Histone serotonylation in dorsal raphe nucleus contributes to stress- and antidepressant-mediated gene expression and behavior.

Mood disorders are an enigmatic class of debilitating illnesses that affect millions of individuals worldwide. While chronic stress clearly increases incidence levels of mood disorders, including major depressive disorder (MDD), stress-mediated disruptions in brain function that precipitate these illnesses remain largely elusive. Serotonin-associated antidepressants (ADs) remain the first line of therapy for many with depressive symptoms, yet low remission rates and delays between treatment and symptomatic alleviation have prompted skepticism regarding direct roles for serotonin in the precipi

Effects of repeated social stress on risk assessment behaviors and response to diazepam in the elevated plus maze in adult male rats.

Anxiety is highly common, and stress is a major trigger for anxiety. Anxiety includes heightened threat assessment and avoidance, but we do not fully understand which components are sensitive to stress. Rodents show a balance of exploration and avoidance that incorporates threat assessment prior to making the relatively risky decision to explore an open area. The purpose of this study was to determine if stress impacts risk assessment and if this is tied to the effects of stress on exploration. The present study used elevated plus maze (EPM) to test the effects of repeated social defeat stress

Adolescent binge drinking causes sex-specific deficits in behavioral reactivity, endocrine recovery from stress and altered expression of hypothalamic KCC2 in males.

RATIONALE: Binge drinking during adolescence is associated with a higher risk of developing Alcohol Use Disorders and impaired stress reactivity in adulthood. The hypothalamic-pituitary-adrenal axis matures during adolescence and is under strict GABAergic control. Yet, how early binge drinking alters GABAergic signaling in this region, and whether these changes contribute to later-life disruptions in stress reactivity, remain unknown. OBJECTIVES: Our goal with these set of experiments was to test the hypothesis that adolescent binge drinking would impair maturation of GABAergic signaling in t

Xiaoyao San, a Chinese herbal formula, ameliorates depression-like behavior in mice through the AdipoR1/AMPK/ACC pathway in hypothalamus.

OBJECTIVE: Depression and metabolic disorders have overlapping psychosocial and pathophysiological causes. Current research is focused on the possible role of adiponectin in regulating common biological mechanisms. Xiaoyao San (XYS), a classic Chinese medicine compound, has been widely used in the treatment of depression and can alleviate metabolic disorders such as lipid or glucose metabolism disorders. However, the ability of XYS to ameliorate depression-like behavior as well as metabolic dysfunction in mice and the underlying mechanisms are unclear. METHODS: An in vivo animal model of depr

G protein-coupled receptor 75 mediates depression-like behaviors and HPA axis hyperactivity through an excitatory arcuate-paraventricular nucleus circuit.

Metabolic dysregulation is a core pathological feature of and a major risk factor for depression. In this study, the role of G protein-coupled receptor 75 (GPR75), a brain-enriched metabolic sensor, in the pathogenesis of depression was explored and its potential role as a therapeutic target was evaluated. In chronic social defeat stress (CSDS) models, GPR75 expression was specifically upregulated in the arcuate nucleus glutamatergic (ARCVglut2) neurons of susceptible mice, a finding corroborated by analyses of postmortem brains from patients with depression. Conditional (GPR75-cKO) or local (

Accumbal Dopamine Responses Are Distinct between Female Rats with Active and Passive Coping Strategies.

There is a gap in existing knowledge of stress-triggered neurochemical and behavioral adaptations in females. This study was designed to explore the short-term consequences of a single social defeat (SD) on accumbal dopamine (DA) dynamics and related behaviors in female Wistar rats. During the SD procedure, rats demonstrated different stress-handling strategies, which were defined as active and passive coping. The "active" subjects expressed a significantly higher level of activity directed toward handling stress experience, while the "passive" ones showed an escalated freezing pattern. Remark

Identification of a Novel Functional Non-synonymous Single Nucleotide Polymorphism in Frizzled Class Receptor 6 Gene for Involvement in Depressive Symptoms.

Although numerous susceptibility loci for depression have been identified in recent years, their biological function and molecular mechanism remain largely unknown. By using an exome-wide association study for depressive symptoms assessed by the Center for Epidemiological Studies Depression (CES-D) score, we discovered a novel missense single nucleotide polymorphism (SNP), rs61753730 (Q152E), located in the fourth exon of the frizzled class receptor 6 gene (FZD6), which is a potential causal variant and is significantly associated with the CES-D score. Computer-based in silico analysis reveale

Putting Your Trauma Lens On.

Trauma in childhood is now understood to cause long-term effects on the brain and body. The pediatric provider, using a "trauma lens," which constitutes observing a child's attachment, resilience, and stress response, is well poised to identify and support children and families at risk. Fortunately, resilience is a dynamic process that can be learned, enhanced, and supported. Familiarity with the most common symptoms of traumatic stress will help the medical provider quickly recognize which children are impacted or FRAYED (Fits, Frets, and Fear; Regulation disorders; Attachment problems; Yawni

Polyphenolic Compounds Alter Stress-Induced Patterns of Global DNA Methylation in Brain and Blood.

SCOPE: Stress is a known contributor to various forms of disease in humans and animals, although mechanisms are still unknown. In animals, psychosocial stress-induced depression/anxiety phenotypes are coincidental with increased inflammation in both brain and blood. The authors recently showed that a novel treatment with a select bioactive polyphenol preparation promotes resilience to stress-mediated depression/anxiety phenotypes mice. Moreover, selective bioactive phenolic compounds within the polyphenol preparation are identified that are effective in mitigating the behavioral effects of bon

Genome-wide methylation patterns associated with chronic stress.

BACKGROUND: Chronic social defeat stress (CSDS) is a validated animal model for depression that produces sustained behavioral and transcriptional changes in the brain, notably the nucleus accumbens (nAcc). RESEARCH DESIGN AND METHODS: We used genome-wide analysis of cytosine methylation patterns in mouse nAcc following CSDS to identify candidate epigenetic mechanisms. RESULTS: CSDS produced extensive differential methylation, increasing CG hypermethylation compared to control conditions; non-CG methylation showed the opposite trend. Highly differentially methylated (DM) regions included seve

Curculigoside Alleviates CSDS-induced Depressive-like Behavior by Modulating Pyramidal Neuron Excitability and Synaptic Transmission.

Curculigoside (CUR), a flavonoid derived from traditional Chinese medicine, has shown potential in alleviating depressive symptoms. However, the mechanisms underlying its antidepressant effects remain unclear. The aim of this study was to investigate the effect of CUR on depressive-like behavior in the chronic social defeat stress (CSDS) model and its potential mechanism of action on pyramidal neuronal excitability and synaptic transmission. We injected 20 mg/kg of CUR intraperitoneally for seven days and found that CUR significantly alleviated emotional deficits in mice subjected to CSDS. CUR

Unravelling the complexity of suicidality: a network analysis of theory-driven and culturally relevant suicide risk factors in a country with high suicide rates.

PURPOSE: This study aimed to explore whether individual theory-based constructs are sufficient in explaining suicidality in a culture marked by high suicide rates and whether, if any, culturally relevant factors need to be considered additionally. METHODS: The study used a community sample of 1873 Lithuanian adults (age M = 43.61 SD = 16.96, 69.1% female). A survey using online and pen-and-paper formats was conducted, measuring suicidality, the main constructs from two predominant theories of suicide (thwarted belongingness, perceived burdensomeness, acquired capability for suicide, and defea

LHPP in Glutamatergic Neurons of the Ventral Hippocampus Mediates Depression-like Behavior by Dephosphorylating CaMKIIα and ERK.

BACKGROUND: LHPP was recently shown to be a risk gene for major depressive disorder. LHPP has been proven to dephosphorylate the residues of histidine, serine, threonine, and tyrosine. However, much remains unknown about how LHPP contributes to depression. METHODS: In the current study, we addressed this issue by integrating approaches of genetics, molecular biology, behavioral testing, and electrophysiology. RESULTS: We found that levels of LHPP were upregulated in glutamatergic neurons of the ventral hippocampus in mice that displayed stress-induced depression-like behaviors. Knockout of L

Icariin reverses corticosterone-induced depression-like behavior, decrease in hippocampal brain-derived neurotrophic factor (BDNF) and metabolic network disturbances revealed by NMR-based metabonomics in rats.

Previously published reports have revealed the antidepressant-like effects of icariin in a chronic mild stress model of depression and in a social defeat stress model in mice. However, the therapeutic effect of icariin in an animal model of glucocorticoid-induced depression remains unclear. This study aimed to investigate antidepressant-like effect and the possible mechanisms of icariin in a rat model of corticosterone (CORT)-induced depression by using a combination of behavioral and biochemical assessments and NMR-based metabonomics. The depression model was established by subcutaneous injec

Is shame the missing link between traumatic experiences and post-traumatic stress disorder in Burundian children living on the streets?

BACKGROUND: Shame is an emotion reflecting an anticipated social devaluation of the self. It is strongly associated with experiences of humiliation and rejection in early life. Individuals suffering from post-traumatic stress disorder (PTSD) often struggle with shame. However, little is known about how shame contributes to the development and maintenance of PTSD symptoms in children. The present study investigated the ways childhood exposure to human-induced traumatic events promotes a coping mechanism of defeat and withdrawal facilitated by the experience of shame. We tested a dose-response r

Stress and brain immunity: Microglial homeostasis through hypothalamus-pituitary-adrenal gland axis and sympathetic nervous system.

Stress has been well documented to bring about various clinical disorders, ranging from neurodegeneration, such as Parkinson's (PD) and Alzheimer's diseases (AD), to metabolic disorders including diabetes mellitus. Importantly, microglia, immunocompetent cells in the brain, have been shown to be involved in these clinical disorders. In the recent studies aiming to clarify the microglial responses, microglia are found to be quite responsive to stressful events, such as acute, subchronic, chronic stress, and social defeat stress. However, the mechanisms of these stress response on microglial act

Burns in pandemic times - The Graz way towards COVID-19 and back.

The first half of the year 2020 has been shaped by quarantines and lock-downs all over the world. The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused a pandemic, that slowed down not only social interactions and economy, but also medical and health care. Governments and hospitals were forced to create ad hoc emergency plans maintaining the balance between an adequate participation in collective response of shutting-down to avoid a further spreading of the virus, while preserving the ongoing acute care and simultaneously being able to react to an imminent overexten

Multimodal MRI signatures and concurrent myelin remodeling alterations in the anterior cingulate cortex correlate with the antidepressant-like effects of 18β-glycyrrhetinic acid in CSDS mice.

BACKGROUND: The anterior cingulate cortex (ACC) regulates emotion, and its dysfunction is associated with depression. 18β-glycyrrhetinic acid (18β-GA) exerts robust antidepressant effects; however, its roles in neural circuit remodeling of the ACC and underlying molecular mechanisms remain unclear. This study investigated its phenotypic changes within the ACC in chronic social defeat stress (CSDS) mice using multimodal MRI, histopathology and molecular assays. METHODS: Male C57BL/6J mice with CSDS depression model received 18β-GA treatment. We detected ACC neuroimaging alterations via multimo
