An ER-targeted electronic-switched cascade probe for synchronous dual-channel imaging of H2S and HClO during liver fibrosis.

BACKGROUND: The endoplasmic reticulum (ER) relies on a delicate redox balance, primarily orchestrated by crucial biomolecules like hydrogen sulfide (H2S) and hypochlorous acid (HClO). The synchronous monitoring of these multiple biomarkers is critical for deciphering the complex redox signaling intricately associated with pathological processes such as liver fibrosis. While simultaneous dual-channel imaging offers a comprehensive view of cellular redox remodeling, developing precise analytical tools remains highly challenging. Traditional cascade-responsive fluorescent probes inherently suffer

Development of Covalent Small-Molecule Fluorescent Probes for DNA Methyltransferase 1 Detection in Cancer Cells and Cervical Exfoliated Cells.

DNA methyltransferase 1 (DNMT1) overexpression is associated with aberrant methylation and tumorigenesis, making its detection vital for tumor diagnosis. In this study, RG108 derivatives bearing cysteine-targeted covalent moieties were constructed as warheads for the DNMT1 detectors. Following affinity assessment by surface plasmon resonance, warheads containing a 2-fluoroacrylamido moiety were selected for preparing fluorescein-labeled probes 20a and 20b. In-gel fluorescence scanning and competitive assays confirmed that the probes can covalently bind to DNMT1 at the S-adenosyl-l-homocysteine

Dual-ligand Zr-based metal-organic framework probe with turn-on fluorescent response for dual-channel live cell imaging of phosphate and glutathione.

In diseases associated with oxidative stress, such as chronic renal failure, alcoholic liver disease, and hyperthyroidism, both phosphate and reduced glutathione (GSH) levels significantly deviate from the normal physiological range. Therefore, simultaneous monitoring of phosphate and GSH levels is of great clinical significance for disease diagnosis and prognosis. Herein, an innovative dual-detection fluorescent probe constructed on a dual-ligand engineered Zr-based metal-organic framework (Zr-MOF) platform is introduced. This probe is designed for the simultaneous detection of phosphate and

Fluorescent Peptide Probe for Specific Detection and In Vivo Imaging of Fusobacterium Nucleatum.

Fusobacterium nucleatum (F. nucleatum), an opportunistic oral anaerobic pathogen, is closely associated with various oral disorders and colorectal cancer (CRC). Current detection methods for this bacterium suffer from technical complexity and labor intensiveness. In this study, we engineered TPAI-YQPVPE, a peptidic fluorescent probe, by conjugating an aggregation-induced emission (AIE) fluorophore (TPAI) with the F. nucleatum-targeting peptide YQPVPE. This probe binds specifically to FomA, a major outer membrane protein of F. nucleatum, enabling its selective accumulation and robust fluorescen

Lipid droplet-targeted and ratiometric fluorescent probe for imaging ClO- in Rheumatoid arthritis model.

BACKGROUND: Rheumatoid arthritis (RA) is a long-term, systemic autoimmune disease that primarily affects the synovial joints, causing chronic inflammation and eventual joint destruction. The underlying cause of RA is strongly associated with the excessive production of hypochlorite (ClO-). Increased levels of ClO- can lead to oxidative damage in cells, playing a crucial role in the development and progression of the disease. Despite advances in research, detecting RA at its early stages remains difficult, especially since existing ratiometric fluorescent probes face significant challenges in a

Investigating the photosensitivity of koneramines for cell imaging and therapeutic applications.

The photophysical properties of the anthracene appended koneramines (LAn) were analyzed and utilized as a chemosensor for the selective detection of Cd2+ and Zn2+. The complexation-induced inhibition of PET (photo-induced electron transfer) from the chelating nitrogen atoms to the excited state of the anthracene moiety resulted in a fluorescence "turn-on" signal upon binding with Cd2+ and Zn2+. The confocal microscopic imaging studies performed on the MCF-7 cells validated that the compound is potentially useful for detecting Cd2+ and Zn2+ inside the cells. The cadmium complex exhibited unique

AIE-active photoresponsive Ir(III) complex as an oxidase mimic for ratiometric ascorbic acid detection.

Ascorbic acid (AA) sustains biological redox homeostasis, hence its rapid and precise detection bears important research value. Conventional nanozyme-based detection strategies are hampered by complicated synthesis, inflexible enzyme-like activity modulation, single detection mode and unsatisfactory anti-interference ability. In this work, an amino-modified Ir(III) complex with aggregation-induced near-infrared emission property was facilely synthesized. The product can self-assemble into photoresponsive oxidase-mimetic nanoparticles. Upon light irradiation, Ir-1 NPs exhibit remarkable catalyt

A Near-Infrared Fluorescent Molecular Rotor Conjugated with the Aβ Self-Assembly Core Sequence for Selective Imaging of Amyloid Plaques In Vivo.

Abnormal aggregation of amyloid-β (Aβ) is a key pathological hallmark of Alzheimer's disease (AD), motivating the development of probes with high sensitivity and selectivity. Most existing fluorescent probes for Aβ42 aggregates are based on molecular rotor scaffolds, typically incorporating N,N-dialkylaniline motifs. Such fluorescent molecular rotors, however, often exhibit a nonspecific response to the β-sheet-rich misfolded protein aggregates, limiting their specificity for Aβ plaque imaging in complex biological environments. Herein, we report a near-infrared (NIR) fluorescent molecular rot

Ratiometric fluorescence detection of aflatoxin B1 in food samples using metal-organic framework confined within mesoporous silica.

Aflatoxin B1 (AFB1) is a highly toxic and carcinogenic mycotoxin that poses significant threats to food safety and human health. A composite material, formed by confining Zr-based metal-organic framework (Zr-LMOF) within aminated mesoporous silica (ASBA-15) and integrating with Ru(bpy)32+, provides a stable and sensitive ratiometric fluorescence-sensing platform for AFB1. The ratiometric detection strategy, based on the fluorescence ratio of 600 nm (Ru(bpy)32+) to 423 nm or 400 nm (Zr-LMOF), significantly improved detection accuracy by suppressing external interferences. The probe achieved a d

Constructing a CA15-3 Ultrasensitive Fluorescent Probe Based on Dual-Ligand Peptides/UiO-66-NH2@AYG Composite Material.

Carbohydrate antigen 15-3 (CA15-3) is an important biomarker for early breast cancer diagnosis and therapeutic monitoring. This work presents an ultrasensitive fluorescent probe based on a dual-ligand peptides/UiO-66-NH2@AYG composite for CA15-3 detection. Dual-ligand peptides (GTTFSNYW and MHYLEYPF) target distinct CA15-3 epitopes, enhancing the binding affinity and specificity. The peptides are immobilized on UiO-66-NH2, whose high surface area supports efficient loading of the fluorescent reporter Acridine Yellow G (AYG). This probe demonstrates exceptional performance with a linear detecti

A CQDs-doped europium-DPA framework for fluorescence and colorimetric dual-mode sensing of amoxicillin and ciprofloxacin.

Amoxicillin (AMO), a penicillin antibiotic, and ciprofloxacin (CIP), a quinolone antibiotic, are widely used to combat bacterial infections and are frequently co-administered to enhance therapeutic efficacy. However, improper use of AMO and CIP may lead to antibiotic residues in the environment, posing serious threats to human health. The visual detection of AMO and CIP remains significant challenges. Herein, the Eu-DPA@CQDs was used for the dual-mode detection of AMO and CIP, which was synthesized by CQDs doped in europium-DPA metal-organic framework. In the colorimetric mode, the catalase-mi

A lipid droplet-targeted fluorescent probe for selective imaging of cysteine fluctuations in drug-induced liver injury.

Cysteine (Cys), an essential endogenous biothiol, plays crucial roles in cellular redox homeostasis, metabolic regulation, and detoxification. Abnormal Cys fluctuations are closely associated with various pathological processes, including liver injury. Lipid droplets are dynamic organelles involved in lipid metabolism and stress responses and are highly relevant to hepatic injury progression. Thus, fluorescent probes capable of both lipid droplet localization and Cys-specific recognition are valuable tools for visualizing Cys fluctuations in drug-induced liver injury-related microenvironments.

Development of a Rhodamine-Based Chemosensor for Cd2+ Ions: A Combined Approach of Colorimetric, Fluorescent Detection, and Computational Modelling.

This paper presents the design and synthesis of a new rhodamine-based Schiff base chemosensor (L) with exceptional selectivity and sensitivity for Cd2+. An analysis of L was conducted using various spectroscopic techniques, thereby validating its structure and activity. The chemosensor exhibited a clear turn-on fluorescence response and significant colour change with Cd2+ ions in acetonitrile. Binding investigations confirmed that L interacts with Cd2+ ions in a 1:1 ratio, as shown by Job's plot and Benesi-Hildebrand (B-H) analysis. The binding constant (Ka = 0.107 × 104) highlights the strong

Machine Learning-Enabled Multidimensional Ratiometric Sensor Array for Accurate Discrimination and Quantification of Tetracyclines.

Extensive use of tetracycline antibiotics (TCs) threatens ecosystems and human health, necessitating sensitive and selective analytical monitoring approaches. Herein, we report an amino-functionalized mesoporous-silica-confined nanosensor (PLNP@SiO2-NH2) that enables self-referenced ratiometric detection and pattern recognition-based discrimination and quantification of TCs. Aminated mesoporous silica effectively enriches and rigidly immobilizes TCs, triggering the turn-on of the intrinsic green fluorescence signal (IG) of TCs. Meanwhile, the strong UV absorption of TCs at 254 nm induces an in

Development of a ratiometric fluorescent probe for selective detection and bioimaging of hydrogen peroxide in HeLa cells and zebrafish.

Because intracellular H2O2 levels vary dynamically and are involved in both redox signaling and oxidative stress linked to human health and disease, quantitative fluorescent detection of H2O2 is essential in living systems. In this work, a carbazole-based ratiometric fluorescent probe, KD-H2O2, was designed and synthesized for the selective detection of H2O2. Performance evaluation demonstrated that KD-H2O2 exhibited excellent sensing properties, including a broad pH tolerance range (pH 3.0-11.0), high detection sensitivity with a low limit of detection (LOD) of 2.74 μM, and favorable biocompa

Rational design of halogen-substituted near-infrared fluorescent probes for butyrylcholinesterase: from drug screening to imaging and therapeutic evaluation in Alzheimer's disease.

Monitoring butyrylcholinesterase (BuChE) activity is crucial for tracking Alzheimer's disease (AD) progression and evaluating therapeutics; however, high-performance near-infrared (NIR) probes with rapid response and clear design principles remain scarce. Here, we report a series of dicyanoisophorone-based NIR fluorescent probes engineered via a "halogen effect" to systematically tune reactivity toward BuChE. Through spectroscopic screening, the CF3-substituted probe DCNC7 emerged as the optimal candidate, benefiting from the strong electron-withdrawing and hydrophobic nature of the trifluorom

Visualization of Cysteine-Mediated Redox Imbalance under Environmental Exposures via π-Bridge-Tailored Fluorescent Probes.

Disturbance of cellular redox homeostasis is often accompanied by aberrant fluctuations in cysteine (Cys) and reactive oxygen species (ROS) levels, and simultaneous monitoring of their fluctuations provides critical insights into redox imbalance mechanisms. However, precise monitoring of such redox disorders under environmental stress remains largely underexplored, limiting understanding of stress-induced physiological disorders and related toxicological mechanisms. Herein, two Cys-activated fluorescent probes featuring furan and thiophene π-bridge modifications, Afd-Cys and Atd-Cys, were desi

Internalized Liquid Crystal Microcapsules for pH Sensing in Living Single Cells.

Liquid crystal (LC) materials provide a unique sensing platform because their long-range molecular order is highly sensitive to interfacial physicochemical changes. However, most LC-based systems operate at extracellular interfaces, limiting their applicability for intracellular analysis in living cells. Here, we report cell-internalized liquid crystal microcapsules for configuration-based monitoring of local intracellular pH-responsive changes. The sensing platform, denoted M-E7PBA, consists of an E7 liquid crystal core doped with pentyl biphenyl carboxylic acid (PBA) and encapsulated within

NIR-II Ratiometric Imaging of SOD Activity with a Superoxide-Responsive Nanoprobe.

Superoxide dismutase (SOD) maintains redox homeostasis, and its activity reflects oxidative stress in aging, inflammation, and diseases. However, real-time monitoring of SOD activity in vivo remains challenging. Here we constructed a superoxide anion radical (O2•-)-responsive NIR-II ratiometric probe based on a "substrate-monitoring and activity-back-calculation" strategy. The probe consists of an O2•--responsive unit (NKO, emission at 950 nm) and a stable internal reference unit (PbS quantum dots, emission at 1665 nm) conjugated via polyethylene glycol, affording robust aqueous dispersibility
