Changes in microbiome composition after fecal microbiota transplantation via oral gavage and magnetic navigation technology-assisted proximal colon/cecum enema in antibiotic knock-down rats: a comparative experimental study.

BACKGROUND: Fecal microbiota transplantation (FMT) transfers fecal matter from a donor into the gastrointestinal tract of a recipient to induce changes to the gut microbiota for therapeutic benefit; however, differences in the composition of gut microbiota after FMT via different donor material delivery routes are poorly understood. In this study, we first developed a novel technique for FMT, magnetic navigation technology(MAT)-assisted proximal colon enemas, in healthy Sprague-Dawley rats. Besides, the difference in fecal microbiota composition after FMT via oral gavage and proximal colon/cec

Wolfberry prevented liver damage caused by anti-tuberculosis drugs associated with the YAP1/FXR pathway through gut microbiota.

BACKGROUND: The prevalence of anti-tuberculosis drug-induced liver injury (AT-DILI) is significant, with severe cases potentially leading to liver failure or mortality. Research indicates that first-line anti-tuberculosis drugs (ATDs), including rifampicin (RIF) and isoniazid (INH), cause a lasting disruption of gut flora, which is significantly associated with drug-induced toxicity. Wolfberry (Lycium barbarum L.) is frequently utilized in traditional Chinese medicine for the treatment of hepatic and renal disorders. The mechanism by which wolfberry prevents AT-DILI remains unclear. This work

Compound of Lactobacillus reuteri and Candida rugosa supplementation alleviates intestinal barrier lesion via improving the gut microbiota in broilers infected with Salmonella Typhimurium.

As a common zoonotic pathogen, Salmonella Typhimurium (S. Typhimurium) impairs the growth performance of broilers and also poses a threat to public health through the food chain. Probiotics as a kind of green microecological agents have been widely used to improve intestinal health. This study investigated the protective effect of combined use of Lactobacillus reuteri (L. reuteri) and Candida rugosa (C. rugosa) on reducing S. Typhimurium infection in broilers. A total of 144 one-day-old Arbor Acres (AA) broilers were randomly assigned to four groups: control (Con) group fed with a basal diet,

Identification of an epilepsy-linked gut microbiota signature in a pediatric rat model of acquired epilepsy.

A dysfunctional gut microbiota-brain axis is emerging as a potential pathogenic mechanism in epilepsy, particularly in pediatric forms of epilepsy. To add new insights into gut-related changes in acquired epilepsy that develops early in life, we used a multi-omics approach in a rat model with a 56% incidence of epilepsy. The presence of spontaneous seizures was assessed in adult rats (n = 46) 5 months after status epilepticus induced by intra-amygdala kainate at postnatal day 13, by 2 weeks (24/7) ECoG monitoring. Twenty-six rats developed epilepsy (Epi) while the remaining 20 rats (No-Epi) di

Bile acids supplementation improves colonic mucosal barrier via alteration of bile acids metabolism and gut microbiota composition in goats with subacute ruminal acidosis (SARA).

Subacute ruminal acidosis (SARA) is a common metabolic disease due to feeding high-concentrate (HC) diets to ruminants, especially dairy cows, in intensive farming system. Long term feeding HC diets commonly induce damages to hindgut barrier, leading to the translocation of harmful substances such as endotoxins (LPS) from lumen to blood, which results in a low-grade inflammation and stress response. Secondary bile acids (SBAs) play an important role in maintaining intestinal homeostasis. However, the function of SBAs on the intestinal epithelial barrier in SARA remains unclear. In this study,

Dietary Triple-Strain Bacillus-Based Probiotic Supplementation Improves Performance, Immune Function, Intestinal Morphology, and Microbial Community in Weaned Pigs.

Probiotics provide health benefits and are used as feed supplements as an alternative prophylactic strategy to antibiotics. However, the effects of Bacillus-based probiotics containing more than two strains when supplemented to pigs are rarely elucidated. SOLVENS (SLV) is a triple-strain Bacillus-based probiotic. In this study, we investigate the effects of SLV on performance, immunity, intestinal morphology, and microbial community in piglets. A total of 480 weaned pigs [initial body weight (BW) of 8.13 ± 0.08 kg and 28 days of age] were assigned to three treatments in a randomized complete b

Calpain-1: a Novel Antiviral Host Factor Identified in Porcine Small Intestinal Mucus.

The thick mucus layer covering of the intestinal epithelium has received increasing attention, owing to its protective role in intestinal infection. However, the exact mechanisms by which the mucus increases intestinal resistance against viral infection remain largely unclear. Here, we identify prominent antiviral activity of the small intestinal mucus and extracted total mucus proteins, as evidenced by their inhibitory effects against porcine epidemic diarrhea virus (PEDV) infection. Of all the extracted mucus proteins, mucin 2 and fraction III (~70 kDa) exhibited potent antiviral activity. W

Effects of three host-associated Bacillus species on mucosal immunity and gut health of Nile tilapia, Oreochromis niloticus and its resistance against Aeromonas hydrophila infection.

Skin and intestinal mucosa lymphoid tissues are known to be the fish's first line of defence since they serve as the first point of contact for pathogens. Only few studies have investigated the influence of host-associated Bacillus on mucosal immunity. In this study, the effects of three host-associated Bacillus species on mucosal immunity, intestinal morphology, intestinal digestive enzymes activity, intestinal microbiome and resistance of Nile tilapia against Aeromonas hydrophila infection was evaluated. The fish were divided into five treatment groups and fed with diets containing no bacter

An integrated methodology for assessing the impact of food matrix and gastrointestinal effects on the biokinetics and cellular toxicity of ingested engineered nanomaterials.

BACKGROUND: Engineered nanomaterials (ENMs) are increasingly added to foods to improve their quality, sensory appeal, safety and shelf-life. Human exposure to these ingested ENMs (iENMS) is inevitable, yet little is known of their hazards. To assess potential hazards, efficient in vitro methodologies are needed to evaluate particle biokinetics and toxicity. These methodologies must account for interactions and transformations of iENMs in foods (food matrix effect) and in the gastrointestinal tract (GIT) that are likely to determine nano-biointeractions. Here we report the development and appli

IQ Motif-Containing GTPase-Activating Protein 2 (IQGAP2) Is a Novel Regulator of Colonic Inflammation in Mice.

IQ motif-containing GTPase-activating protein 2 (IQGAP2) is a multidomain scaffolding protein that plays a role in cytoskeleton regulation by juxtaposing Rho GTPase and Ca2+/calmodulin signals. While IQGAP2 suppresses tumorigenesis in liver, its role in pathophysiology of the gastrointestinal tract remains unexplored. Here we report that IQGAP2 is required for the inflammatory response in colon. Mice lacking Iqgap2 gene (Iqgap2-/- mice) were resistant to chemically-induced colitis. Unlike wild-type controls, Iqgap2-/- mice treated with 3% dextran sulfate sodium (DSS) in water for 13 days displ

Dimethyl itaconate attenuates dextran sulfate sodium-induced ulcerative colitis in BALB/c mice: investigations on the TXNIP/NLRP3 inflammasome signaling pathway.

BACKGROUND: Ulcerative colitis (UC) involves recurrent colonic mucosal inflammation and impaired epithelial repair. Current therapies are limited due to safety concerns. The TXNIP-NLRP3 inflammasome axis serves as a critical link between oxidative stress and inflammation in UC. The present study evaluated the protective role of dimethyl itaconate (DMI), a cell-permeable itaconate derivative, in a chronic dextran sulfate sodium (DSS)-induced ulcerative colitis model with verapamil as a positive control. METHODOLOGY: Chronic colitis was induced in male BALB/c mice with repeated cycles of 3% DSS

GABABR activation alleviates Streptococcus equi subsp. zooepidemicus-induced acute lung injury by suppressing inflammation and oxidative stress in pulmonary macrophages via IRAK3.

UNLABELLED: Streptococcus equi subsp. zooepidemicus (SEZ) infection poses a significant threat to animals and humans, causing acute lung injury driven by excessive inflammation and tissue destruction. The gamma-aminobutyric acid type B receptor (GABABR) has been implicated in modulating inflammatory responses, but its specific role in SEZ-induced acute lung injury remains unexplored. This study aimed to investigate the protective effects of pharmacological GABABR activation against SEZ-triggered pulmonary damage. Employing a mouse model of SEZ infection and MH-S alveolar macrophages, we demons

Multi-omics elucidation of HDRSD alleviating allergic rhinitis via IL-1β/TRPV1 pathway regulation.

ETHNOPHARMACOLOGICAL RELEVANCE: Han-Duo-Re-Shao Decoction (HDRSD), a traditional Korean medicine, effectively treats cold-pattern allergic rhinitis (AR). Its mechanism is rooted in addressing the lung-spleen-kidney qi-yang deficiency triad, which heightens rhinitis susceptibility. Guided by the principle of 'warming yang to dissipate cold', HDRSD fortifies body resistance and applies the 'cultivating earth to generate metal' theory to nourish the lungs. This study integrates ethnopharmacology with network pharmacology to elucidate HDRSD's "multi-component, multi-target, multi-pathway" mechanis
