Proteomic analysis of peritoneal fluid identified COMP and TGFBI as new candidate biomarkers for endometriosis.

Endometriosis is a common non-malignant gynecological disease that significantly compromises fertility and quality of life of the majority of patients. The gold standard for diagnosis is visual inspection of the pelvic organs by surgical laparoscopy and there are no biomarkers that would allow non-invasive diagnosis. The pathogenesis of endometriosis is not completely understood, thus analysis of peritoneal fluid might contribute in this respect. Our prospective case-control study included 58 patients undergoing laparoscopy due to infertility, 32 patients with peritoneal endometriosis (cases)

TGFBI R124H mutant allele silencing in granular corneal dystrophy type 2 using topical siRNA delivery.

In recent years, success has been achieved in treating several eye conditions with oligonucleotide-based therapies. Herein, we outline the experimentation involved in progressing selection and development of a lead therapeutic siRNA for R124H mutation of TGFBI gene which causes Granular Corneal Dystrophy Type 2 (GCD2/Avellino CD). Firstly, a series of siRNA designs, generated by a gene walk across the R124H TGFBI mutation site, were tested and a lead siRNA identified. The lead siRNA was delivered into an immortalised human corneal epithelial cell line to assess on-target efficacy and off-targe

TGFBI functions similar to periostin but is uniquely dispensable during cardiac injury.

Extracellular matrix production and accumulation stabilize the heart under normal conditions as well as form a protective scar after myocardial infarction injury, although excessive extracellular matrix accumulation with long-standing heart disease is pathological. In the current study we investigate the role of the matricellular protein, transforming growth factor beta-induced (TGFBI), which is induced in various forms of heart disease. Additionally, we sought to understand whether TGFBI is functionally redundant to its closely related family member periostin, which is also induced in the dis

Altered protein conformation and lower stability of the dystrophic transforming growth factor beta-induced protein mutants.

PURPOSE: Transforming growth factor beta-induced protein (TGFBIp) is a widely expressed extracellular matrix protein that plays roles in cell adhesion and migration, differentiation, apoptosis, bone morphogenesis, and carcinogenesis. Mutations of TGFBIp have been linked to stromal corneal dystrophies, a group of protein conformational diseases characterized by abnormal protein aggregations in the cornea. However, the underlying pathogenic mechanism remains elusive due to a lack of insight into the molecular properties of the disease-causing mutants. In the current study, we applied spectroscop

NICD3 mediates pro-angiogenic effects through SMAD3/TGFBI axis in colorectal cancer.

The consensus molecular subtype 4 (CMS4) of colorectal cancer (CRC) represents an aggressive, mesenchymal phenotype associated with the poorest clinical prognosis. Although angiogenesis is fundamental to CRC progression, current anti-VEGF therapies are frequently compromised by primary or acquired resistance, creating an urgent need to identify alternative vascular drivers. NOTCH3 is significantly enriched in CMS4 CRC, yet its direct role in modulating the tumor vasculature remains elusive. Here, integrating multi-center transcriptomic datasets with clinical validation, we identified NOTCH3 as

TGFBI Inhibits the Pyroptosis of Macrophages to Ameliorate Septic Shock.

Septic shock is one of the leading causes of morbidity and mortality in hospital patients. The present study aimed to investigate the potential of transforming growth factor β induced (TGFBI) in macrophage functions and progression of septic shock. Mice were treated with caecal ligation and puncture (CLP) to establish a septic shock model in vivo. Histopathologic analysis was performed using haematoxylin and eosin (HE) staining. Gene expression was detected using reverse transcription-quantitative PCR and Western blot. Cytokine release was detected using an enzyme-linked immunosorbent assay. T

TGFBI promotes liver fibrosis through remodeling the profibrotic microenvironment by a positive feedback regulatory loop.

Liver fibrosis is a major global health burden with limited treatment options. Transforming growth factor-beta-induced protein (TGFBI) is crucial in fibrotic diseases and tumors, however, its precise mechanism in liver fibrosis remains unclear. Here we show that TGFBI promotes liver fibrosis in male C57BL/6 mice. TGFBI is upregulated in fibrotic livers and derived from non-parenchymal cells. Genetic TGFBI deficiency alleviates liver fibrosis in both CCl4 (carbon tetrachloride) injection and bile duct ligation (BDL) models. Mechanistically, PDGFRβ is identified via RNA sequencing as a key downs

MiR-21 plays an important role in radiation induced carcinogenesis in BALB/c mice by directly targeting the tumor suppressor gene Big-h3.

Dysregulation of certain microRNAs (miRNAs) in cancer can promote tumorigenesis, metastasis and invasion. However, the functions and targets of only a few mammalian miRNAs are known. In particular, the miRNAs that participates in radiation induced carcinogenesis and the miRNAs that target the tumor suppressor gene Big-h3 remain undefined. Here in this study, using a radiation induced thymic lymphoma model in BALB/c mice, we found that the tumor suppressor gene Big-h3 is down-regulated and miR-21 is up-regulated in radiation induced thymic lymphoma tissue samples. We also found inverse correlat

A Novel Loss-of-Function Variant in TGFBI Associated with Non-Syndromic Orofacial Clefts in a Chinese Pedigree.

Background/Objectives: Non-syndromic orofacial clefts (NSOFCs) are among the most common congenital craniofacial anomalies, with genetic factors playing a major role in their etiology. Transforming growth factor-β-induced (TGFBI) protein is an extracellular matrix protein implicated in cell adhesion and apoptosis. Although experimental studies have implicated TGFBI in palatal fusion, its contribution to human NSOFCs remains unclear. This study investigated the genetic basis of hereditary NSOFCs in a Chinese family. The functional consequences of the identified TGFBI variant were subsequently e

BIGH3 is a mediator of TGFβ-Induced collagen formation in fibrosis and pancreatic cancer and a potential therapeutic target.

Fibroblast-driven deposition of extracellular matrix, particularly type III collagen, is linked to poor outcomes in patients with liver fibrosis, fibrotic solid tumors such as hepatocellular carcinoma and pancreatic cancer as well as in patients with other fibrotic diseases. The pro-peptide of type III collagen (PRO-C3) serves as a serum biomarker of active collagen formation. PRO-C3 is elevated and prognostic in patients with liver fibrosis and pancreatic cancer, suggesting that targeting mechanisms behind PRO-C3 elevation could benefit these individuals. In this study, we sought to identify
