Biofilm inhibition and antagonism of Klebsiella pneumoniae by probiotic lactic acid bacteria (LAB) isolated from raw cow milk.

Lactic acid bacteria (LAB) with their potential health benefits are naturally prevalent in dairy and fermented food products. This probiotic microbiota can be an alternative biological tool for controlling other pathogenic bacteria. The study aimed to isolate lactic acid bacteria (LAB) from raw cow milk and evaluate their probiotic potential. Twelve gram-positive isolates showing tolerance to bile salt, acid, and low pH were identified by 16S rRNA sequencing, which revealed the isolates belong to the genera including Lactococcus, Enterococcus, Streptococcus, Bacillus, and Weissella. In case of

Designing and Cloning of the Gene Vaccine Carrying the Viral Haemorrhagic Septicaemia Multi-Epitope Gene in the pNZ8121 Secretion Vector.

The viral haemorrhagic septicaemia virus (VHSV) is considered a very significant disease affecting fish. The objective of this project is to develop a multi-epitope vaccine targeting viral hemorrhagic fever using the reverse vaccinology approach. Epitope prediction was conducted using the immunogenic components of VHSVV. A multi-epitope vaccine was developed by combining the immunogenic proteins' most potent B- and T-cell epitopes with the adjuvant. Following that, a comprehensive evaluation was carried out on many facets of the formulated vaccine, including its physicochemical properties, ant

Biological functional identification of holins from Salmonella pullorum phages and their therapeutic efficacy against pullorum disease in chickens.

Salmonella pullorum (S.pullorum) is a significant pathogen that seriously endangers the poultry industry. However, its prevention and control mainly rely on antibiotics, which leads to the wide spread of drug-resistant genes and poses a serious public health risk. Phage lytic proteins have received extensive attention in recent years as potential alternatives to antibiotics. In this study, whole genome sequencing and phylogenetic analysis demonstrated that Salmonella Pullorum phage vB_SpuM_23 is most closely related to Salmonella phages SWJM-02 and SalM SPJ41. Two holin proteins (Holin-1 and H

Nisin lantibiotic prevents NAFLD liver steatosis and mitochondrial oxidative stress following periodontal disease by abrogating oral, gut and liver dysbiosis.

Oral microbiome dysbiosis mediates chronic periodontal disease, gut microbial dysbiosis, and mucosal barrier disfunction that leads to steatohepatitis via the enterohepatic circulation. Improving this dysbiosis towards health may improve liver disease. Treatment with antibiotics and probiotics have been used to modulate the microbial, immunological, and clinical landscape of periodontal disease with some success. The aim of the present investigation was to evaluate the potential for nisin, an antimicrobial peptide produced by Lactococcus lactis, to counteract the periodontitis-associated gut d

Soluble Lactobacillus delbrueckii subsp. bulgaricus 92059 PrtB proteinase derivatives for production of bioactive peptide hydrolysates from casein.

The proteinase-encoding prtB gene of Lactobacillus (Lb.) delbrueckii (d.) subsp. bulgaricus 92059 was cloned and sequenced. Two soluble, secreted, C-terminally His-tagged derivatives were constructed and expressed in Lactococcus lactis by means of the NICE® Expression System. In both obtained derivatives PrtBb and PrtB2, the C-terminal, cell wall-binding domain was deleted. In addition, in derivative PrtB2, the C-terminal part of the B domain was deleted and the signal sequence was replaced by a lactococcal export signal. The affinity-purified derivatives were both proteolytically active. Pept

Novel particulate vaccines utilizing polyester nanoparticles (bio-beads) for protection against Mycobacterium bovis infection - a review.

Bovine tuberculosis (TB) continues to be a major health problem in cattle and development of a safe effective vaccine to control TB in cattle would be very useful. This paper reviews progress and provides new data in development of a TB bio-bead vaccine based on polyester nanoparticle inclusions which were produced by bioengineered bacteria. Polyhydroxybutyrate (PHB) biopolyester nanoparticles (bio-beads) have been produced which displayed mycobacterial antigens, Ag85A and ESAT-6, on the surface of the bio-beads for use as vaccines for the control of tuberculosis. Bio-beads were purified from

Genomic and surface proteomic analysis of the canine pathogen Staphylococcus pseudintermedius reveals proteins that mediate adherence to the extracellular matrix.

Cell wall-associated (CWA) proteins made by Gram-positive pathogens play a fundamental role in pathogenesis. Staphylococcus pseudintermedius is a major animal pathogen responsible for the canine skin disease bacterial pyoderma. Here, we describe the bioinformatic analysis of the family of 18 predicted CWA proteins encoded in the genome of S. pseudintermedius strain ED99 and determine their distribution among a phylogenetically diverse panel of S. pseudintermedius clinical isolates and closely related species of the Staphylococcus intermedius group. In parallel, we employed a proteomic approach

Bioactive peptides encrypted in milk proteins: proteolytic activation and thropho-functional properties.

The bioactivities of peptides encrypted in major milk proteins are latent until released and activated by enzymatic proteolysis, e.g. during gastrointestinal digestion or food processing. The proteolytic system of lactic acid bacteria can contribute to the liberation of bioactive peptides. In vitro, the purified cell wall proteinase of Lactococcus lactis was shown to liberate oligopeptides from beta- and alpha-caseins which contain amino acid sequences present in casomorphins, casokinines, and immunopeptides. The further degradation of these peptides by endopeptidases and exopeptidases of lact

Oral probiotic promotes indoleamine 2,3-dioxygenase- and TGF-β-Producing plasmacytoid dendritic cells to initiate protection against type 1 diabetes.

Colonization factor antigen I (CFA/I) fimbria, an adhesin from enterotoxigenic Escherichia coli, confers protection in murine autoimmune models for type 1 diabetes (T1D), multiple sclerosis, and rheumatoid arthritis. Although CFA/I fimbriae's initial mode of action is in a bystander or in an antigen (Ag)-independent fashion, protection is ultimately dependent upon the induction and/or activation of auto-Ag-specific regulatory T cells (Tregs). However, little is known about how protection transitions from bystander suppression to Ag-specific Tregs. Since dendritic cells (DCs) play an integral r

[Sweet protein brazzein as a promising sweetener].

The excessive consumption of sugar-containing foods contributes to the development of a number of diseases, including obesity, diabetes mellitus, etc. As a substitute for sugar, people with diabetes mellitus and obesity most often use sweeteners. Sweet proteins, in particular brazzein, are an alternative to synthetic sweeteners that have natural origin, are broken down in the intestines along with food proteins, and do not affect blood sugar and insulin levels. The purpose of the review was to analyze the available data on the sweet protein brazzein, its physical and chemical properties, exist

New potentially antihypertensive peptides liberated in milk during fermentation with selected lactic acid bacteria and kombucha cultures.

Compounds with the ability to inhibit angiotensin-converting enzyme (ACE) are used medically to treat human hypertension. The presence of such compounds naturally in food is potentially useful for treating the disease state. The goal of this study was to screen lactic acid bacteria, including species commonly used as dairy starter cultures, for the ability to produce new potent ACE-inhibiting peptides during milk fermentation. Strains of Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus helveticus, Lactobacillus paracasei, Lactococcus lact

Combined metagenomic and metabolomic analyses reveal gut microbiota dysbiosis and metabolic dysfunction in pediatric neurodevelopmental disorders.

INTRODUCTION: Neurodevelopmental disorders (NDDs) are chronic brain diseases linked to innate immune signaling abnormalities, affecting children with complex gut-brain axis etiologies and limited targeted therapies. While infant microbes/metabolites may predict childhood NDDs, their landscape and host-metabolism interactions in NDDchildren remain unclear. METHODS: This study enrolled 40 NDDchildren (mean age: 5.18 ± 1.77, F:M = 11:29) and 60 healthy controls (HCs; mean age:5.11 ± 1.42, F:M = 25:35) from Gansu Province Hospital Rehabilitation Center. Spinal Cord Injury Changes the Structure and Functional Potential of Gut Bacterial and Viral Communities.

Emerging data indicate that gut dysbiosis contributes to many human diseases, including several comorbidities that develop after traumatic spinal cord injury (SCI). To date, all analyses of SCI-induced gut dysbiosis have used 16S rRNA amplicon sequencing. This technique has several limitations, including being susceptible to taxonomic "blind spots," primer bias, and an inability to profile microbiota functions or identify viruses. Here, SCI-induced gut dysbiosis was assessed by applying genome- and gene-resolved metagenomic analysis of murine stool samples collected 21 days after an experiment

Dynamic alterations of the intestinal microbiome and metabolome during transmissible gastroenteritis virus infection in weaned pigs.

Transmissible gastroenteritis virus (TGEV) infection induces diarrhea in piglets by targeting the small intestine, especially the jejunum and ileum. However, dynamic changes in the gut microbiota and metabolome during TGEV infection remain unclear. This study investigated these alterations and their association with intestinal damage in weaned pigs during early TGEV infection. Thirty 4-week-old pigs were allocated randomly into TGEV-inoculated and mock groups. On days 3, 5, and 7 postinoculation, intestinal tissue and fecal samples were collected. Full-length 16 S rRNA sequencing and ultrahigh

Avidumicin, a novel cyclic bacteriocin, produced by Cutibacterium avidum shows anti-Cutibacterium acnes activity.

The prevalence of antimicrobial-resistant Cutibacterium acnes in acne patients has increased owing to inappropriate antimicrobial use. Commensal skin bacteria may play an important role in maintaining the balance of the skin microbiome by producing antimicrobial substances. Inhibition of Cu. acnes overgrowth can prevent the development and exacerbation of acne vulgaris. Here, we evaluated skin bacteria with anti-Cu. acnes activity. Growth inhibition activity against Cu. acnes was tested using 122 strains isolated from the skin of healthy volunteers and acne patients. Comparative genomic analys

Different Group A Streptococcus pili lead to varying proinflammatory cytokine responses and virulence.

The human pathogen Streptococcus pyogenes, or Group A Streptococcus (GAS), is associated with a variety of diseases ranging from mild skin and soft tissue infections to invasive diseases and immune sequelae such as rheumatic heart disease. We have recently reported that one of the virulence factors of this pathogen, the pilus, has inflammatory properties and strongly stimulates the innate immune system. Here we used a range of nonpathogenic Lactococcus lactis gain-of-function mutants, each expressing one of the major pilus types of GAS, to compare the immune responses generated by various type
