Restriction endonuclease analysis and ribotyping differentiate Pasteurella haemolytica serotype A1 isolates from cattle within a feedlot.

Pasteurella haemolytica serotype A1 isolates were collected from cattle within a feedlot during an outbreak of bovine respiratory disease. Genetic heterogeneity among the isolates was examined by restriction endonuclease analysis (REA), ribotyping, and analysis of plasmid content. The susceptibilities of isolates to several antibiotics were also examined. Five different REA patterns and three different ribotypes were observed among the isolates. Fifty percent of the isolates had an identical REA type, ribotype, and plasmid profile. Examination of the plasmid content of the isolates revealed th

Evaluating the potential of third generation metagenomic sequencing for the detection of BRD pathogens and genetic determinants of antimicrobial resistance in chronically ill feedlot cattle.

BACKGROUND: Bovine respiratory disease (BRD) is an important cause of morbidity and mortality and is responsible for most of the injectable antimicrobial use in the feedlot industry. Traditional bacterial culture can be used to diagnose BRD by confirming the presence of causative pathogens and to support antimicrobial selection. However, given that bacterial culture takes up to a week and early intervention is critical for treatment success, culture has limited utility for informing rapid therapeutic decision-making. In contrast, metagenomic sequencing has the potential to quickly resolve all

Pyogenic cervical spondylitis caused by Pasteurella haemolytica attributed to excessive contact with dogs.

The bacterium Pasteurella haemolytica is resident in the oral cavities of dogs and cats and is often a cause of zoonotic infection. However, it is rare for it to be the pathogenic bacteria behind pyogenic spondylitis, and few studies have been conducted on it. This article reports a case of pyogenic cervical spondylitis thought to be caused by excessive contact with pet dogs.A 52-year-old man reported neck pain and pain in the right upper limb. He was admitted after plain radiographs and magnetic resonance imaging suggested pyogenic spondylitis. Pasteurella haemolytica was detected by needle a

Evolutionary genetics of Pasteurella haemolytica isolates recovered from cattle and sheep.

Genetic diversity and relationships among 194 Pasteurella haemolytica isolates, which were recovered predominantly from cattle (39%) and sheep (58%) suffering from pneumonic pasteurellosis in the United Kingdom, Germany, and the United States, were estimated by examination of allelic variation at 18 enzyme-encoding loci detected by multilocus enzyme electrophoresis. The isolates formed two major divisions. One included 178 Pasteurella haemolytica sensu stricto strains representing serotypes A1, A2, A5 to A9, A12 to A14, and A16; the other was composed of 16 isolates belonging to the A11 taxon.

Pasteurella haemolytica leukotoxin induces bovine leukocytes to undergo morphologic changes consistent with apoptosis in vitro.

Infection of the bovine lung with Pasteurella haemolytica results in an acute respiratory disorder known as pneumonic pasteurellosis. One of the key virulence determinants used by this bacterium is secretion of an exotoxin that is specific for ruminant leukocytes (leukotoxin). At low concentrations, the leukotoxin can activate ruminant leukocytes, whereas at higher concentrations, it inhibits leukocyte functions and is cytolytic, presumably as a result of pore formation and subsequent membrane permeabilization. We have investigated the possibility that the activation-inhibition paradox is expl

Effect of copper deficiency on tissue, blood characteristics, and immune function of calves challenged with infectious bovine rhinotracheitis virus and Pasteurella hemolytica.

Fourteen Holstein steers, averaging 30 d of age, were fed a semipurified diet (1.5 mg of Cu/kg) supplemented with 0 (-Cu) or 10 mg of Cu/kg of diet (+Cu) for 5 mo. Calves were then challenged by consecutive exposure to aerosol preparations of infectious bovine rhinotracheitis virus (IBRV) and Pasteurella hemolytica on d 0 and 7, respectively, of the 30-d study. Serum ceruloplasmin and plasma copper were higher in +Cu calves throughout the challenge period and increased in +Cu calves after microbial challenge. Heart weights were higher in -Cu calves, although weights of liver, spleen, and thymu

Increased citrullination and expression of peptidylarginine deiminases independently of P. gingivalis and A. actinomycetemcomitans in gingival tissue of patients with periodontitis.

BACKGROUND: A relationship between rheumatoid arthritis (RA) and periodontitis has been suggested from findings that individuals with RA are prone to have advanced periodontitis and vice versa. In search of possible common pathogenetic features of these two diseases, we investigated the presence of citrullinated proteins and expression of endogenous peptidylarginine deiminases (PAD2 and PAD4), in periodontal tissue of individuals with periodontitis and healthy controls, in relation to the periodontal pathogens Porphyromonas gingivalis (P. gingivalis) and Aggregatibacter actinomycetemcomitans (

A novel multiplex PCR-electronic microarray assay for rapid and simultaneous detection of bovine respiratory and enteric pathogens.

Respiratory and enteric diseases continue to be two major causes of economic losses to the cattle industry worldwide. Despite their multifactorial etiology, the currently available diagnostic tests for bovine respiratory disease complex (BRDC) and bovine enteric disease (BED) are single-pathogen-tests. DNA microarray when combined with multiplex polymerase chain reaction (PCR) is a powerful tool in detection and differentiation of multiple pathogens in a single sample. This study reports development and initial validation of two independent highly sensitive and specific multiplex PCR-electroni

The calf holobiont under challenge: longitudinal microbiota-pathogen dynamics and respiratory health.

The bovine respiratory disease complex (BRDC) is a major health and welfare challenge driven by multifactorial interactions among pathogens, the host, the microbiota, and environmental pressures, reflecting the holobiont nature of the calf. Many implicated bacterial pathogens are also respiratory tract commensals, complicating diagnosis, risk assessment, and prevention. Although the potential association of the respiratory and gut microbiotas in BRDC is only beginning to be recognized, their integrated effects have not yet been investigated. Here, we dynamically followed the microbiota, pathog

[Mechanisms of pathogenicity and adhesion in Pasteurella haemolytica].

Pasteurella haemolytica is one of the bacteria most commonly isolated from pneumonic cases in ruminants. Some of the mechanisms and factors involved in the pathogenesis of the disease are partially documented; and the early stages of bacterial colonization have not been totally clarified. Therefore a review is presented in this paper, particularly related with the mechanisms of bacterial pathogenicity responsible of pulmonary damage to ruminants, as well as a detailed analysis of the adherence process.

Enhancement of neutrophil-mediated injury to bovine pulmonary endothelial cells by Pasteurella haemolytica leukotoxin.

In this study, we used an in vitro coculture system to determine which virulence factor from Pasteurella haemolytica A1 was responsible for augmenting bovine polymorphonuclear neutrophil (PMN)-mediated killing of bovine pulmonary artery endothelial cells (BPAEC). A 51Cr release cytotoxicity assay was used as a measure of BPAEC killing. The mechanisms associated with this BPAEC killing were also studied. Our results demonstrated that the leukotoxin and not the lipopolysaccharide from P. haemolytica was responsible for augmenting the PMN-mediated killing of BPAEC. Furthermore, this augmented kil

Use of an indwelling bronchial catheter model of bovine pneumonic pasteurellosis for evaluation of therapeutic efficacy of various compounds.

A model of bovine pneumonic pasteurellosis, using an indwelling bronchial catheter for inoculation and subsequent lavage of a single main stem bronchus of the lung, was evaluated in a preliminary efficacy trial of an experimental therapeutic compound. Inoculation of 10(7) Pasteurella haemolytica organisms into the bronchus consistently induced a focal pneumonic lesion with typical morphology of pneumonic pasteurellosis in the left or right caudal lung lobe. The experimental treatment caused significant (P less than 0.05) reduction in lung lesion volume, compared with that of a saline-treated c

Occurrence of respiratory and enteric bacteria and viruses in calves in Iceland.

BACKGROUND: Respiratory and gastrointestinal diseases in calves pose a significant challenge to the cattle industry due to their impact on mortality rates, economic losses, animal welfare, and antimicrobial usage. Understanding which pathogens are involved is crucial for optimizing both treatment and preventive measures. In Iceland, bovine respiratory disease in calves is relatively rare, whereas neonatal calf diarrhea is more frequent. However, limited diagnostic resources in the country have resulted in a lack of knowledge about the specific pathogens involved. This study aimed to investigat

Comparison of Bacterial Community in the Jejunum, Ileum and Cecum of Suckling Lambs During Different Growth Stages.

Given that suckling lambs with immature rumen development rely on intestinal microbiota for nutrient utilization, investigating the composition and functional characteristics of their intestinal microbiota is therefore of paramount importance. In this study, 16S rRNA gene amplicon sequencing technology was adopted to characterize and analyze the diversity and composition of the jejunum, ileum and cecum bacterial communities of lambs at 0, 7 and 28 days of age, and to predict the functions of the bacterial communities. The α-diversity analysis results revealed that in the jejunum of lambs, the
