A bigenic mouse model of FSGS reveals perturbed pathways in podocytes, mesangial cells and endothelial cells.

Focal segmental glomerulosclerosis is a major cause of end stage renal disease. Many patients prove unresponsive to available therapies. An improved understanding of the molecular basis of the disease process could provide insights leading to novel therapeutic approaches. In this study we carried out an RNA-seq analysis of the altered gene expression patterns of podocytes, mesangial cells and glomerular endothelial cells of the bigenic Cd2ap+/-, Fyn-/- mutant mouse model of FSGS. In the podocytes we observed upregulation of many genes related to the Tgfβ family/pathway, including Gdnf, Tgfβ1,

The Human FSGS-Causing ANLN R431C Mutation Induces Dysregulated PI3K/AKT/mTOR/Rac1 Signaling in Podocytes.

BACKGROUND: We previously reported that mutations in the anillin (ANLN) gene cause familial forms of FSGS. ANLN is an F-actin binding protein that modulates podocyte cell motility and interacts with the phosphoinositide 3-kinase (PI3K) pathway through the slit diaphragm adaptor protein CD2-associated protein (CD2AP). However, it is unclear how the ANLN mutations cause the FSGS phenotype. We hypothesized that the R431C mutation exerts its pathogenic effects by uncoupling ANLN from CD2AP. METHODS: We conducted in vivo complementation assays in zebrafish to determine the effect of the previously

MAGI-1 Interacts with Nephrin to Maintain Slit Diaphragm Structure through Enhanced Rap1 Activation in Podocytes.

MAGI-1 is a multidomain cytosolic scaffolding protein that in the kidney is specifically located at the podocyte slit diaphragm, a specialized junction that is universally injured in proteinuric diseases. There it interacts with several essential molecules, including nephrin and neph1, which are required for slit diaphragm formation and as an intracellular signaling hub. Here, we show that diminished MAGI-1 expression in cultured podocytes reduced nephrin and neph1 membrane localization and weakened tight junction integrity. Global magi1 knock-out mice, however, demonstrated normal glomerular

Fyn Mediates High Glucose-Induced Actin Cytoskeleton Reorganization of Podocytes via Promoting ROCK Activation In Vitro.

Fyn, a member of the Src family of tyrosine kinases, is a key regulator in cytoskeletal remodeling in a variety of cell types. Recent studies have demonstrated that Fyn is responsible for nephrin tyrosine phosphorylation, which will result in polymerization of actin filaments and podocyte damage. Thus detailed involvement of Fyn in podocytes is to be elucidated. In this study, we investigated the potential role of Fyn/ROCK signaling and its interactions with paxillin. Our results presented that high glucose led to filamentous actin (F-actin) rearrangement in podocytes, accompanied by paxillin

Effects of interleukin-13 and montelukast on the expression of zonula occludens-1 in human podocytes.

PURPOSE: The aim of this study was to investigate whether pathologic changes in zonula occludens-1 (ZO-1) are induced by interleukin-13 (IL-13) in the experimental minimal-change nephrotic syndrome (MCNS) model and to determine whether montelukast, a leukotriene receptor antagonist, has an effect on ZO-1 restoration in cultured human podocytes. MATERIALS AND METHODS: Human podocytes cultured on bovine serum albumin-coated plates were treated with different doses of IL-13 and montelukast and then examined for distribution using confocal microscopy and for ZO-1 protein levels using Western blot

mTORC1 activation in podocytes is a critical step in the development of diabetic nephropathy in mice.

Diabetic nephropathy (DN) is among the most lethal complications that occur in type 1 and type 2 diabetics. Podocyte dysfunction is postulated to be a critical event associated with proteinuria and glomerulosclerosis in glomerular diseases including DN. However, molecular mechanisms of podocyte dysfunction in the development of DN are not well understood. Here we have shown that activity of mTOR complex 1 (mTORC1), a kinase that senses nutrient availability, was enhanced in the podocytes of diabetic animals. Further, podocyte-specific mTORC1 activation induced by ablation of an upstream negati

Clinical significance of fibroblast-specific protein-1 expression on podocytes in patients with focal segmental glomerulosclerosis.

BACKGROUNDS/AIMS: We previously reported that fibroblast-specific protein 1 (FSP1) is a marker of epithelial-mesenchymal transition (EMT) in tubulointerstitial fibrosis. The EMT-like changes observed in podocytes are reportedly associated with podocyte detachment which may cause focal glomerulosclerosis. METHODS: In cross-sectional studies, we analyzed podocyte expression of FSP1 immunohistochemically using renal biopsy specimens from 31 patients with focal segmental glomerulosclerosis (FSGS) and 39 patients with minimal change disease (MCD). We also semiquantitatively analyzed glomerular exp

Podocytes are the major source of IL-1 alpha and IL-1 beta in human glomerulonephritides.

To address the question of in situ production of IL-1 alpha and IL-1 beta in proliferative and non-proliferative forms of human glomerulonephritis (GN), we performed immunocytochemical and in situ hybridization studies on renal biopsies from patients with mesangial IgA-GN (N = 38), idiopathic membranous GN (MGN; N = 12), minimal change disease (MCD; N = 9), focal segmental glomerulosclerosis (FSGS; N = 5) and acute endocapillary GN (AGN; N = 3). Normal kidneys (N = 10) served as controls. Concomitantly, the expression of IL-1 receptor type I (IL-1 RI), IL-1 receptor type II (IL-1 RII) and of I

Targeting DPP9 attenuates podocyte injury by regulating NRF2 antioxidant signaling.

INTRODUCTION: Podocyte injury is a critical event in the progression of chronic kidney disease (CKD), with oxidative stress as a central pathogenic mechanism. Despite the efficacy of finerenone in mitigating CKD progression, its precise cytoprotective mechanisms within podocytes remain incompletely defined. This study aimed to elucidate the role of dipeptidyl peptidase 9 (DPP9) in podocyte injury and determine whether finerenone exerts its renoprotective effects via DPP9-mediated antioxidant effects. METHODS: Integrated bioinformatic analyses of single-cell and bulk transcriptomic databases (

Nanotechnology-mediated podocyte injury repair: mechanistic exploration and therapeutic prospects.

Podocyte injury serves as a central pathological driver in chronic kidney diseases (CKD), including diabetic kidney disease (DKD) and IgA nephropathy (IgAN). However, conventional therapies are still limited by poor targeting efficacy and systemic side effects. Nanotechnology provides transformative strategies via tunable nanomedicines, which enables precise podocyte repair through targeted drug delivery, antioxidant and anti-inflammatory effects, as well as gene regulation. This review exclusively focuses on the preclinical research progress in this field. Three categories of nanomaterials: p

Emerging roles of S100 proteins in podocyte biology and disease: A mini review.

The glomerular podocyte is the final barrier preventing urinary protein loss, relying on a resilient ultrastructure exposed to shear stress and hydrostatic forces. While often affected secondarily by systemic disease, podocytes are primary targets in conditions such as nephrotic syndrome. S100 proteins, involved in diverse intra- and extracellular functions, are linked to several diseases, yet their roles in podocytes remain poorly defined. The available podocyte-specific literature remains sparse, but emerging studies suggest that selected S100 proteins may be associated with podocyte injury

IgM Hyposialylation Modulates Podocyte Vulnerability in Patients With Idiopathic Nephrotic Syndrome.

INTRODUCTION: Altered Ig glycosylation has been implicated in antibody-mediated podocytopathies; however, the functional relevance of IgM sialylation remains poorly defined. Previous evidence suggests that circulating cationic or hyposialylated IgM may contribute to podocyte vulnerability in idiopathic nephrotic syndrome (iNS). METHODS: Serum IgM from 86 pediatric and adult patients with podocytopathies, 20 patients with membranous nephropathy (MN), 20 patients with lupus nephritis (LN), and 30 healthy controls were analyzed by lectin-based enzyme-linked immunosorbent assay (ELISA) using biot

A novel redox signalling axis mediates nicotine-induced podocyte injury.

INTRODUCTION: Nicotine has been shown to induce Nlrp3 (nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3) inflammasome activation, thereby contributing to podocyte injury. However, the molecular mechanisms underlying nicotine-induced Nlrp3 inflammasome activation and subsequent podocyte dysfunction remain largely unknown. The present study investigated whether membrane raft (MR)-associated redox signalling plays a critical role in nicotine-induced Nlrp3 inflammasome activation and podocyte injury. METHODS AND RESULTS: Nicotine treatment induced membrane

The podocyte contactome in lupus nephritis: Current evidence and a framework for disease progression.

Lupus nephritis (LN) is initiated by immune-complex deposition, complement activation, and inflammation, but these mechanisms do not fully explain why some glomerular lesions resolve whereas others progress to podocyte depletion, glomerulosclerosis, and fibrosis. We present a conceptual spatial framework in which graded disruption of the podocyte organelle-contact network may contribute to this transition. The framework does not assume that contact-site abundance is uniformly protective or injurious. Contactome remodeling denotes an axis-specific change in contact distance, length, frequency,

AMPK Activation by 2'-Hydroxy-2,4,5-Trimethoxychalcone Derivatives in Podocyte Cells.

Activating AMP-activated protein kinase (AMPK) using chalcones has emerged as a potential therapeutic strategy for managing diabetes mellitus (DM) and diabetic nephropathy. This research focuses on discovering new chalcone derivatives with a stronger ability to stimulate AMPK in podocytes compared to 2'-hydroxychalcones 1, 2, and 3 reported from the earlier research. The results show that hydrogenated products (4-6) cannot exhibit considerably improved activity. Additionally, 2'-hydroxychalcone bearing 2,4,5-triethoxy groups on the B-ring (10) and 2,4,5-trimethoxychalcones bearing 2',4'-, 2',5

Longitudinal Study of the Vascular Glycocalyx and Urinary Podocyte Markers During Pregnancy.

BACKGROUND: We aimed to assess the longitudinal changes of glycocalyx, a proteoglycan/glycoprotein layer that lines and protects endothelial cells in normotensive pregnancies (NPs); to correlate glycocalyx parameters with kidney injury markers (podocin-positive to nephrin-positive [P/N] ratio); and to explore changes in P/N ratio in preeclampsia. METHODS: Plasma and urine samples and glycocalyx measurements using side-stream darkfield imaging were collected at each trimester from pregnant women (n=31). Plasma glycocalyx components were measured by ELISA, and urinary P/N-positive extracellular

Activation of IRF2 signaling networks facilitates podocyte pyroptosis in lupus nephritis.

Lupus nephritis (LN), a severe and often debilitating complication of systemic lupus erythematosus (SLE), is marked by renal inflammation and structural damage. While interferon regulatory factor 2 (IRF2) has been implicated in various immune-mediated pathologies, its precise role in LN pathogenesis remains elusive. In this study, we demonstrate a significant upregulation of IRF2 in renal biopsy specimens from patients with advanced LN, as well as in the MRL/lpr murine model of LN, with expression levels correlating positively with disease severity. Genetic ablation of IRF2 in MRL/lpr mice res

Podocyte, tubular epithelial-immune cell interplay in the pathogenesis of lupus nephritis.

Lupus nephritis (LN), a severe complication of systemic lupus erythematosus (SLE), is associated with increased morbidity and mortality. The pathogenesis of LN involves complex immune-mediated mechanisms that alter the biology of renal resident epithelial cells. Emerging evidence highlights the bidirectional interactions between immune cells and renal epithelial cells-including podocytes and tubular epithelial cells(TECs)-as critical contributors to disease progression. These interactions shape local immune responses, drive inflammatory injury, and disrupt renal function. However, the molecula

Podocyte-specific Nup160 knockout mice develop nephrotic syndrome and glomerulosclerosis.

More than 60 monogenic genes mutated in steroid-resistant nephrotic syndrome (SRNS) have been identified. Our previous study found that mutations in nucleoporin 160 kD (NUP160) are implicated in SRNS. The NUP160 gene encodes a component of the nuclear pore complex. Recently, two siblings with homozygous NUP160 mutations presented with SRNS and a nervous system disorder. However, replication of nephrotic syndrome (NS)-associated phenotypes in a mammalian model following loss of Nup160 is needed to prove that NUP160 mutations cause SRNS. Here, we generated a podocyte-specific Nup160 knockout (Nu
