Tonic immobility and phenomenal consciousness in animals: a review.

Tonic immobility (TI) is an innate, last-resort response to the presence of a predator, commonly referred to as feigning death or thanatosis. However, it is critical to distinguish death feigning from TI; the former encompasses a complex series of behaviors, with TI representing its final aspect. Given this complexity, death feigning is hypothesized to manifest a higher-order intentionality in animals. Considering that third-order and higher intentional states are correlated with some form of phenomenal consciousness, their presence in animals has significant implications for their conscious e

Dorsal Forebrain-Specific Deficiency of Reelin-Dab1 Signal Causes Behavioral Abnormalities Related to Psychiatric Disorders.

Reelin-Dab1 signaling is involved in brain development and neuronal functions. The abnormalities in the signaling through either reduction of Reelin and Dab1 gene expressions or the genomic mutations in the brain have been reported to be associated with psychiatric disorders. However, it has not been clear if the deficiency in Reelin-Dab1 signaling is responsible for symptoms of the disorders. Here, to examine the function of Reelin-Dab1 signaling in the forebrain, we generated dorsal forebrain-specific Dab1 conditional knockout mouse (Dab1 cKO) and performed a behavioral test battery on the D

Role of hippocampal 5-HT1A receptors in the antidepressant-like phenotype of mice expressing RGS-insensitive Gαi2 protein.

A single base mutation in the Gαi2 protein (G184S) renders this Gα subunit insensitive to the negative modulatory effects of Regulator of G-protein Signaling (RGS) proteins. Mice expressing this RGS insensitive (RGSi) variant of Gαi2 (RGSi Gαi2) display a spontaneous antidepressant-like phenotype that is reversible by treatment with the 5-HT1A receptor (5-HT1AR) antagonist WAY100635. Here we test the hypothesis that increased activity of 5-HT1ARs in the hippocampus of RGSi Gαi2 knock-in mice is responsible for the expression of the observed antidepressant-like behavior. We administered the 5-H

Sex-dependent influence of chronic mild stress (CMS) on voluntary alcohol consumption; study of neurobiological consequences.

Alcohol use disorder and depression are highly comorbid, and both conditions exhibit important sexual dimorphisms. Here, we aimed to investigate voluntary alcohol consumption after 6weeks of chronic mild stress (CMS) in Wistar rats - employed as an animal model of depression. Male and female rats were investigated, and changes in several molecular markers were analysed in frontal cortex (FCx) and hippocampal formation (HF). CMS induced depressive-like responses in the forced swimming test - increased immobility time - in male and female animals, without affecting anhedonia (sucrose preference

Behavioral Diversity Across Classic Rodent Models Is Sex-Dependent.

Symptoms of trauma and stressor related disorders such as post-traumatic stress disorder (PTSD) often develop well after the traumatic experience has occurred, and so identifying early predictors of risk or resilience is important for the implementation of interventional therapies. For example, passive coping strategies such as tonic immobility and peritraumatic dissociation during the trauma itself are risk factors for the developments of PTSD, especially in women. However, discrete, sex-specific coping responses that predict later outcomes in animal models have not been rigorously defined. R

Characterization of two Indian native chicken breeds for production, egg and semen quality, and welfare traits.

The present study was conducted to characterize 2 important native chicken breeds from India and compare them on growth, production, egg and semen quality, and welfare traits. The Aseel breed showed (P < 0.001) higher BW at different ages; higher shank, radius, and toe lengths at 40 wk of age; and greater egg weights at 28, 32, and 40 wk of age than did the Kadaknath breed. The Kadaknath breed reached sexual maturity at an early age, and it had higher 40-wk egg production (P < 0.001). Higher egg specific gravity (P < 0.05) and higher albumen (P < 0.001) and shell (P < 0.009) percentages in the

NMDA receptor blockade by ketamine abrogates lipopolysaccharide-induced depressive-like behavior in C57BL/6J mice.

We have previously demonstrated that lipopolysaccharide (LPS) induces depressive-like behavior by activating indoleamine 2,3 dioxygenase (IDO; O'Connor et al, 2009c). IDO degrades tryptophan along the kynurenine pathway. Using mass-spectrometry (LC-MS) analysis of kynurenine metabolites in the brain of mice injected at the periphery with 1 mg/kg LPS, we show that LPS activates the kynurenine 3-monooxygenase pathway that ultimately degrades kynurenine into quinolinic acid. As quinolinic acid acts as an N-methyl-D-aspartate (NMDA) receptor agonist, we used the NMDA receptor antagonist ketamine t

Developmental motor deficits induced by combined fetal exposure to lipopolysaccharide and early neonatal hypoxia/ischemia: a novel animal model for cerebral palsy in very premature infants.

A critical issue in animal models of perinatal brain injury is to adapt the pertinent pathophysiological scenarios to their corresponding developmental window in order to induce neuropathological and behavioral characteristics reminiscent to perinatal cerebral palsy (CP). A major problem in most of these animal models designed up to now is that they do not present motor deficits characteristic of CP. Using a unique rat paradigm of prenatal inflammation combined to an early postnatal hypoxia-ischemia pertinent to the context of very early premature human newborns, we were interested in finding

Early-Life Social Isolation-Induced Depressive-Like Behavior in Rats Results in Microglial Activation and Neuronal Histone Methylation that Are Mitigated by Minocycline.

Early-life stress is a potent risk factor for development of psychiatric conditions such as depression. The underlying mechanisms remain poorly understood. Here, we used the early-life social isolation (ESI) model of early-life stress in rats to characterize development of depressive-like behavior, the role of microglia, levels of histone methylation, as well as expression of glutamate receptor subunits in the hippocampus. We found that depressive-like behavior was induced after ESI as determined by sucrose preference and forced swimming tests. Increased expression of microglial activation mar

Research note: approach rather than avoidance: behavioral responses of broiler chicks repeatedly exposed to an autonomous mobile robot.

Previous experience shapes animal behavior, and repeated exposure to non-aversive stimuli can reduce responses to subsequent challenges. Autonomous robots that navigate commercial broiler houses are being developed for environmental monitoring and have been shown to increase bird locomotion during their passage. However, whether this increased activity primarily reflects displacement or fear-induced avoidance, and how robot exposure influences behavioral responses, remains unclear. We assessed behavioral responses of broilers repeatedly exposed to the AviSense robot (Appelie Robotics) compared

Effects of drop height, conveyor belt speed, and acceleration on the welfare of broiler chickens in early and later life.

During automated processing in commercial hatcheries, day-old chicks are subjected to a range of possible mental and physical stressors. Three determinants of the processing line seem to have the potential to affect the birds in particular: drop height from one conveyor belt to another, conveyor belt speed, and acceleration. The aim of this study was to evaluate the effects of these 3 factors on chicken health and welfare in early and later life. In a first trial, chickens were tested on an experimental processing line that was adjusted to different levels of drop heights, belt speeds, and acc

Components of feed efficiency in broiler breeding stock: is improved feed conversion associated with increased docility and lethargy in broilers?

Two trials were conducted to test the hypothesis that broilers classified as good rather than poor feed converters (low vs. high feed-to-gain ratios, respectively) would show decreased activity (increased lethargy) and reduced fear of human caretakers. In both trials birds were reared to juvenile selection age when the 192 males with the "best" breast and leg conformation were placed into individual bird cages (46 x 60 cm), and their feed conversion ratio (FCR) was measured. In trial 1, birds remained in the cages for 7 d for assessment of feed conversion. During the feed conversion testing pe

Genetic, pharmacological and lesion analyses reveal a selective role for corticohippocampal GLUN2B in a novel repeated swim stress paradigm.

Glutamate and N-methyl-d-aspartate receptor (NMDAR) dysfunction is strongly implicated in the pathophysiology of mood and anxiety disorders. Treatment with NMDAR antagonists has antidepressant efficacy in treatment-resistant depressives. In preclinical rodent models, NMDAR antagonist administration reduces anxiety- and stress-related behaviors in concert with increases in prefrontal cortical (PFC) dendritic spinogenesis and synaptic proteins. While these effects have been attributed to actions at the NMDAR GluN2B subunit, the precise role of cortical GluN2B in mediating emotional behaviors and

Increase in cortical pyramidal cell excitability accompanies depression-like behavior in mice: a transcranial magnetic stimulation study.

Clinical evidence suggests that cortical excitability is increased in depressives. We investigated its cellular basis in a mouse model of depression. In a modified version of forced swimming (FS), mice were initially forced to swim for 5 consecutive days and then were treated daily with repetitive transcranial magnetic stimulation (rTMS) or sham treatment for the following 4 weeks without swimming. On day 2 through day 5, the mice manifested depression-like behaviors. The next and last FS was performed 4 weeks later, which revealed a 4 week maintenance of depression-like behavior in the sham m

Frequent range visits further from the shed relate positively to free-range broiler chicken welfare.

Little is known about the implications of accessing an outdoor range for broiler chicken welfare, particularly in relation to the distance ranged from the shed. Therefore, we monitored individual ranging behaviour of commercial free-range broiler chickens and identified relationships with welfare indicators. The individual ranging behaviour of 305 mixed-sex Ross 308 broiler chickens was tracked on a commercial farm from the second day of range access to slaughter age (from 16 to 42 days of age) by radio frequency identification (RFID) technology. The radio frequency identification antennas wer

Integrated Microbiome and Metabolomics Analysis Reveals Altered Aggressive Behaviors in Broiler Chickens Showing Different Tonic Immobility.

Tonic immobility (TI) serves as an indicator of innate stress response recovery in poultry. Broilers with different TI phenotypes exhibit varying levels of aggressive behavior, which can significantly impact their welfare. However, the influences of TI phenotypes on broiler aggression remain largely unexplored. In this study, broiler chickens were stratified into two distinct phenotypic groups based on the TI duration: short TI (STI) and long TI (LTI). The impacts of TI phenotypes on broiler aggression were investigated by analyzing cecal intestinal morphology, cecal bacteria, plasma metabolit

Maternally derived egg hormones, antibodies and antimicrobial proteins: common and different pathways of maternal effects in Japanese quail.

Avian eggs contain a variety of maternally-derived substances that can influence the development and performance of offspring. The levels of these egg compounds vary in relation to environmental and genetic factors, but little is known about whether there are correlative links between maternal substances in the egg underlying common and different pathways of maternal effects. In the present study, we investigated genetically determined variability and mutually adjusted deposition of sex hormones (testosterone-T, androstenedione-A4 and progesterone-P4), antibodies (IgY) and antimicrobial protei

Acute D-serine treatment produces antidepressant-like effects in rodents.

Research suggests that dysfunctional glutamatergic signalling may contribute to depression, a debilitating mood disorder affecting millions of individuals worldwide. Ketamine, a N-methyl-D-aspartate (NMDA) receptor antagonist, exerts rapid antidepressant effects in approximately 70% of patients. Glutamate evokes the release of D-serine from astrocytes and neurons, which then acts as a co-agonist and binds at the glycine site on the NR1 subunit of NMDA receptors. Several studies have implicated glial deficits as one of the underlying facets of the neurobiology of depression. The present study t
