αA-crystallin [MAAC]) retains some chaperone activity, with hydrophobic interactions hypothesized to mediate c

Enterovirus 71 (EV71) can cause severe hand-foot-and-mouth disease with neurological complications. It has evolved multiple mechanisms to compromise the host type I interferon (IFN-I) response. In neuronal cells, EV71-mediated IFN-I antagonism may be associated with neural precursor cell-expressed developmentally downregulated 4-like (Nedd4L), the E3 ubiquitin ligase that can interact with alphaB-crystallin (CRYAB) in the regulation of Nav1.5 stability. Here, we investigated the effect of CRYAB stability on IFN-β promoter activity in neuronal SH-SY5Y cells infected with EV71, and its relations

αB-crystallin, a member of the small heat shock protein (sHSP) family, is expressed in diverse tissues, including the eyes, brain, muscles, and heart. This protein plays a crucial role in maintaining eye lens transparency and exhibits holdase chaperone and anti-apoptotic activities. Therefore, structural and functional changes caused by genetic mutations in this protein may contribute to the development of disorders like cataract and cardiomyopathy. Recently, the substitution of arginine 123 with tryptophan (p.R123W mutation) in human αB-crystallin has been reported to trigger cardiomyopathy.

Fiber morphology and crystalline structure of poly[(R)-3-hydroxybutyrate] (P(3HB)) and stereocomplexed poly(lactide) (PLA) nanofibers were investigated by using scanning and transmission electron microscopies and X-ray and electron diffractions. In the P(3HB) nanofibers spun from less than 1 wt% 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) solution, planar zigzag conformation (beta-form) as well as 2(1) helix conformation (alpha-form) structure was formed. Based on the electron diffraction measurement of single P(3HB) nanofiber, it was revealed that the molecular chains of P(3HB) align parallel to

Oxidative aggregation of γ-crystallins induced by copper in aged lens increases the lens opacity and causes cataract formation. Therefore, chelation of free Cu2+ by small molecules can inhibit metal-mediated aggregation of γ-crystallin. In this work, the inhibition potency of several naturally occurring flavonoid compounds was studied against aggregation of human γD-crystallin (HGD) mediated by copper ions. Among them, rutin demonstrated ~20% inhibition of HGD aggregation induced by Cu2+ through its metal chelation ability. Not only that, the chaperone activity of lens chaperone, human αA-crys

The factors influencing the formation, organizations, and temperature ranges of the smectic phases of a structurally diverse family of phosphonium salts have been examined. The salts consist of one short group and three long n-alkyl chains attached to a positively charged phosphorus atom and either a free or covalently attached counterion, the latter resulting in zwitterionic salts. Of the 61 salts investigated, of which 37 have not been synthesized previously, most pack in lamellae within their solid phases. Single-crystal X-ray structures of two of amidomethyl-tri-n-tetradecylphosphonium bro

Pediatric cataract of the congenital type is the most common form of childhood blindness and it is clinically and genetically heterogeneous. Mutations in 22 different genes have been identified to be associated with congenital cataracts, and among them, eight mutants belong to αA-crystallin. To explain how mutations in αA-crystallin lead to the development of cataract, quaternary structural parameters, and chaperone function have been investigated in αA-wt and in the following mutants: R12C, R21L, R21W, R49C, R54C, R116C, and R116H. Average molar mass, mass at the RI peak, mass across the peak

Human αB-crystallin is a small heat shock protein that functions as a chaperone and anti-apoptotic protein to maintain cellular protein integrity. A specific mutation (p.R163C) in the C-terminal domain has been linked to dilated cardiomyopathy (DCM). However, the impact of this mutation on the protein's structure, activity, stability, and amyloidogenic properties remains unclear. Here, we introduced the mutation, expressed and purified the protein, and used spectroscopic and microscopic techniques to conduct a comprehensive investigation of the mutant protein. The p.R163C mutation in αB-crysta

Arginine 54 (R54) in αA-Crystallin (αA-Cry) is highly conserved within different species. Recently, three missense mutations at this hot spot position have been reported to cause congenital cataract disorders. To investigate the impact of charge on structural and functional aspects of αA-Cry, R54 was individually substituted with lysine and aspartate. Replacement of R54 with the positively and negatively charged residues led to structural alteration and reduction in the protein conformational and proteolytic stability. Also, these mutations resulted in important increase in the amyloidogenic p

Though the term "inflammation" is traditionally defined as proliferation or infiltration of lymphatic cells of the lymphatic immune system and macrophages or as immunoreactive proteins including cytokines, interleukins and major histocompatibility complexes, recently recognized reactions to tissue injury also are inflammation, often occurring in the central nervous system in conditions where they previously were not anticipated and where they may play a role in both pathogenesis and repair. We highlight 4 such novel inflammatory conditions revealed by neuropathologic studies: (1) inflammatory

The substitution of leucine to proline at position 39 (p.P39L) in human αB-crystallin (αB-Cry) has been associated with conflicting interpretations of pathogenicity in cataracts and cardiomyopathy. This study aimed to investigate the effects of the p.P39L mutation on the structural and functional features of human αB-Cry. The mutant protein was expressed in Escherichia coli (E. coli) and purified using anion exchange chromatography. We employed a wide range of spectroscopic analyses, gel electrophoresis, transmission electron microscopy (TEM), and atomic force microscopy (AFM) techniques to in

d-β-aspartyl (Asp) residue has been found in a living body such as aged lens crystallin, although l-α-amino acids are constituents in natural proteins. Isomerization from l-α- to d-β-Asp probably modulates structures to affect biochemical reactions. At Asp residue, isomerization and peptide bond cleavage compete with each other. To gain insight into how fast each reaction proceeds, the analysis requires the consideration of both pathways simultaneously and independently. No information has been provided, however, about these competitive processes because each reaction has been studied separate

AlphaB-crystallin is a small heat shock protein that is found in the eye lens, retina, heart, brain, skin, and skeletal muscle. Mutations of αB-crystallin lead to the development of cataracts, human muscle and heart disorders. The role of non-enzymatic glycation of αB-crystallin in the development of pathological processes has been little studied. The aim of this study was to evaluate the influence of enhanced methylglyoxal (MGO) level on the functions of αB-crystallin. We modified recombinant αB-crystallin (aB-Cr) by MGO and investigated the effect of this modification on different functions

Aggregation of the α-Synuclein (αSyn) protein in neurons is responsible for synucleinopathies such as Parkinson's disease. In healthy cells, αSyn is primarily present as monomers. Under pathological conditions, oligomers and fibrils are formed, leading to neuronal toxicity and death. No treatment prevents fatal synucleinopathies. We designed small peptidomimetics based on the structure of αSyn aggregates and on its chaperone protein αB-Crystallin. Interestingly, a relationship between the impact of peptidomimetics on the αSyn aggregation process, their sequences, and secondary conformation has

Found from bacteria to humans, small heat shock proteins (sHSPs) are the least understood protein chaperones. HSPB5 (or αB-crystallin) is among the most widely expressed of the 10 human sHSPs, including in muscle, brain, and eye lens where it is constitutively present at high levels. A high content of disorder in HSPB5 has stymied efforts to uncover how its structure gives rise to function. To uncover its mechanisms of action, we compared human HSPB5 and two disease-associated mutants, R120G and D109H. Expecting to learn how the mutations lead to loss of function, we found instead that the mut

Lens proteins become increasingly cross-linked through nondisulfide linkages during aging and cataract formation. One mechanism that has been implicated in this cross-linking is glycation through formation of advanced glycation end products (AGEs). Here, we found an age-associated increase in stiffness in human lenses that was directly correlated with levels of protein-cross-linking AGEs. α-Crystallin in the lens binds to other proteins and prevents their denaturation and aggregation through its chaperone-like activity. Using a FRET-based assay, we examined the stability of the αA-crystallin-γ

Antisera have been made to synthetic peptides corresponding to the expected tryptic fragments from the C-terminal region of human alpha A2 crystallin (T19 corresponds to residues 158-163; T20 corresponds to residues 164-173). These antisera were used on conjunction with a sensitive radioimmunoassay, to identify the elution times of peptides resolved on a C18 column from a tryptic digest of water soluble and water insoluble proteins from the human lens. Isolation and purification of the peptides reactive with the anti-peptide sera, followed by the use of tandem mass spectrometry to determine th
