Immunochemical characterization of the deficiency of the alpha-ketoglutarate dehydrogenase complex in thiamine-deficient rat brain.

Mitochondrial dysfunction is a common feature of many neurodegenerative disorders. The metabolic encephalopathy caused by thiamine deficiency (TD) is a classic example in which an impairment of cerebral oxidative metabolism leads to selective cell death. In experimental TD in rodents, a reduction in the activity of the thiamine diphosphate-dependent, mitochondrial enzyme alpha-ketoglutarate dehydrogenase complex (KGDHC) occurs before the onset of pathologic lesions and is among the earliest biochemical deficits found. To understand the molecular basis and the significance of the deficiency of

[Posttraumatic regeneration of the skeletal muscles in certain forms of hypovitaminosis B1].

Light microscopy was employed to study the development of posttraumatic aseptic inflammation and subsequent regeneration in the affected musculus soleus in different patterns of hypovitaminosis B1. Alimentary deficiency of thiamine entailed pronounced retardation of the post-traumatic inflammation. The growth of macrophage density and formation of the pool of the regenerating cells were also retarded. On day 21 the regeneration pool showed an elevated content of the connective tissue. During long-term administration of oxythiamine in a dose of 10 mg/kg with a 12-hour interval, the development

IL21-STAT3 controls the pentose phosphate pathway to support metabolic reprogramming and tumor progression in chronic lymphocytic leukemia.

Studying how microenvironmental cues influence metabolic reprogramming can uncover mechanisms driving tumor progression. Using an in vitro model with proliferative stimuli of the in vivo lymph node niche (LN)-including interleukin-21 (IL-21)-we examined metabolic rewiring in chronic lymphocytic leukemia (CLL) cells. We found that the metabolic intermediates of upper glycolysis and its branching pathways are key in fulfilling metabolic demands of proliferating CLL cells. Among branching pathways, the pentose phosphate pathway (PPP) was the most transcriptionally upregulated in proliferating CLL

Use of dihydroxyacetone synthase for the assimilation of methanol in E. coli.

Dihydroxyacetone synthase (Das) is a key enzyme of the xylulose monophosphate (XuMP) pathway, catalysing the assimilation of formaldehyde in methylotrophic yeasts. In this study, we first performed a systematic screening of twenty-five Das variants in combination with a methanol dehydrogenase (Mdh) from Acinetobacter gerneri to identify the Das variants supporting the most efficient methanol assimilation.13C-methanol labelling experiments revealed marked differences in 13C incorporation into central carbon metabolism among the various Das variants and enabled the identification of the Pichia a

Clinical characterization, genetic mapping and whole-genome sequence analysis of a novel autosomal recessive intellectual disability syndrome.

We identified six patients presenting with a strikingly similar clinical phenotype of profound syndromic intellectual disability of unknown etiology. All patients lived in the same village. Extensive genealogical work revealed that the healthy parents of the patients were all distantly related to a common ancestor from the 17th century, suggesting autosomal recessive inheritance. In addition to intellectual disability, the clinical features included hypotonia, strabismus, difficulty to fix the eyes to an object, planovalgus in the feet, mild contractures in elbow joints, interphalangeal joint

Biosynthesis of the benzylpyrrolidine precursor in anisomycin by a unique ThDP-dependent enzyme.

Anisomycin (compound 1), a multifunctional pyrrolidine antibiotic, primarily inhibits protein biosynthesis by binding to the ribosome. Upon binding to the ribosome, the para-phenol moiety of anisomycin inserts completely into the hydrophobic crevice of the A-site and blocks the access of the incoming aminoacyl-tRNAs, disrupting peptide bond formation. Hence, the para-methoxyphenyl group serves as a starting point for developing novel anisomycin analogs with potent antifungal and insecticidal properties. However, the activation and condensation mechanism of phenylpyruvic acid has not yet been e
