PURPOSE: Analysis of aqueous humor from patients with primary open-angle glaucoma (POAG) revealed marked increases in the content of endothelin-1 (ET-1) and transforming growth factor-beta (TGF-β). We determined the consequences of TGF-β signaling on ET-1 expression and secretion by human trabecular meshwork (TM) cells.
METHODS: Primary or transformed (NTM5 and GTM3) human TM cells conditioned in serum-free media were incubated in the absence or presence of TGF-β1 or -β2. Relative changes in preproendothelin (ppET)-1 mRNA content and secreted ET-1 peptide were quantified by real-time PCR and

Corneal endothelial cells respond to a circular freeze wound by undergoing actin cytoskeletal reorganization that is mainly characterized by the disappearance of circumferential microfilament bundles (CMBs) and the subsequent appearance of distinct stress fibers. This cytoskeletal rearrangement is associated with changes in cell shape as migrating cells lose their polyhedral appearance, spread out, and assume a stellate morphology with cell processes extending outward into the injured area. We report here that in the presence of low concentrations (0.01-0.l mM) of the anti-metabolite 5-fluorou

Actin-based motility (ABM) is a mechanism for intercellular spread that is utilized by vaccinia virus and the invasive bacteria within the genera Rickettsia, Listeria, and Shigella. Within the Rickettsia, ABM is confined to members of the spotted fever group (SFG), such as Rickettsia rickettsii, the agent of Rocky Mountain spotted fever. Infection by each agent induces the polymerization of host cell actin to form the typical F (filamentous)-actin comet tail. Assembly of the actin tail propels the pathogen through the host cytosol and into cell membrane protrusions that can be engulfed by neig

Effects of T8993G mutation in mitochondrial DNA (mtDNA), associated with neurogenical muscle weakness, ataxia and retinitis pigmentosa (NARP), on the cytoskeleton, mitochondrial network and calcium homeostasis in human osteosarcoma cells were investigated. In 98% NARP and rho(0) (lacking mtDNA) cells, the organization of the mitochondrial network and actin cytoskeleton was disturbed. Capacitative calcium entry (CCE) was practically independent of mitochondrial energy status in osteosarcoma cell lines. The significantly slower Ca(2+) influx rates observed in 98% NARP and rho(0), in comparison t

Objectives: Critical-sized bone defects continue to represent a substantial challenge in orthopedic clinical practice. Decellularized bone matrix (DBM) possesses favorable osteoconductive properties due to its retention of native extracellular matrix architecture and collagen components. However, its limited osteogenic bioactivity restricts its application in complex bone defect repair. This study aimed to construct a dendritic mesoporous silica (DMSN)-modified DBM composite scaffold loaded with teriparatide (DBM-DMSN@TPTD) and to systematically evaluate its physicochemical properties, drug re

BACKGROUND: Achieving rapid bone fusion is critical for preventing complications in Extreme Lateral Interbody Fusion (XLIF), yet harvesting sufficient autologous bone presents surgical limitations. Platelet-rich fibrin (PRF), an autologous biomaterial rich in osteogenic growth factors, offers potential as a bone graft enhancer. This study evaluated the efficacy of PRF combined with allogeneic bone in promoting interbody fusion using an XLIF-simulated rabbit model.
METHODS: In vitro, bone marrow mesenchymal stem cells were cultured with PRF, allogeneic bone, or PRF/allogeneic bone composites.

BACKGROUND: Diabetic vascular remodeling is the most important pathological basis of diabetic cardiovascular complications. The accumulation of advanced glycation end products (AGEs) caused by elevated blood glucose promotes the proliferation and migration of vascular smooth muscle cells (VSMCs), leading to arterial wall thickening and ultimately vascular remodeling. Therefore, the excessive proliferation and migration of VSMCs is considered as an important therapeutic target for vascular remodeling in diabetes mellitus. However, due to the lack of breakthrough in experiments, there is current

Tetrandrine (TET) is a bisbenzylisoquinoline alkaloid with promising anticancer activity, although its limited selectivity toward malignant over non-cancerous cells restricts its therapeutic potential. A series of mono- and bis-N-substituted tetrandrine derivatives was therefore investigated to determine whether N-functionalization could improve the cytotoxic selectivity of the parent scaffold. The series included the two new compounds MU1NT and DPBT. Following initial screening in non-cancerous ARPE-19 retinal epithelial cells, selected derivatives were evaluated against 22Rv1 prostate carcin

ETHNOPHARMACOLOGICAL RELEVANCE: Depression has emerged as a significant public health concern in recent years. Evidence suggests that aberrant hippocampal synaptic plasticity plays a central role in the cognitive impairment associated with depression. Nevertheless, the precise underlying mechanisms remain poorly understood. Although Jiawei Sini San (JWSNS) has shown significant clinical efficacy in treating depression, its specific role in improving cognitive function requires further investigation.
AIM OF THE STUDY: To elucidate the role and mechanism of JWSNS in enhancing cognitive function

This study aimed to elucidate the mechanism of Shenmai Injection(SMI) in inducing immediate hypersensitivity reaction by activating the Ras homolog gene family member A(RhoA)/Rho-associated coiled-coil forming protein kinase(ROCK) signaling pathway. In the in vivo experiment, ICR mice were randomly divided into a negative control group, SMI groups of low-dose(7.5 mL·kg~(-1)), medium-dose(15 mL·kg~(-1)), and high-dose(30 mL·kg~(-1)), as well as a positive control group. The effect of SMI on vascular leakage was detected by mouse auricular blue staining, and histopathological changes in the lung

Stochastic optical reconstruction microscopy (STORM) is a single-molecule localization microscopy technique that enables visualization of cellular structures beyond the diffraction limit. This approach has revealed previously inaccessible ultrastructural details in a wide range of cellular components, including the actin cytoskeleton, clathrin-coated pits, mitochondria, and bacterial nucleoid-associated proteins. STORM relies on the sequential emission of single photons from photosensitive fluorophores, which are precisely localized before entering a dark state or undergoing photobleaching. By

BACKGROUND: Circular RNAs (circRNAs), characterized by their covalently closed-loop structures, constitute a distinct class of non-coding RNAs. They play pivotal regulatory roles within cells and are intricately associated with the progression of malignant tumors. However, their roles and the underlying mechanisms in nasopharyngeal carcinoma (NPC) progression have yet to be fully uncovered and comprehensively understood.
METHODS: Employing RNA sequencing technology, high-abundance circular RNAs in NPC were identified. Expression analysis of circCLASP2 in NPC tissues was conducted using quanti

OBJECTIVE: Diabetic cardiomyopathy (DCM) is the most common cardiovascular complication of type 2 diabetes mellitus (T2DM). Patients affected with DCM face a notably higher risk of progressing to congestive heart failure compared to other populations. Myocardial hypertrophy, a clearly confirmed pathological change in DCM, plays an important role in the development of DCM, with abnormal Ca2+ homeostasis serving as the key signal to induce myocardial hypertrophy. Therefore, investigating the mechanism of Ca2+ transport is of great significance for the prevention and treatment of myocardial hyper

UNLABELLED: Osteogenesis imperfecta (OI) is a collagen-related disorder associated to dominant, recessive or X-linked transmission, mainly caused by mutations in type I collagen genes or in genes involved in type I collagen metabolism. Among the recessive forms, OI types VII, VIII, and IX are due to mutations in CRTAP, P3H1, and PPIB genes, respectively. They code for the three components of the endoplasmic reticulum complex that catalyzes 3-hydroxylation of type I collagen α1Pro986. Under-hydroxylation of this residue leads to collagen structural abnormalities and results in moderate to letha

PURPOSE: To investigate histamine-induced changes in the phosphorylation of myosin light chain (MLC) and its influence on the barrier integrity of corneal endothelial cells through altered contractility of the actin cytoskeleton.
METHODS: Experiments were performed in cultured bovine corneal endothelial cells (BCECs). Phosphorylation of MLC, which increases contractility of the actin cytoskeleton through actomyosin interaction, was assessed by urea-glycerol gel electrophoresis and Western blot analysis. Immunocytochemistry was used to locate phosphorylated MLC in relation to tight junctions.
