The cGAS-STING signaling pathway: A central regulator and novel therapeutic target in skeletal muscle pathophysiology.

The cGAS-STING signaling pathway is a central component of the innate immune system. Skeletal muscle, the body's largest metabolic and endocrine organ, is essential for overall health, and maintaining its homeostasis is critically important. This review systematically elaborates on the central position and "double-edged sword" role of the cGAS-STING pathway in skeletal muscle pathophysiology. We detail how, under various pathological stimuli-such as metabolic stress, physical injury, aging, toxin exposure, and systemic diseases-cytoplasmic DNA accumulation aberrantly activates the cGAS-STING p

The Role of the cGAS-STING Signaling Pathway in Oral Mucosal Diseases: Mechanisms and Therapeutic Implications.

The oral mucosa maintains a delicate immunological equilibrium amidst constant microbial and environmental challenges. Within this unique microenvironment, the cytosolic DNA-sensing cGAS-STING signaling pathway has emerged as a central innate immune hub characterized by critical duality across a spectrum of pathologies. While transient STING activation is vital for protective anti-microbial defenses and driving anti-tumor immunity in oral squamous cell carcinoma, chronic, maladaptive signaling-often driven by self-DNA accumulation-fuels pathology in autoimmune and inflammatory conditions. The Murine Antimicrobial Peptide CRAMP Alleviates Neuroinflammation By Downregulating the cGAS-STING Signalling Pathway.

Facial nerve injury (FNI) is common, yet therapeutic outcomes are often suboptimal due to axonal degeneration, neuronal death, and persistent neuroinflammation. Microglia, the primary immune cells of the central nervous system, exacerbate neuroinflammation when polarized to the pro-inflammatory M1 state. Cathelicidin-related antimicrobial peptide (CRAMP in mice and rats; LL-37 in humans) has been shown to mitigate neuroinflammation by inhibiting microglial M1 polarization via suppression of the cGAS-STING signalling pathway, suggesting a potential therapeutic strategy for FNI. This study emplo

The cGAS-STING signaling pathway contributes to the inflammatory response and autophagy in Aspergillus fumigatus keratitis.

Fungal keratitis is a serious corneal infection, which can lead to significant visual impairment and blindness. The cGAS-STING signaling pathway has emerged as a key player in innate immunity by sensing of invading pathogens. However, the role of the cGAS-STING pathway in Aspergillus fumigatus (A. fumigatus) keratitis is still unknown. In this study, we showed that the cGAS-STING signaling pathway was activated in human corneal epithelial cells (HCECs) and in mouse corneas infected with A. fumigatus. Knockdown of cGAS reduced A. fumigatus-induced production of pro-inflammatory cytokines, inclu

cGAS-STING signaling pathway: a central pathological mechanism and emerging therapeutic target for postoperative cognitive dysfunction.

Postoperative cognitive dysfunction (POCD) is a prevalent neurological complication in older patients following surgery. However, the upstream molecular triggers of perioperative neuroinflammation, a key factor in its pathogenesis, remain insufficiently understood. This review systematically examines the emerging evidence implicating the cGAS-STING signaling pathway as a potentially central mediator in the pathological progression of POCD. Integrating recent advancements, we outline a critical pathological cascade in POCD: perioperative stressors, including anesthesia and surgical trauma, indu

Decursin protects against DSS-induced experimental colitis in mice by inhibiting the cGAS-STING signaling pathway.

While studies have shown that Angelica gigas Nakai (A. gigas) can alleviate ulcerative colitis in mice, the therapeutic role of its main active ingredient, decursin, is uncertain. Therefore, we aimed to investigate the protective effect and mechanism of decursin against inflammatory bowel disease (IBD) in vivo using mice. IBD was simulated via induction with 3% dextran sodium sulfate (DSS), with or without daily treatment with decursin (10 mg/kg or 20 mg/kg) or 5-amino salicylic acid (5-ASA; 100 mg/kg) for 14 days. Mice were weighed and monitored daily for disease activity index (DAI) scoring.

NLRC4 promotes the cGAS-STING signaling pathway by facilitating CBL-mediated K63-linked polyubiquitination of TBK1.

TANK-binding kinase 1 (TBK1) is crucial in producing type Ⅰ interferons (IFN-Ⅰ) that play critical functions in antiviral innate immunity. The tight regulation of TBK1, especially its activation, is very important. Here we identify NLRC4 as a positive regulator of TBK1. Ectopic expression of NLRC4 facilitates the activation of the IFN-β promoter, the mRNA levels of IFN-β, ISG54, and ISG56, and the nuclear translocation of interferon regulatory factor 3 induced by cGAS and STING. Consistently, under herpes simplex virus-1 (HSV-1) infection, knockdown or knockout of NLRC4 in BJ cells and primary

Porphyromonas gingivalis induces an inflammatory response via the cGAS-STING signaling pathway in a periodontitis mouse model.

Periodontitis is an inflammatory disease initiated by periodontopathogenic bacteria in the dental plaque biofilms. Understanding the role of Porphyromonas gingivalis (P. gingivalis), a keystone pathogen associated with chronic periodontitis, in the inflammatory response is crucial. Herein, we investigated whether P. gingivalis infection triggers the expression of the type I IFN gene and various cytokines and leads to activation of the cGAMP synthase-stimulator of IFN genes (cGAS-STING) pathway both in vitro and in a mouse model. Additionally, in an experimental model of periodontitis using P.

Navigating the cGAS-STING signaling pathway in breast cancer: Reinterpreting the paradox of antitumor and pro-metastatic.

The cGAS-STING signaling pathway exhibits functions in breast cancer that include both antitumor immunity and pro-metastatic inflammation, transcending traditional linear switch models. To address this cognitive bottleneck, this paper proposes the conceptual framework of "cGAS-STING pathway-guided signal flow." It attributes pathway outcomes to multi-level fine-tuning, aiming to decipher initial immunogenic/pathogenic signals in the upstream phase based on intensity, duration, and origin. It elucidates how the STING protein, as a central hub, integrates and programs signals through a complex n

Role of the cGAS-STING signaling pathway in diabetes mellitus and its complications: from mechanisms to therapeutics.

Diabetes mellitus and its complications represent a major global public health challenge, with their pathogenesis closely linked to chronic, low-grade, non-infectious metabolic inflammation. The cGAS-STING signaling pathway, a crucial innate immune sensor of cytosolic DNA, has recently emerged as a central hub linking metabolic stress to sterile inflammation. This review systematically elucidates the mechanisms and therapeutic potential of the cGAS-STING pathway in diabetes mellitus and its associated complications. Under diabetic metabolic stress conditions-such as hyperglycemia and lipotoxic

The cGAS-STING pathway contributes to cisplatin-induced skeletal muscle atrophy through altered proteostasis and myogenic signaling.

BACKGROUND: Cisplatin chemotherapy is widely used for cancer treatment but frequently induces skeletal muscle atrophy, which compromises physical function and patient outcomes. The molecular mechanisms underlying this process remain incompletely understood. The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) signaling pathway, classically involved in innate immune responses, has recently been implicated in cellular stress and tissue dysfunction. Whether cGAS-STING signaling contributes to cisplatin-induced skeletal muscle atrophy remains unclear. METHODS: We employed both

The bidirectional immunoregulatory effects of Traditional Chinese Medicine on the cGAS-STING signaling pathway and their translational prospects.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a central component of innate immunity, mediating host defense against pathogens, antitumor responses, and inflammation. Aberrant activation of this pathway is implicated in a spectrum of diseases, including autoimmune disorders, chronic inflammation, organ injury, and metabolic dysregulation. Traditional Chinese Medicine (TCM), with its multi-component and multi-target characteristics, presents a promising strategy for modulating cGAS-STING signaling. Emerging evidence indicates that bioactive compounds, single

African Swine Fever Virus EP364R and C129R Target Cyclic GMP-AMP To Inhibit the cGAS-STING Signaling Pathway.

African swine fever virus (ASFV) is a highly pathogenic swine DNA virus with high mortality that causes African swine fever (ASF) in domestic pigs and wild boars. For efficient viral infection, ASFV has developed complex strategies to evade key components of antiviral innate immune responses. However, the immune escape mechanism of ASFV remains unclear. Upon ASFV infection, cyclic GMP-AMP (2',3'-cGAMP) synthase (cGAS), a cytosolic DNA sensor, recognizes ASFV DNA and synthesizes the second messenger 2',3'-cGAMP, which triggers interferon (IFN) production to interfere with viral replication. [Activation of cGAS/STING signaling pathway and its immunological role in the progression of nonalcoholic fatty liver disease].

Today, nonalcoholic fatty liver disease remains the most dominant chronic liver disease. Cyclic guanosine monophosphate-adenosine monosphosphate synthase (cGAS) is a cytosolic DNA sensor that catalyzes the synthesis of cyclic guanosine monophosphate, activates stimulator of interferon genes (STING), and releases type-I interferon cytokines to trigger immune responses. Exogenous or endogenous DNA acts as a cGAS ligand to activate the cGAS-STING signaling pathway, which plays a role in hepatitis, nonalcoholic fatty liver disease, liver cancer and other diseases, and affects liver disease progres
