Changes in Palatability Processing across the Estrous Cycle Are Modulated by Hypothalamic Estradiol Signaling.

Consumption varies across the stages (metestrus, diestrus, proestrus, estrus) of a rat's estrous cycle, changing in ways that might be expected to reflect, in part, a direct impact of hormones on taste palatability. Evidence regarding this hypothesis has been mixed, however, and critical within-subject experiments comparing consumption of multiple tastes with distinct valences across all estrous phases have been few. Here, we assayed female rats' licking of palatable (saccharin, sucrose, NaCl) and aversive (quinine-HCl, citric acid) tastes in brief-access trials, while tracking their estrous c

Ovarian hormonal state at exercise initiation interacts with nigrostriatal circuitry to determine long-term voluntary exercise behavior.

Despite the well-established health benefits of exercise, adherence to physical activity remains low, highlighting the need to identify biological factors that regulate the initiation and persistence of exercise behavior. Here, we tested whether ovarian hormone state at the onset of voluntary wheel running (VWR) influences both immediate and long-term exercise behavior in female rats. Females that initiated VWR during proestrus (Pro) ran farther, spent more time running, and ran at higher speeds on the first day of wheel access than females initiating VWR outside of Pro. Remarkably, initiating

Sex-Based Differences in Severe Trauma and Hemorrhagic Shock: A Systematic Review of Pre-Clinical and Veterinary Animal Studies.

PURPOSE: To systematically review preclinical studies to determine whether biological sex alters outcomes after severe trauma with hemorrhagic shock (T/HS), identify the mechanistic drivers of any sex dimorphism, and define key gaps that limit translation to critical care. METHODS: Following PRISMA guidance, we searched PubMed and Google Scholar through December 2025 for original animal or veterinary T/HS studies, comparing biological sexes or manipulating sex hormones. Exclusions included non-hemorrhagic shock, traumatic brain injury, and reviews. Two reviewers independently screened 1,450 r

Application of Machine Learning Models to Identify Differences in Neural Electrophysiological Properties Across Estrous Cycle Phases.

Machine learning models (MLMs) have been used to classify neuron subtypes based upon electrophysiological attributes. What has been less explored is using MLMs to assess differences within a neuron type, especially in the context of subtle changes induced by neuromodulation such as the rodent estrous cycle. Previous research found that the estrous cycle shifts rat nucleus accumbens (NAc) medium spiny neuron (MSN) electrophysiology, including action potential (AP), passive, and miniature excitatory post-synaptic current (mEPSC) properties. This plasticity provides a model system to investigate

Obesity inhibits hypothalamic activation and luteinizing hormone dynamics through Sox-2-dependent demyelination.

Obesity is a well - recognized cause of hypothalamic - pituitary hypogonadism. However, the activation patterns of whole hypothalamic neurons, the dynamic secretion of luteinizing hormone (LH) pulses and surges, and the underlying mechanisms remain unclear. We combined c-fos-based TetTag labeling, tissue clearing, and 3D imaging to map global hypothalamic neuronal activation in diet-induced obese mice. Serial blood sampling revealed how metabolic stress disrupts LH secretion dynamics-specifically its pulsatility across the estrous cycle and preovulatory surge. We further profiled transcription

Differentiating the estrous cycle phases using vaginal vestibule pH and cytology in Asian elephants (Elephas maximus) in human care.

Reproductive dysfunction in elephants is often confirmed through blood hormonal assays, as behavioral observations alone are insufficient. While fecal and saliva hormonal analyses offer welfare-friendly alternatives, significant resources are required, limiting practicality in field applications. To improve reproductive management and ensure animal welfare, this study aimed to investigate pH measurements and cytological analysis of the vaginal vestibule to differentiate estrous cycle phases in female elephants, both those with regular and irregular cycling. A total of 106 vaginal vestibule cyt

Determining the best timing of insemination based on salivary fern pattern and its association with conception in buffalo.

Salivary fern patterns (SFP) offer a simple, non-invasive, and cost-effective alternative to conventional estrus detection methods, addressing challenges such as silent heat, inaccurate heat detection, and improper timing of artificial insemination (AI). This study evaluated the utility of SFP in determining best timing for AI and its association with conception in buffaloes. Two experiments were conducted. In Experiment 1, twelve cyclic buffaloes in follicular phase were treated with standard Ovsynch protocol and 168 saliva samples were systematically collected to establish reference SFP, its

Hypothalamic KNDy neuron expression in streptozotocin-induced diabetic female rats.

Kisspeptin neurons, i.e. KNDy neurons, in the arcuate nucleus (ARC) coexpress neurokinin B and dynorphin and regulate gonadotropin-releasing hormone/luteinizing hormone (LH) pulses. Because it remains unclear whether these neurons are associated with reproductive dysfunction in diabetic females, we examined the expression of KNDy neurons detected by histochemistry in streptozotocin (STZ)-induced diabetic female rats 8 weeks after STZ injection. We also evaluated relevant metabolic parameters - glucose, 3-hydroxybutyrate, and non-esterified fatty acids - as indicators of diabetes progression. S

Involvement of a neutrophil-mast cell axis in the effects of Piper malacophyllum (C. PESL) C. DC extract and its isolated compounds in a mouse model of dysmenorrhoea.

The effects of Piper malacophyllum (C. Pesl) C. DC extracts and its isolated compounds were analysed in a mouse model of primary dysmenorrhoea (PD). Female Swiss mice (6-8 weeks old) on proestrus were intraperitoneally treated with estradiol benzoate for 3 days, to induce PD. Twenty-four hours later, animals were treated 24 h later with vehicle, plant extract, gibbilimbol B, 4,6-dimethoxy-5-E-phenylbutenolide, mixture of 4,6-dimethoxy-5-E-phenylbutenolide and 4,6-dimethoxy-5-Z-phenylbutenolide, or ibuprofen. Identification of a novel distension-evoked motility pattern in the mouse uterus.

The dynamic changes in uterine contractility in response to distension are incompletely understood. Rhythmic, propagating contractions of nonpregnant uterine smooth muscle occur in the absence of nerve activity (i.e., myogenic), events that decline during pregnancy and reemerge at parturition. We therefore sought to determine how myogenic contractions of the nonpregnant uterus are affected by distension, which might provide mechanistic clues underlying distension-associated uterine conditions such as preterm birth. Uteri isolated from nulliparous adult female mice in proestrus were video image

Enhanced responsiveness to hypoxic panicogenic challenge in female rats in late diestrus is suppressed by short-term, low-dose fluoxetine: Involvement of the dorsal raphe nucleus and the dorsal periaqueductal gray.

BACKGROUND: Acute hypoxia, which is panicogenic in humans, also evokes panic-like behavior in male rats. Panic disorder is more common in women and susceptibility increases during the premenstrual phase of the cycle. AIMS: We here investigated for the first time the impact of hypoxia on the expression of panic-like escape behavior by female rats and its relationship with the estrous cycle. We also evaluated functional activation of the midbrain panic circuitry in response to this panicogenic stimulus and whether short-term, low-dose fluoxetine treatment inhibits the hyper-responsiveness of fe

Progesterone dose during synchronization treatment alters luteinizing hormone receptor and steroidogenic enzyme mRNA abundances in granulosa cells of Nellore heifers.

The objective was to investigate effects of progesterone (P4) dose on abundance of luteinizing hormone receptor (LHCGR), aromatase (CYP19A1), 3β-hydroxysteroid dehydrogenase (HSD3B1), and other steroidogenic mRNA transcripts in granulosa cells from dominant follicles. Nellore heifers were assigned to one of six groups: new, first-use controlled internal drug release device (CIDR1) inserted for 5 days (Large-P4-dose-D5; n = 7) or 6 days (Large-P4-dose-D6; n = 8), prostaglandin (PG)F2α administered on D0 and 1 previously-used CIDR (CIDR3) inserted for 5 days (Small- P4-dose-D5; n = 8) or 6 days

Female Offspring From Chronic Hyperandrogenemic Dams Exhibit Delayed Puberty and Impaired Ovarian Reserve.

Female offspring of many species exposed to high doses of androgens in utero experience endocrine dysfunction during adulthood. The phenotype of offspring from females with prepregnancy hyperandrogenemia and impaired ovulation, however, has not been examined. We developed a mouse model of hyperandrogenemia by implanting a low-dose dihydrotestosterone (DHT) pellet 15 days before conception. Female offspring born to dams with hyperandrogenemia (DHT daughters) had delayed puberty (P < 0.05) with first estrus on postnatal day (PND) 41 compared with daughters from dams with physiological levels of

Molecular Plasticity of Male and Female Murine Gonadotropes Revealed by mRNA Sequencing.

Gonadotropes in the anterior pituitary gland are of particular importance within the hypothalamic-pituitary-gonadal axis because they provide a means of communication and thus a functional link between the brain and the gonads. Recent results indicate that female gonadotropes may be organized in the form of a network that shows plasticity and adapts to the altered endocrine conditions of different physiological states. However, little is known about functional changes on the molecular level within gonadotropes during these different conditions. In this study we capitalize on a binary genetic s

Ghrelin and peptide YY (PYY) profiles in gastrointestinal tissues and the circulation of the rat during pregnancy and lactation.

Plasma and tissue profiles of gastrointestinal hormones ghrelin and peptide YY (PYY) were investigated in different female rat reproductive states. Neither plasma nor tissue ghrelin concentrations were suppressed during pregnancy despite elevated leptin. The highest concentrations of stomach ghrelin were measured in late pregnancy. PYY concentrations in plasma, descending colon and rectum tissues were increased (P<0.001) throughout pregnancy and lactation. PYY peaked at day 5 of lactation in plasma, as well as descending colon and rectum tissues (proestrus vs day 5 of lactation: 25+/-3.0 pmol/

Relationship between opposite changes of vaginal and vestibular impedance during estrous cycle in sows.

Several bioimpedance techniques have been developed for noninvasive monitoring of reproductive events occurring in cyclic gilts and sows. Our objective was to compare the changes of vaginal and vestibular impedance during the porcine estrous cycle (experiment 1). In addition, we examined the causes of impedance variations in the vaginal vestibule during periestrus (experiment 2). The vaginal and vestibular impedance were measured with specially designed instruments. Sows were monitored for estrus via exposure to a sexually mature boar. The impedance in the vagina decreased gradually after wean

Mechanism of salutary effects of estradiol on organ function after trauma-hemorrhage: upregulation of heme oxygenase.

A growing body of evidence indicates that heme degradation products may counteract the deleterious consequences of hypoxia and/or ischemia-reperfusion injury. Because heme oxygenase (HO)-1 induction after adverse circulatory conditions is known to be protective, and because females in the proestrus cycle (with high estrogen) have better hepatic function and less hepatic damage than males after trauma-hemorrhage, we hypothesized that estrogen administration in males after trauma-hemorrhage will upregulate HO activity and protect the organs against dysfunction and injury. To test this hypothesis

Asymmetric antibodies (AAb) in the female reproductive tract.

Vaginal mucosa has been shown to play an important role in fertility, since several changes during the estrous cycle determine fertility and pregnancy outcome. The contribution of vaginal fluid IgG antibodies (Abs) to these changes is not fully characterized. Asymmetric Abs (AAb) are a subpopulation of IgG Abs bearing a carbohydrate residue in only one Fab region of the molecule, being therefore functionally univalent and unable to trigger immunological mechanisms tending to destroy the antigens. Here, we investigated the presence of AAb in vaginal secretions of virgin mice. Vaginal fluids wer

The Nhlh2 transcription factor is required for female sexual behavior and reproductive longevity.

Nhlh2 is a member of the basic helix-loop-helix (bHLH) transcription factor family and is expressed in developing and adult neuroendocrine tissues such as the pituitary and hypothalamus. Targeted deletion of Nhlh2 (N2KO) in mice results in hypogonadism and obesity. While gonadally intact male N2KO mice are infertile and lack male sexual behavior, female N2KO mice can become pregnant and carry litters to full term. Unlike normal females in which fertility averages 8-12 months with approximately one pregnancy per month, N2KO females have a shorter reproductive span with most females supporting o
