Staphylococcus biofilm dynamics and antibiotic resistance: insights into biofilm stages, zeta potential dynamics, and antibiotic susceptibility.

Staphylococcus spp. infections often involve biofilms, but standard antibiotic minimum inhibitory concentration (MIC) testing used to determine treatment evaluates planktonic bacterial growth only and does not account for biofilm presence, strength, or growth stage. To aid in determining a cost-effective method to solve this issue, we built upon in vitro methods initially published by Stepanovic et al. used to determine weak and strong biofilm formations. First, we determined 115 unique S. aureus isolate biofilms at 2, 4, 6, 8, 16, and 24 h to classify the hourly stages of biofilm development

Dental plaque biofilm transforms host-derived β2-microglobulin into polymorphic fibrils for integration into the biofilm matrix.

Dental plaque is a polymicrobial biofilm formed on tooth surfaces despite continuous exposure to variable host-derived antimicrobial factors. To date, the underlying mechanisms remain nebulous. This study aimed to determine whether dental plaque biofilms affect the major salivary antimicrobial protein β2-microglobulin (B2M). Immunostaining of human dental plaque specimens with an anti-B2M antibody revealed that B2M exists as elongated fibers, punctate structures, and amorphous aggregates. Fractionation of dental plaque suspensions revealed that B2M was present in both the soluble and insoluble

Biofilm volume and acidification within initial biofilms formed in situ on buccally and palatally exposed bracket material.

PURPOSE: Acidification by bacterial biofilms at the bracket/tooth interface is one of the most common problems in fixed orthodontic treatments, which can lead to white spot lesions (WSL) and caries. As lingual brackets were shown to exhibit reduced WSL formation clinically, the aim of this in situ study was to compare initial intraoral biofilm formation and acidification on bracket-like specimens placed buccally and palatally in the upper jaw as a possible cause for this observation. METHODS: Intraoral biofilm was collected from splints equipped with buccally and palatally exposed test specim

Small cyclic dipeptide produced by Lactobacillus rhamnosus with anti-biofilm properties against Streptococcus mutans biofilm.

The human oral cavity harbors many bacterial species collectively termed the oral microbiome and is integral for maintaining oral health. Dysbiosis of oral microbiota leads to common oral diseases, including dental caries, gingivitis, and periodontitis. Streptococcus mutans is the primary causative agent of dental caries. Studies have explored the use of probiotic Lactobacillus spp. to mitigate S. mutans biofilms. In the present study, we have tested the use of Lactobacillus rhamnosus extracts/metabolites for anti-biofilm properties. A small organic compound/metabolite was isolated from the ce

Inhibition of Listeria monocytogenes Cocktail Culture Biofilms on Crab and Shrimp Coupons and the Expression of Biofilm-Related Genes.

Listeria monocytogenes, a bacterium that is transmitted by tainted food, causes the infection listeriosis. In this study, quercetin was tested for its antibacterial properties and effectiveness as a food additive in preventing the growth of L. monocytogenes cocktail (ATCC19117, ATCC19113, and ATCC15313) biofilms on crabs and shrimps. Quercetin showed the least bactericidal activity and no discernible microbial growth at a minimum inhibitory concentration (MIC) of 250 µg/mL. The biofilm inhibition was performed at sub-MICs (1/2, 1/4, and 1/8 MIC). There was no quercetin added to the control gro

Natural and synthetic plant compounds as anti-biofilm agents against Escherichia coli O157:H7 biofilm.

Escherichia coli is a common gram-negative bacterium found in the gut and intestinal tract of warm-blooded animals including humans. An evolved seropathotype E. coli O157:H7 (STEC) came into existence in 1982, since then it has been evolved as a stronger and more robust drug-resistant pathotype of E. coli. This drug resistance is due to horizontal gene transfer, natural gene evolution for survival, and most of the cases due to the ability of STEC to switch to the biofilm growth mode from planktonic lifestyle. During the growth in biofilm mode, Escherichia coli O157:H7 opts more robust ability

Anti-biofilm activity of a semi-synthetic molecule obtained from resveratrol against Candida albicans biofilm.

Candida albicans can form biofilm on tissues and medical devices, becoming, in that case, less susceptible to antifungal agents. Treatment of candidiasis associated with the formation of C. albicans biofilms is restricted to echinocandins and lipid forms of amphotericin B. This study investigated the activity of micafungin and resveratrol modified molecule (EB487) against C. albicans biofilms. The anti-biofilm growth (Bgrowth) and anti-preformed biofilm (Bpreformed) activities of micafungin (0 to 3.94 μM) and EB487 (0 to 20.32 mM) were comparatively studied separately and combined, using XTT,

Chitosan disrupts biofilm formation and promotes biofilm eradication in Staphylococcus species isolated from bovine mastitis.

Staphylococci are the main pathogens associated with hard-to-control intramammary infections in dairy cattle, and bacterial biofilms are suspected to be responsible for the antimicrobial resistance and persistence of this disease. Biofilms have the ability to resist to higher levels of antibiotics and reduce their efficacy. It is thus necessary to develop strategies targeted to bacterial biofilm infections. Chitosan is a polysaccharide with a proven broad spectrum of antimicrobial activity against fungi and bacteria. The aim of this study was assess the effect of low molecular weight (LMW) chi

Relative Abundances of Candida albicans and Candida glabrata in In Vitro Coculture Biofilms Impact Biofilm Structure and Formation.

Candida is a member of the normal human microbiota and often resides on mucosal surfaces such as the oral cavity or the gastrointestinal tract. In addition to their commensality, Candida species can opportunistically become pathogenic if the host microbiota is disrupted or if the host immune system becomes compromised. An important factor for Candida pathogenesis is its ability to form biofilm communities. The two most medically important species-Candida albicans and Candida glabrata-are often coisolated from infection sites, suggesting the importance of Candida coculture biofilms. In this wor

Breaking the biofilm barrier: herbomineral nanocomposites to control Staphylococcus epidermidis through oxidative response and biofilm disruption.

The growing prevalence of antibiotic resistance and biofilm-forming dental pathogens necessitates the development of alternative antimicrobial strategies. Staphylococcus epidermidis is an opportunistic pathogen associated with dental biofilms, oral microbial colonization, and implant-related infections, contributing to persistent infections and treatment challenges. In this study, a herbomineral nanocomposite (HMNCs) was fabricated using the traditional Siddha formulation Kalnar Parpam and silver nitrate, and its antibacterial and antibiofilm activities were evaluated against S. epidermidis is

CApEsid biOfilm: a suggested pipeline for clinical phage microbiology for biofilm infections based on comparative method study.

The rise of antibiotic-resistant infections, particularly those involving biofilms, presents a significant global health threat. Phage therapy, the use of bacteriophages as antimicrobial agents, offers promising solutions to this crisis. A critical component of phage therapy is the assessment of phage efficacy, in both the presence and absence of antibiotics, prior to clinical application. While considerable progress has been made using planktonic bacterial cultures, there remains an urgent need for standardized methods to evaluate phage efficacy against biofilms. In this study, we address thi

Lysyl-phosphatidylglycerol promotes cell-to-cell contact and biofilm formation of Staphylococcus aureus as a biofilm matrix component.

Staphylococcus aureus biofilms contribute significantly to persistent infections and antibiotic resistance, supported by a complex extracellular matrix. While their proteins, polysaccharides, and extracellular DNA have been well studied, the role of phospholipids in biofilm architecture remains underexplored. Here, we identify extracellular phospholipids within the biofilm matrix, particularly lysyl-phosphatidylglycerol (Lys-PG), as critical structural elements in S. aureus biofilms. Bacterial phospholipase A1 (PLA1), which hydrolyzes phospholipid acyl ester bonds, effectively dispersed pre-fo

Prevention and Treatment of Staphylococcus aureus Biofilms Using Promising Agr-QS-Targeting Anti-Biofilm Agents.

Staphylococcus aureus (S. aureus), a leading cause of nosocomial infections, contributes significantly to increased morbidity and mortality, especially when it forms biofilms on medical devices. This pathogen, specifically methicillin-resistant S. aureus (MRSA), remains a challenge to treat due to its ability to form biofilms and rapidly develop resistance against antibiotics. Biofilm formation allows bacteria to adhere to biotic and abiotic surfaces, creating a protective matrix that shields them from immune responses and antibiotic therapies. The widespread prevalence of multidrug-resistant

The landscape of biofilm models for phage therapy: mimicking biofilms in diabetic foot ulcers using 3D models.

Diabetic foot ulcers (DFU) affect up to 15-25% of patients suffering from diabetes and are considered a global health concern. These ulcers may result in delayed wound healing and chronic infections, with the potential to lead to amputations. It has been estimated that 85% of diabetes-related amputations are preceded by a diagnosis of DFU. A critical factor in the persistence of this disease is the presence of polymicrobial biofilms, which generally include Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli. The involvement of diabetic comorbidities such as ischemia, hyperglyc

Natural TPs inhibit biofilm formation by Multidrug-resistant Acinetobacter baumannii and biofilm-induced pulmonary inflammation.

Multidrug-resistant Acinetobacter baumannii (MDRAB) infections cause elevated rates of patient deaths in intensive care units owing to the high antibiotic resistance of the clinical isolates. The advent of multidrug-resistant A. baumannii (MDRAB) strains and the formation of their biofilms are cause for concern. Tea polyphenols (TPs), which exhibit antimicrobial activity, is an ideal alternative strategy for lowering the incidence of nosocomial bacterial infections. This study was conducted to determine the effects of TPs on MDRAB. The antimicrobial and anti-biofilm activities of TPs against M

PCV13 vaccine prevents pneumococcal biofilms without affecting Staphylococcus aureus population within the polymicrobial biofilm.

In respiratory pathogens such as Streptococcus pneumoniae, biofilm formation is associated with the colonization of the nasopharynx and chronic respiratory infection. Previous data have shown that pneumococcal conjugate vaccines (PCVs) had an impact on S. pneumoniae colonization and a potential replacement by other respiratory pathogens such as Staphylococcus aureus. The objective of this work was to evaluate the evasion of the immune system by monospecific biofilms and by S. aureus-S. pneumoniae mixed biofilms. We performed opsonophagocytosis assays (OPA) using human HL-60 against previously
