β-Adrenergic receptors suppress Rap1B prenylation and promote the metastatic phenotype in breast cancer cells.

A greater understanding of the molecular basis of breast cancer metastasis will lead to identification of novel therapeutic targets and better treatments. Rap1B is a small GTPase that suppresses the metastasis of breast cancer cells by increasing cell-cell adhesion. In breast cancer, a decrease in Rap1B prenylation and subsequent loss of Rap1B at the plasma membrane decreases cell-cell adhesion and increases cell scattering, which promotes the metastatic phenotype. Protein kinase A (PKA) was recently found to phosphorylate Rap1B and inhibit its prenylation. PKA is activated by G protein-couple

Rap1b regulates B cell development, homing, and T cell-dependent humoral immunity.

Rap1 is a small GTPase that belongs to Ras superfamily. This ubiquitously expressed GTPase is a key regulator of integrin functions. Rap1 exists in two isoforms: Rap1a and Rap1b. Although Rap1 has been extensively studied, its isoform-specific functions in B cells have not been elucidated. In this study, using gene knockout mice, we show that Rap1b is the dominant isoform in B cells. Lack of Rap1b significantly reduced the absolute number of B220(+)IgM(-) pro/pre-B cells and B220(+)IgM(+) immature B cells in bone marrow. In vitro culture of bone marrow-derived Rap1b(-/-) pro/pre-B cells with I

Overexpression of Rap1B alleviate central precocious puberty and neurodevelopmental damage.

INTRODUCTION: The central precocious puberty (CPP) in children and adolescents can adversely impact their overall well-being, affecting both their physical and psychological health. Whether Rap1B can mitigate the development of CPP remains unknown. METHODS: The expression of Rap1B was investigated through a comprehensive bioinformatics analysis. Precocious puberty model in rats was established through high-fat feeding (HFD), and further injected AAV-Rap1B. Then, HE staining, vaginal exfoliated cell smear and other methods were used to explore the incidence of precocious puberty in rats in eac

Somatic RAP1B gain-of-function variant underlies isolated thrombocytopenia and immunodeficiency.

The ubiquitously expressed small GTPase Ras-related protein 1B (RAP1B) acts as a molecular switch that regulates cell signaling, cytoskeletal remodeling, and cell trafficking and activates integrins in platelets and lymphocytes. The residue G12 in the P-loop is required for the RAP1B-GTPase conformational switch. Heterozygous germline RAP1B variants have been described in patients with syndromic thrombocytopenia. However, the causality and pathophysiological impact remained unexplored. We report a boy with neonatal thrombocytopenia, combined immunodeficiency, neutropenia, and monocytopenia cau

Role of MIR-101-3P/RAP1B Axis and Insulin Protection in Glucose-Induced Stress in HK-2 Cell Model for Diabetic Kidney Disease.

Proximal tubular cell (PTC) injury is a critical driver of diabetic kidney disease (DKD). This study aimed to characterize the molecular response of HK-2 PTCs to acute glucotoxicity, focusing on the miR-101-3p/RAP1B axis, insulin-mediated protection, and the primary mechanisms of cell death. HK-2 cells were exposed to varying glucose concentrations (5.5–128 mM) with or without insulin (10 mg/L) for 48 h. We assessed cell viability, mitochondrial membrane potential (ΔΨm), and intracellular reactive oxygen species (ROS). Expression of miR-101-3p, RAP1B, insulin receptor substrates (IRS1/2), and

The tuberous sclerosis 2 gene is expressed at high levels in the cerebellum and developing spinal cord.

Tuberous sclerosis (TS) is an autosomal dominant multisystem disorder characterized by the widespread development of hamartomas in many tissues and organs. TSC2 is predicted to encode a 1784-amino acid tumor suppressor protein that may function, in part, as a GTPase-activating protein for Rap1. Given the high incidence of central nervous system abnormalities in individuals affected with tuberous sclerosis, the expression of TSC2 in developing and adult nervous system tissues was examined. Reverse transcription-PCR, Northern blot, and in situ hybridization analyses demonstrated high levels of e

A fibroblast-specific miRNA functional cluster in remote exosomes aggravates myocardial ischaemia/reperfusion injury via the Rap1b/ERK1/2 axis.

BACKGROUND AND PURPOSE: Communication between various cardiac cells by paracrine factors and exosomes has an important role in myocardial ischaemia/reperfusion (I/R) injury. It remains unclear whether exosomes derived from healthy cardiac fibroblasts affect I/R injury and, if so, what are the underlying mechanisms? EXPERIMENTAL APPROACH: Cardiac fibroblasts were isolated from neonatal rats, adult rats and adult rats subjected to I/R. Their exosomes were designated as follows: neonatal cardiac fibroblasts (N-Exo), adult rat cardiac fibroblasts (A-Exo) and cardiac fibroblasts in a remote non-is

The G protein-coupled receptor GALR2 promotes angiogenesis in head and neck cancer.

Squamous cell carcinoma of the head and neck (SCCHN) is an aggressive disease with poor patient survival. Galanin receptor 2 (GALR2) is a G protein-coupled receptor that induces aggressive tumor growth in SCCHN. The objective of this study was to investigate the mechanism by which GALR2 promotes angiogenesis, a critical oncogenic phenotype required for tumor growth. The impact of GALR2 expression on secretion of proangiogenic cytokines in multiple SCCHN cell lines was investigated by ELISA and in vitro angiogenesis assays. Chemical inhibitor and genetic knockdown strategies were used to unders

Comparative analysis and genomic structure of the tuberous sclerosis 2 (TSC2) gene in human and pufferfish.

Germ-line mutations of the TSC2 tumour suppressor gene have been identified in humans with tuberous sclerosis and in the Eker rat. Tuberin, the human TSC2 gene product, has a small region of homology with rap1GAP and stimulates rap1 GTPase activity in vitro, suggesting that one of its cellular roles is to function as a GTPase activating protein (GAP). We have undertaken a comparative analysis of the TSC2 gene in human and the pufferfish, Fugu rubripes. In addition to the GAP domain, three other regions of the proteins are highly conserved (peptide sequence similarity > 80%). These regions are

Inactivation Rap2a in Endothelial Cell Prevents Pulmonary Fibrosis by Regulating Immune Microenvironment Through MAP4K4-VCAM1 Signaling.

Idiopathic pulmonary fibrosis (IPF) is characterized by progressive fibrotic remodeling accompanied by persistent endothelial activation and leukocyte infiltration. Although endothelial dysfunction is increasingly recognized as a key contributor to fibrogenesis, the intracellular signaling pathways that couple inflammatory cues to endothelial-immune interactions remain incompletely defined. Ras-related protein Rap2a (RAP2A), a small GTPase implicated in stress and inflammatory signaling, has not been systematically investigated in pulmonary endothelial cells during fibrotic lung injury. Here,

Overexpression of FMOD in Thyroid Carcinoma Triggers M1-Like Tumor-Associated Macrophage Polarization by Targeting Rap1B.

Thyroid carcinoma, with limited efficacy of current treatment, influences the lives and health of many people. It is important to explore potential therapeutic targets for thyroid carcinoma. Fibromodulin (FMOD) has been indicated to be connected with the progression of different kinds of tumors, with unknown functions in thyroid carcinoma. In this study, the potential candidate therapeutic targets for thyroid carcinoma were identified by bioinformatics methods, and FMOD was screened out for verification. Cell counting kit-8, wound healing, transwell, and flow cytometry assays were conducted to

Cloning and characterization of Rap GTPase from the Chinese white shrimp Fenneropenaeus chinensis.

Ras-related protein Rap GTPase has been implicated in cell adhesion, cell proliferation, and cell junction formation. The first shrimp Rap cDNA (FcRap) was recently identified from the Chinese white shrimp Fenneropenaeus chinensis. The full length of FcRap is 1013 bp, with a 561 bp open reading frame that encodes a 186 amino acid protein. FcRap has a calculated molecular mass of 20.90 kDa and pI of 6.37. Phylogenetic analysis shows that FcRap and other Rap proteins are clustered into one group. Results from the quantitative real-time polymerase chain reaction show that FcRap could be detected

The Epac1 signaling pathway regulates Cl- secretion via modulation of apical KCNN4c channels in diarrhea.

The apical membrane of intestinal epithelia expresses intermediate conductance K(+) channel (KCNN4), which provides the driving force for Cl(-) secretion. However, its role in diarrhea and regulation by Epac1 is unknown. Previously we have established that Epac1 upon binding of cAMP activates a PKA-independent mechanism of Cl(-) secretion via stimulation of Rap2-phospholipase Cε-[Ca(2+)]i signaling. Here we report that Epac1 regulates surface expression of KCNN4c channel through its downstream Rap1A-RhoA-Rho-associated kinase (ROCK) signaling pathway for sustained Cl(-) secretion. Depletion of

Rap1 signal modulators control the maintenance of hematopoietic progenitors in bone marrow and adult long-term hematopoiesis.

Adult long-term hematopoiesis depends on sustaining hematopoietic stem/progenitor cells (HSPC) in bone marrow (BM) niches, where their balance of quiescence, self-renewal, and hematopoietic differentiation is tightly regulated. Although various BM stroma cells that produce niche factors have been identified, regulation of the intrinsic responsiveness of HSPC to the niche factors remains elusive. We previously reported that mice deficient for Sipa1, a Rap1 GTPase-activating protein, develop diverse hematopoietic disorders of late onset. Here we showed that transplantation of BM cells expressing

The cAMP effectors, Rap2b and EPAC, are involved in the regulation of the development of the Coxiella burnetii containing vacuole by altering the fusogenic capacity of the vacuole.

Cyclic Adenosine 3',5'-monophosphate (cAMP) is a key second messenger known to directly regulate not only the protein kinase A (PKA) activity but also other important molecules such as the exchange protein activated by cAMP (EPAC), which is as a guanine nucleotide exchange factor (GEF) of the low molecular weight GTPase, Rap2. Coxiella burnetii is a Gram negative bacterium that survives and grows in a large Coxiella replicative vacuole (CRV), which displays lysosomal and autophagic features. In this report, we present evidence that both, EPAC and its downstream effector Rap2b, were recruited t

Cloning and evaluation of RALGDS as a candidate for the tuberous sclerosis gene TSC1.

RALGDS is a 115 kDa protein which was identified by its ability to enhance guanine nucleotide exchange for the ras family member ral. It also binds to activated ras and rap1, and appears to function as part of a signalling complex in downstream events following rap1 activation. Here we report the identification of full-length cDNA clones for human RALGDS, isolated from a brain cDNA library. The predicted protein has strong sequence homology to rat and murine isoforms of RALGDS in the N- and C-terminal regions, but an internal region (aa 250-380) shows relatively high divergence with only 42% i

Activation of the Drosophila C3G leads to cell fate changes and overproliferation during development, mediated by the RAS-MAPK pathway and RAP1.

The cellular signal transduction pathways by which C3G, a RAS family guanine nucleotide exchange factor, mediates v-crk transformation are not well understood. Here we report the identification of Drosophila C3G, which, like its human cognate, specifically binds to CRK but not DRK/GRB2 adaptor molecules. During Drosophila development, constitutive membrane binding of C3G, which also occurs during v-crk transformation, results in cell fate changes and overproliferation, mimicking overactivity of the RAS-MAPK pathway. The effects of C3G overactivity can be suppressed by reducing the gene dose of

Influence of integrins on thrombus formation: a road leading to the unravelling of DVT.

Integrins are a group of transmembrane glycoprotein receptors that are responsible for platelet activation through bidirectional signalling. These receptors have left their footprints in various cellular events and have intrigued many groups of scientists that have led to some significant discoveries. A lot of the recent understanding of haemostasis has been possible due to the integrins filling the gaps in between several cellular mechanism. Apart from this, other important functions carried out by integrins are growth and maturation of cardiomyocytes, mechano-transduction, and interaction wi

Fibronectin 1 Aggravates Colon Cancer Metastasis by Regulating RAP1B Protein Stability Through Akt/CREB Signalling Pathway.

Metastasis is one of the important factors leading to poor prognosis in patients with colon cancer. However, the molecular mechanism contributing to this cellular behaviour remains largely unknown. Here, RNA-seq analysis suggested that Fibronectin 1 (FN1) was significantly increased in metastatic colon cancer, which was associated with poor prognosis. FN1 enhanced colon cancer cell migration, invasion, and epithelial to mesenchymal transition (EMT) in vitro and promoted liver and lung metastasis of colon cancer in nude mice through RAP1B. Mechanistically, FN1 could interact with RAP1B by suppr
