Recombinant BCG strains expressing the SIVmac251nef gene induce proliferative and CTL responses against nef synthetic peptides in mice.

CTL responses are known to be important for the control of HIV and SIV infections. Such responses are targeted against various components of these viruses including regulatory proteins like Nef. The SIVmac251nef gene was cloned in Mycobacterium bovis BCG under the control of P(AN), a promoter from Mycobacterium paratuberculosis. Nef was expressed as a fused polypeptide with ORF2, an open reading frame adjacent to P(AN). Mice inoculated with rBCG(SIVmac251nef) exhibited proliferative and CD8+ cytotoxic T-cell (CTL) responses against several Nef synthetic peptides. A mapping of the epitopes reco

A Literature Review of Selected Bacterial Diseases in Alpacas and Llamas-Epidemiology, Clinical Signs and Diagnostics.

The breeding of South American Camelids (SACs), particularly llamas and alpacas, is becoming increasingly popular in regions that are not their natural habitat, including Europe. These animals are considered to be relatively disease resistant. However, due to their growing popularity, special attention should be given to infections in llamas and alpacas. Knowledge of bacterial infections is very important to veterinarians and breeders. Many of these diseases also have zoonotic potential, so these animals must be considered as sources of potential zoonotic infections. Due to the limited informa

ALSUntangled #66: antimycobacterial antibiotics.

Several infections have been associated with motor neuron diseases resembling ALS, including species of viruses, bacteria, and parasites. Mycobacterium avium subspecies paratuberculosis (MAP), most known for its probable etiologic association with Crohn's disease, has been suggested as another possible infectious cause of motor neuron disease. Two published case reports describe the successful treatment of ALS-like symptoms with antimycobacterial antibiotics. Both cases had atypical features. Based on these, we believe it would be reasonable to begin performing chest imaging in PALS who have f

Long Non-Coding RNAs and Their "Discrete" Contribution to IBD and Johne's Disease-What Stands out in the Current Picture? A Comprehensive Review.

Non-coding RNAs (ncRNA) have paved the way to new perspectives on the regulation of gene expression, not only in biology and medicine, but also in associated fields and technologies, ensuring advances in diagnostic means and therapeutic modalities. Critical in this multistep approach are the associations of long non-coding RNA (lncRNA) with diseases and their causal genes in their networks of interactions, gene enrichment and expression analysis, associated pathways, the monitoring of the involved genes and their functional roles during disease progression from one stage to another. Studies ha

The role of infection in inflammatory bowel disease: initiation, exacerbation and protection.

Inflammatory bowel disease, a collective term for ulcerative colitis and Crohn's disease, is a chronic, immune-mediated disease of the gastrointestinal tract that develops in genetically susceptible individuals. The role of infection in the development of inflammatory bowel disease is underscored by various clinical observations, such as the delayed age of onset, suggesting that childhood exposure to pathogens is essential, and the clinical improvement that follows decreasing bacterial intestinal load. Over the years, many a pathogen has been linked to the development and exacerbation of infla

Relationship between Vaccine Application and Climate Factors in Sheep and Goat Farms in Greece.

The objectives of the present study were (a) to evaluate the importance of climate-related variables in the vaccination patterns applied in sheep and goat farms in Greece and (b) to assess potential interactions between these factors and previously established important health management- and human resources-related factors applied in the farms. Vaccination patterns against chlamydial abortion, clostridial infections, contagious agalactia, contagious ecthyma, foot-rot, paratuberculosis, pneumonia or staphylococcal mastitis were assessed. Climatic variables (2010-2019 and 2018-2019) were obtain

Pathogens of zoonotic and biological importance in roe deer (Capreolus capreolus): Seroprevalence in an agro-system population in France.

Antibody prevalence for several infectious and parasitic diseases in a population of roe deer (Capreolus capreolus) inhabiting a mixed agricultural landscape (south of France) has been analyzed. Serological analyses with ELISA in 245 animals captured from 2008 to 2012 has been performed. We found a high prevalence of Toxoplasma gondii (46.4%), Chlamydophila abortus (17.27%) and Coxiella burnetii (11.26%) compared to other studies in Europe. Seroprevalence varied strongly among years for T. gondii (27-91%), C. abortus (0-42%) and C. burnetii (0-27%). T. gondii prevalence was lower in juvenile f

From Viral Fossils to β-Cell Futures: A Retro-Exposome Framework for Type 1 Diabetes.

Type 1 diabetes affects over 8.4 million individuals worldwide, with approximately 0.5 million new cases annually, and continues to impose substantial morbidity and mortality, particularly in settings where delayed diagnosis is common. Diabetic ketoacidosis occurs at presentation in 20-80% of newly diagnosed children, underscoring persistent gaps in early detection and prevention. Recent evidence linking antibody signatures against human endogenous retroviruses and Mycobacterium avium subspecies paratuberculosis provides a mechanistically provocative perspective on disease pathogenesis. The use of a single complement fixation test technique in bovine brucellosis, Johne's disease, dourine, equine piroplasmosis and Q fever serology.

The same techniques may be used in the complement fixation test (CFT) for the serological diagnosis of bovine brucellosis, Johne's disease (paratuberculosis), dourine, equine piroplasmosis and Q fever (caused by Coxiella burnetii). The reproducibility of results is excellent, falling for the most part within the twofold range and never exceeding the fourfold range. Agreement with other laboratories is excellent (i.e. within twofold) in the case of brucellosis and equine piroplasmosis antibody titres. A good correlation between the occurrence of the disease and serological reactions is found on

Conflicting Role of Mycobacterium Species in Multiple Sclerosis.

Mycobacterium is a genus of aerobic and acid-fast bacteria, which include several pathogenic organisms that cause serious diseases in mammals. Previous studies have associated the immune response against mycobacteria with multiple sclerosis (MS), a chronic demyelinating disease of the central nervous system with unknown etiology. The role of mycobacteria in the pathological process has been controversial and often conflicting. We provide a detailed review of the mycobacteria that have been linked to MS over the last three decades, with a focus on Mycobacterium bovis bacille Calmette-Guérin vac

Inter- and intra-specific exposure to parasites and pathogens via the faecal-oral route: a consequence of behaviour in a patchy environment.

Livestock herbivores are at risk of inter- and intra-specific exposure to parasites/pathogens via the faecal-oral route during grazing. Each contact between livestock and faeces in the environment is a potential parasite/pathogen transmission event. Cattle grazing contact with faeces varies in relation to the species depositing the faeces and the distribution of the faeces. We used a foraging model to simulate the grazing behaviour of beef cattle in two grazing systems to compare the relative inter-specific and intra-specific exposure risks to parasites/pathogens. Overall, there is a greater l

Herd-level seroprevalence and risk-mapping of bovine hypodermosis in Belgian cattle herds.

Our objective was to determine the seroprevalence of Hypoderma spp. and to develop a spatial model describing the risk surface of warble-fly infection in Belgian cattle herds (adjusting for herd size, herd type, local temperature, rainfall, relative air humidity and land-cover). This survey was carried out in 390 selected herds of all types (dairy, mixed and beef) from December 1997 to March 1998, which were included in a national infectious bovine rhinotracheitis and paratuberculosis (Johne's-disease) survey. All animals >24 months old were blood sampled and an ELISA was used on pooled serum

Recombinant BCG expressing the leishmania surface antigen Gp63 induces protective immunity against Leishmania major infection in BALB/c mice.

We have cloned and expressed the gp63 gene of Leishmania major in BCG to develop a recombinant vaccine against zoonotic cutaneous leishmaniasis. Two different expression systems were investigated. The first system consists of pAN, a Mycobacterium paratuberculosis promoter, which drives expression of ORF2, an open reading frame in IS900. This system allows the production of heterologous polypeptides as hybrids with the ORF2 gene product. The second expression system relies on the production of antigenic fragments as fusion proteins with the N-terminal region of Mycobacterium fortuitum beta-lact

The application of food safety interventions in primary production of beef and lamb: a review.

The production of safe red meat depends on effective control of pathogenic microorganisms at all stages of the "farm-to-fork" chain. Eight microorganisms have been selected as the focus of the PathogenCombat project: Shiga toxin producing Escherichia coli (STEC), Mycobacterium avium subspecies paratuberculosis (Map), Listeria monocytogenes, Campylobacter jejuni, Penicillium nordicum, invasive variants of Saccharomyces cerevisiae, hepatitis E virus and tick borne encephalitis virus. The need and potential for coordinated control of the selected food-borne pathogens by on-farm interventions is a

Causality and gastrointestinal infections: Koch, Hill, and Crohn's.

The gastrointestinal tract contains a complex mix of microorganisms. Therefore, the finding of a particular microbe in the gastrointestinal tract or stools of a patient with intestinal disease does not necessarily indicate that the clinical state resulted from the presence of this organism. For this reason, extending from epidemiological association to causality is particularly challenging for gastrointestinal diseases. Most established agents of acute bacterial gastroenteritis (eg, Escherichia coli, Campylobacter jejuni) have been shown to fulfil Koch's postulates of causality in human volunt

Disposable silicon-based all-in-one micro-qPCR for rapid on-site detection of pathogens.

Rapid screening and low-cost diagnosis play a crucial role in choosing the correct course of intervention when dealing with highly infectious pathogens. This is especially important if the disease-causing agent has no effective treatment, such as the novel coronavirus SARS-CoV-2, and shows no or similar symptoms to other common infections. Here, we report a disposable silicon-based integrated Point-of-Need transducer (TriSilix) for real-time quantitative detection of pathogen-specific sequences of nucleic acids. TriSilix can be produced at wafer-scale in a standard laboratory (37 chips of 10 ×
