Multimerin1 and not Galectin-8 tempers WNT signaling to promote gastric chief cell differentiation.

Galectins are a family of proteins that bind galactose-containing glycans. One member, galectin-8, preferentially binds galactose that contains a terminal sulfate. Aberrant expression and secretion of sulfated glycosylation epitopes, such as 3'-Sulfo-LeA/C, is a feature of high-risk human foregut metaplasias. In addition, recent work has demonstrated that 3'-Sulfo-LeC is a marker of mature murine zymogenic chief cells of the stomach and that 3'-Sulfo-LeC epitope is secreted via cathartocytosis during the cellular transition to a metaplastic state. Based on those findings, we used Lgals8-/- mic

Galectin-3 in sepsis: A multifaceted regulator of innate immunity, vascular dysfunction, and organ injury.

Sepsis is a life-threatening syndrome characterized by dysregulated host responses, endothelial injury, microcirculatory disturbance, and progressive organ dysfunction. Galectin-3 (Gal-3), a β-galactoside-binding lectin with diverse intracellular and extracellular activities, has emerged as an important mediator in inflammatory and fibrotic diseases. Increasing evidence suggests that Gal-3 is involved in multiple pathophysiological processes relevant to sepsis, rather than acting within a single pathway. In this review, we summarize the structural and functional features of Gal-3 and discuss i

Microglial galectin-3 disrupts parvalbumin interneurons and hippocampal synchrony, driving cognitive deficits.

Sepsis-associated encephalopathy (SAE), a devastating neurological complication of systemic inflammation, affects approximately 70% of patients with sepsis. It not only increases mortality but also leaves survivors with persistent cognitive deficits. However, the mechanisms underlying SAE progression remain incompletely understood. Here, using a lipopolysaccharide (LPS)-induced mouse model of SAE, we identify microglial galectin-3 (Gal-3) as a central pathogenic mediator driving systemic inflammation-induced cognitive impairment. Mechanistically, systemic LPS challenge robustly upregulates mic

SOX9 represses the human galectin-3 promoter in SW1353 cells: potential implications for osteoarthritis.

Galectin-3 (Gal-3), a member of the β-galactoside-binding protein family, is critically involved in inflammation, extracellular matrix remodelling, and cartilage degeneration in osteoarthritis (OA). This study aims to elucidate the regulation of the human galectin-3 gene (LGALS3) promoter in SW1353 cells and its control by SOX transcription factors, known to be dysregulated during OA pathogenesis. We sought to identify key sequence elements in the LGALS3 promoter responsible for its transcriptional activity and relevant transcription factors (TFs). Using luciferase reporter assays, we examined

Galectin-3 is Necessary for Selective Cathartocytosis, which Expedites the Development of Proliferative Gastric SPEM.

The expression and secretion of sulfated colonic-type mucins is a feature of high-risk metaplasias of the gastrointestinal foregut (Barrett's esophagus, type III intestinal metaplasia of the stomach, and pancreatic intraepithelial neoplasia). Galectin-3 is a lectin that preferentially associates with galactose modified by a 3'-O-sulfate relative to its unmodified counterparts and is upregulated as the tissue transitions to high-risk metaplasia, dysplasia, and cancer. Since both galectin-3 and sulfated glycotopes are aberrantly and concurrently overexpressed in high-risk premalignant and malign

Galectin-1 Signaling Dysregulation Impairs Stromal Senescence and Decidualization in Recurrent Implantation Failure.

Endometrial decidualization is indispensable for embryo implantation, and moderate cellular senescence has recently been recognized as a functional component of this process. However, how this senescence response is regulated upstream and whether its disruption contributes to recurrent implantation failure (RIF) remain unclear. Here, we identify Galectin-1 (GAL1) as a potential regulator of senescence-associated decidual responses in human endometrial stromal cells. Moderate senescence induction in human endometrial stromal cells and primary decidual stromal cells (DSCs) was accompanied by enh

Galectin-8 binds HIV envelope glycoproteins with high affinity and promotes viral infectivity.

Target cell entry of HIV-1 is dependent on the binding of gp120, the outer component of the viral envelope glycoprotein complex (Env), to CD4 and a coreceptor, preferentially CCR5 or CXCR4. Still, other interactions may also contribute to the infectivity of the virus. One such interaction is between the highly glycosylated gp120 and carbohydrate-binding proteins, such as galectins. Here, we studied the interaction between HIV-1 Env and a panel of galectins and found that galectin-8 (Gal-8), bound with highest affinity (KD < 1µM) and also interacted with soluble CD4. Detailed analysis using pro

Galectin-9-driven immunosuppressive macrophage population shapes the lymph node metastatic microenvironment.

BACKGROUND: Lymph node (LN) metastasis is not only a prognostic indicator but also a driver of systemic immune suppression. However, the mechanisms of immune escape within LNs remain unclear. METHODS: Our pan-cancer single-cell transcriptomic analysis integrated public and in-house datasets from 11 cancer types, comprising 859,744 cells across 132 samples. We subsequently applied unsupervised clustering and cell-type annotation to identify dominant immunosuppressive cell populations within metastatic lymph nodes. Focused transcriptomic and pathway enrichment analyses were used to characterize

Nuclear Galectin-1 promotes KRAS-dependent activation of pancreatic cancer stellate cells.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive cancers, primarily due to its complex tumor microenvironment (TME), which drives both disease progression and therapy resistance. Understanding the molecular mechanisms governing TME dynamics is essential for developing new treatment strategies for this devastating disease. In this study, we uncover an oncogenic role for Galectin-1 (Gal1), a glycan-binding protein abundantly expressed by activated pancreatic stellate cells (PSCs), a key component of the PDAC TME that orchestrates tumor progression. Our findings reveal that G

Galectin-9 promotes colon cancer development by polarizing macrophages toward the M2 phenotype.

Galectin-9 plays multiple roles in various tumors and exerts immune regulation within the tumor microenvironment. It is closely associated with tumor prognosis. This study aimed to analyze the expression of galectin-9 in the colon tumor microenvironment. The results indicated that galectin-9 expression is higher in the colon tumor microenvironment compared to adjacent normal tissues. Furthermore, high expression of galectin-9 was related to M2-type macrophages, which promote tumor development by increasing tumor cell viability, migration, and invasion. Notably, high expression of galectin-9 in

Expression of co-signaling molecules TIM-3/Galectin-9 at the maternal-fetal interface.

INTRODUCTION: During early pregnancy, fetal placental tissue implants into maternal decidual tissue, forming a unique interface where maternal immune cells do not reject the invading fetal cells. Given the roles of Galectin-9 and Tim-3 in tumor immune regulation, studying their distribution and function at this interface may provide insights into recurrent pregnancy loss. METHODS: This study uses single-cell transcriptomics, spatial transcriptomics, and multiplex immunohistochemistry to examine the expression and localization of Galectin-9 and TIM-3. Galectin-3 gene deletion alters lipid profile in healthy and cardiomyopathy mouse hearts.

Galectin-3 (Gal3) is known to interact with glycans of proteins and lipids. In cardiovascular disease, the elevated expression of Gal3 mediates inflammation, hypertrophy and fibrosis. We explored the effect of Gal3 on the cardiac lipid profile in healthy mice or in mice with dilated cardiomyopathy (DCM). Using lipidomics and gene-targeted mouse models, we studied the influence of Gal3 gene deletion on cardiac lipid profiles in the healthy mice or mice with DCM. Cardiac-restricted transgenic activation of Hippo pathway led to DCM phenotype and Gal3 upregulation. DCM mice were cross-bred with Ga

Galectin-3 facilitates helicobacter pylori-induced apoptosis independently of sensing lysosomal damage.

Helicobacter pylori is a prevalent gastric pathogen that modulates host cell signaling pathways and represents a major risk factor for chronic gastritis, peptic ulcers, and gastric cancer. Galectin-3 is a host factor that contributes to immune regulation and cell death responses. However, its precise role in epithelial cell fate during H. pylori infection remains unclear. In this study, we demonstrate in AGS epithelial cells that H. pylori infection induces the accumulation of cytosolic galectin-3 around lysosomes damaged by the infection, detectable as puncta formation, and this process requi

Neuroprotective effects of galectin‑1 on cerebral ischemia/reperfusion injury by regulating oxidative stress.

Oxidative stress contributes to the pathology of cerebral ischemia/reperfusion (I/R) injury. Galectin-1 has shown an anti-oxidative stress effect. The present study investigated whether this anti-oxidative stress effect can account for the neuroprotective actions of galectin-1 induced by cerebral I/R injury. A cerebral I/R injury model was created in C57Bl/6 mice by transient occlusion of the middle cerebral artery, after which the mice were treated with galectin-1 for 3 days. Infarct volumes were measured. A rotarod test and neurological deficit score assessment was performed to evaluate the
