Cholesterol gallstone formation occurs in three stages. First, the bile must be saturated with cholesterol, thereby allowing cholesterol crystals to form. Then, nucleation and growth of the gallstone can occur, although little is known about these latter two stages. Therapy for dissolution of gallstones is directed at desaturating the bile. Chenodeoxycholic acid (CDCA), the most extensively tested agent, is successful in dissolving 60 per cent of radiolucent gallstones; however, long-term safety remains to be demonstrated. Ursodeoxycholic acid (UDCA), the 7 beta epimer of CDCA, is a promising

BACKGROUND: Hypercholesterolemia is closely associated with an increased risk of cardiovascular diseases. L-Arabinose exhibited hypocholesterolemia properties, but underlying mechanisms have not been sufficiently investigated. This study aimed to elucidate the mechanisms of L-arabinose on hypocholesterolemia involving the enterohepatic circulation of bile acids.
METHODS: Thirty six-week-old male mice were randomly divided into three groups: the control group and the high-fat-high-sucrose diet (HFHSD)-fed group were gavaged with distilled water, and the L-arabinose-treated group were fed HFHSD

Biological markers (biomarkers) are used to recognize, characterize and monitor treatment-related responses following exposure to xenobiotics. Biomarkers serve three primary applications in toxicology: 1) to confirm exposure to a deleterious agent, 2) to provide a system for monitoring individual susceptibility to a toxicant, and 3) to quantitatively assess deleterious effects of a toxicant to an organism or individual. Because the liver is a general target for adverse effects of drugs and other chemicals, biomarkers of untoward hepatic response to xenobiotics are of particular interest to the

Background: Hemolytic jaundice, characterized by excessive bilirubin production and hepatic dysfunction, currently lacks effective therapeutic strategies targeting its underlying mechanisms. Methods: This study evaluated the therapeutic potential of the aqueous extract of Inonotus obliquus (IO) in ameliorating hemolytic jaundice through modulation of the gut-liver axis. A murine model of hemolytic jaundice was induced by phenylhydrazine (PHZ) administration, and IO was orally administered as the intervention. Antibiotic depletion and FMT verified gut microbiota dependence. Liver injury, serum

INTRODUCTION: The human CRB1 gene encodes the CRB1 protein, primarily expressed in retinal Muller cells and photoreceptors, where it regulates apical-basal polarity and cellular signalling through its role in adherence junctions and maintaining the outer limiting membrane barrier. Dysfunction of CRB1 results in a range of retinal phenotypes with few systemic implications reported. It has been suggested that disrupted Crb1 expression in the gastrointestinal epithelium of rd8 mouse models (Crb1 -/-) results in barrier dysfunction permitting translocation of bacteria to the retina.

BACKGROUND Cholecystectomy alters enterohepatic bile circulation, impairing fat-soluble vitamin absorption and reducing bioavailability of vitamin D and vitamin K, required for hepatic synthesis of coagulation factors II, VII, IX, and X. Despite the high global frequency of this procedure, evidence regarding clinically significant postoperative changes in fat-soluble vitamin status remains inconsistent. This retrospective single-center study compared preoperative and postoperative serum 25-hydroxyvitamin D (25[OH]D) levels and vitamin K-dependent coagulation profiles in patients undergoing cho

Intestinal microorganisms regulate bile acid (BA) homeostasis through bile salt hydrolase (BSH)-mediated deconjugation and a series of biotransformation reactions that generate secondary BAs with potent signaling functions. Antibiotic exposure profoundly disrupts these microbial processes, leading to alterations in BA composition, diversity, and enterohepatic circulation. Recent multi-omics studies have demonstrated that antibiotics reduce the abundance of key BA-transforming bacteria, including members of the Lachnospiraceae, Ruminococcaceae, Lactobacillaceae, Clostridiaceae, and Bacteroidace

PURPOSE: Intermittent skin-to-skin care (SSC) during phototherapy for neonatal jaundice improves breast milk consumption and reduces enterohepatic circulation. The primary objective of this study was to evaluate the effect of SSC on the rate of fall in bilirubin in neonatal jaundice. The secondary objectives were maximum TSB, duration of phototherapy, duration of hospital stay, weight gain, feeding and stooling pattern.
METHODS: In this randomised controlled trial, otherwise healthy late preterm and term neonates of birth weight ≥ 2 kg, who required phototherapy (AAP 2004 guidelines) and if t

BACKGROUND AND AIMS: Previous studies have suggested that cholecystectomy may lead to an elevated risk of metabolic dysfunction-associated fatty liver disease (MAFLD). However, there is a dearth of knowledge concerning the precise effects and underlying mechanisms through which cholecystectomy influences hepatic lipid metabolism, necessitating further investigation.
METHODS: Post-cholecystectomy (PC) model mice were established under both normal and high-fat diet conditions. Liver lipid metabolism, inflammation, bile acid profiles, and immune cell alterations were evaluated. Hepatocytes mimic

Background: The gut is a target organ of thyroid hormone (TH) that exerts its action via the nuclear thyroid hormone receptor α1 (TRα1) expressed in intestinal epithelial cells. THs are partially metabolized via hepatic sulfation and glucuronidation, resulting in the production of conjugated iodothyronines. Gut microbiota play an important role in peripheral TH metabolism as they produce and secrete enzymes with deconjugation activity (β-glucuronidase and sulfatase), via which TH can re-enter the enterohepatic circulation. Summary: Intestinal epithelium homeostasis (the finely tuned balance be

Colorectal cancer (CRC) risk is predominantly driven by environmental factors, in particular diet. A high intake of dietary fat has been implicated as a risk factor inducing the formation of pre-neoplastic lesions (e.g., adenomatous polyps) and/or exacerbating colonic tumorigenesis. Recent data attributed the tumor-promoting activity of high-fat diets to their effects on gut microbiota composition and metabolism, in particular with regard to bile acids. Bile acids are synthesized in the liver in response to dietary fat and facilitate lipid absorption in the small intestine. The majority of bil

The Aryl hydrocarbon receptor (AhR) is primarily known as one of the xenosensors and regulators of drug-metabolizing genes. Bile acids (BAs) are synthesized in the liver, and undergo several enterohepatic recirculations in which the liver removes BAs from the portal blood, minimizing the BAs that spill over into the systemic circulation. Previous studies revealed a lifelong patent ductus venosus (PDV) in AhR-null mice. Increased concentration of total BAs (Σ-BAs) in AhR-null mice is known; however, the impact of PDV on BA homeostasis in liver and bile remains unclear. This work investigated th

The farnesoid X receptor (FXR, NR1H4) is a bile acid (BA)-activated transcription factor, which is essential for BA homeostasis. FXR and its hepatic and intestinal target genes, small heterodimer partner (SHP, NR0B2) and fibroblast growth factor 15/19 (Fgf15 in mice, FGF19 in humans), transcriptionally regulate BA synthesis, detoxification, secretion, and absorption in the enterohepatic circulation. Furthermore, FXR modulates a large variety of physiological processes, such as lipid and glucose homeostasis as well as the inflammatory response. Targeted deletion of FXR renders mice highly susce

The metabolism of 7-ethyl- and 7-propyl-chenodeoxycholic acids was studied in hamsters. Both bile acid analogs were absorbed efficiently by the intestine and secreted into the bile at rates similar to those of chenodeoxycholic acid. After intraduodenal administration into bile fistula hamsters, the 7-alkyl analogs were present in bile as the glycine and taurine conjugates. The glycine/taurine ratios were: chenodeoxycholic acid, 1.9; 7-ethyl analog, 0.3; and 7-propyl analog, 0.2. After oral administration, during a 21-day feeding experiment, the 14C-labeled analogs were recovered quantitatively

The object of the present study was to improve the accuracy of measurements of ileal function obtained by abdominal scanning and fecal counting after oral administration of the gamma-labeled bile acid 75seleno-homocholic acid-taurine (75SeHCAT), as current techniques do not distinguish between retention of the bile acid within the enterohepatic circulation from retention within the colon, and are also affected by incomplete stool collection when using the fecal method. We have therefore introduced the following modifications: (a) simultaneous ingestion of 51CrCl3 as a nonabsorbable correction

BACKGROUND: Crohn's disease (CD) exhibits substantial heterogeneity in disease behavior and therapeutic outcomes across distinct disease locations. Specific alterations in the gut microbiota and metabolites may drive this variation. This review aims to characterize the distinctive microbial and metabolic signatures across CD phenotypes based on disease location.
METHODS: Electronic databases were searched from inception to December 2025 for studies that observed alterations in gut microbiota and metabolites in CD patients with different disease locations.
RESULTS: Forty-eight studies includi
