Molecular detection and phylogenetic characterization of Anaplasma phagocytophilum in brown bears (Ursus arctos) in Slovakia.

Wild animals play a crucial role in the maintenance of tick-borne pathogens, acting as reservoir and sentinel hosts within natural ecosystems. The ongoing expansion of tick populations across Europe has intensified the circulation of zoonotic tick-borne pathogens, including Anaplasma phagocytophilum. Although the epidemiology of this bacterium has been extensively investigated in domestic animals and small wild mammals, the contribution of large omnivores such as the brown bear (Ursus arctos) remains poorly understood, particularly with respect to their exposure to genetically distinct lineage

Biotransformed bear bile powder ameliorates diet-induced nonalcoholic steatohepatitis in mice through modulating arginine biosynthesis via FXR/PXR-PI3K-AKT-NOS3 axis.

NASH is a highly prevalent metabolic syndrome that has no specific approved agents up to now. BBBP, which mainly contains bile acids, possess various pharmacological properties and some bile acids are available for NASH treatment. Herein, the therapeutic effects and underlying mechanisms of BBBP against NASH were systemically evaluated. In this study, mice received an HFHS diet over a 20-week period to induce NASH with or without BBBP intervention were used to evaluate the effect and underlying mechanisms of BBBP against NASH. Our results demonstrated that BBBP attenuated hepatic steatosis, re

Usefulness of MRI Slices Parallel to the Optic Chiasma in a Case with Traumatic Optic Nerve Avulsion after a Bear Attack.

Optic nerve avulsion is an exceedingly rare condition. Here, we describe a case of optic nerve avulsion in a 74-year-old man with temporal hemianopia in the contralateral eye after a bear attack. Magnetic resonance imaging (MRI) revealed separation of the optic nerve distal to the optic chiasma, whereas the high signal in diffusion-weighted imaging suggested nerve injury from the left side of the optic chiasma to the left optic tract. MRI slices parallel to the optic chiasma were obtained and used for evaluating the site of optic nerve avulsion and nerve injury, which were responsible for temp

Insights from the Den: How Hibernating Bears May Help Us Understand and Treat Human Disease.

Hibernating brown bears (Ursus arctos) and black bears (Ursus americanus) spend half of the year in a physically inactive state inside their winter dens without food intake and defecating and no or little urination. Under similar extreme conditions, humans would suffer from loss of lean body mass, heart failure, thrombosis, azotemia, osteoporosis, and more. However, bears exit the den in the spring strong without organ injuries. Translational animal models are used in human medicine but traditional experimental animals have several shortcomings; thus, we believe that it is time to systematical

Himalayan black bear mauling: offense or defense?

The Asiatic Black Bear (Ursus thibetanus or Selenarctos thibetanus), also known as the Tibetan black bear, the Himalayan black bear, or the moon bear is a omnivorous mammal. This animal is declared threatened animal and rarely comes in human contact. Recent decrease in forest area has, however, increased the chances of bear-human interaction, hence causing injuries to humans. There is only one published report in English literature on Himalayan black bear mauling. We present 5 cases referred to our department over a period of 1 year.

Parasites in grizzly bears from the central Canadian Arctic.

Standardized flotation techniques were used to survey 56 grizzly bear (Ursus arctos) fecal samples for parasites. The samples were collected during the spring and autumn of 1995 and 1996 in the central Arctic of the Northwest Territories (Canada). Parasites of the genera Nematodirus, gastrointestinal coccidia, and an unidentified first stage protostrongylid larva are reported for the first time from grizzly bear feces in North America. Parasites of the genera Diphyllobothrium and Baylisascaris also were collected. Prevalence of gastrointestinal parasites were significantly different between th

Trichinosis surveillance, United States, 1986.

Trichinella spiralis is a parasite of carnivorous animals that causes the disease trichinosis. In the United States, people become infected by eating poorly cooked pork products or wild animal meat that is infected with the parasite. Although fewer than 100 cases per year are reported to CDC, trichinosis continues to persist as a public health problem in this country. Public health officials believe that the reported cases represent only a fraction of the total number of cases, since many of the mild or asymptomatic infections are undetected or are misdiagnosed unless they are related to more

A treatment strategy with nifedipine versus labetalol for women with pregnancy hypertension: study protocol for a randomised controlled trial (Giant PANDA).

BACKGROUND: Approximately one in ten women have high blood pressure during pregnancy. Hypertension is associated with adverse maternal and perinatal outcomes, and as treatment improves maternal outcomes, antihypertensive treatment is recommended. Previous trials have been unable to provide a definitive answer on which antihypertensive treatment is associated with optimal maternal and neonatal outcomes and the need for robust evidence evaluating maternal and infant benefits and risks remains an important, unanswered question for research and clinical communities. METHODS: The Giant PANDA study

Hibernating bear serum hinders osteoclastogenesis in-vitro.

Bears do not suffer from osteoporosis during hibernation, which is associated with long-term inactivity, lack of food intake, and cold exposure. However, the mechanisms involved in bone loss prevention have scarcely been elucidated in bears. We investigated the effect of serum from hibernating Japanese black bears (Ursus thibetanus japonicus) on differentiation of peripheral blood mononuclear cells (PBMCs) to osteoclasts (OCs). PBMCs collected from 3 bears were separately cultured with 10% serum of 4 active and 4 hibernating bears (each individual serum type was assessed separately by a bear P

Elevated skull fractures in pediatric age group: report of two cases.

Elevated fractures of the skull, which are rarely reported in the literature, are always compound, have maximal neurological deficits at presentation and have been reported only in adults. We report two cases of elevated skull fractures in the pediatric age group, one of which was a simple elevated fracture and presented with delayed neurological deterioration. The etiologies were a fall in first case and an animal attack (bear maul) in the second case as reported for the first time. One of the cases presented with delayed onset of left focal hemispheric signs. The first case underwent debride

Immobilization of black bears (Ursus americanus) with a combination of butorphanol, azaperone, and medetomidine.

Sixteen captive and five free-ranging black bears (Ursus americanus) were immobilized with a combination of butorphanol, azaperone, and medetomidine (BAM). The BAM drug combination was premixed using 0.5 ml butorphanol (30 mg/ml), 0.25 ml azaperone (50 mg/ml), and 0.25 ml medetomidine (20 mg/ml) per milliliter to yield a final mix of (15 mg butorphanol+12.5 mg azaperone+5 mg medetomidine)/ml. This combination, dosed at 0.4 ml BAM/approximately 23 kg estimated body weight, provided a mean induction time of 10 min (95% confidence interval [CI]=2 min), consistent anesthesia without apparent adver

Spatial and temporal trends and effects of contaminants in the Canadian Arctic marine ecosystem: a review.

Recent studies have added substantially to our knowledge of spatial and temporal trends of persistent organic pollutants and heavy metals in the Canadian Arctic marine ecosystem. This paper reviews the current state of knowledge of contaminants in marine biota in the Canadian Arctic and where possible, discusses biological effects. The geographic coverage of information on contaminants such as persistent organochlorines (OCs) (PCBs, DDT- and chlordane-related compounds, hexachlorocyclohexanes, toxaphene) and heavy metals (mercury, selenium, cadmium, lead) in tissues of marine mammal and sea bi

PCR assays for detection of Baylisascaris procyonis eggs and larvae.

The objective of this study was to develop polymerase chain reaction (PCR) assays for detection of Baylisascaris procyonis eggs and larvae in fecal, environmental, and tissue samples. We have optimized conventional and real-time PCR assays for B. procyonis using the mitochondrial cytochrome oxidase 2 gene as the target for amplification. The lower limit of detection of the parasite genomic DNA was 10 pg in the conventional PCR and 100 fg in the real-time PCR. In both PCR assays, specific amplification of a 146 bp product was achieved with DNA extracted from a single in vitro hatched B. procyon

Bear bile: dilemma of traditional medicinal use and animal protection.

Bear bile has been used in Traditional Chinese Medicine (TCM) for thousands of years. Modern investigations showed that it has a wide range of pharmacological actions with little toxicological side effect and the pure compounds have been used for curing hepatic and biliary disorders for decades. However, extensive consumption of bear bile made bears endangered species. In the 1980's, bear farming was established in China to extract bear bile from living bears with "Free-dripping Fistula Technique". Bear farming is extremely inhumane and many bears died of illness such as chronic infections and

Serologic survey of trichinellosis in wild mammals kept in a Mexico City Zoo.

A serologic survey of Trichinella infection was carried out to determine the prevalence of this parasitosis among wild mammals kept in captivity at the Chapultepec Zoo. This was prompted by the necropsy finding of a heavy Trichinella infection in a Canadian polar bear (Ursus maritimus) that had been kept at the Zoo for more than 11 years. The parasites recovered were identified as T. nativa (T2). A serologic study based on ELISA and Western blot analysis was performed in serum samples from two polar bears (U. maritimus), six wolves (Canis lupus); nine foxes (Urocyon cinereoargenteus); seven co

Whole-genome analysis of escherichia coli isolated from captive giant pandas (ailuropoda melanoleuca) at the Dujiangyan base of the China conservation and research center for the giant panda, Sichuan, China.

The giant panda (Ailuropoda melanoleuca) is a rare and vulnerable species facing threats from bacterial infections. The extensive use of antibiotics in disease prevention has raised concerns about antibiotic resistance, which reduces treatment efficacy and poses environmental and public health risks. This study aims to analyze the resistance profiles of Escherichia coli (E. coli) in captive pandas, providing insights into the antibiotic resistance genes within their gut microbiota. Twenty-two E. coli isolates were obtained from the feces of 22 captive giant pandas, and whole-genome sequencing

The first detection of Babesia species DNA from Japanese black bears (Ursus thibetanus japonicus) in Japan.

In this study, we tried to detect protozoan blood parasites from the liver or blood of 156 Japanese black bears (Ursus thibetanus japonicus) in Iwate Prefecture of Japan by polymerase chain reaction. Two amplicons (approximately 540 bp and 480 bp) were detected by amplification for V4 hyper-variable regions of the 18S rRNA gene. Approximately 540-bp products were obtained in 119 samples (76.3%) and were considered to be DNA of Hepatozoon ursi. Approximately 480-bp products were obtained in 22 samples (14.1%) and were considered to be DNA of Babesia species. The nucleotide sequences (1635 bp) o

Comparison and optimization of ancient DNA extraction.

Ancient DNA analyses rely on the extraction of the tiny amounts of DNA remaining in samples that are hundreds to tens of thousands of years old. Despite the critical role extraction efficiency plays in this field of research, no study has comprehensively compared ancient DNA extraction techniques to date. There are a wide range of methods currently in use, which rely on such disparate principles as spin columns, alcohol precipitation, or binding to silica. We have compared a number of these methods using quantitative PCR and then optimized each step of the most promising method. We found that
