Modeling the Impact of Case Finding for Tuberculosis: The Role of Infection Dynamics.

BACKGROUND: Many individuals previously infected with Mycobacterium tuberculosis (Mtb) may clear their infections or experience substantially reduced progression risks over time. Such dynamics suggest that recent transmission is more important in driving TB incidence in high-burden settings than previously estimated; thus, the impact of interventions to reduce transmission (eg, community-based active case finding) may also be greater than previously thought. METHODS: We constructed 2 models of Mtb transmission that differed only in the inclusion of a clearance mechanism. We calibrated these m

Review of human behavior integration in COVID-19 modeling studies.

BACKGROUND: Human behavior influences the spread of infectious diseases, making it essential to integrate behavioral processes into epidemiological models. This became particularly evident during the COVID-19 pandemic, as many models did not incorporate behavior in response to policies. METHODS: We reviewed modeling analyses of population dynamics in response to interventions intended to mitigate the spread of COVID-19 from February 2020 to February 2023. Key characteristics of each study were extracted, including the behavioral aspects integrated within the models and utilized databases. RE

Global dynamics of a compartmental model to assess the effect of transmission from deceased.

During several epidemics, transmission from deceased people significantly contributed to disease spread, but mathematical analysis of this transmission has not been seen in the literature numerously. Transmission of Ebola during traditional burials was the most well-known example; however, there are several other diseases, such as hepatitis, plague or Nipah virus, that can potentially be transmitted from disease victims. This is especially true in the case of serious epidemics when healthcare is overwhelmed and the operative capacity of the health sector is diminished, such as seen during the

New measures of effect size in moderation analysis.

Measures of explained variance, ΔR2 and f,2 are routinely used to evaluate the size of moderation effects. However, they suffer from several drawbacks: (a) Not all the variance components of the outcome variable Y are related to the effect of moderation, and so an effect size with the total variance of Y as the denominator cannot accurately characterize the moderation effect; (b) moderation and interaction are conflated; and (c) the assumption of homoscedasticity might be violated when moderation exists. By arguing that measures for the size of moderation effect should be based on the variance

Nonsteroidal anti-inflammatory drugs in experimental parkinsonian models and Parkinson's disease.

A number of experimental studies using parkinsonian models have revealed that nonsteroidal anti-inflammatory drugs (NSAIDs) have neuroprotective properties against dopaminergic neurotoxicity not only by their cyclooxygenase-inhibiting effect but also by other specific properties or some unknown pharmacological effects. This article reviews heterogeneous pharmacological properties of NSAIDs including inhibitory effect against nitric oxide synthesis, agonistic action for peroxisome proliferator-activated receptor gamma, or possible suppressive effects against dopamine quinone generation, and als

Cannabinoids and morphine differentially affect HIV-1 expression in CD4(+) lymphocyte and microglial cell cultures.

The influence of substances of abuse on the progression of HIV-1 infection is controversial, and pharmacologic factors have been postulated as a potential explanation for conflicting data arising from epidemiological studies and animal models. In the present study, cell culture models of HIV-1 infection were used to test this hypothesis. The synthetic cannabinoid WIN 55,212-2 was found to potently inhibit HIV-1 expression in a concentration- and time-dependent manner in CD4(+) lymphocyte and microglial cell cultures. In sharp contrast, morphine either inhibited or stimulated viral expression,

Epidemiological inference for emerging viruses using segregating sites.

Epidemiological models are commonly fit to case and pathogen sequence data to estimate parameters and to infer unobserved disease dynamics. Here, we present an inference approach based on sequence data that is well suited for model fitting early on during the expansion of a viral lineage. Our approach relies on a trajectory of segregating sites to infer epidemiological parameters within a Sequential Monte Carlo framework. Using simulated data, we first show that our approach accurately recovers key epidemiological quantities under a single-introduction scenario. We then apply our approach to S

Comparative analysis of the costs and benefits of alternative disease control strategies: vector control versus human case finding and treatment.

Recent estimates of the costs of detection and treatment and vector control for human trypanosomiasis are summarized and the main factors influencing their levels discussed. In order to proceed to an economic analysis a benefit unit is defined as equivalent to one year's infection avoided due to the control strategy for one person. Different monetary values could be assigned to this unit and it has the advantage of being easily integrated into epidemiological models, being represented by the difference in prevalence with and without control. A simple spreadsheet-based economic model was develo

Not everyone panics alike: When heterogeneous behavioral responses change epidemic outcomes.

Compartmental epidemic models increasingly capture demographic and contact heterogeneities, yet behavioral responses are typically treated as uniform: as cases or deaths rise, an average person perceives greater risk and increases compliance with non-pharmaceutical interventions (e.g., masking), reducing transmission. But when is treating societal behavior as a single average feedback loop a safe simplification? We develop a two-group compartmental behavioral epidemic model in which groups differ in combinations of infection fatality ratio, susceptibility, and contact rates, which leads to dis

Dynamics and Persistence of a Generalized Multi-strain SIS Model.

Autonomous differential equation compartmental models hold broad utility in epidemiology and public health. However, these models typically cannot account explicitly for myriad factors that affect the trajectory of infectious diseases, with seasonal variations in host behavior and environmental conditions as noteworthy examples. Fortunately, using non-autonomous differential equation compartmental models can mitigate some of these deficiencies, as the inclusion of time-varying parameters can account for temporally varying factors. The inclusion of these temporally varying factors does come at

Integrating information from historical data into mechanistic models for influenza forecasting.

Seasonal influenza causes significant annual morbidity and mortality worldwide. In France, it is estimated that, on average, 2 million individuals consult their GP for influenza-like-illness (ILI) every year. Traditionally, mathematical models used for epidemic forecasting can either include parameters capturing the infection process (mechanistic or compartmental models) or rely on time series analysis approaches that do not make mechanistic assumptions (statistical or phenomenological models). While the latter make extensive use of past epidemic data, mechanistic models are usually independen

A note on parameter estimation for variant Creutzfeldt-Jakob disease epidemic models.

A recent series of papers has raised issues regarding estimation of the key epidemiological parameters of variant Creutzfeldt-Jakob disease (vCJD) from fitting survival models to case data. In particular, it was stated that the scale of the epidemic cannot be estimated and must be fixed in any analysis. We show that this problem is an artefact of the approximate likelihood used in these papers to facilitate model-fitting, and is not a concern if estimation is based on the full likelihood. We also address comments in these papers regarding the choice of incubation period distribution.

Potential role of endocrine-disrupting chemicals in preeclampsia: A hypothesis based on integration of epidemiological and experimental evidence.

Preeclampsia (PE) is a complex hypertensive disorder and a leading cause of maternal and perinatal morbidity worldwide. Emerging evidence suggests that exposure to endocrine-disrupting chemicals (EDCs) may contribute to the etiology of PE, yet this association remains underexplored. This review aimed to investigate epidemiological and experimental studies assessing the potential link between EDC exposure and PE development. A literature search was conducted across PubMed, ScienceDirect, and Google Scholar for original articles published in the last ten years. Forty studies were selected, inclu

Mathematical modelling of Wolbachia replacement in Aedes aegypti for dengue control: a scoping review.

Wolbachia replacement in dengue virus vectors is a promising tool for controlling transmission, and models can explore its potential in novel and complex scenarios. We analyse how research questions in Wolbachia replacement modelling have developed over time and in response to empirical findings. A scoping review was conducted of Wolbachia replacement models. For each study, we extracted research questions, key findings and modelling methodology and thematically categorized research questions. From 726 search results, 115 studies met inclusion criteria. Four themes were identified: identifying

A fractional SEIVRB model for aquatic diseases dynamics with stability analysis and numerical solution.

This research presents a fractional epidemiological model utilizing the standard incidence rate to characterize the transmission dynamics of aquatic infectious diseases like cholera. The framework incorporates a vaccinated compartment to assess preventive measures. We analytically derive both disease-free and endemic equilibrium states and compute the basic reproduction number. The human population is modeled as a time-dependent variable, and we construct a positively invariant domain for the solution space, demonstrating boundedness and nonnegativity of solutions. For the disease-free equilib

Estimating HIV incidence from case-report data: method and an application in Colombia.

OBJECTIVE: Quantifying HIV incidence is essential for tracking epidemics but doing this in concentrated epidemic can be a particular challenge because of limited consistent high-quality data about the size, behaviour and prevalence of HIV among key populations. Here, we examine a method for estimating HIV incidence from routinely collected case-reporting data. METHODS: A flexible model of HIV infection, diagnosis and survival is constructed and fit to time-series data on the number of reported cases in a Bayesian framework. The time trend in the hazard of infection is specified by a penalized

Improved estimation of controlled direct effects in the presence of unmeasured confounding of intermediate variables.

Adjusting for a causal intermediate is a common analytic strategy for estimating an average causal direct effect (ACDE). The ACDE is the component of the total exposure effect that is not relayed through the specified intermediate. Even if the total effect is unconfounded, the usual ACDE estimate may be biased when an unmeasured variable affects the intermediate and outcome variables. Using linear programming optimization to compute non-parametric bounds, we develop new ACDE estimators for binary measured variables in this causal structure, and use root mean square confounding bias (RMSB) to c

In-vivo rodent models for the experimental investigation of prenatal immune activation effects in neurodevelopmental brain disorders.

Based on the epidemiological association between maternal infection during pregnancy and enhanced risk of neurodevelopmental brain disorders in the offspring, a number of in-vivo models have been established in rats and mice in order to study this link on an experimental basis. These models provide indispensable experimental tools to test the hypothesis of causality in human epidemiological associations, and to explore the critical neuroimmunological and developmental factors involved in shaping the vulnerability to infection-induced neurodevelopmental disturbances in humans. Here, we summariz

Best practice assessment of disease modelling for infectious disease outbreaks.

During emerging disease outbreaks, public health, emergency management officials and decision-makers increasingly rely on epidemiological models to forecast outbreak progression and determine the best response to health crisis needs. Outbreak response strategies derived from such modelling may include pharmaceutical distribution, immunisation campaigns, social distancing, prophylactic pharmaceuticals, medical care, bed surge, security and other requirements. Infectious disease modelling estimates are unavoidably subject to multiple interpretations, and full understanding of a model's limitatio
